  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Comparative Study of Dayside Pulsating Auroras Induced by Ultralow-Frequency Waves

    • Pulsating auroras are usually observed with ultralow-frequency (ULF) waves in the Pc 3-5 band (period 10-600 s). These auroras are thought to result from interactions between energetic electrons and chorus waves, but their relationship with ULF waves remains an open question. In this study, we investigated this question by conducting a comparative study on two ULF wave events with pulsating auroras observed near the magnetic footprints. Conjugate observations from the Magnetospheric Multiscale mission and the Chinese Yellow River Station were used. In both events, lower-band chorus waves were observed, which were suggested to be connected with the auroral pulsations by wavelet analysis. The intensity of these waves oscillates at the period of the ULF waves, but the physics laid behind them differs by events. During the event of 22 January 2019, compressional ULF waves changed the threshold for the whistler anisotropy instability periodically, affecting the emission of chorus waves. In the event on 10 January 2016, poloidal ULF waves modulated the chorus wave generation by regulating electron temperature anisotropy through drift resonance. ULF waves in these events may originate from perturbations in the solar wind. We highlight the role of ULF waves in the solar wind-magnetosphere-ionosphere coupling, which requires further study.

  • Interleukin-22 regulates B3GNT7 expression to induce fucosylation of glycoproteins in intestinal epithelial cells.

    • Interleukin (IL)-22 is a cytokine that plays a critical role in intestinal epithelial homeostasis. Its downstream functions are mediated through interaction with the heterodimeric IL-22 receptor and subsequent activation of signal transducer and activator of transcription 3 (STAT3). IL-22 signaling can induce transcription of genes necessary for intestinal epithelial cell proliferation, tissue regeneration, tight junction fortification, and antimicrobial production. Recent studies have also implicated IL-22 signaling in the regulation of intestinal epithelial fucosylation in mice. However, whether IL-22 regulates intestinal fucosylation in human intestinal epithelial cells and the molecular mechanisms that govern this process are unknown. Here, in experiments performed in human cell lines and human-derived enteroids, we show that IL-22 signaling regulates expression of the B3GNT7 transcript, which encodes a β1-3-N-acetylglucosaminyltransferase that can participate in the synthesis of poly-N-acetyllactosamine (polyLacNAc) chains. Additionally, we find that IL-22 signaling regulates levels of the α1-3-fucosylated Lewis X (Lex) blood group antigen, and that this glycan epitope is primarily displayed on O-glycosylated intestinal epithelial glycoproteins. Moreover, we show that increased expression of B3GNT7 alone is sufficient to promote increased display of Lex-decorated carbohydrate glycan structures primarily on O-glycosylated intestinal epithelial glycoproteins. Together, these data identify B3GNT7 as an intermediary in IL-22-dependent induction of fucosylation of glycoproteins and uncover a novel role for B3GNT7 in intestinal glycosylation.

  • BODIPY-linked conjugated porous polymers for dye wastewater treatment

    • As a new family of functional porous materials, conjugated porous polymers (CPPs) with tuneable porosities and molecular architectures show great potentials in adsorption, light harvesting, and catalysis applications. This paper describes the incorporation of strong visible-light absorbing BODIPY molecules into the skeleton of CPPs via Sonogashira coupling reactions. The obtained CPPs displayed integrated properties of high surface area, hierarchical porous structures and strong visible-light absorption. As a result, the CPPs showed relatively high adsorption capacity and high photocatalytic degradation efficiency towards organic dyes. Mechanism studies revealed that the CPPs enabled the generation of singlet oxygen species under light irradiation, accounting for the main driving force for the dye degradation. This study provides a new route for the development of organic adsorbents and photocatalysts for water treatment and purification.

    • Genetic variants within nearly 1000 loci are known to contribute to modulation of blood lipid levels. However, the biological pathways underlying these associations are frequently unknown, limiting understanding of these findings and hindering downstream translational efforts such as drug target discovery.To expand our understanding of the underlying biological pathways and mechanisms controlling blood lipid levels, we leverage a large multi-ancestry meta-analysis (N=1,654,960) of blood lipids to prioritize putative causal genes for 2286 lipid associations using six gene prediction approaches. Using phenome-wide association (PheWAS) scans, we identify relationships of genetically predicted lipid levels to other diseases and conditions. We confirm known pleiotropic associations with cardiovascular phenotypes and determine novel associations, notably with cholelithiasis risk. We perform sex-stratified GWAS meta-analysis of lipid levels and show that 3-5% of autosomal lipid-associated loci demonstrate sex-biased effects. Finally, we report 21 novel lipid loci identified on the X chromosome. Many of the sex-biased autosomal and X chromosome lipid loci show pleiotropic associations with sex hormones, emphasizing the role of hormone regulation in lipid metabolism.Taken together, our findings provide insights into the biological mechanisms through which associated variants lead to altered lipid levels and potentially cardiovascular disease risk.

    • Fatty liver disease (FLD) is a growing health issue with burdening unmet clinical needs. FLD has a genetic component but, despite the common variants already identified, there is still a missing heritability component. Using a candidate gene approach, we identify a locus (rs71519934) at the Pleckstrin and Sec7 domain-containing 3 (PSD3) gene resulting in a leucine to threonine substitution at position 186 of the protein (L186T) that reduces susceptibility to the entire spectrum of FLD in individuals at risk. PSD3 downregulation by short interfering RNA reduces intracellular lipid content in primary human hepatocytes cultured in two and three dimensions, and in human and rodent hepatoma cells. Consistent with this, Psd3 downregulation by antisense oligonucleotides in vivo protects against FLD in mice fed a non-alcoholic steatohepatitis-inducing diet. Thus, translating these results to humans, PSD3 downregulation might be a future therapeutic option for treating FLD.

  • A nonlinear prediction model of the debonding process of an FRP-concrete interface under fatigue loading

    • Externally bonded Fiber Reinforced Polymer (FRP) strengthening has been proven to be an efficient and reliable method for structural strengthening of reinforced concrete (RC) members. However, the beneficial effects of this method can be diminished due to the debonding of the FRP laminates. The mechanism of FRP debonding still requires further research, especially for strengthened members under fatigue loading. To understand and predict the FRP fatigue debonding process better, eleven FRP-concrete joint specimens were tested under static or fatigue loading. Both the theoretical derivation and the experimental study indicated that the debonding growth rate of the FRP laminate depended not only on the mean level (), but also the amplitude () of the applied fatigue load. In addition, the debonded portion of the FRP laminate had a significant impact on the following debonding process due to the friction and mechanical interaction between the debonded FRP and the concrete surface. Therefore, a new nonlinear prediction model is proposed in this paper. The proposed model explicitly took into account the amplitude and the mean level of the fatigue loading, which enabled the effect of both to be modelled. Meanwhile, a correction term was also introduced into the model to account for the influence of the previously debonded FRP laminate. The predicted results of the debonding growth rate and the debonding length agreed well with the experimental results.

  • An experimental study on fatigue debonding growth of RC beams strengthened with prestressed CFRP plates

    • Externally bonded Fiber Reinforced Polymer (FRP) laminates are increasingly used to strengthen Reinforced Concrete (RC) members. However, FRP debonding remains a major drawback of this strengthening method. To better understand the mechanisms of FRP debonding, six RC beams strengthened with prestressed or non-prestressed Carbon Fiber Reinforced Polymer (CFRP) plates were subjected to static and fatigue loading. CFRP plate debonding was observed in both cases. However, the mechanism of debonding differed: under cyclical fatigue loading, debonding was initiated under both loading points simultaneously and propagated synchronously towards the nearest support whereas in static tests debonding began under a single loading point and progressed suddenly towards its adjacent support. The results also showed that stress redistribution induced coupling between accumulated fatigue damage in the steel reinforcement and fatigue debonding of the CFRP plate, accelerating the fatigue failure of the specimens.

  • Dual-functional ionic porous organic framework for palladium scavenging and heterogeneous catalysis

    • Porous organic frameworks (POFs) with predesigned structures and tunable porosities have been widely studied in adsorption and heterogeneous catalysis. Introducing ionic structure into the framework endows POFs with new functionalities that may extend their applications. Here, we report new applications for a guanidinium-based ionic POF (IPOF-Cl) in palladium scavenging and heterogeneous catalysis. Due to the ionic framework and the porous structure, the IPOF-Cl displays fast adsorption kinetics and high adsorption capacities (up to 754 mg g−1) of Na2PdCl4 in aqueous solutions via a chemisorption (ion exchange) process. Significantly, it shows excellent scavenging activity towards trace amount of [PdCl4]2− in aqueous solution. More importantly, the loaded [PdCl4]2− species on the IPOF substrate are further reduced into ultrafine Pd nanoparticles with size of ∼2–5 nm. The obtained IPOF-Pd(0) nanocomposite containing uniformly distributed Pd nanoparticles and hierarchical porous structure demonstrates high activity in catalyzing a range of Suzuki coupling reactions. This study provides new routes for the development of ionic porous organic materials for applications in metal scavenging and catalysis.
