  • X-radiation inhibits histone deacetylase 1 and 2, upregulates Axin expression and induces apoptosis in non-small cell lung cancer

    • BackgroundHistone deacetylase (HDAC) plays an important role in the deacetylation of histone, which can alter gene expression patterns and affect cell behavior associated with malignant transformation. The aims of this study were to investigate the relationships between HDAC1, HDAC2, clinicopathologic characteristics, patient prognosis and apoptosis, to clarify the mechanism of upregulation of the Axis inhibitor Axin (an important regulator of the Wnt pathway) by X-radiation and to elucidate the effect of siRNA on radiation therapy of non-small cell lung cancer (NSCLC).MethodsHDAC1 and HDAC2 expression levels were measured by immunohistochemistry and reverse transcription PCR. Apoptosis was determined by terminal deoxynucleotidyl transferase-mediated dUTP-nick end labeling and fluorescence activated cell sorting. BE1 cells expressing Axin were exposed to 2 Gy of X-radiation.ResultsExpression of HDAC1 and that of HDAC2 were correlated, and significantly higher in NSCLC tissues than in normal lung tissues (P < 0.05). HDAC1 and HDAC2 expression was correlated with pTNM stage and negatively correlated with differentiation of NSCLC and apoptotic index (P < 0.05). The prognosis of patients with low expression of HDAC1 and HDAC2 was better than that of those with high expression. X-radiation and siRNA inhibited HDAC1 and HDAC2 expression in NSCLC cells and Axin levels were significantly higher in BE1 cells.ConclusionsX-radiation and siRNA inhibit expression of HDAC1 and HDAC2, weaken the inhibitory effect of HDAC on Axin, upregulate Axin expression and induce apoptosis of lung cancer cells. Inhibition of HDAC1 and HDAC2 is a means of enhancing the radiosensitivity of NSCLC.

  • Experimental Test of the State Estimation-Reversal Tradeoff Relation in General Quantum Measurements

    • When a measurement has limited strength, only partial information, regarding the initial state, is extracted, and, correspondingly, there is a probability to reverse its effect on the system and retrieve the original state. Recently, a clear and direct quantitative description of this complementary relationship, in terms of a tradeoff relation, was developed by Y. K. Cheong and S. W. Lee. [Phys. Rev. Lett. 109, 150402 (2012)]. Here, this tradeoff relation is experimentally verified using polarization-encoded single photons from a quantum dot. Measurement operators representing a complete range, from not affecting the system to a projection to a single polarization state, are realized. In addition, for each measurement operator, an optimal reversal operator is also implemented. The upper bound of the tradeoff relation is mapped to experimental parameters representing the measurement strength. Our results complement the theoretical work and provide a hands-on characterization of general quantum measurements.

    • Ultra wideband technology becomes more widely used for near-field object detection. We present an aperture synthetic radar (SAR) system using ultra-wideband (UWB) signals for such applications. One of the problems with UWB time-domain SAR imaging algorithm is the artefact originated from scattered field of adjacent objects. In this paper an algorithm based on the Range Point Migration (RPM) method is used to alleviate this problem. This method estimates a possibility distribution of the optimal Direction of Arrival (DOA) of the scattered field from objects. A weighting factor based on this possibility is used to suppress the effect of artifact scattered fields. The results in the paper show that the UWB SAR accompanied with RPM can produce an image of an object with a significant suppression of undesired artifact from scattered field of adjacent objects.

  • Are the elderly more vulnerable to psychological impact of natural disaster? A population-based survey of adult survivors of the 2008 Sichuan earthquake

    • BACKGROUND: The association between ages and psychological impact of natural disasters has not been well characterized. A population-based study was conducted 15 months after the 2008 Sichuan earthquake to assess whether elderly survivors were more likely to develop posttraumatic stress disorder (PTSD) and general psychiatric morbidity.METHODS: A population-based survey of 327 survivors (152 elders, 175 younger adults) was conducted in severely affected areas by the earthquake, using a multi-stage systematic sampling design.RESULTS: Compared with the younger adult survivors, the elderly were more likely to have symptoms of PTSD (22.5% vs. 8.0%, p = 0.001) and general psychiatric morbidity (42.0% vs. 25.4%, p = 0.003). Risk factors, such as being elderly, having been in serious danger, having lost family members, and having felt guilt concerning one's death or injury were significantly associated with developing PTSD; being elderly, having family members or friends seriously injured, and having felt guilt concerning one's death or injury were significantly associated with developing general psychiatric morbidity. Utilization of mental health services is strongly associated with the decreased risk for developing both of the symptoms.CONCLUSION: Compared with the younger adults, the elderly survivors were more likely to develop PTSD and general psychiatric morbidity. More mental health services should be distributed to the elderly and groups at particular risk, to ensure their smooth mental health reconstruction after the earthquake.

    • Detection and recognition of metallic targets based on Ultra Wideband electromagnetic transients have been widely studied throughout the years. Due to the complexity of the scattering phenomena, such techniques have not been widely applied for dielectric targets. In this paper, the potential of detection and characterization of dielectric target is investigated. Instead of using single co-polarized linear responses, Ultra wideband polarimetric responses are considered. Numerical example of a breast cancer detection scenario will be used to demonstrate the feasibility of using wideband polarimetric data to characterize dielectric target.

    • Mooring systems for wave energy converters (WEC) have to be designed to survive the cyclic loads and motions they are subjected to as a result of the wave load-WEC interaction and the motions of the WEC in the random elevation of the sea surface. The current study compares four simulation procedures for the analysis of fatigue of WEC moorings. The objective is to recommend the type of simulation procedure that can be used to make reliable fatigue design of WEC mooring systems at a reasonable computational effort. A cylindrical floating WEC with four spread mooring lines is chosen for case study. The mooring dynamics of the WEC is simulated using coupled and de-coupled approaches in the time domain. In total, four types of simulation procedures are compared using the commercial simulation software DNV DeepC and an in-house code MOODY. A parameter sensitivity analysis of environmental conditions, model and numerical parameters is presented. The results are compared with respect to fatigue damage calculated using a stress-based approach and the rainflow counting method. It is found that a de-coupled approach, using DNV DeepC to simulate the buoy’s motions and cable response, is recommended since (i) it gives reliable results in terms of motion and stress responses of the buoy, mooring lines and accumulated fatigue damage, (ii) it requires the least model preparation by the engineer and the computation time is reasonable.

    • The fast development of new organic sensitizers leads to the need for a better understanding of the complexity and significance of their adsorption processes on TiO2 surfaces. We have investigated a prototype of the triphenylamine-cyanoacrylic acid (donor-acceptor) on rutile TiO2 (110) surface with special attention on the monolayer region. This molecule belongs to the type of dye, some of which so far has delivered the record efficiency of 10%-10.3% for pure organic sensitizers [W. Zeng, Y. Cao, Y. Bai, Y. Wang, Y. Shi, M. Zhang, F. Wang, C. Pan, and P. Wang, Chem. Mater. 22, 1915 (2010)]. The molecular configuration of this dye on the TiO2 surface was found to vary with coverage and adopt gradually an upright geometry, as determined from near edge x-ray absorption fine structure spectroscopy. Due to the molecular interaction within the increasingly dense packed layer, the molecular electronic structure changes systematically: all energy levels shift to higher binding energies, as shown by photoelectron spectroscopy. Furthermore, the investigation of charge delocalization within the molecule was carried out by means of resonant photoelectron spectroscopy. A fast delocalization (similar to 1.8 fs) occurs at the donor part while a competing process between delocalization and localization takes place at the acceptor part.
