  • ZnT8 loss-of-function accelerates functional maturation of hESC-derived β cells and resists metabolic stress in diabetes

    • Human embryonic stem cell-derived β cells (SC-β cells) hold great promise for treatment of diabetes, yet how to achieve functional maturation and protect them against metabolic stresses such as glucotoxicity and lipotoxicity remains elusive. Our single-cell RNA-seq analysis reveals that ZnT8 loss of function (LOF) accelerates the functional maturation of SC-β cells. As a result, ZnT8 LOF improves glucose-stimulated insulin secretion (GSIS) by releasing the negative feedback of zinc inhibition on insulin secretion. Furthermore, we demonstrate that ZnT8 LOF mutations endow SC-β cells with resistance to lipotoxicity/glucotoxicity-triggered cell death by alleviating endoplasmic reticulum (ER) stress through modulation of zinc levels. Importantly, transplantation of SC-β cells with ZnT8 LOF into mice with preexisting diabetes significantly improves glycemia restoration and glucose tolerance. These findings highlight the beneficial effect of ZnT8 LOF on the functional maturation and survival of SC-β cells that are useful as a potential source for cell replacement therapies.

  • Humidity-Dependent Phase State of Gasoline Vehicle Emission-Related Aerosols

    • The phase states of primarily emitted and secondarily formed aerosols from gasoline vehicle exhausts were investigated by quantifying the particle rebound fraction (f). The rebound behaviors of gasoline vehicle emission-related aerosols varied with engines, fuel types, and photochemical aging time, showing distinguished differences from biogenic secondary organic aerosols. The nonliquid-to-liquid phase transition of primary aerosols emitted from port fuel injection (PFI) and gasoline direct injection (GDI) vehicles started at a relative humidity (RH) = 50 and 60%, and liquefaction was accomplished at 60 and 70%, respectively. The RH at which f declined to 0.5 decreased from 70 to 65% for the PFI case with 92# fuel, corresponding to the photochemical aging time from 0.37 to 4.62 days. For the GDI case, such RH enhanced from 60 to 65%. Our results can be used to imply the phase state of traffic-related aerosols and further understand their roles in urban atmospheric chemistry. Taking Beijing, China, as an example, traffic-related aerosols were mainly nonliquid during winter with the majority ambient RH below 50%, whereas they were mostly liquid during the morning rush hour of summer, and traffic-related secondary aerosols fluctuated between nonliquid and liquid during the daytime and tended to be liquid at night with increased ambient RH.

  • Significant effects of transport on nanoparticles during new particle formation events in the atmosphere of Beijing

    • The mechanisms of new particle formation (NPF) events that occurred under high aerosol loadings ("polluted" NPF) in the atmosphere have been unclear, which has inhibited the precision of particle pollution control. To deepen the understanding of how the "polluted" NPF events occur, a one-month comprehensive measurement was conducted in the atmosphere of Beijing during the summer of 2016. The "clean" NPF events (frequency = 22%) (condensation sink, CS < 0.015 s-1) were found to be caused by local nucleation and growth. The "polluted" NPF events (frequency = 28%) (CS > 0.015 s-1) were influenced by both local nucleation-growth and regional transport, and the contributions from the two factors to 6-25 nm particle number concentration were 60% and 40%, respectively. This study emphasized the importance of the transport for nanoparticles in relatively polluted atmospheres, and for that the regional joint particle pollution control would be an essential policy.   • Evidence of a causal effect of genetic tendency to gain muscle mass on uterine leiomyomata

    • Uterine leiomyomata (UL) are the most common tumours of the female genital tract and the primary cause of surgical removal of the uterus. Genetic factors contribute to UL susceptibility. To add understanding to the heritable genetic risk factors, we conduct a genome-wide association study (GWAS) of UL in up to 426,558 European women from FinnGen and a previous UL meta-GWAS. In addition to the 50 known UL loci, we identify 22 loci that have not been associated with UL in prior studies. UL-associated loci harbour genes enriched for development, growth, and cellular senescence. Of particular interest are the smooth muscle cell differentiation and proliferation-regulating genes functioning on the myocardin-cyclin dependent kinase inhibitor 1A pathway. Our results further suggest that genetic predisposition to increased fat-free mass may be causally related to higher UL risk, underscoring the involvement of altered muscle tissue biology in UL pathophysiology. Overall, our findings add to the understanding of the genetic pathways underlying UL, which may aid in developing novel therapeutics.

  • CONCERTO : Extracting the power spectrum of the [CII] emission line

    • Context. CONCERTO is the first experiment to perform a [CII] line intensity mapping (LIM) survey on the COSMOS field to target z > 5.2. Measuring the [CII] angular power spectrum allows us to study the role of dusty star-forming galaxies in the star formation history during the epochs of Reionization and post-Reionization. The main obstacle to this measurement is the contamination by bright foregrounds: the dust continuum emission and atomic and molecular lines from foreground galaxies at z ≲ 3.Aims. We evaluate our ability to retrieve the [CII] signal in mock observations of the sky using the Simulated Infrared Dusty Extragalactic Sky (SIDES), which covers the mid-infrared to millimetre range. We also measure the impact of field-to-field variance on the residual foreground contamination.Methods. We compared two methods for dealing with the dust continuum emission from galaxies (i.e. the cosmic infrared background fluctuations): the standard principal component analysis (PCA) and the asymmetric re-weighted penalized least-squares (arPLS) method. For line interlopers, the strategy relies on masking low-redshift galaxies using the instrumental beam profile and external catalogues. As we do not have observations of CO or deep-enough classical CO proxies (such as LIR), we relied on the COSMOS stellar mass catalogue, which we demonstrate to be a reliable CO proxy for masking. To measure the angular power spectrum of masked data, we adapted the P of K EstimatoR (POKER) from cosmic infrared background studies and discuss its use on LIM data.Results. The arPLS method achieves a reduction in the cosmic infrared background fluctuations to a sub-dominant level of the [CII] power at z ∼ 7, a factor of > 70 below our fiducial [CII] model. When using the standard PCA, this factor is only 0.7 at this redshift. The masking lowers the power amplitude of line contamination down to 2 × 10−2 Jy2 sr−1. This residual level is dominated by faint undetected sources that are not clustered around the detected (and masked) sources. For our [CII] model, this results in a detection at z = 5.2 with a power ratio [CII]/(residual interlopers) = 62 ± 32 for a 22% area survey loss. However, at z = 7, [CII]/(residual interlopers) = 2.0 ± 1.4, due to the weak contrast between [CII] and the residual line contamination. Thanks to the large area covered by SIDES-Uchuu, we show that the power amplitude of line residuals varies by 12–15% for z = 5.2 − 7, which is less than the field-to-field variance affecting [CII] power spectra.Conclusions. We present an end-to-end simulation of the extragalactic foreground removal that we ran to detect the [CII] at high redshift via its angular power spectrum. We show that cosmic infrared background fluctuations are not a limiting foreground for [CII] LIM. On the contrary, the CO and [CI] line contamination severely limits our ability to accurately measure the [CII] angular power spectrum at z ≳ 7.

    • Ocean acidification and hypoxia have become increasingly severe in coastal areas, and their co-occurrence poses emerging threats to coastal ecosystems. Here, we investigated the combined effects of ocean acidification and hypoxia on the reproductive capacity of the thick-shelled mussel Mytilus coruscus. Our results demonstrated low pH but not low oxygen induced decreased gonadosomatic index (GSI) in mussels. Male mussels had a lower level of sex steroids (estradiol, testosterone, and progesterone) when kept at low pH. Expression of genes related to reproduction were also impacted by low pH with a downregulation of genes involved in gonad development in males (beta-catenin and Wnt-7b involved in males) and an upregulation of testosterone synthesis inhibition-related gene (Wnt-4) in females. Overall, our results suggest that ocean acidification has an impact on the gonadal development through an alternation of gene expression and level of steroids while hypoxia had no significant effect.
