  • Guidelines for the use and interpretation of assays for monitoring autophagy
  • 2012
  • Ingår i: Autophagy. - : Informa UK Limited. - 1554-8635 .- 1554-8627. ; 8:4, s. 445-544
  • Forskningsöversikt (refereegranskat)abstract
    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Near-infrared broadband absorber with film-coupled multilayer nanorods
  • 2013
  • Ingår i: Optics Letters. - : Optical Society of America. - 0146-9592 .- 1539-4794. ; 38:13, s. 2247-2249
  • Tidskriftsartikel (refereegranskat)abstract
    • Turning the surfaces of noble metals (metasurfaces) into black (highly absorptive) surfaces can be potentially applied in thermophotovoltaics, sensing, tailoring thermal emissivity, etc. Here we demonstrate an extremely broadband absorber for the 900-1600 nm wavelength range with robust high absorption efficiency. The inexpensive droplet evaporation method is implemented to create patterns of nanoparticles dispersed on a gold film spaced by a thin dielectric layer. The diversity of the complicated random stacking of the chemically synthesized gold nanorods is the major factor for the broad absorption band. Such a metamaterial absorber may pave the way for cost-effective manufacture of large-area black metasurfaces.

  • Ordered Au nanocrystals on a substrate formed by light-induced rapid annealing
  • 2014
  • Ingår i: Nanoscale. - : Royal Society of Chemistry (RSC). - 2040-3364 .- 2040-3372. ; 6:3, s. 1756-1762
  • Tidskriftsartikel (refereegranskat)abstract
    • Light-induced rapid annealing (LIRA) is a widely used method to modify the morphology and crystallinity of noble metal nanoparticles, and the nanoparticles generally evolve into nanospheres. It is rather challenging to form faceted Au nanocrystals on a substrate using LIRA. Here the formation of spatially ordered Au nanocrystals using a continuous wave infrared laser is reported, assisted by a metamaterial perfect absorber. Faceted Au nanocrystals in truncated-octahedral or multi-twinned geometries can be obtained. The evolution of morphology and crystallinity of the Au nanoparticles during laser annealing is also revealed, where the crystal grain growth and the surface melting are shown to play key roles in nanocrystal formation. The evolution of morphology also gives the freedom of tuning the absorption spectrum of the metamaterial absorber. These findings provide a novel way for tailoring the morphology and crystallinity of metallic nanoparticles and may pave the way to fabricate refined nano-devices in many potential applications for optics, electronics, catalysis, surface-chemistry and biology.

  • Gold nanoparticle transfer through photothermal effects in a metamaterial absorber by nanosecond laser
  • 2014
  • Ingår i: Scientific Reports. - : Springer Science and Business Media LLC. - 2045-2322. ; 4, s. 6080-
  • Tidskriftsartikel (refereegranskat)abstract
    • A non-complicated, controllable method of metallic nanoparticle fabrication at low operating light power is proposed. The method is based on laser-induced forward transfer, using a metamaterial absorber as the donor to significantly enhance the photothermal effect and reduce the operating light fluence to 35 mJ/cm(2), which is much lower than that in previous works. A large number of metallic nanoparticles can be transferred by one shot of focused nanosecond laser pulses. Transferred nanoparticles exhibit good size uniformity and the sizes are controllable. The optical properties of transferred particles are characterized by dark-field spectroscopy and the experimental results agree with the simulation results.

  • Omnidispersible poly(ionic liquid)-functionalized cellulose nanofibrils : surface grafting and polymer membrane reinforcement
  • 2014
  • Ingår i: Chemical Communications. - : Royal Society of Chemistry (RSC). - 1359-7345 .- 1364-548X. ; 50:83, s. 12486-12489
  • Tidskriftsartikel (refereegranskat)abstract
    • We report a facile one-step route to graft poly(ionic liquid)s (PILs) onto cellulose nanofibrils (CNFs). The dispersibility of the PIL-functionalized CNFs in water and various organic solvents could be tuned by the choice of the PIL-binding anion. We demonstrate that such omnidispersible PIL@CNF hybrids can be used to reinforce porous poly(ionic liquid) membranes.

  • A population-based survey of the epidemiology of symptom-defined gastroesophageal reflux disease : the Systematic Investigation of Gastrointestinal Diseases in China
  • 2010
  • Ingår i: BMC Gastroenterology. - 1471-230X .- 1471-230X. ; 10, s. 94-
  • Tidskriftsartikel (refereegranskat)abstract
    • Background: The epidemiology of gastroesophageal reflux disease (GERD) has yet to be investigated using the symptomatic threshold criteria recommended by the Montreal Definition. This study aimed to determine the prevalence of symptom-defined GERD across five regions of China, and to investigate variables associated with GERD. Methods: A representative sample of 18 000 adults (aged 18-80 years) were selected equally from rural and urban areas in each region (n = 1800). According to the Montreal Definition, GERD is present when mild symptoms of heartburn and/ or regurgitation occur on >= 2 days a week, or moderate-to-severe symptoms of heartburn and/or regurgitation occur on >= 1 day a week. Results: In total, 16 091 participants completed the survey (response rate: 89.4%) and 16 078 responses were suitable for analysis. Applying the Montreal criteria, the prevalence of symptom-defined GERD was 3.1% and varied significantly (p < 0.001) among the five regions (from 1.7% in Guangzhou to 5.1% in Wuhan) and between rural and urban populations (3.8% vs 2.4%). Factors significantly associated with GERD included living in a rural area and a family history of gastrointestinal diseases. Conclusions: This population-based survey found that the prevalence of symptom-defined GERD in China was 3.1%, which is lower than that found in Western countries.

  • Photothermal Enhancement in Core-Shell Structured Plasmonic Nanoparticles
  • 2014
  • Ingår i: Plasmonics. - : Springer Science and Business Media LLC. - 1557-1955 .- 1557-1963. ; 9:3, s. 623-630
  • Tidskriftsartikel (refereegranskat)abstract
    • Plasmonic nanoparticles (NPs) with photothermal effects can be exploited as efficient heat sources in various applications. Here, the photothermal properties in core-shell structured plasmonic NPs, including metal/silica NP, silica/metal NP, and metal/silica/metal NP, are investigated. Compared with bare metal NPs, the core-shell plasmonic NPs not only exhibit extremely agile tunability in the surface plasmon resonances but also show considerably enhanced photothermal effects in terms of the maximum temperature rise. For metal/silica NPs and metal/silica/metal NPs, the SiO2 shells function as effective thermal-protective layers for enhanced photothermal effect. For silica/metal NPs, the SiO2 core and the metal shell show uniform temperature rise. These findings are essential for applying the core-shell structured plasmonic NPs on photothermal imaging, nanofluidics, etc.

  • Suspended InAsnanowire gate-all-around field-effect transistors
  • 2014
  • Ingår i: Applied Physics Letters. - : AIP Publishing. - 0003-6951 .- 1077-3118. ; 105:11
  • Tidskriftsartikel (refereegranskat)abstract
    • Gate-all-around field-effect transistors are realized with thin, single-crystalline, pure-phase InAs nanowires grown by molecular beam epitaxy. At room temperature, the transistors show a desired high on-state current I-on of similar to 10 mu A arid an on-off current ratio I-on/I-off of as high as 10(6) at sourcedrain bias voltage of 50 mV and gate length of 1 mu m with a gate underlap spacing of 1 mu m from the source and from the drain. At low temperatures, the on-state current I-on is only slightly reduced, while the ratio I-on/I-off is increased to 10(7). The field-effect mobility in the nanowire channels is also investigated and found to be similar to 1500 cm(2)/V s at room temperature and similar to 2000 cm(2)/V s at low temperatures. The excellent performance of the transistors is explained in terms of strong electrostatic and quantum confinements of carriers in the nanowires. (C) 2014 A1P Publishing LLC.
