  • Effect of protein release rates from tablet formulations on the immune response after sublingual immunization

    • Dry vaccine formulations for sublingual administration would provide great advantages for public health use, especially in developing countries, since they are easy to administer and might also have improved stability properties. This study investigates the influence of protein release rate from mucoadhesive twolayer tablets on the elicited antibody responses after sublingual immunization. Two fast release tablets, one based on a mixture of lactose and microcrystalline cellulose (MCC) and one protein coated ethylcellulose (EC) tablet, and three hydrophilic matrix tablets with extended release (ER) properties based on HPMC 90 SH 100000 or Carbopol® 974-P NF were tested. The in vitro release profiles of the model protein ovalbumin (OVA) from these tablets were characterized and correlated to the in vivo potential of the tablets to induce an immune response after sublingual immunization in BALB/c mice. It could be concluded that a tablet with fast protein release elicits antibody titres not significantly different from titres obtained with OVA in solution, whereas low immune responses were observed with a slow release of OVA from the ER formulations. Thus, an ER tablet seems not favorable for vaccine delivery to the sublingual mucosa. Thus, we can present a fast releasing tablet formulation with attractive features for sublingual immunization, whereas the use of ER formulations for sublingual vaccination has to be investigated more in detail.

  • Preparation and evaluation of a freeze-dried oral killed cholera vaccine formulation

    • Different oral liquid cholera vaccines have proved to be safe and effective, but their formulations present problems for use in low-income countries, since large package volumes have to be transported and cold chain maintenance is required. A solid state formulation would here be more advantageous, and consequently, the possibility to develop a dry cholera vaccine formulation by freeze-drying was investigated. The ability of sucrose, trehalose and mannitol to provide process stabilization during freeze-drying was tested on a formalin-killed whole-cell Vibrio cholerae model vaccine. A matrix of sucrose or trehalose prevented bacterial aggregation, preserved cell morphology and maintained practically completely the protective lipopolysaccharide (LPS) antigen on the cell surface and its reactivity with specific antibody in vitro. After reconstitution, this formulation also retained the capacity to elicit a strong serum and gut mucosal anti-LPS antibody response in orally immunized mice, as compared to the corresponding liquid vaccine formulation. The full preservation of the in vivo immunogenicity was also maintained when the internationally widely licensed oral cholera vaccine Dukoral (TM), which comprises a cocktail of inactivated V. cholerae together with cholera toxin B-subunit (CTB), was freeze-dried using sucrose for stabilization. Thus, we present a process generating a dry oral inactivated whole-cell cholera vaccine formulation with attractive features for public health use in cholera-afflicted settings.

    • Within a project called “The sustainable greenhouse as a complement to buildings” financed by the Delegation of sustainable cities in Sweden, researchers, consultants and students have during 2013 developed, tested and evaluated various solutions for cultivating under roofs in or close to dwellings. We included solutions such as cultivating a plot in a rooftop greenhouse, a circular cultivation balcony or to cultivate in one’s kitchen or living room using a fish and vegetable furniture, a green wall, a green lamp or a green frame attached to the wall. The practical trials were preceded by a literature survey that described good examples of cultivation under roofs in various cities around the world.The results show that here are many inspiring examples of cultivating under roofs in cities and that such cultivation has gained some momentum. Our practical trials show among others that urban agriculture seems challenging and appealing for many but that the solutions that we have tested seem to be too expensive and partly also too time-consuming. Those who like to cultivate do so because it is fun and they can get products of good quality. To lessen the environmental impacts are also a reason but here our results that productivity in urban farming has to be higher than in our trials in order for the products to be considered as “climate-friendly”. Growing fish at home showed to be rather unpopular and many thought that slaughtering the fish by themselves would be nasty. Others results from the project are a water cooled LED lamp built in four copies and successfully installed in a fish and vegetable production unit as well as drawings and costs estimates for a cultivation bench.Proposals for further studies include analysis of flight and nutrient requirements for cultivation under roofs in cities as well as measurements of productivity. More low cost solutions for cultivating under glass should also be developed. To expand and maintain interest and knowledge for cultivation under roofs in cities is not only important for increasing social sustainability and eventually the environmental one as well but also because of the need to enhance preparedness.

  • Quantification of protein concentration by the Bradford method in the presence of pharmaceutical polymers

    • We investigated how the Bradford assay for measurements of protein released from a drug formulation may be affected by a concomitant release of a pharmaceutical polymer used to formulate the protein delivery device. The main result is that polymer-caused perturbations of the Coomassie dye absorbance at the Bradford monitoring wavelength (595 nm) can be identified and corrected by recording absorptionspectra in the region of 350–850 mm. The pharmaceutical polymers Carbopol and chitosan illustrate two potential types of perturbations in the Bradford assay, whereas the third polymer, hydroxypropylmethylcellulose (HPMC), acts as a nonperturbing control. Carbopol increases the apparent absorbance at 595 nm because the polymer aggregates at the low pH of the Bradford protocol, causing a turbidity contribution that can be corrected quantitatively at 595 nm by measuring the sample absorbance at 850 nm outside the dye absorption band. Chitosan is a cationic polymer under Bradford conditions and interacts directly with the anionic Coomassie dye and perturbs its absorption spectrum, including 595 nm. In this case, the Bradford method remains useful if the polymer concentration is known but should be used with caution in release studies where the polymer concentration may vary and needs to be measured independently.

  • Protein kinase Cα suppresses the expression of STC1 in MDA-MB-231 breast cancer cells

    • Several protein kinase C (PKC) isoforms have been shown to influence different cellular processes that may contribute to the malignancy of breast cancer cells. To obtain insight into mechanisms mediating the PKC effects, global gene expression was analyzed in MDA-MB-231 breast cancer cells in which PKCα, PKCδ or PKCε had been down-regulated with siRNA. Gene set enrichment analyses revealed that hypoxia-induced genes were enriched among genes that increased in PKCα-down-regulated cells. The STC1 mRNA, encoding stanniocalcin 1, was particularly up-regulated following depletion of PKCα and was also induced by hypoxia. Both hypoxia and PKCα down-regulation also led to increased STC1 protein levels. The results demonstrate that PKCα suppresses the expression of STC1 in breast cancer cells

  • Linking the Klein-Bell ADL Scale to the International Classification of Functioning, Disability, and Health (ICF) : Linking an ADL scale to the ICF

    • Objective: The main objective of this study was to link the Klein-Bell Activities of Daily Living (KB) Scale to the International Classification of Functioning, Disability and Health (ICF), in order to validate the KB Scale content-wise. Design: A qualitative approach was used with directed content analysis. Methods: Concepts in the KB Scale items were linked to ICF categories according to established rules. This was followed by 4 analyses: examination of the linked categories' frequency distribution; comparison of these categories with Core Sets for spinal cord injury (SCI) and occupational therapists; calculation of content density, content diversity and range of linked categories; and calculation of agreement between two independent linkage versions. Results: All except one identified KB Scale concept could be linked to ICF categories. The occupational therapists Core Sets were most consistent with linked categories in the KB Scale. Content density, content diversity and range varied between the different KB Scale dimensions. Agreement was reliable for the whole KB Scale and for 5 of 6 dimensions. Conclusion: The ICF has provided a valuable reference to identify and quantify the concepts in the KB Scale. Furthermore, comparison between the KB Scale and ICF Core Sets provides insights into areas covered by these instruments.

  • Theory of mind in Children with Severe Speech and Physical Impairments

    • The development of a person's ability to understand other's thoughts and feelings, so called "theory of mind" (ToM), is subject to study. Children with communicative disabilities have exhibited problems in this respect, highlighting the role of language in the development of ToM. In this study, ToM was studied in children with cerebral palsy and severe speech impairments. Two tasks, differently dependent on verbal abilities, were used. The results were compared to those of a mental age matched group. The groups differed significantly on the verbally dependent task while difference in performance did not reach significance on the less verbally dependent one. The results are discussed in terms of a delayed development of ToM in children with severe speech and physical impairments, dependent on verbal abilities.
