  • Millimeter-wave beam scattering and induced broadening by plasma turbulence in the TCV tokamak

    • The scattering of millimeter-wave beams from electron density fluctuations and the associated beam broadening are experimentally demonstrated. Using a dedicated setup, instantaneous deflection and (de-)focusing of the beam due to density blobs on the beam path are shown to agree with full-wave simulations. The detected time-averaged wave power transmitted through the turbulent plasma is reproduced by the radiative-transfer model implemented in the WKBeam code, which predicts a ∼50% turbulence-induced broadening of the beam cross-section. The role of core turbulence for the considered geometry is highlighted.

  • Monitoring the magnetar SGR 1935+2154 with the MAGIC telescopes

    • The Galactic magnetar SGR 1935+2154 was associated with a bright, millisecond-timescale fast radio burst (FRB) which occured in April 2020, during a flaring episode. This was the first time an FRB was unequivocally associated with a Galactic source, and the first FRB for which the nature of the emitting source was identified. Moreover, it was the first FRB with a counterpart at another wavelength correlated in time, an atypical, hard X-ray burst, which provides clear evidence for accompanying non-thermal processes. The MAGIC Telescopes are Imaging Air Cherenkov Telescopes (IACTs) sensitive to very-high-energy (VHE, E>100 GeV) gamma rays. Located at the center of the camera lies the MAGIC Central pixel, a single fully-modified photosensor-toreadout chain to measure millisecond-duration optical signals, displaying a maximum sensitivity at a wavelength of 350 nm. This allows MAGIC to operate simultaneously both as a VHE gammaray and a fast optical telescope. The MAGIC telescopes have monitored SGR 1935+2154 in a multiwavelength campaign involving X-ray, radio and optical facilities. In this contribution, we will show the results on the search for the VHE counterpart of the first SGR-FRB source in this multiwavelength context, as well as the search for fast optical bursts with the MAGIC Central Pixel.

  • Imitation of β-lactam binding enables broad-spectrum metallo-β-lactamase inhibitors

    • Carbapenems are vital antibiotics, but their efficacy is increasingly compromised by metallo-β-lactamases (MBLs). Here we report the discovery and optimization of potent broad-spectrum MBL inhibitors. A high-throughput screen for NDM-1 inhibitors identified indole-2-carboxylates (InCs) as potential β-lactamase stable β-lactam mimics. Subsequent structure-activity relationship studies revealed InCs as a new class of potent MBL inhibitor, active against all MBL classes of major clinical relevance. Crystallographic studies revealed a binding mode of the InCs to MBLs that, in some regards, mimics that predicted for intact carbapenems, including with respect to maintenance of the Zn(II)-bound hydroxyl, and in other regards mimics binding observed in MBL-carbapenem product complexes. InCs restore carbapenem activity against multiple drug-resistant Gram-negative bacteria and have a low frequency of resistance. InCs also have a good in vivo safety profile, and when combined with meropenem show a strong in vivo efficacy in peritonitis and thigh mouse infection models.

  • 4D and 5D phase-space tomography using slowing-down physics regularization

    • We compute reconstructions of 4D and 5D fast-ion phase-space distribution functions in fusion plasmas from synthetic projections of these functions. The fast-ion phase-space distribution functions originating from neutral beam injection (NBI) at TCV and Wendelstein 7-X (W7-X) at full, half, and one-third injection energies can be distinguished and particle densities of each component inferred based on 20 synthetic spectra of projected velocities at TCV and 680 at W7-X. Further, we demonstrate that an expansion into a basis of slowing-down distribution functions is equivalent to regularization using slowing-down physics as prior information. Using this technique in a Tikhonov formulation, we infer the particle density fractions for each NBI energy for each NBI beam from synthetic measurements, resulting in six unknowns at TCV and 24 unknowns at W7-X. Additionally, we show that installing 40 LOS in each of 17 ports at W7-X, providing full beam coverage and almost full angle coverage, produces the highest quality reconstructions.

  • The global distribution of plants used by humans

    • Plants sustain human life. Understanding geographic patterns of the diversity of species used by people is thus essential for the sustainable management of plant resources. Here, we investigate the global distribution of 35,687 utilized plant species spanning 10 use categories (e.g., food, medicine, material). Our findings indicate general concordance between utilized and total plant diversity, supporting the potential for simultaneously conserving species diversity and its contributions to people. Although Indigenous lands across Mesoamerica, the Horn of Africa, and Southern Asia harbor a disproportionate diversity of utilized plants, the incidence of protected areas is negatively correlated with utilized species richness. Finding mechanisms to preserve areas containing concentrations of utilized plants and traditional knowledge must become a priority for the implementation of the Kunming-Montreal Global Biodiversity Framework.

    • Integrating efficient connectivity, positioning and sensing functionalities into 5G New Radio (NR) and beyond mobile cellular systems is one timely research paradigm, especially at mm-wave and sub-THz bands. In this article, we address the radio-based sensing and environment mapping prospect with specific emphasis on the user equipment (UE) side. We first describe an efficient $\ell_1$-regularized least-squares (LS) approach to obtain sparse range--angle charts at individual measurement or sensing locations. For the subsequent environment mapping, we then introduce a novel state model for mapping diffuse and specular scattering, which allows efficient tracking of individual scatterers over time using interacting multiple model (IMM) extended Kalman filter and smoother. Also the related measurement selection and data association problems are addressed. We provide extensive numerical indoor mapping results at the 28~GHz band deploying OFDM-based 5G NR uplink waveform with 400~MHz channel bandwidth, covering both accurate ray-tracing based as well as actual RF measurement results. The results illustrate the superiority of the dynamic tracking-based solutions, compared to static reference methods, while overall demonstrate the excellent prospects of radio-based mobile environment sensing and mapping in future mm-wave networks.

  • Small molecule inhibitor of OGG1 blocks oxidative DNA damage repair at telomeres and potentiates methotrexate anticancer effects

    • The most common oxidative DNA lesion is 8-oxoguanine which is mainly recognized and excised by the 8-oxoG DNA glycosylase 1 (OGG1), initiating the base excision repair (BER) pathway. Telomeres are particularly sensitive to oxidative stress (OS) which disrupts telomere homeostasis triggering genome instability. In the present study, we have investigated the effects of inactivating BER in OS conditions, by using a specific inhibitor of OGG1 (TH5487). We have found that in OS conditions, TH5487 blocks BER initiation at telomeres causing an accumulation of oxidized bases, that is correlated with telomere losses, micronuclei formation and mild proliferation defects. Moreover, the antimetabolite methotrexate synergizes with TH5487 through induction of intracellular reactive oxygen species (ROS) formation, which potentiates TH5487-mediated telomere and genome instability. Our findings demonstrate that OGG1 is required to protect telomeres from OS and present OGG1 inhibitors as a tool to induce oxidative DNA damage at telomeres, with the potential for developing new combination therapies for cancer treatment.
