A common coding variant in CASP8 is associated with breast cancer risk

- The Breast Cancer Association Consortium (BCAC) has been established to conduct combined case-control analyses with augmented statistical power to try to confirm putative genetic associations with breast cancer. We genotyped nine SNPs for which there was some prior evidence of an association with breast cancer: CASP8 D302H (rs1045485), IGFBP3 -202 C --> A (rs2854744), SOD2 V16A (rs1799725), TGFB1 L10P (rs1982073), ATM S49C (rs1800054), ADH1B 3' UTR A --> G (rs1042026), CDKN1A S31R (rs1801270), ICAM5 V301I (rs1056538) and NUMA1 A794G (rs3750913). We included data from 9-15 studies, comprising 11,391-18,290 cases and 14,753-22,670 controls. We found evidence of an association with breast cancer for CASP8 D302H (with odds ratios (OR) of 0.89 (95% confidence interval (c.i.): 0.85-0.94) and 0.74 (95% c.i.: 0.62-0.87) for heterozygotes and rare homozygotes, respectively, compared with common homozygotes; P(trend) = 1.1 x 10(-7)) and weaker evidence for TGFB1 L10P (OR = 1.07 (95% c.i.: 1.02-1.13) and 1.16 (95% c.i.: 1.08-1.25), respectively; P(trend) = 2.8 x 10(-5)). These results demonstrate that common breast cancer susceptibility alleles with small effects on risk can be identified, given sufficiently powerful studies.

- The third-order nonlinearity is measured for TeO2-ZnO-Na2O (TZN) glasses codoped with BaO, Nb2O5, and La2O3. The results for the sign and magnitude of the nonlinearity were obtained using a combination of the eclipse Z-scan with thermal nonlinearity managed Z-scan, whereas the Kerr shutter technique was employed to obtain the electronic time response of the nonlinearity, all performed with 76 MHz repetition rate 150 fs pulses at 800 nm. The results show a fast response (< 200 fs) and a nonlinear refractive index varying from 1.5 to 3.5x10(-15) cm(2)/W, depending upon glass composition. At the peak power level employed, nonlinear absorption was negligible. The results obtained are in good agreement with other tellurite compositions reported, confirming the electronic origin of the nonlinearity.

- We discuss two dimensional N-extended supersymmetry in Euclidean signature and its R-symmetry. For N = 2, the R-symmetry is SO(2) x SO(1, 1), so that only an A-twist is possible. To formulate a B-twist, or to construct Euclidean N = 2 models with H-flux so that the target geometry is generalised Kahler, it is necessary to work with a complexification of the sigma models. These issues are related to the obstructions to the existence of non-trivial twisted chiral superfields in Euclidean superspace.

- We investigate the topological theory obtained by twisting the N = (2, 2) supersymmetric nonlinear sigma model with target a bihermitian space with torsion. For the special case in which the two complex structures commute, we show that the action is a Q-exact term plus a quasi-topological term. The quasi-topological term is locally given by a closed two-form which corresponds to a flat gerbe-connection and generalises the usual topological term of the A-model. Exponentiating it gives a Wilson surface, which can be regarded as a generalization of a Wilson line. This makes the quantum theory globally well-defined.

- White matter damage and inflammatory response are important secondary outcomes after acute neural disorders. Nevertheless, a few studies addressed the temporal outcomes of these pathological events using non-traumatic models of acute brain injury. In the present study, we describe acute inflammatory response and white matter neuropathology between 1 and 7 days after acute excitotoxic striatal damage. Twenty micrometer sections were stained by hematoxylin and eosin technique for gross histopathological analysis and immunolabed for neutrophils (anti-mbs-1), activated macrophages/microglia (anti-ed1), astrocytes (anti-gfap), damaged axons (anti-beta app) and myelin basic protein (MBP). Recruitment peak of neutrophils and macrophages occurred at 1 and 7 days post-nmda injection, respectively. Diffuse damaged axons (beta-app + end-bulbs) were apparent at 7 days, concomitant with progressive myelin impairment and astrocytosis. Further studies using electron microscopy and blockers of inflammatory response and glutamatergic receptors should be performed to confirm and address the mechanisms of white matter damage following an excitotoxic lesion.
