  • AURKA F31I polymorphism and breast cancer risk in BRCA1 and BRCA2 mutation carriers: A consortium of investigators of modifiers of BRCA1/2 study

    • The AURKA oncogene is associated with abnormal chromosome segregation and aneuploidy and predisposition to cancer. Amplification of AURKA has been detected at higher frequency in tumors from BRCA1 and BRCA2 mutation carriers than in sporadic breast tumors, suggesting that overexpression of AURKA and inactivation of BRCA1 and BRCA2 cooperate during tumor development and progression. The F31I polymorphism in AURKA has been associated with breast cancer risk in the homozygous state in prior studies. We evaluated whether the AURKA F31I polymorphism modifies breast cancer risk in BRCA1 and BRCA2 mutation carriers from the Consortium of Investigators of Modifiers of BRCA1/2. Consortium of Investigators of Modifiers of BRCA1/2 was established to provide sufficient statistical power through increased numbers of mutation carriers to identify polymorphisms that act as modifiers of cancer risk and can refine breast cancer risk estimates in BRCA1 and BRCA2 mutation carriers. A total of 4,935 BRCA1 and 2,241 BRCA2 mutation carriers and 11 individuals carrying both BRCA1 and BRCA2 mutations was genotyped for F31I. Overall, homozygosity for the 311 allele was not significantly associated with breast cancer risk in BRCA1 and BRCA2 carriers combined [hazard ratio (HR), 0.91; 95% confidence interval (95% CI), 0.77-1.061. Similarly, no significant association was seen in BRCA1 (HR, 0.90; 95% Cl, 0.75-1.08) or BRCA2 carriers (HR, 0.93; 95% CI, 0.67-1.29) or when assessing the modifying effects of either bilateral prophylactic oophorectomy or menopausal status of BRCA1 and BRCA2 carriers. In summary, the F31I polymorphism in AURKA is not associated with a modified risk of breast cancer in BRCA1 and BRCA2 carriers.

  • Assessment of inorganic content of PM(2.5) particles sampled in a rural area north-east of Hanoi, Vietnam.

    • Atmospheric aerosols from seven rural sites in northern Vietnam, east of Hanoi, were sampled and analyzed. The aim of the study was to evaluate trace elemental and black carbon (BC) concentrations in fine particles (PM(2.5)) and to investigate the influence of the Pha Lai power plant and other pollution sources on regional air quality. Seven measurement stations were set up at selected rural sites and a campaign consisting of 12 two-day measurement periods was conducted from the end of May until the end of October 2000. At each location a pair of samplers was installed consisting of a cyclone loaded with Teflon filters and a modified Millipore air monitoring cartridge loaded with glass fibre filters. The obtained samples were analyzed for trace elements by Energy Dispersive X-ray Fluorescence (EDXRF), while a black smoke detector was used for BC analysis. Seventeen trace elements were analyzed; Br, Ca, Cl, Cr, Cu, Fe, K, Mn, Ni, Pb, Rb, S, Se, Sr, Ti, V and Zn and their concentrations evaluated. The results showed that BC, Ca, Cl, Fe, K and S dominated in the sampled atmospheric aerosols. The measured concentrations of the potentially hazardous trace elements Cr, Mn, Ni and Pb were all below the limits defined by Vietnamese standards of ambient air quality. Statistical evaluations indicated that coal and heavy fuel oil combustion were major sources of atmospheric pollutants in the area and that biomass burning and road transport had a marked influence on regional air quality. It was concluded that the Pha Lai power plant was the major source of coal combustion emissions. Trace element emissions originating from river transportation were suggested as another major source of atmospheric pollutants. The results indicate that the elemental concentrations in PM(2.5) are strongly influenced by seasonal variations. Further measurements are required to identify the impact of long-range transported continental air masses on the air quality of the investigated rural area.

  • Understanding the 3D structure of GaAs nanowires

    • The effects of lamellar twinning in epitaxial particle-assisted GaAs <111>B nanowires are investigated in an extensive high resolution electron microscopy (HRTEM) study of the low index zones <-100>, <-1-10>, <1-1-1> and <1-1-2>. As these directions are non-parallel to the (-1-1-1) twin planes we find that the twin segments exhibit two different zone axes as a consequence of twinning. In the first three cases the alternative zones were found to be <1-2-2> <11-4> and <11-5>. These findings are supported by a comparison of experimental HRTEM images and multi-slice simulations along with fast Fourier transform mapping. From the appearance of non-overlapping regions we conclude that the nanowires are bound by {111} facets only. The twin formation and the development of the stable side facets are discussed

  • On-line detection of microbial contaminations in animal cell reactor cultures using an electronic nose device

    • An electronic nose (EN) device was used to detect microbial and viral contaminations in a variety of animal cell culture systems. The emission of volatile components from the cultures accumulated in the bioreactor headspace, was sampled and subsequently analysed by the EN device. The EN, which was equipped with an array of 17 chemical gas sensors of varying selectivity towards the sampled volatile molecules, generated response patterns of up to 85 computed signals. Each 15 or 20 min a new gas sample was taken generating a new response pattern. A software evaluation tool visualised the data mainly by using principal component analysis. The EN was first used to detect microbial contaminations in a Chinese hamster ovary (CHO) cell line producing a recombinant human macrophage colony stimulating factor (rhM-CSF). The CHO cell culture was contaminated by Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Candida utilis which all were detected. The response patterns from the CHO cell culture were compared with monoculture references of the microorganisms. Second, contaminations were studied in an Sf-9 insect cell culture producing another recombinant protein (VP2 protein). Contaminants were detected from E. coli, a filamentous fungus and a baculovirus. Third, contamination of a human cell line, HEK-293, infected with E. coli exhibited comparable results. Fourth, bacterial contaminations could also be detected in cultures of a MLV vector producer cell line. Based on the overall experiences in this study it is concluded that the EN method has in a number of cases the potential to be developed into a useful on-line contamination alarm in order to support safety and economical operation for industrial cultivation. © Springer 2005.

  • NOVA V2362 CYGNI (NOVA CYGNI 2006): SPITZER, SWIFT, AND GROUND-BASED SPECTRAL EVOLUTION

    • Nova V2362 Cygni has undergone a number of very unusual changes. Ground-based spectroscopy initially revealed a normal sequence of events: the object faded and its near-infrared emission lines gradually shifted to higher excitation conditions until about day 100 when the optical fading reversed and the object slowly brightened. This was accompanied by a rise in the Swift X-ray telescope flux and a sudden shift in excitation of the visible and IR spectrum back to low levels. The new lower excitation spectrum revealed broad line widths and many P-Cygni profiles, all indicative of the ejection of a second shell. Eventually, dust formed, the X-ray brightness—apparently unaffected by dust formation—peaked and then declined, and the object faded at all wavelengths. The Spitzer dust spectra revealed a number of solid-state emission features that, at this time, are not identified.

  • New Escherichia coli outer membrane proteins identified through prediction and experimental verification

    • Many new Escherichia coli outer membrane proteins have recently been identified by proteomics techniques. However, poorly expressed proteins and proteins expressed only under certain conditions may escape detection when wild-type cells are grown under standard conditions. Here, we have taken a complementary approach where candidate outer membrane proteins have been identified by bioinformatics prediction, cloned and overexpressed, and finally localized by cell fractionation experiments. Out of eight predicted outer membrane proteins, we have confirmed the outer membrane localization for five-YftM, YaiO, YfaZ, CsgF, and YliI - and also provide preliminary data indicating that a sixth -YfaL- may be an outer membrane autotransporter.
