  • Disease-related variations of the glycosylation of haptoglobin in the dog

    • Haptoglobin phenotypes have been shown in human medicine to be related to the prevalence of various diseases. Furthermore, abnormal glycosylation of haptoglobin has been reported as a consequence of liver disease, cancer and immunological disorders in man. To our knowledge, similar findings have not, so far, been reported in canine disease. The present paper describes a method for investigation of canine haptoglobin phenotypes and of microheterogeneity caused by altered glycosylation. The method consisted of isoelectric focusing (IEF) of dog serum, followed by immunoblotting. The results indicated the existence of only one canine haptoglobin phenotype with a characteristic microheterogeneity pattern in healthy dogs. Changes in this pattern were found in serum from dogs with liver disease, predominantly chronic progressive hepatitis, and with different kinds of anaemia. Pretreatment of serum with neuraminidase or glycopeptidase F (PNGase F) resulted in identical IEF patterns of haptoglobin from healthy and diseased dogs. Moreover, a fucose-specific lectin was capable of binding to some of the abnormal haptoglobin fractions, mainly those found in association with anaemia. The changes described were interpreted as alterations of the carbohydrate content, with or without fucosylation, of some haptoglobin fractions.

  • Process analytical methods for monitoring of the film coating on pharmaceutical pellets and tablets using NIR spectrometry and image analysis

    • Pharmaceutical tablets and pellets are often covered with a coating. There is a need for methods that enable monitoring of the growth of such films during processing. Such analyses can be performed using process analytical chemical methods based on near infrared (NIR) diffuse reflectance spectrometry. It provides with analyses that take only ca. 8 seconds in total analysis time, including physical sampling. The use of chemometrics is the key to handle the complexity of the NIR spectra. The chemometric methods of principal component analysis (PCA) and partial least squares (PLS) were used to study and to calibrate NIR-spectra to the amount of coating. An at-line method using NIR-spectra for determination of the amount of coating of film applied on pharmaceutical tablets was developed. Limitations for quantitation of the film were determined by the 'information depth', ca 0.1 - 0.2 mm. Novel tools to determine the coating thickness and its variations on pharmaceutical pellets were based on image analysis and NIR diffuse reflectance spectrometry. Data from the images were used to predict variations in the release rate of the active substance due to geometrical variations. The image analysis was further used as a reference method for thickness measurements. Together with a theoretical film thickness growth model with physical relevance to underlying process mechanisms, an in-line process analytical chemical method based on NIR was enabled for analysis of the coating thickness on pharmaceutical pellets (RMSEP = 2.2 µm, range tested 0 - 50 µm). Indeed, such tools have not been combined previously, and through use of a multivariate batch calibration, the thickness values delivered from the NIR spectrometer became less disturbed by commonly varied process conditions. Additionally, variations in the coating thickness could be indicated by NIR . The spectral artefacts encountered for samples moving during acquisition of a scan were characterised in a separate study using multivariate methods. These artefacts could be detected by PCA.

  • Structural, thermal and thermodynamic properties of some Ln-Pd oxides and their behaviour as catalyst precursors

    • The structural, thermal and thermodynamic properties of some Ln-Pd oxides of the Ln4Pd07 type are presented. The main results are; (i) Reduction of Ln4Pd07, either ex-situ by CO or in-situ by a synthetic car exhaust gas, results in a materialwith catalytic activity for car exhaust gas clean-up reactions. The obtained material consists of Pd nanoparticles situated on the surface of micron-sized grains of Ln2O3 Upon heat treatment of the reduced material in air, Lu4Pd07 i srapidly re-formed. This property May be utilised for reactivation/conditioning of aged Ln4Pd07-based catalysts; (ii) The Ln4Pd07 compounds with Ln = Nd, Sm, Eu and Gd dare isostructural and have a structure which is a triclinic distortion of the monoclinic La4Pd07 type; (iii) ΔH°, and ΔS ° f values for La4Pd07 and Nd4Pd07 have been calculated by a combination of data obtained from dissolution calorimetry and thermogravimetric analysis studies; ( iv ) Carbon nanotubes, having the so-called fishbone structure, can be prepared by heat treatment o f La4Pd07 in a 20% CO/He gas mixture for an extended period of time. Nanotubes have been extracted from the obtained La203/Pd/nanotube mixture, using a wet-chemical method.

    • Strips of rat soleus muscle were incubated in media containing a superoxide generating system and/or the nitric oxide donor sodium nitroprusside (SNP) before the maximal catalytic activities of aconitase, citrate synthase, and oxoglutarate dehydrogenase were measured. The maximal activities of aconitase and oxoglutarate dehydrogenase were both decreased by 25-30% by superoxide anions; however, only the maximal activity of aconitase was decreased, by approximately 50%, by incubation of muscles with SNP. Furthermore, when both superoxide and NO were present in the medium, aconitase activity was decreased by 70%. The maximal activity of citrate synthase was not affected by any of the treatments. This is the first time that superoxide anions or NO has been shown to inactivate aconitase and oxoglutarate dehydrogenase in skeletal muscle. It is suggested that these effects may be responsible for some alterations in skeletal muscle metabolism, and these possibilities are discussed.

  • Crank Angle Resolved HC Detection Using LIF in the Exhausts of Small Two-Stroke Engines Running at High Engine Speed

    • In order to separate the HC-emissions from two-stroke engines into short-circuit losses and emissions due to incomplete combustion, Laser Induced Fluorescence (LIF) measurements were performed on the exhaust gases just outside the exhaust ports of two engines of different designs. The difference between the two engines was the design of the transfer channels. One engine had "finger" transfer channels and one had "cup handle" transfer channels. Apart from that they were similar. The engine with "finger" transfer channels was earlier known to give more short-circuiting losses than the other engine, and that behavior was confirmed by these measurements. Generally, the results show that the emission of hydrocarbons has two peaks, one just after exhaust port opening and one late in the scavenging phase. The spectral information shows differences between the two peaks and it can be concluded that the latter peak is due to short-circuiting and the earlier due to incomplete combustion. The flow outside the exhaust port was measured with Laser Doppler Velocimetry (LDV). These measurements confirm the occurrence of two emission peaks.

  • Using design-of-experiments techniques for an efficient finite element study of the influence of changed parameters in design

    • All designs are marred by uncertainties and tolerances in dimen- sions, load levels etc. Traditionally, one has often over-dimensioned to take these uncertainties into account. The demand for optimized designs with high quality and reliability increases, which means that more sophisticated methods have been developed, see e.g. Lochner and Matar (1990). By describing the fluctuations in design parame- ters in terms of distributions with expectation and variance, the design can be examined with statistical methods, which results in a more op-timized design. This treatment of the design often demands several experiments, and to plan these experiments Design Of Experiments (DOE) techniques, see e.g. Montgomery (1991), are often used. By using DOE methods the design variables are systematically altered, which minimizes the number of experiments needed. The output of the experiments is the results of a specified response function, giving an indication of the influence of design variable fluctuations. A FEM system is a suitable tool when performing repeated, similar analyses. Examples exist where the DOE process has been performed external- ly and then transferred to the FEM system in the form of parameter sets defining the analysis cases that are to be solved, see e.g. Summers et al. (1996) and Billings (1996). This paper describes a statistical DOE module based on Taguchi’s method that works within ANSYS. The module plans the FEM anal-ysis and calculates the standard statistical moments of the FEM result. This module serves as a powerful tool for the engineering designer or analysts when examining the influence of variance and mean value of different design variables. It also serves as an exploration of where to concentrate an optimization process.

    • Optical properties of human whole blood were measured in vitro, at 633 nm with a double integrating sphere set-up, The blood was kept at constant flow through a flow cell while slowly heating the blood from approximately 25 degrees C to 55 degrees C in a heat exchanger, The results show a small but distinct decrease in the g-factor of 1.7 +/- 0.6\% and a similar increase in the scattering coefficient, mu(s), of 2.9 +/- 0.6\% at approximately 45-46 degrees C. When studying the thermal effect on the blood cells under a white-light transmission microscope, the changes in the scattering properties could be correlated to a sudden change in the shape of the red blood cells, from disc-shaped to spherical, at approximately the same temperature. Furthermore, a continuous manifest increase in the absorption coefficient, mu(a), was seen with temperature rise, on average 83.8 +/- 68.1\% when reaching the temperature 50 degrees C. This might be due to heat-induced haemolysis of the red blood cells, resulting in free light absorbing haemoglobin in the surrounding plasma and thus higher effective light absorption.

    • It is demonstrated that it is possible to investigate details of the electronic structure of an internal atomic monolayer using soft-x-ray-emission spectroscopy. The local and partial density of states of one monolayer and three monolayers of Si, embedded deep below a GaAs(001) surface, was extracted. Clear differences to the density of states for bulk Si were observed.
