  • Kelb - A Real-Time Programming Environment for the Sony Aibo

    • Kelb is a new real-time programming environment developed at Uppsala University for the Sony AIBO ERS-210. It is aimed to provide efficiency by introducing a notion of light-weight tasks executing according to well-known real-time scheduling algorithms and resource protocols, while still allowing applications to be developed in a high-level abstract programming language. In this paper we give an overview of the design of Kelb and describe the status of the environment, currently including: a real-time programming language and compiler extending gcc for MIPS with support for time- and event-triggered tasks, a runtime library with support for static and dynamic preemptive scheduling algorithms (e.g. fixed priority and earliest deadline first), and a prototype connection to the Times tool allowing Kelb designs to be analysed for schedulabilty.

  • A Multivariate Screening Strategy for Investigating Metabolic Effects of Strenuous Physical Exercise in Human Serum

    • A novel hypothesis-free multivariate screening methodology for the study of human exercise metabolism in blood serum is presented. Serum gas chromatography/time-of-flight mass spectrometry (GC/TOFMS) data was processed using hierarchical multivariate curve resolution (H-MCR), and orthogonal partial least-squares discriminant analysis (OPLS-DA) was used to model the systematic variation related to the acute effect of strenuous exercise. Potential metabolic biomarkers were identified using data base comparisons. Extensive validation was carried out including predictive H-MCR, 7-fold full cross-validation, and predictions for the OPLS-DA model, variable permutation for highlighting interesting metabolites, and pairwise t tests for examining the significance of metabolites. The concentration changes of potential biomarkers were verified in the raw GC/TOFMS data. In total, 420 potential metabolites were resolved in the serum samples. On the basis of the relative concentrations of the 420 resolved metabolites, a valid multivariate model for the difference between pre- and post-exercise subjects was obtained. A total of 34 metabolites were highlighted as potential biomarkers, all statistically significant (p < 8.1E-05). As an example, two potential markers were identified as glycerol and asparagine. The concentration changes for these two metabolites were also verified in the raw GC/TOFMS data.The strategy was shown to facilitate interpretation and validation of metabolic interactions in human serum as well as revealing the identity of potential markers for known or novel mechanisms of human exercise physiology. The multivariate way of addressing metabolism studies can help to increase the understanding of the integrative biology behind, as well as unravel new mechanistic explanations in relation to, exercise physiology.

  • Vedlevande arters ekologi : kunskaper för skötsel av död ved

    • Betydelsen av död ved i skogen för biologisk mångfald är väl dokumenterad. Sveriges riksdag har fastställt 15 miljömål och varje mål är uppdelat i flera delmål. I ett av dessa mål, Levande skogar, konstateras att kvantiteten av hård död ved måste öka med åtminstone 40 procent i hela landet och betydligt mer i områden, där biologisk mångfald är speciellt hotad. Men än så länge finns inget detaljerat långsiktigt mål. Syftet med den här studien var att undersöka: 1. Vilken kvalitet och kvantitet av död ved behövs i skogen för att bevara mångfalden? 2. Var i landskapet är det viktigast att öka mängden död ved? 3. Hur når vi målet? Arbetet har baserats på litteraturöversikter och frågeformulär till markägare, avverkningsentreprenörer och tjänstemän inom skogsnäringen. Olika scenarier baserade på litteraturgranskningen framställdes via datorsimulationer, där mängden och spridningen av död ved varierade med en beräknad ökningstakt. Utifrån dessa studier kan vi dra följande slutsatser: 1. Somliga arter är specialiserade och kräver ett rikligt utbud av död ved (50 m3/ha eller mer). Dessa arter kan endast bevaras i skyddade områden. 2. Brukade skogar utanför skyddade områden med 20 m3 död ved/ha tycks vara högkvalitativa habitat för många arter. 3. I områden dominerade av skog bör sådana habitat täcka mellan 10 och 30 procent av landskapet. 4. Dessa områden bör lokaliseras i anslutning till skyddade områden och områden med sällsynta arter. 5. Om den här strategin antas, kommer medelvärdet för hela Sverige vara 10 m3 död ved/ha. 6. För att kunna nå det målet 2030 måste mängden död ved öka med 40 procent varje årtionde (vilket innebär en exponentiell ökning). 7. Men ännu viktigare än att nå ett visst medelvärde för hela Sverige är, att så snabbt som möjligt öka förekomsten av död ved i anslutning till områden med hotade arter. 8. Det gäller att så snabbt som möjligt identifiera dessa hotade områden/ biotoper och för det ändamålet behöver regionala bristanalyser tas fram. 9. Men död ved behövs även utanför speciellt rika områden, som en resurs för kommande generationer samt som spridningskorridor mellan olika “hot-spots”. Där kan mängden död ved vara mindre och ökningsgraden lägre. 10. Förutom ökningen av död ved är det viktigt med en större variation av olika kvaliteter samt en kontinuerlig tillförsel. 11. Att öka mängden död ved snabbt i hotade områden (hot-spots) enbart med hjälp av frivilliga åtgärder, kommer troligen att bli svårt. Därför kan olika nya slags politiska åtgärder vara nödvändiga (såsom ekonomisk kompensation). 12. För att definiera målet mer i detalj, behövs mer kunskap om den kritiska tröskeln, spridningsmöjligheter, habitat och substratanspråk samt populationsbiologi för några arter.

  • Core-needle biopsy performed by the cytopathologist : a technique to complement fine-needle aspiration of soft tissue and bone lesions

    • BACKGROUND: Fine-needle aspiration cytology (FNAC) is gaining increased popularity in the diagnosis of musculoskeletal lesions; and, in many patients, a definitive diagnosis can be rendered from aspiration smears alone. The main limitation of FNAC of soft tissue and bone neoplasms is in the evaluation of tissue architecture. In addition cytologic specimens are not always adequate for ancillary studies.METHODS: A consecutive series of 130 patients with soft tissue and bone lesions was examined by core-needle biopsy (CNB) performed by a cytopathologist in conjunction with FNAC. The findings of this combined diagnostic approach were compared with histologic diagnoses made on surgical biopsies and resected specimens from 86 patients. Adequate follow-up was available in all patients.RESULTS: FNAC combined with CNB correctly could identify 77 of 78 malignant lesions and 50 of 52 benign lesions. Only seven patients underwent incisional biopsy. The tumor subtype was determined correctly in 30 of 39 patients (77%) and the malignancy grade was determined in 35 of 39 patients (90%) with primary soft tissue and bone sarcomas compared with the biopsy or operative specimens.CONCLUSIONS: FNAC of musculoskeletal tumors/lesions complemented with CNB combined cytomorphology with tissue architecture and ancillary procedures. In the current study, obtaining FNAC as well as CNB at the same clinic visit and by the cytopathologist made preliminary diagnosis on the day of referral possible. This speeded diagnosis increased the number of correct diagnoses and usually enabled correct subtyping and malignancy grading of sarcomas.

    • In many nuclear power plants, areas of susceptible material in the reactor systems are replaced or mitigated. Many of the areas where the nickel-based weld metal Alloy 182 have been used, are not replaceable but need to be mitigated. One possibility to mitigate is to make known susceptible material non-accessible for the reactor coolant water by covering it with less susceptible materials. One such possibility that has been utilized frequently in the Swedish Boiling Water Reactor (BWR) fleet is in-lay welding of butt welds in the main circulation and feed water loops with the less susceptible Alloy 82, which has fewer reported failure cases under these conditions. The study focuses on the development of a Factor of Improvement between Alloy 182 and the replacement, Alloy 82 material. As part of this, a disposition curve under conditions relevant for Normal Water Chemistry, NWC, in the Swedish BWRs is presented.

  • The meaning of oral health-related quality of life for elderly persons with dementia

    • Objective. Studies of oral health developments increasingly include self-reported assessments of how oral health affects quality of life (QoL), referred to as “oral health-related QoL”. People with dementia are often excluded in studies of oral health-related QoL and thus our aim in this study was to explore this area in elderly persons with dementia. Material and methods. Eighteen elderly individuals (aged between 78 and 94 years) with dementia of varying degrees of severity were interviewed with the aid of an interview guide; pictures and objects were used as stimulus material (triggers). The material was analyzed using grounded theory as point of departure, and a professional assessment of the oral health of the participants was used as reference. Results. Four categories were identified: the ability to chew and eat, independence, oral problems, and teeth are important. These factors are largely consistent with those that have emerged in earlier studies of the elderly, but in some cases less pronounced in persons with dementia. Conclusion. The use of triggers is a positive way to communicate oral health-related QoL among persons suffering from dementia, although the material used in this study needs further evaluation and development.

  • An experimental study of the velocity field during filling of an ingot mould

    • In the present study the velocity field in a 1:3 scale water model of a 4.2-tonne ingot mould was determined using Laser Doppler Anemometry (LDA). The velocity was measured in the horizontal and vertical directions at several different locations along the centre plane of the model. The effect of different volumetric flow rates and water temperatures was also investigated. The reproducibility of the measurements was found to be satisfactory, since the mean velocity at any measurement location had an average difference of around 10% between two fillings. The effect of different volumetric flow rates showed that while decreasing the flow rate, the mean velocity at the bottom turned from an upward direction to a downward direction. At the top of the model the difference between the mean velocities for the different flow rates was less pronounced. An influence of the temperature on the mean velocity could be observed. However, further studies are required to verify this result.

    • Biologically active recombinant human Flt3 ligand was expressed and isolated from transgenic barley seeds. Its expression is controlled by a tissue specific promoter that confines accumulation of the recombinant protein to the endosperm tissue of the seed. The recombinant Flt3 ligand variant expressed in the seeds contains an HQ-tag for affinity purification on immobilized metal ion affinity chromatography (IMAC) resin. The tagged protein was purified from seed extracts to near homogeneity using sequential chromatography on IMAC affinity resin and cation exchange resin. We also show that the recombinant Flt3 ligand protein undergoes posttranslational modifications: it is a glycoprotein containing alpha-1,3-fucose and alpha-1,2-xylose. The HQ-tagged Flt3 ligand variant exhibits comparable biological activity to commercial Flt3 ligand. This is the first report showing expression and accumulation of recombinant human growth factor in barley seeds with a yield of active protein similar to a bacterial expression system. The present results demonstrate that plant molecular farming is a viable approach for the bioproduction of human-derived growth factors.

  • A Mathematical Model of an Impinging Air Jet on a Water Surface

    • A fundamental mathematical model of the flow field and surface deformation caused by an impinging jet in a top blown reactor has been developed. The results have been validated against water model experiments. More specifically, the predicted penetration depth has been found to agree well with surface deformation measurements and predictions using analytical equations. Furthermore, the predictions of the location of a vortex have been found to agree fairly well with PIV measurements. Calculations were also done to compare the widely used standard k-ε model against the realizable extension of the standard k-ε model to calculate the turbulent conditions of the flow. It was found that the penetration depth caused by the impinging jet on the liquid surface is relatively unaffected by the choice of turbulence model employed. However, when the main re-circulation loop in the bath was investigated there was a clear distinction in the flow fields produced when the two different turbulence models were used.

    • A novel modeling approach is presented where a computational fluid dynamics software is coupled to thermodynamic databases to obtain dynamic simulations of metallurgical process phenomena. The modeling approach has been used on a fundamental model of a top-blown converter. Reactions between gas-steel, gas-slag, steel-slag and gas-steel-slag have been considered. The results show that the mass transport in the surface area is totally controlled by convection. Also, that a large amount of CO produced during the decarburization might slow down the rate of decarburization in droplets ejected from the bath. For the present simulation conditions reflecting laboratory experiments, it was also seen that the amount of slag (FeO and/or SiO2) created is close to zero, i.e. only gas (CO+CO2) is created as the oxygen jet hits the steel bath. It was also illustrated how an extrapolation of the decarburization rate, sampled from a few seconds of simulation, could be done to get a rough estimate of the carbon content at a later stage in the process as long as the carbon content is relatively high. The overall conclusion is that it is possible to make a dynamic coupling of the Thermo-Calc databases and a CFD software to make dynamic simulations of metallurgical processes such as a top-blown converter.
