  • Statistical analysis of whistler precursors upstream of foreshock transient shocks : MMS observations

    • Using the high-time-resolution data from the Magnetospheric Multiscale mission, precursor waves upstream of foreshock transient (FT) shocks are statistically investigated using the four-spacecraft timing method. The wave frequencies and wave vectors determined in the plasma rest frame (PRF) are shown to follow the cold plasma dispersion relation for whistler waves. Combining with the feature of the right-hand polarization in the PRF, the precursors are identified as whistler-mode waves around the lower hybrid frequency. The occurrence of whistler precursors is independent of the Alfvén Mach number and the FT shock normal angle. More importantly, all the whistler precursors have group velocities pointing upstream in the shock frame, suggesting the dispersive radiation to be a possible generation mechanism. The study improves the understanding of not only the whistler precursors but also the overall FT shock dynamics.

  • Deep Learning for Additive Screening in Perovskite Light-Emitting Diodes

    • Additive engineering with organic molecules is of critical importance for achieving high-performance perovskite optoelectronic devices. However, experimentally finding suitable additives is costly and time consuming, while conventional machine learning (ML) is difficult to predict accurately due to the limited experimental data available in this relatively new field. Here, we demonstrate a deep learning method that can predict the effectiveness of additives in perovskite light-emitting diodes (PeLEDs) with a high accuracy up to 96 % by using a small dataset of 132 molecules. This model can maximize the information of the molecules and significantly mitigate the duplicated problem that usually happened with previous models in ML for molecular screening. Very high efficiency PeLEDs with a peak external quantum efficiency up to 22.7 % can be achieved by using the predicated additive. Our work opens a new avenue for further boosting the performance of perovskite optoelectronic devices.

  • A Transcriptome-Wide Association Study Identifies Novel Candidate Susceptibility Genes for Pancreatic Cancer

    • Background: Although 20 pancreatic cancer susceptibility loci have been identified through genome-wide association studies in individuals of European ancestry, much of its heritability remains unexplained and the genes responsible largely unknown. Methods: To discover novel pancreatic cancer risk loci and possible causal genes, we performed a pancreatic cancer transcriptome-wide association study in Europeans using three approaches: FUSION, MetaXcan, and Summary-MulTiXcan. We integrated genome-wide association studies summary statistics from 9040 pancreatic cancer cases and 12 496 controls, with gene expression prediction models built using transcriptome data from histologically normal pancreatic tissue samples (NCI Laboratory of Translational Genomics [n = 95] and Genotype-Tissue Expression v7 [n = 174] datasets) and data from 48 different tissues (Genotype-Tissue Expression v7, n = 74-421 samples). Results: We identified 25 genes whose genetically predicted expression was statistically significantly associated with pancreatic cancer risk (false discovery rate < .05), including 14 candidate genes at 11 novel loci (1p36.12: CELA3B; 9q31.1: SMC2, SMC2-AS1; 10q23.31: RP11-80H5.9; 12q13.13: SMUG1; 14q32.33: BTBD6; 15q23: HEXA; 15q26.1: RCCD1; 17q12: PNMT, CDK12, PGAP3; 17q22: SUPT4H1; 18q11.22: RP11-888D10.3; and 19p13.11: PGPEPI) and 11 at six known risk loci (5p15.33: TERT, CLPTMIL, ZDHHCIIB; 7p14.1: INHBA; 9q34.2: ABO; 13q12.2: PDX1; 13q22.1: KLF5; and 16q23.1: WDR59, CFDP1, BCAR1, TMEM170A). The association for 12 of these genes (CELA3B, SMC2, and PNMT at novel risk loci and TERT, CLPTMIL, INHBA, ABO, PDX1, KLF5, WDR59, CFDP1, and BCAR1 at known loci) remained statistically significant after Bonferroni correction. Conclusions: By integrating gene expression and genotype data, we identified novel pancreatic cancer risk loci and candidate functional genes that warrant further investigation.

    • Recent Unmanned Combat Air Vehicle (UCAV) configurations have not-so-slender wings with moderate leading-edge sweepangles of 45◦ to 60◦. Planforms vary from pure delta to diamond and even lambda more or less blended wing bodies with a relatively small thickness ratio at the inner wing/fuselage region. The performance and low-observable signature considerations require a compromise between a small radar cross-section and lifting surface shapes for long range and sufficient agility. The airflow is governed largely by the progression of vortices shed from the wing leading edge which interact and produce non-linear aerodynamics. Like early swept wing fighters like the Saab 32 the UCAV may exhibit undesirable flying characteristics over part of the envelope with tip stall and pitch-up. Such problems were mitigated by vortex control "devices" like leading edge fences and notches which violate the low observable requirement. The AVT-181 SACCON is a well studied case. We attempt to improve its stalling characteristics by mimicking the leading edge fence action by a configuration of very small vortex generators and study their effect by CFD.

    • The mechanisms that link hyperandrogenism and insulin resistance to the increased miscarriage rate in women with polycystic ovary syndrome (PCOS) remain elusive. Previous studies demonstrate that increased uterine and placental ferroptosis is associated with oxidative stress-induced fetal loss in a pre-clinical PCOS-like rat model. Here, we investigated the efficacy and molecular mechanism of action of the antioxidant N-acetylcysteine (NAC) in reversing gravid uterine and placental ferroptosis in pregnant rats exposed to 5α-dihydrotestosterone (DHT) and insulin. Molecular and histological analyses showed that NAC attenuated DHT and insulin-induced uterine ferroptosis, including dose-dependent increases in anti-ferroptosis gene content. Changes in other molecular factors after NAC treatment were also observed in the placenta exposed to DHT and insulin, such as increased glutathione peroxidase 4 protein level. Further, increased apoptosis inducing factor mitochondria associated 2 mRNA expression was seen in the placenta but not in the uterus. Additionally, NAC was not sufficient to rescue DHT+insulin-induced mitochondria-morphological abnormalities in the uterus, whereas the same treatment partially reversed such abnormalities in the placenta. Finally, we demonstrated that NAC selectively normalized uterine leukemia inhibitory factor, osteopontin/secreted phosphoprotein 1, progesterone receptor, and homeobox A11 mRNA expression and placental estrogen related receptor beta and trophoblast specific protein alpha mRNA expression. Collectively, our data provide insight into how NAC exerts beneficial effects on differentially attenuating gravid uterine and placental ferroptosis in a PCOS-like rat model with fetal loss. These results indicate that exogenous administration of NAC represents a potential therapeutic strategy in the treatment of hyperandrogenism and insulin resistance-induced uterine and placental dysfunction.

    • Genome-scale mutagenesis, phenotypic screening, and tracking the causal mutations is a powerful approach for genetic analysis. However, classic mutagenesis approaches require extensive effort to identify causal mutations. It is desirable to demonstrate a powerful approach for rapid trackable mutagenesis. Here, we mapped the distribution of nonhomologous end joining (NHEJ)-mediated integration for the first time and demonstrated that it can be used for constructing the genome-scale trackable mutagenesis library in Yarrowia lipolytica. The sequencing of 9.15 x 10(5) insertions showed that NHEJ-mediated integration inserted DNA randomly across the chromosomes, and the transcriptional regulatory regions exhibited integration preference. The insertions were located in both nucleosome-occupancy regions and nucleosome-free regions. Using NHEJ-mediated integration to construct the genome-scale mutagenesis library, the new targets that improved beta-carotene biosynthesis and acetic acid tolerance were identified rapidly. This mutagenesis approach is readily applicable to other organisms with strong NHEJ preference and will contribute to cell factory construction.

  • Fast Catalytic Esterification Using a Hydrophobized Zr-MOF with Acidic Ionic Liquid Linkers

    • A Zr-MOF with an acidic ionic liquid linker ([bpyH][CF3SO3]) were externally hydrophobized with polydimethylsiloxane (PDMS) via a vapor deposition method (denoted as PDMS@UiO-67-CF3SO3). The morphology of UiO-67-CF3SO3 was well maintained after PDMS coating, and PDMS had little effect on the chemical properties of UiO-67-CF3SO3, while the hydrophobicity of the surface of UiO-67-CF3SO3 was improved greatly as demonstrated by water contact angle measurement. The derived hydrophobized PDMS@UiO-67-CF3SO3 exhibited excellent catalytic performance in the esterification of acetic acid and isooctyl alcohol, it only needed 4 h to reach the reaction equilibrium, while for UiO-67-CF3SO3, it was 18 h. The decrease in reaction time was mainly attributed to the accelerated the removal of water and isooctyl acetate, resulting the increase rate in the forward reaction. In addition, in the other esterification of acids and alcohols, PDMS@UiO-67-CF3SO3 also showed good catalytic performance.

    • In this paper, an automation process to perform Reynolds-Averaged Navier-Stokes (RANS) computational fluid dynamics (CFD) analysis is developed to carry out aerodynamic design and optimization. The aircraft model/geometry is defined by a Common Parametric Aircraft Configuration Schema (CPACS) file, and the analyses are facilitated using high performance computers (HPC). As the computational capability of the available HPC systems is a limiting factor in the complexity of analyses that can be performed, a detailed performance analysis of the open source CFD code SU2 is undertaken and the profiling and performance analyses for large simulations are carried out.
