  • Spectral and atmospheric characterization of 51 Eridani b using VLT/SPHERE
  • 2017
  • Ingår i: Astronomy and Astrophysics. - : EDP Sciences. - 0004-6361 .- 1432-0746. ; 603
  • Tidskriftsartikel (refereegranskat)abstract
    • Context. 51 Eridani b is an exoplanet around a young (20 Myr) nearby (29.4 pc) F0-type star, which was recently discovered by direct imaging. It is one of the closest direct imaging planets in angular and physical separation (similar to 0.5 '', similar to 13 au) and is well suited for spectroscopic analysis using integral field spectrographs. Aims. We aim to refine the atmospheric properties of the known giant planet and to constrain the architecture of the system further by searching for additional companions. Methods. We used the extreme adaptive optics instrument SPHERE at the Very Large Telescope (VLT) to obtain simultaneous dual-band imaging with IRDIS and integral field spectra with IFS, extending the spectral coverage of the planet to the complete Y-to H-band range and providing additional photometry in the K12-bands (2.11, 2.25 mu m). The object is compared to other known cool and peculiar dwarfs. The posterior probability distributions for parameters of cloudy and clear atmospheric models are explored using MCMC. We verified our methods by determining atmospheric parameters for the two benchmark brown dwarfs Gl 570D and HD 3651B. We used archival VLT-NACO (L') Sparse Aperture Masking data to probe the innermost region for additional companions. Results. We present the first spectrophotometric measurements in the Y and K bands for the planet and revise its J-band flux to values 40% fainter than previous measurements. Cloudy models with uniform cloud coverage provide a good match to the data. We derive the temperature, radius, surface gravity, metallicity, and cloud sedimentation parameter f(sed). We find that the atmosphere is highly super-solar ([Fe/H] = 1.0 +/- 0.1 dex), and the low f(sed) = 1.26(-0.29)(+0.36) value is indicative of a vertically extended, optically thick cloud cover with small sized particles. The model radius and surface gravity estimates suggest higher planetary masses of M-gravity = 9.1(-3.3)(+4.9) M-J. The evolutionary model only provides a lower mass limit of > 2 M-J (for pure hot-start). The cold-start model cannot explain the luminosity of the planet. The SPHERE and NACO/SAM detection limits probe the 51 Eri system at solar system scales and exclude brown-dwarf companions more massive than 20 M-J beyond separations of similar to 2.5 au and giant planets more massive than 2 M-J beyond 9 au.

  • Global Opportunities to Increase Agricultural Independence Through Phosphorus Recycling
  • 2019
  • Ingår i: Earth's Future. - : AMER GEOPHYSICAL UNION. - 2328-4277. ; 7:4, s. 370-383
  • Tidskriftsartikel (refereegranskat)abstract
    • Food production hinges largely upon access to phosphorus (P) fertilizer. Most fertilizer P used in the global agricultural system comes from mining of nonrenewable phosphate rock deposits located within few countries. However, P contained in livestock manure or urban wastes represents a recyclable source of P. To inform development of P recycling technologies and policies, we examined subnational, national, and global spatial patterns for two intersections of land use affording high P recycling potential: (a) manure-rich cultivated areas and (b) populous cultivated areas. In turn, we examined overlap between P recycling potential and nation-level P fertilizer import dependency. Populous cultivated areas were less abundant globally than manure-rich cultivated areas, reflecting greater segregation between crops and people compared to crops and livestock, especially in the Americas. Based on a global hexagonal grid (290-km(2) grid cell area), disproportionately large shares of subnational hot spots for P recycling potential occurred in India, China, Southeast Asia, Europe, and parts of Africa. Outside of China, most of the remaining manure-rich or pulous cultivated areas occurred within nations that had relatively high imports of P fertilizer (net P import:consumption ratios >= 0.4) or substantial increases in fertilizer demand between the 2000s (2002-2006) and 2010s (2010-2014). Manure-rich cultivated grid cells (those above the 75th percentiles for both manure and cropland extent) represented 12% of the global grid after excluding cropless cells. Annually, the global sum of animal manure P was at least 5 times that contained in human excreta, and among cultivated cells the ratio was frequently higher (median = 8.9). The abundance of potential P recycling hot spots within nations that have depended on fertilizer imports or experienced rising fertilizer demand could prove useful for developing local P sources and maintaining agricultural independence.

  • High occurrence of transportation and logistics occupations among vascular dementia patients : An observational study
  • 2019
  • Ingår i: Alzheimer's Research and Therapy. - : Springer Science and Business Media LLC. - 1758-9193. ; 11:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Background: Growing evidence suggests a role of occupation in the emergence and manifestation of dementia. Occupations are often defined by complexity level, although working environments and activities differ in several other important ways. We aimed to capture the multi-faceted nature of occupation through its measurement as a qualitative (instead of a quantitative) variable and explored its relationship with different types of dementia. Methods: We collected occupational information of 2121 dementia patients with various suspected etiologies from the Amsterdam Dementia Cohort (age 67 ± 8, 57% male; MMSE 21 ± 5). Our final sample included individuals with Alzheimer's disease (AD) dementia (n = 1467), frontotemporal dementia (n = 281), vascular dementia (n = 98), Lewy body disease (n = 174), and progressive supranuclear palsy/corticobasal degeneration (n = 101). Within the AD group, we used neuropsychological data to further characterize patients by clinical phenotypes. All participants were categorized into 1 of 11 occupational classes, across which we evaluated the distribution of dementia (sub)types with χ 2 analyses. We gained further insight into occupation-dementia relationships through post hoc logistic regressions that included various demographic and health characteristics as explanatory variables. Results: There were significant differences in the distribution of dementia types across occupation groups (χ 2 = 85.87, p <.001). Vascular dementia was relatively common in the Transportation/Logistics sector, and higher vascular risk factors partly explained this relationship. AD occurred less in Transportation/Logistics and more in Health Care/Welfare occupations, which related to a higher/lower percentage of males. We found no relationships between occupational classes and clinical phenotypes of AD (χ 2 = 53.65, n.s.). Conclusions: Relationships between occupation and dementia seem to exist beyond the complexity level, which offers new opportunities for disease prevention and improvement of occupational health policy.

  • Comment : The FAIR Guiding Principles for scientific data management and stewardship
  • 2016
  • Ingår i: Scientific Data. - : Springer Science and Business Media LLC. - 2052-4463. ; 3
  • Tidskriftsartikel (refereegranskat)abstract
    • There is an urgent need to improve the infrastructure supporting the reuse of scholarly data. A diverse set of stakeholders-representing academia, industry, funding agencies, and scholarly publishers-have come together to design and jointly endorse a concise and measureable set of principles that we refer to as the FAIR Data Principles. The intent is that these may act as a guideline for those wishing to enhance the reusability of their data holdings. Distinct from peer initiatives that focus on the human scholar, the FAIR Principles put specific emphasis on enhancing the ability of machines to automatically find and use the data, in addition to supporting its reuse by individuals. This Comment is the first formal publication of the FAIR Principles, and includes the rationale behind them, and some exemplar implementations in the community.

  • Combining traditional dietary assessment methods with novel metabolomics techniques: Present efforts by the Food Biomarker Alliance
  • 2017
  • Ingår i: Proceedings of the Nutrition Society. - 0029-6651 .- 1475-2719. ; 76:4, s. 619-627
  • Tidskriftsartikel (refereegranskat)abstract
    • FFQ, food diaries and 24 h recall methods represent the most commonly used dietary assessment tools in human studies on nutrition and health, but food intake biomarkers are assumed to provide a more objective reflection of intake. Unfortunately, very few of these biomarkers are sufficiently validated. This review provides an overview of food intake biomarker research and highlights present research efforts of the Joint Programming Initiative 'A Healthy Diet for a Healthy Life' (JPI-HDHL) Food Biomarkers Alliance (FoodBAll). In order to identify novel food intake biomarkers, the focus is on new food metabolomics techniques that allow the quantification of up to thousands of metabolites simultaneously, which may be applied in intervention and observational studies. As biomarkers are often influenced by various other factors than the food under investigation, FoodBAll developed a food intake biomarker quality and validity score aiming to assist the systematic evaluation of novel biomarkers. Moreover, to evaluate the applicability of nutritional biomarkers, studies are presently also focusing on associations between food intake biomarkers and diet-related disease risk. In order to be successful in these metabolomics studies, knowledge about available electronic metabolomics resources is necessary and further developments of these resources are essential. Ultimately, present efforts in this research area aim to advance quality control of traditional dietary assessment methods, advance compliance evaluation in nutritional intervention studies, and increase the significance of observational studies by investigating associations between nutrition and health.

  • Assessment of the appropriate use criteria for amyloid PET in an unselected memory clinic cohort : The ABIDE project
  • 2019
  • Ingår i: Alzheimer's and Dementia. - : Wiley. - 1552-5260.
  • Tidskriftsartikel (refereegranskat)abstract
    • Introduction: The objective of this study was to assess the usefulness of the appropriate use criteria (AUC) for amyloid imaging in an unselected cohort. Methods: We calculated sensitivity and specificity of appropriate use (increased confidence and management change), as defined by Amyloid Imaging Taskforce in the AUC, and other clinical utility outcomes. Furthermore, we compared differences in post–positron emission tomography diagnosis and management change between “AUC-consistent” and “AUC-inconsistent” patients. Results: Almost half (250/507) of patients were AUC-consistent. In both AUC-consistent and AUC-inconsistent patients, post–positron emission tomography diagnosis (28%–21%) and management (32%–17%) change was substantial. The Amyloid Imaging Taskforce's definition of appropriate use occurred in 55/507 (13%) patients, detected by the AUC with a sensitivity of 93%, and a specificity of 56%. Diagnostic changes occurred independently of AUC status (sensitivity: 57%, specificity: 53%). Discussion: The current AUC are not sufficiently able to discriminate between patients who will benefit from amyloid positron emission tomography and those who will not.

  • Modeling phosphorus in rivers at the global scale: recent successes, remaining challenges, and near-term opportunities
  • 2019
  • Ingår i: Current Opinion in Environmental Sustainability. - : ELSEVIER SCI LTD. - 1877-3435 .- 1877-3443. ; 36, s. 68-77
  • Forskningsöversikt (refereegranskat)abstract
    • Understanding and mitigating the effects of phosphorus (P) overenrichment of waters globally, including the evaluation of the global Sustainability Development Goals, requires the use of global models. Such models quantitatively link land use, global population growth and climate to aquatic nutrient loading and biogeochemical cycling. Here we describe, compare, and contrast the existing global models capable of predicting P transport by rivers at a global scale. We highlight important insights gained from the development and application of these models, and identify important near-term opportunities for model improvements as well as additional insight to be gained through new model analysis.

  • 2015
  • Ingår i: Publications of the Astronomical Society of the Pacific. - : IOP Publishing. - 0004-6280 .- 1538-3873. ; 127:953, s. 584-594
  • Tidskriftsartikel (refereegranskat)abstract
    • MIRI (the Mid-Infrared Instrument for the James Webb Space Telescope [JWST]) operates from 5 to 28: 5 mu m and combines over this range: (1) unprecedented sensitivity levels; (2) subarcsecond angular resolution; (3) freedom from atmospheric interference; (4) the inherent stability of observing in space; and (5) a suite of versatile capabilities including imaging, low- and medium-resolution spectroscopy (with an integral field unit), and coronagraphy. We illustrate the potential uses of this unique combination of capabilities with various science examples: (1) imaging exoplanets; (2) transit and eclipse spectroscopy of exoplanets; (3) probing the first stages of star and planet formation, including identifying bioactive molecules; (4) determining star formation rates and mass growth as galaxies are assembled; and (5) characterizing the youngest massive galaxies.

  • The extended law of corresponding states when attractions meet repulsions
  • 2018
  • Ingår i: Soft Matter. - : Royal Society of Chemistry (RSC). - 1744-683X .- 1744-6848. ; 14:19, s. 3704-3715
  • Tidskriftsartikel (refereegranskat)abstract
    • Short-range attractive colloids show well-defined phase behaviour in the absence of repulsions, and highly intriguing equilibrium gelation in the presence of long-range repulsions. We present the state diagram of short-range attractive colloids with repulsions that range from fully screened to intermediately ranged, i.e. longer-ranged than the attractions, but shorter ranged than the colloid size. We demonstrate that although the macroscopic phase behaviour does not change perceptibly, there is a dramatic increase of inhomogeneities once the repulsions become longer-ranged than the attractions. The interaction potentials are characterized with small angle neutron scattering, and used to renormalize the state diagram with the minimum in the interaction potential, min[U(r)], and with the reduced second virial coefficient, B2∗. We find that the extended law of corresponding states captures the onset of phase separation for shorter ranged repulsions, but fails for longer ranged repulsions. Instead, for a given model of U(r), the transition from visually homogeneous fluid to phase separation and/or gelation can be rescaled with min[U(r)] over the full range of repulsions. Finally, we suggest a generic state diagram to describe the effect of repulsions on short-range attractive systems.

  • The BRCA1-Δ11q alternative splice isoform bypasses germline mutations and promotes therapeutic resistance to PARP inhibition and cisplatin
  • 2016
  • Ingår i: Cancer Research. - 0008-5472. ; 76:9, s. 2778-2790
  • Tidskriftsartikel (refereegranskat)abstract
    • Breast and ovarian cancer patients harboring BRCA1/2 germline mutations have clinically benefitted from therapy with PARP inhibitor (PARPi) or platinum compounds, but acquired resistance limits clinical impact. In this study, we investigated the impact of mutations on BRCA1 isoform expression and therapeutic response. Cancer cell lines and tumors harboring mutations in exon 11 of BRCA1 express a BRCA1-Δ11q splice variant lacking the majority of exon 11. The introduction of frameshift mutations to exon 11 resulted in nonsense-mediated mRNA decay of full-length, but not the BRCA1-Δ11q isoform. CRISPR/Cas9 gene editing as well as overexpression experiments revealed that the BRCA1-Δ11q protein was capable of promoting partial PARPi and cisplatin resistance relative to full-length BRCA1, both in vitro and in vivo. Furthermore, spliceosome inhibitors reduced BRCA1- Δ11q levels and sensitized cells carrying exon 11 mutations to PARPi treatment. Taken together, our results provided evidence that cancer cells employ a strategy to remove deleterious germline BRCA1 mutations through alternative mRNA splicing, giving rise to isoforms that retain residual activity and contribute to therapeutic resistance.
