    • Recently, artificially designed metamaterials have become of considerable interests, because they exhibit extraordinary optical characteristics that do not exist in nature and promise many potential applications, such as negative refraction, subwavelength imaging, and electromagnetic invisibility cloaking. Although creating metamaterials at the optical frequency range faces numerous technological challenges, such materials with particular properties have been realized gradually based on new device concepts. In this talk, we present our efforts to employ specific metamaterials to manipulate the polarization states of incident lights, in both microwave [1, 2] and optical frequency regimes [3]. Experimental results reveal that the maximum polarization conversion ratio (PGR) value can reach 100% in microwave regime (see left figure below) and 92% in optical frequency (see right figure blow) under certain conditions. Theoretical studies combined with numerical simulations show that the governing physics is dominated by the unique reflection properties of the metamaterials

  • Conditional overexpression of transgenes in megakaryocytes and platelets in vivo

    • Megakaryocyte (MK)-specific transgene expression has proved valuable in studying thrombotic and hemostatic processes. Constitutive expression of genes, however, could result in altered phenotypes due to compensatory mechanisms or lethality. To circumvent these limitations, we used the tetracycline/doxycycline (Tet)-off system to conditionally over-express genes in megakaryocytes and platelets in vivo. We generated 3 transactivator transgenic lines expressing the Tet transactivator element (tTA), under the control of the MK-specific platelet factor 4 promoter (PF4-tTA-VP16). Responder lines were simultaneously generated, each with a bidirectional minimal cytomegalovirus (CMV)-tTA responsive promoter driving prokaryotic β-galactosidase gene, as a cellular reporter, and a gene of interest (in this case, the mitotic regulator Aurora-B). A transactivator founder line that strongly expressed PF4-driven tTA-viral protein 16 (VP16) was crossbred to a responder line. The homozygous double-transgenic mouse line exhibited doxycycline-dependent transgene overexpression in MKs and platelets. Using this line, platelets were conveniently indicated at sites of induced stress by β-galactosidase staining. In addition, we confirmed our earlier report on effects of constitutive expression of Aurora-B, indicating a tight regulation at protein level and a modest effect on MK ploidy. Hence, we generated a new line, PF4-tTA-VP16, that is available for conditionally overexpressing genes of interest in the MK/platelet lineage in vivo.

    • A novel, cartridge-based procedure for the efficient and irreversible detritylation of oligonucleotides is reported. This method, combined with a process for the elimination of depurinated fragments produces, in a highly parallel fashion, oligonucleotides with better purity than those traditionally obtained using reversed-phase high-performance liquid chromotography purification. Our combined detritylation and purification methodology compares favorably with commercial cartridge-based purification systems. The benefits of working with pure oligonucleotides, with regard to higher signal and better signal linearity, are shown in array-based hybridization experiments.

    • Six centrosymmetric D-(pi-A)(3) structural triphenylamine derivatives that can be used as two- photon photopolymerization and optical data storage chromophores, tris[ 4-( 4- pyridylethenyl) phenyl] amine ( 1), tris[ 4-( 2- pyridylethenyl) phenyl] amine ( 2), tris( 4- cyanoethenylphenyl) amine ( 3), tris[ 4- butylacrylatephenyl] amine ( 4), tris[ 4- methylacrylatephenyl] amine ( 5) and tris[ 4- acrylicethenylphenyl] amine ( 6), have been successfully synthesized via a triple palladium-catalyzed Heck coupling reaction, and the novel chromophores were fully characterized by elemental analysis, IR, (1)H-NMR and ESIMS. The structure for 3 was determined by single crystal X-ray diffraction study. One- and two-photon absorption and fluorescence in various solvents were experimentally investigated. Two-photon initiated polymerization microfabrication and optical data recording experiments were carried out under 780 nm laser radiation, and the possible polymerization mechanism is discussed based on theoretical calculations. All the six chromophores have relatively large two-photon absorption crosssections, and exhibit optical memory and highly efficient two-photon initiated polymerization abilities.

  • Strain-insensitive and high-temperature long-period gratings inscribed in photonic crystal fiber

    • We fabricate and demonstrate strain-insensitive and high-temperature long-period gratings in endlessly single-mode photonic crystal fiber by use of focused pulses of a CO2 laser and a periodic stress relaxation technique without geometrical deformation and elongation of the fiber. The thermal dependence of mode coupling at 1299.59 nm is 10.9 pm/°C from 24 to 992 °C, whereas the coefficient of strain sensitivity is −0.192 pm/??-0.192 pm/−0.192 pm/?? up to the maximum strain of 2.74%?2.74%−0.192 pm/?. It is found for what is believed to be the first time that, in contrast with the traditional fiber case, the coupling resonance shifts toward shorter wavelengths under applied strain, indicating that the refractive index of the core is decreased as a result of the rebuilding of tension attributed to the stress-elastic effect, and the cladding modes is highly dispersive because of airholes arranged in the fiber cladding.
