Adrenocorticotropin and cortisol response to lysine vasopressin in relation to the outcome of the dexamethasone suppression test in major depressive disorder

- The pathophysiology behind the abnormalities of the hypothalamic pituitary adrenal cortex axis found in patients with major depressive disorder was studied by the use of the vasopressin test. The response of plasma adrenocorticotropin (ACTH) and cortisol to the injection of 10 IU lysine-vasopressin (LVP) was investigated in 18 patients meeting the DSM-III criteria for major depressive episode. The response was correlated to the outcome of the dexamethasone suppression test (DST) with the use of two different cut-off points, 139 nmol/l and 200 nmol/l respectively. The results show that no significant difference was found in ACTH or cortisol response between patients having a normal or abnormal DST. The results do not seem to support the hypothesis that the abnormalities of the hypothalamic pituitary adrenal cortex axis involve a hypersecretion of corticotropin-releasing factor (CRF) and a subsequent desensitization of the corticotrophs to CRF-stimulated ACTH release.

- Intramuscular ceruletide or placebo was given in a randomized double-blind crossover design to 12 non-obese and 12 obese individuals, 30 min before a palatable lunch meal. No significant effects were found on the amount of food intake or the hunger ratings in any group. Although rapid CCK or ceruletide infusions have been found in some studies to reduce food intake in man and animals, slow infusions have increased food intake. Under the present study conditions, the moderate rate of release of ceruletide from intramuscular depots did not affect the food intake.

En perspektiv- och problemfokuserad arbetsmodell framläggs.

- Folsomia fimetaria (L.) were added (0, 5, 10, 20 animals) to 0.100 g barley straw which had been inoculated 10 days (244 h) earlier with a natural soil microflora. Respiration (CO2 evolution) was monitored continuously. Mass loss, fungal standing crop (total and FDA-active), bacterial and protozoan biomass were estimated 42 days (1,000 h) after microbial inoculation. The degree of surface cover by hyphae was surveyed at regular intervals. No significant differences (P>0.05) were found in respiration, mass loss or microbial biomass, but the density of surface hyphae were reduced by addition of Collembola. Fungal production was low, less than 5% of the estimated microbial production, and could not account for all collembolan growth during incubation. F. fimetaria appeared to consume mainly bacteria and protozoa, and had little impact on carbon mineralization.

- Data are presented on exclusive ρ0 and φ{symbol} production in deep inelastic muon scattering from a target consisting mainly of nitrogen. The ratio of the total cross sections for ρ0 and φ{symbol} production is found to be 9:(1.6±0.4) at 〈Q2〉=7.5 GeV2, consistent with the SU(3) prediction of 9:2. The t dependence for exclusive ρ0 production is found to become shallover as Q2 increases and, for large Q2, the t dependence is typical of that for a hard scattering process. Furthermore, the ratio of the cross sections for coherent: incoherent production from nitrogen is found to decrease rapidly with Q2. Such behaviour indicates that even for exclusive vector meson production the virtual photon behaves predominantly as an electromagnetic probe.

- Intrinsic hepatic glycogenolysis was examined after hypovolemic stress. Hemorrhagic hypotension of 70 (P70) and 40 mm Hg (P40) for 60 min was inflicted for two postprandial groups and of 70 mm Hg (S70) in a 24-h starved group. The results were compared with three control groups; one postprandial (Pc), one 24-h starved (Sc), and one starved for 9 h (Sc: 9) to mimic the glycogen depletion produced by 70 mm Hg hemorrhagic hypotension. Glucose output was studied in vitro using av recirculating isolated liver perfusion system with a perfusate free of glucose and endocrine stimulation. Liver glycogen determination was made before perfusion start. Although the glycogen stores were decreased after hemorrhage glucose yield was increased (P70) and unchanged (P40) as compared to controls (Pc and Sc: 9). Both starved groups delivered small amounts of glucose, but the released fraction of the S70 group was more than twice that from the Sc group. These data suggest a liver enzyme activation with increased velocity of the enzymesubstrate reactions responsible for glycogen degradation, induced during in vivo hemorrhage and persisting for at least 30 min in vitro perfusion.

Microbial biomass as a fraction of potentially mineralizable nitrogen in soils from long-term field experiments

- Aerobic long-term incubations (40-wk) were employed to measure the potentially mineralizable nitrogen (N0) in five 30-yr old cropping systems. The cropping systems consisted of: (1) bare fallow; (2) cropping with no additions; (3) cropping with 80 kg N ha-1 y-1 as Ca(NO3)2; (4) cropping with 80 kg N ha-1 yr-1 as Ca(NO3)2 plus 1800kg C ha-1 yr-1 as straw; and (5) cropping with 80 kg N ha-1 yr-1 plus 1800 kg C ha-1 yr-1 as farmyard manure. The amounts of N mineralized during the 40-wk incubations were between 93 and 168 μg g-1 (302-543 kg N ha-1 down to 25cm depth) with the lowest value for the fallow and the highest for the farmyard manure treatment. Microbial biomass-C and -N were measured on four occasions during the incubations. The biomass-C showed a rapid decrease to week 4 (to 36% of the initial mass), a slower decrease to week 9 (to 23% of initial mass) and a very slow decline to the final determination at the end of the incubation (to 8% of initial mass). The biomass-N displayed a similar pattern. Two related models were employed to describe the kinetics of N-mineralization during incubation: (1) a two-component first-order; and (2) a simplified special case of the two-component model. In all cases except the straw-amended soil, the simplified two-component model offered the best description of the curves of accumulated mineral-N. The available fraction, Na, of soil organic-N had mineralization rate constants similar to those for mineralization of microbial biomass.

- Isoproterenol (50 nM) potentiated the effects of angiotensin (1-50 nM) on 86Rb efflux and 45Ca efflux from guinea pig hepatocytes. This effect occurred in the presence or absence of extracellular Ca2+ and required the simultaneous presence of both isoproterenol and angiotensin. Neither the divalent cationophore, A23187, nor 4 beta-phorbol dibutyrate could substitute for angiotensin. The effects of isoproterenol were greatest with submaximal concentrations of angiotensin, whereas maximal concentrations of angiotensin were affected little. Isoproterenol did not substantially increase the formation of [3H]inositol triphosphate or the ratio of isomers [3H]inositol 1,4,5-trisphosphate and [3H]inositol 1,3,4-trisphosphate formed in response to angiotensin. Isoproterenol also enhanced the phase of Ca2+ mobilization involving Ca2+ entry which is consistent with the previously proposed functional linkage between receptor-regulated Ca2+ release and Ca2+ entry. These findings suggest that isoproterenol may act by increasing the sensitivity of the endoplasmic reticulum to the Ca2+-releasing action of inositol 1,4,5-trisphosphate.

- The existence and significance of a hormone-sensitive, rapidly mobilizable intracellular pool of Ca2+ in hamster brown-fat cells was investigated with 45Ca2+-labelling techniques. It was shown that such a pool existed and was probably located within the abundant mitochondria. It was rapidly mobilized by norepinephrine (median effective concentration 50 nM) through alpha-adrenergic mechanisms. The mobilization of Ca2+ from the intracellular stores (mitochondria) required the presence of extracellular Na+, but not of Ca2+, K+ or Mg2+. It is concluded that the experiments are in agreement with a hypothesis linking the mobilization of intracellular Ca2+ pools with an alpha-adrenergically-induced increase in plasma membrane Na+ permeability (observed as a membrane depolarization), and a subsequent activation of the mitochondrial Na+/Ca2+ exchange, leading to mobilization of mitochondrial Ca2+ and the mediation of alpha-adrenergic effects as a result of an elevated cytosolic Ca2+ level.
