    • Intrachromosomal amplification of chromosome 21 (iAMP21) defines a distinct cytogenetic subgroup of childhood B-cell precursor acute lymphoblastic leukaemia (BCP-ALL). To date, fluorescence in situ hybridisation (FISH), with probes specific for the RUNX1 gene, provides the only reliable detection method (five or more RUNX1 signals per cell). Patients with iAMP21 are older (median age 9 years) with a low white cell count. Previously, we demonstrated a high relapse risk when these patients were treated as standard risk. Recent studies have shown improved outcome on intensive therapy. In view of these treatment implications, accurate identification is essential. Here we have studied the cytogenetics and outcome of 530 iAMP21 patients that highlighted the association of specific secondary chromosomal and genetic changes with iAMP21 to assist in diagnosis, including the gain of chromosome X, loss or deletion of chromosome 7, ETV6 and RB1 deletions. These iAMP21 patients when treated as high risk showed the same improved outcome as those in trial-based studies regardless of the backbone chemotherapy regimen given. This study reinforces the importance of intensified treatment to reduce the risk of relapse in iAMP21 patients. This now well-defined patient subgroup should be recognised by World Health Organisation (WHO) as a distinct entity of BCP-ALL.

    • © 2014, American Meteorological Society. All rights reserved. In 2013, the vast majority of the monitored climate variables reported here maintained trends established in recent decades. ENSO was in a neutral state during the entire year, remaining mostly on the cool side of neutral with modest impacts on regional weather patterns around the world. This follows several years dominated by the effects of either La Niña or El Niño events. According to several independent analyses, 2013 was again among the 10 warmest years on record at the global scale, both at the Earth’s surface and through the troposphere. Some regions in the Southern Hemisphere had record or near-record high temperatures for the year. Australia observed its hottest year on record, while Argentina and New Zealand reported their second and third hottest years, respectively. In Antarctica, Amundsen-Scott South Pole Station reported its highest annual temperature since records began in 1957. At the opposite pole, the Arctic observed its seventh warmest year since records began in the early 20th century. At 20-m depth, record high temperatures were measured at some permafrost stations on the North Slope of Alaska and in the Brooks Range. In the Northern Hemisphere extratropics, anomalous meridional atmospheric circulation occurred throughout much of the year, leading to marked regional extremes of both temperature and precipitation. Cold temperature anomalies during winter across Eurasia were followed by warm spring temperature anomalies, which were linked to a new record low Eurasian snow cover extent in May. Minimum sea ice extent in the Arctic was the sixth lowest since satellite observations began in 1979. Including 2013, all seven lowest extents on record have occurred in the past seven years. Antarctica, on the other hand, had above-average sea ice extent throughout 2013, with 116 days of new daily high extent records, including a new daily maximum sea ice area of 19.57 million km2 reached on 1 October. ENSO-neutral conditions in the eastern central Pacific Ocean and a negative Pacific decadal oscillation pattern in the North Pacific had the largest impacts on the global sea surface temperature in 2013. The North Pacific reached a historic high temperature in 2013 and on balance the globally-averaged sea surface temperature was among the 10 highest on record. Overall, the salt content in nearsurface ocean waters increased while in intermediate waters it decreased. Global mean sea level continued to rise during 2013, on pace with a trend of 3.2 mm yr-1 over the past two decades. A portion of this trend (0.5 mm yr-1) has been attributed to natural variability associated with the Pacific decadal oscillation as well as to ongoing contributions from the melting of glaciers and ice sheets and ocean warming. Global tropical cyclone frequency during 2013 was slightly above average with a total of 94 storms, although the North Atlantic Basin had its quietest hurricane season since 1994. In the Western North Pacific Basin, Super Typhoon Haiyan, the deadliest tropical cyclone of 2013, had 1-minute sustained winds estimated to be 170 kt (87.5 m s-1) on 7 November, the highest wind speed ever assigned to a tropical cyclone. High storm surge was also associated with Haiyan as it made landfall over the central Philippines, an area where sea level is currently at historic highs, increasing by 200 mm since 1970. In the atmosphere, carbon dioxide, methane, and nitrous oxide all continued to increase in 2013. As in previous years, each of these major greenhouse gases once again reached historic high concentrations. In the Arctic, carbon dioxide and methane increased at the same rate as the global increase. These increases are likely due to export from lower latitudes rather than a consequence of increases in Arctic sources, such as thawing permafrost. At Mauna Loa, Hawaii, for the first time since measurements began in 1958, the daily average mixing ratio of carbon dioxide exceeded 400 ppm on 9 May.   • Navier-Stokes Simulations of Fluid Flow Through a Rock Fracture

    • A surface profilometer was used to measure fracture profiles every 10 microns over the surfaces of a replica of a fracture in a red Permian sandstone, to within an accuracy of a few microns. These surface data were used as input to two finite element codes that solve the Navier-Stokes equations and the Reynolds equation, respectively. Numerical simulations of flow through these measured aperture fields were carried out at different values of the mean aperture, corresponding to different values of the relative roughness. Flow experiments were also conducted in casts of two regions of the fracture. At low Reynolds numbers, the Navier-Stokes simulations yielded transmissivities for the two fracture regions that were closer to the experimental values than were the values predicted by the lubrication model. In general, the lubrication model overestimated the transmissivity by an amount that varied as a function of the relative roughness, defined as the standard deviation of the aperture divided by the mean aperture. The initial deviations from linearity, for Reynolds numbers in the range 1-10, were consistent with the "weak inertia" model developed by Mei and Auriault for porous media, and with the results obtained computationally by Skjetne et al in 1999 on a two-dimensional self-affine fracture. In the regime 10 < Re < 40, both the computed and measured transmissivities could be fit very well to a Forchheimer-type equation, in which the additional pressure drop varies quadratically with the Reynolds number.

    • Biocomposite hydrogels with carboxymethylated, nanofibrillated cellulose (c-NFC) powder were prepared by UV polymerization of N-vinyl-2-pyrrolidone with Tween 20 trimethacrylate as a crosslinking agent for replacement of the native, human nucleus pulposus (NP) in intervertebral discs. The swelling ratios and the moduli of elasticity in compression of neat and biocomposite hydrogels were evaluated in dependence of c-NFC concentration (ranging from 0 to 1.6% v/v) and degree of substitution (DS, ranging from 0 to 0.23). The viscoelastic properties in shear and the material relaxation behavior in compression were measured for neat and biocomposite hydrogels containing 0.4% v/v of fibrils (DS ranging from 0 to 0.23) and their morphologies were characterized by cryo-scanning electron microscopy (cryo-SEM). The obtained results show that the biocomposite hydrogels can successfully mimic the mechanical and swelling behavior of the NP. In addition, the presence of the c-NFC show lower strain values after cyclic compression tests and consequently create improved material relaxation properties, compared to neat hydrogels. Among the tested samples, the biocomposite hydrogel containing 0.4% v/v of c-NFC with a DS of 0.17 shows the closest behavior to native NP. Further investigation should focus on evaluation and improvement of the long-term relaxation behavior.

  • Assessing the Uncertainty in Projecting Local Mean Sea Level from Global Temperature

    • The process of moving from an ensemble of global climate model temperature projections to local sea level projections requires several steps. Sea level was estimated in Olympia, Washington (a city that is very concerned with sea level rise because parts of downtown are barely above mean highest high tide), by relating global mean temperature to global sea level; relating global sea level to sea levels at Seattle, Washington; and finally relating Seattle to Olympia. There has long been a realization that accurate assessment of the precision of projections is needed for science-based policy decisions. When a string of statistical and/or deterministic models is connected, the uncertainty of each individual model needs to be accounted for. Here the uncertainty is quantified for each model in the described system and the total uncertainty is assessed in a cascading effect throughout the system. The projected sea level rise over time and its total estimated uncertainty are visualized simultaneously for the years 2000-2100, the increased uncertainty due to each of the component models at a particular projection year is identified, and estimates of the time at which a certain sea level rise will first be reached are made.

    • Inkjet ink analysis is the best way to discriminate between printed documents, or even though more difficult, to connect an inkjet printed document with a brand or model of printers. Raman spectroscopy and laser desorption mass spectrometry (LDMS) have been demonstrated as powerful tools for dyes and pigments analysis, which are ink components. The aim of this work is to evaluate the aforementioned techniques for inkjet inks analysis in terms of discriminating power, information quality, and nondestructive capability. So, we investigated 10 different inkjet ink cartridges (primary colors and black), 7 from the HP manufacturer and one each from Epson, Canon and Lexmark. This paper demonstrates the capabilities of three methods: Raman spectroscopy, LDMS and MALDI-MS. Raman spectroscopy, as it is preferable to try the nondestructive approach first, is successfully adapted to the analysis of color printed documents in most cases. For analysis of color inkjet inks by LDMS, we show that a MALDI matrix (9-aminoacridine, 9AA) is needed to desorb and to ionize dyes from most inkjet inks (except Epson inks). Therefore, a method was developed to apply the 9AA MALDI matrix directly onto the piece of paper while avoiding analyte spreading. The obtained mass spectra are very discriminating and lead to information about ink additives and paper compositions. Discrimination of black inkjet printed documents is more difficult because of the common use of carbon black as the principal pigment. We show for the first time the possibility to discriminate between two black-printed documents coming from different, as well as from the same, manufacturers. Mass spectra recorded from black inks in positive ion mode LDMS detect polyethylene glycol polymers which have characteristic mass distributions and end groups. Moreover, software has been developed for rapid and objective comparison of the low mass range of these positive mode LDMS spectra which have characteristic unknown peaks.

  • Impact of stress on solute transport in a fracture network : A comparison study

    • This paper compares numerical modeling of the effect of stress on solute transport (advection and matrix diffusion) in fractured rocks in which fracture apertures are correlated with fracture lengths. It is mainly motivated by the performance and safety assessments of underground radioactive waste repositories. Five research teams used different approaches to model stress/deformation, flow and transport processes, based on either discrete fracture network or equivalent continuum models. The simulation results derived by various teams generally demonstrated that rock stresses could significantly influence solute transport processes through stress-induced changes in fracture apertures and associated changes in permeability. Reasonably good agreement was achieved regarding advection and matrix diffusion given the same fracture network, while some observed discrepancies could be explained by different mechanical or transport modeling approaches.
