    • We describe the main ideas and some experimental outcomes of two experiments, both aimed to study quantum tunneling effects in high-T C structures. In the first experiment, macroscopic quantum tunneling is demonstrated in grain boundary YBaCuO Josephson junctions revealing dissipation levels lower than expected and opening novel perspectives for quantum circuitry. In the second, the low temperature dissipation is dominated by quantum tunneling of individual Pearl vortices in ultra-thin CaBaCuO systems characterized by extremely long Pearl lengths. © 2006 Elsevier B.V.     • OBJECTIVE: Systemic lupus erythematosus (SLE) is the prototypical systemic autoimmune disorder, with complex etiology and a strong genetic component. Recently, gene products involved in the interferon pathway have been under intense investigation in terms of the pathogenesis of SLE. STAT-1 and STAT-4 are transcription factors that play key roles in the interferon and Th1 signaling pathways, making them attractive candidates for involvement in SLE susceptibility. METHODS: Fifty-six single-nucleotide polymorphisms (SNPs) across STAT1 and STAT4 on chromosome 2 were genotyped using the Illumina platform, as part of an extensive association study in a large collection of 9,923 lupus patients and control subjects from different racial groups. DNA samples were obtained from the peripheral blood of patients with SLE and control subjects. Principal components analyses and population-based case-control association analyses were performed, and the P values, false discovery rate q values, and odds ratios with 95% confidence intervals were calculated. RESULTS: We observed strong genetic associations with SLE and multiple SNPs located within STAT4 in different ethnic groups (Fisher's combined P = 7.02 x 10(-25)). In addition to strongly confirming the previously reported association in the third intronic region of this gene, we identified additional haplotypic association across STAT4 and, in particular, a common risk haplotype that is found in multiple racial groups. In contrast, only a relatively weak suggestive association was observed with STAT1, probably due to its proximity to STAT4. CONCLUSION: Our findings indicate that STAT4 is likely to be a crucial component in SLE pathogenesis in multiple racial groups. Knowledge of the functional effects of this association, when they are revealed, might improve our understanding of the disease and provide new therapeutic targets.

  • Eck-Like Resonances in High-T-c Long Faceted Josephson Junctions

    • By means of numerical simulations we study the dynamics of the phase difference in faceted long Josephson junctions. We compare our simulations with experiments on a sample of high quality high-Tc biepitaxial junctions. Adjusting junction length and dissipation to reproduce the observed data we found a significant agreement, which permits to identify the observed resonances mostly as Eck steps. Some considerations are also made on the requirements to observe flux-flow or Fiske resonances in these junctions.

    • We report on the fabrication and properties of sub-micron YBa2Cu3O7-x off-axis biepitaxial junctions. The vision behind this work is to set up a reliable nanotechnology processing to improve junctions' performances and reproducibility through the reduction of their size. We will illustrate the fabrication procedure employed and show the results obtained on samples where the transport parameters were tuned in a predictable way. Also, proofs of a clean barrier structure with few facets and low disorder were obtained.

    • Research in autophagy continues to accelerate,1 and as a result many new scientists are entering the field. Accordingly, it is important to establish a standard set of criteria for monitoring macroautophagy in different organisms. Recent reviews have described the range of assays that have been used for this purpose.2,3 There are many useful and convenient methods that can be used to monitor macroautophagy in yeast, but relatively few in other model systems, and there is much confusion regarding acceptable methods to measure macroautophagy in higher eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers of autophagosomes versus those that measure flux through the autophagy pathway; thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from fully functional autophagy that includes delivery to, and degradation within, lysosomes (in most higher eukaryotes) or the vacuole (in plants and fungi). Here, we present a set of guidelines for the selection and interpretation of the methods that can be used by investigators who are attempting to examine macroautophagy and related processes, as well as by reviewers who need to provide realistic and reasonable critiques of papers that investigate these processes. This set of guidelines is not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to verify an autophagic response.

  • P Cygni Profiles of Molecular Lines Toward Arp 220 Nuclei

    • We report ~100 pc (0farcs3) resolution observations of (sub)millimeter HCO+ and CO lines in the ultraluminous infrared galaxy Arp 220. The lines peak at two merger nuclei, with HCO+ being more spatially concentrated than CO. Asymmetric line profiles with blueshifted absorption and redshifted emission are discovered in HCO+(3-2) and (4-3) toward the two nuclei and in CO(3-2) toward one nucleus. We suggest that these P Cygni profiles are due to ~100 km s–1 outward motion of molecular gas from the nuclei. This gas is most likely outflowing from the inner regions of the two nuclear disks rotating around individual nuclei, clearing the shroud around the luminosity sources there.
