  • The prognostic role of KRAS, BRAF, PIK3CA and PTEN in colorectal cancer

    • Background Mutations in KRAS, BRAF, PIK3CA and PTEN expression have been in focus to predict the effect of epidermal growth factor receptor-blocking therapy in colorectal cancer (CRC). Here, information on these four aberrations was collected and combined to a Quadruple index and used to evaluate the prognostic role of these factors in CRC. Patients We analysed the mutation status in KRAS, BRAF and PIK3CA and PTEN expression in two separate CRC cohorts, Northern Sweden Health Disease Study (NSHDS; n = 197) and Colorectal Cancer in Umea Study (CRUMS; n = 414). A Quadruple index was created, where Quadruple index positivity specifies cases with any aberration in KRAS, BRAF, PIK3CA or PTEN expression. Results Quadruple index positive tumours had a worse prognosis, significant in the NSHDS but not in the CRUMS cohort (NSHDS; P = 0.003 and CRUMS; P = 0.230) in univariate analyses but significance was lost in multivariate analyses. When analysing each gene separately, only BRAF was of prognostic significance in the NSHDS cohort (multivariate HR 2.00, 95% CI: 1.16-3.43) and KRAS was of prognostic significance in the CRUMS cohort (multivariate HR 1.48, 95% CI: 1.02-2.16). Aberrations in PIK3CA and PTEN did not add significant prognostic information. Conclusions Our results suggest that establishment of molecular subgroups based on KRAS and BRAF mutation status is important and should be considered in future prognostic studies in CRC.

  • Fractional domain walls from on-site softening in dipolar bosons

    • We study dipolar bosons in a 1D optical lattice and identify a region in parameter space-strong coupling but relatively weak on-site repulsion-hosting a series of stable charge-density-wave (CDW) states whose low-energy excitations, built from fractional domain walls, have remarkable similarities to those of non-Abelian fractional quantum Hall states. Here, a conventional domain wall between translated CDW's may be split by inserting strings of degenerate, but inequivalent, CDW states. Outside these insulating regions, we find numerous supersolids as well as a superfluid regime. The mentioned phases should be accessible experimentally and, in particular, the fractional domain walls can be created in the ground state using single-site addressing, i.e., by locally changing the chemical potential.

  • Non-abelian quantum Hall states and fractional charges in one dimension

    • The fractional quantum Hall effect has, since its discovery around 30 years ago, been a vivid field of research—both experimentally and theoretically. In this thesis we investigate certain non-abelian quantum Hall states by mapping the two-dimensional system onto a thin torus, where the problem becomes effectively one-dimensional and hopping is suppressed, meaning that the classical electrostatic interaction dominates. The approach assists with a simplified view of ground states and their degeneracies, as well as of the nature of the fractionally charged, minimal excitations of the corresponding quantum Hall states. Similar models are also relevant for cold atoms trapped in one-dimensional optical lattices, where interaction parameters are available for tuning, which opens up for realizing interesting lattice states in controllable environments. The diverse applicability of the one-dimensional electrostatic lattice hamiltonian motivates the exploration of the systems and models treated in this work.In the absence of hopping or tunneling, the low-energy behavior of the one-dimensional lattice system is ultimately dependent on the nature of the electrostatic interaction present. For ordinary interactions such as Coulomb, the ground state at particle filling fraction ν= p/q has a well-known q-fold center-of-mass degeneracy and the elementary excitations are domain walls of fractional charge e* = ±e/q. These appear in abelian quantum Hall systems and are known since earlier. In this work, we show how other types of interaction give rise to increased ground state degeneracies and, as a result, to the emergence of split fractional charges recognized from non- abelian quantum Hall systems.

  • Configuration with architectural objects in industrialised house-building

    • The construction industry is facing the dilemma of simultaneously reducing costs and increasing quality. An increasing control and standardization of processes and products is strived for, while at the same time regarding customer and society requirements for architectural variation. Industrialised building and development of house building platforms are examples of this development. The research presented here shows a way to analyze the results of the development of a platform for a multi-storey house building system, specifically with regard to the flexibility of the platform. The analysis is based on the use of architectural objects as design interface in different design levels. The method is shown to complement the traditional QFD methodology, and shows the relative dependence of design decision levels.

    • The paper presents the intermediate results of an ongoing research project with the aim to develop concepts and tools for architectural design in industrialized house-building. Architectural design aims to enable different “situations” in the built environment involving people, behaviour, experience and environment, with desired technical, functional and aesthetic properties. "Situations” may be represented by configurable modular “architectural objects” that also reflect the variability of an industrialized building system. In three case studies the relevance of the concept of architectural object is investigated; in architectural design, and through modularization, development and organization of technical platforms. The results however also show a need for further research concerning the implementation in a BIM environment.

    • There are two main routes for customers of industrialized house building to follow: either rigid platforms offering preconfigured products, often called ‘type houses’, or more flexible platforms open to customization. The problem in customization is to link customer requirements with system opportunities. In axiomatic design processes, this problem is understood as the transformation of customer requirements in the architectural view into design parameters that are supported by the system. A new method is proposed that allows requirements and constraints of an industrialized building platform to be handled fully in the architectural view through configuration with so-called architectural objects. It is based upon established theories and complies with the rationale of separating platform development from product customization. An experimental study, using standard building information modelling software, shows that a hierarchical structure of architectural objects can support both the development, as well as the configuration process, of the platform. Modularization in the architectural view is believed to be an important first step to capturing user requirements in the development of platforms for a specific market segment. It is also anticipated that if these requirements can be conceptualized as architectural objects the capabilities in different configurator solutions can be made more transparent to the customer.
