    • Candida albicans is the principal opportunistic fungal pathogen in nosocomial settings and resistance to antifungal drugs is on the rise. Antimicrobial peptides from natural sources are promising novel therapeutics against C. albicans. OsDef2 defensin was previously found to be active against only Gram-positive bacteria, whereas derived fragments Os and its cysteine-free analogue, Os-C, are active against Gram-positive and Gram-negative bacteria at low micromolar concentrations. In this study, OsDef2-derived analogues and fragments were screened for anticandidal activity with the aim to identify peptides with antifungal activity and in so doing obtain a better understanding of the structural requirements for activity and modes of action. Os, Os-C and Os(11-22)NH2 , a Os-truncated carboxy-terminal-amidated fragment, had the most significant antifungal activities, with minimum fungicidal concentrations (MFCs) in the micromolar range (6-28 μM). C. albicans killing was rapid and occurred within 30-60 min. Further investigations showed all three peptides interacted with cell wall derived polysaccharides while both Os and Os(11-22)NH2 permeabilized fungal liposomes. Confocal laser scanning microscopy confirmed that Os-C and Os(11-22)NH2 could enter the cytosol of live cells and subsequent findings suggest that the uptake of Os and Os-C, in contrast to Os(11-22)NH2 , is energy dependent. Although Os, Os-C and Os(11-22)NH2 induced the production of reactive oxygen species (ROS), co-incubation with ascorbic acid revealed that only ROS generated by Os-C and to a lesser extent Os(11-22)NH2 resulted in cell death. Overall, Os, Os-C and Os(11-22)NH2 are promising candidacidal agents.

    • Designers of context-sensitive program analyses need to take special care of the memory consumption of the analysis results. In general, they need to sacrifice accuracy to cope with restricted memory resources. We introduce χ-terms as a general data structure to capture and manipulate context-sensitivity analysis results. A χ-term is a compact representation of arbitrary forward program analysis distinguishing the effects of different control-flow paths. While χ-terms can be represented by trees, we propose a memory efficient representation generalizing ordered binary decision diagrams (OBDDs).

    • We present radial velocities for five member stars of the recently discovered young (age similar or equal to 100-150 Myr) stellar system Price-Whelan 1 (PW1), which is located far away in the Galactic Halo (D similar or equal to 29 kpc, Z similar or equal to 15 kpc), and that is probably associated with the leading arm (LA) of the Magellanic Stream. We measure the systemic radial velocity of PW 1, V-r = 275 +/- 10 km s(-1), significantly larger than the velocity of the LA gas in the same direction. We re-discuss the main properties and the origin of this system in the light of these new observations, computing the orbit of the system and comparing its velocity with that of the HI in its surroundings. We show that the bulk of the gas at the velocity of the stars is more than 10 deg (5 kpc) away from PW 1 and the velocity difference between the gas and the stars becomes larger as gas closer to the stars is considered. We discuss the possibilities that (1) the parent gas cloud was dissolved by the interaction with the Galactic gas, and (2) that the parent cloud is the high-velocity cloud (HVC) 287.5+22.5 + 240, lagging behind the stellar system by similar or equal to 25 km s(-1) and similar or equal to 10 deg similar or equal to 5 kpc. This HVC, which is part of the LA, has metallicity similar to PW 1, displays a strong magnetic field that should help to stabilize the cloud against ram pressure, and shows traces of molecular hydrogen. We also show that the system is constituted of three distinct pieces that do not differ only by position in the sky but also by stellar content.
