  • Semantic Integration of Plug-and-Play Software Components for Industrial Edges Based on Microservices

    • The industrial cyber-physical system enables collaboration between distributed nodes across industrial clouds and edge devices. Flexibility and interoperability could be enhanced significantly by introducing the service-oriented architecture to industrial edge devices. From the industrial edge computing perspective, software components shall be dynamically composed across heterogeneous edge devices to perform various functionalities. In this paper, a knowledge-driven Microservice-based architecture to enable plug-and-play software components is proposed for industrial edges. These software components can be dynamically configured based on the orchestration of microservices with the support of the knowledge base and the reasoning process. These semantically enhanced plug-and-play microservices could provide rapid online reconfiguration without any programming efforts. The use of the plug-and-play software components is demonstrated by an assembly line example.

    • Because of the diurnal thermal cycle and the irregular shape of the nucleus, gas outflow of comet 67P/Churyumov-Gerasimenko could be highly anisotropic as indicated by the colliminated dust jet structures on the sunlit side. Based on the OSIRIS imaging observations of the outgassing effect, a simple model of surface sublimation can be constructed by taking into account the dependence on the solar insolation. With preliminary information on the time variability of the global gas production rate, a sequence of gas coma models can be generated at different epochs before and after perihelion. We also investigate different patterns of dust particle dynamics under the influences of nuclear rotation and gas drag. From these considerations, a consistent picture of the spatial distribution of dusty materials across the surface of comet 67P as it moves around the perihelion can be developed. It is found that because of the redeposition of the ejected dust from the Southern hemisphere to the Northern hemisphere during the southern summer season the Hapi region could gain up to 0.4 m while the Wosret region would lose up to 1.8 m of dust mantle per orbit.

  • A Robust, Non-Cooperative Localization Algorithm in the Presence of Outlier Measurements in Ocean Sensor Networks

    • As an important means of multidimensional observation on the sea, ocean sensor networks (OSNs) could meet the needs of comprehensive information observations in large-scale and multifactor marine environments. In what concerns OSNs, accurate location information is the basis of the data sets. However, because of the multipath effect-signal shadowing by waves and unintentional or malicious attacks-outlier measurements occur frequently and inevitably, which directly degrades the localization accuracy. Therefore, increasing localization accuracy in the presence of outlier measurements is a critical issue that needs to be urgently tackled in OSNs. In this case, this paper proposed a robust, non-cooperative localization algorithm (RNLA) using received signal strength indication (RSSI) in the presence of outlier measurements in OSNs. We firstly formulated the localization problem using a log-normal shadowing model integrated with a first order Taylor series. Nevertheless, the problem was infeasible to solve, especially in the presence of outlier measurements. Hence, we then converted the localization problem into the optimization problem using squared range and weighted least square (WLS), albeit in a nonconvex form. For the sake of an accurate solution, the problem was then transformed into a generalized trust region subproblem (GTRS) combined with robust functions. Although GTRS was still a nonconvex framework, the solution could be acquired by a bisection approach. To ensure global convergence, a block prox-linear (BPL) method was incorporated with the bisection approach. In addition, we conducted the Cramer-Rao low bound (CRLB) to evaluate RNLA. Simulations were carried out over variable parameters. Numerical results showed that RNLA outperformed the other algorithms under outlier measurements, notwithstanding that the time for RNLA computation was a little bit more than others in some conditions.

  • Quantification for total demethylation potential of environmental samples utilizing the EGFP reporter gene

    • Abstract The demethylation potential of pollutants is arguably an innate component of their toxicity in environmental samples. A method was developed for determining the total demethylation potential of food samples (TDQ). The demethylation epigenetic toxicity was determined using the Hep G2 cell line transfected with pEGFP-C3 plasmids containing a methylated promoter of the EGFP reporter gene. The total demethylation potential of the sample extracts (the 5-AZA-CdR demethylation toxic equivalency) can be quantified within one week by using a standard curve of the 5-AZA-CdR demethylation agent. To explore the applicability of TDQ for environmental samples, 17 groundwater samples were collected from heavy polluted Kuihe river and the total demethylation potentials of the sample extracts were measured successfully. Meaningful demethylation toxic equivalencies ranging from 0.00050 to 0.01747 μM were found in all groundwater sample extracts. Among 19 kinds of inorganic substance, As and Cd played important roles for individual contribution to the total demethylation epigenetic toxicity. The TDQ assay is reliable and fast for quantifying the DNA demethylation potential of environmental sample extracts, which may improve epigenetic toxicity evaluations for human risk assessment, and the consistent consuming of groundwater alongside the Kuihe river pose unexpected epigenetic health risk to the local residents.

  • Integrin Based Isolation Enables Purification of Murine Lineage Committed Cardiomyocytes

    • In contrast to mature cardiomyocytes which have limited regenerative capacity, pluripotent stem cells represent a promising source for the generation of new cardiomyocytes. The tendency of pluripotent stem cells to form teratomas and the heterogeneity fromvarious differentiation stages and cardiomyocyte cell sub-types, however, are major obstacles to overcome before this type of therapy could be applied in a clinical setting. Thus, the identification of extracellularmarkers for specific cardiomyocyte progenitors and mature subpopulations is of particular importance. The delineation of cardiomyocyte surfacemarker patterns not only serves as a means to derive homogeneous cell populations by FACS, but is also an essential tool to understand cardiac development. By using single-cell expression profiling in early mouse embryonic hearts, we found that a combination of integrin alpha-1, alpha-5, alpha-6 and N-cadherin enables isolation of lineage committed murine cardiomyocytes. Additionally, we were able to separate trabecular cardiomyocytes from solid ventricular myocardium and atrial murine cells. These cells exhibit expected subtype specific phenotype confirmed by electrophysiological analysis. We show that integrin expression can be used for the isolation of living, functional and lineage-specific murine cardiomyocytes.
