    • Being related to law and state-of-the-art technology, digital forensics needs more discipline than traditional forensics. The variety of types of crimes, distribution of networks and complexity of information and communication technology, add to the complexity of the process or digital investigations. A rigorous and flexible process model is needed to overcome challenges and obstacles in this area. In this paper we propose a digital forensics process, called "Two-Dimensional Evidence Reliability Amplification Process Model", which presents a detailed digital forensic process model in five main phases and different roles to perform it. At the same time, this iterative process addresses four essential tasks as the umbrella activities that are applicable across all phases and subphases. We have also developed a hypothetical solution based on intersection of events and exploit mathematical operations and symbols for making an algorithm to increase the reliability of evidence. This process model is detailed enough to describe the investigation process so that it could possibly provide a guideline that investigators can take advantage of it during a forensics investigation process.

  • Cosmology intertwined : A review of the particle physics, astrophysics, and cosmology associated with the cosmological tensions and anomalies

    • The standard Λ Cold Dark Matter (ΛCDM) cosmological model provides a good description of a wide range of astrophysical and cosmological data. However, there are a few big open questions that make the standard model look like an approximation to a more realistic scenario yet to be found. In this paper, we list a few important goals that need to be addressed in the next decade, taking into account the current discordances between the different cosmological probes, such as the disagreement in the value of the Hubble constant H0, the σ8–S8 tension, and other less statistically significant anomalies. While these discordances can still be in part the result of systematic errors, their persistence after several years of accurate analysis strongly hints at cracks in the standard cosmological scenario and the necessity for new physics or generalisations beyond the standard model. In this paper, we focus on the 5.0σ tension between the Planck CMB estimate of the Hubble constant H0 and the SH0ES collaboration measurements. After showing the H0 evaluations made from different teams using different methods and geometric calibrations, we list a few interesting new physics models that could alleviate this tension and discuss how the next decade's experiments will be crucial. Moreover, we focus on the tension of the Planck CMB data with weak lensing measurements and redshift surveys, about the value of the matter energy density Ωm, and the amplitude or rate of the growth of structure (σ8,fσ8). We list a few interesting models proposed for alleviating this tension, and we discuss the importance of trying to fit a full array of data with a single model and not just one parameter at a time. Additionally, we present a wide range of other less discussed anomalies at a statistical significance level lower than the H0–S8 tensions which may also constitute hints towards new physics, and we discuss possible generic theoretical approaches that can collectively explain the non-standard nature of these signals. Finally, we give an overview of upgraded experiments and next-generation space missions and facilities on Earth that will be of crucial importance to address all these open questions.

  • Analysis of 153 115 patients with hematological malignancies refines the spectrum of familial risk

    • Estimating familial cancer risks is clinically important in being able to discriminate between individuals in the population at differing risk for malignancy. To gain insight into the familial risk for the different hematological malignancies and their possible inter-relationship, we analyzed data on more than 16 million individuals from the Swedish Family-Cancer Database. After identifying 153 115 patients diagnosed with a primary hematological malignancy, we quantified familial relative risks (FRRs) by calculating standardized incident ratios (SIRs) in 391 131 of their first-degree relatives. The majority of hematological malignancies showed increased FRRs for the same tumor type, with the highest FRRs being observed for mixed cellularity Hodgkin lymphoma (SIR, 16.7), lymphoplasmacytic lymphoma (SIR, 15.8), and mantle cell lymphoma (SIR, 13.3). There was evidence for pleiotropic relationships; notably, chronic lymphocytic leukemia was associated with an elevated familial risk for other B-cell tumors and myeloproliferative neoplasms. Collectively, these data provide evidence for shared etiological factors for many hematological malignancies and provide information for identifying individuals at increased risk, as well as informing future gene discovery initiatives.

  • Evolvability and evolutionary rescue

    • The survival prospects of threatened species or populations can sometimes be improved by adaptive change. Such evolutionary rescue is particularly relevant when the threat comes from changing environments, or when long-term population persistence requires range expansion into new habitats. Conservation biologists are therefore often interested in whether or not populations or lineages show a disposition for adaptive evolution, that is, if they are evolvable. Here, we discuss four alternative perspectives that target different causes of evolvability and outline some of the key challenges those perspectives are designed to address. Standing genetic variation provides one familiar estimate of evolvability. Yet, the mere presence of genetic variation is often insufficient to predict if a population will adapt, or how it will adapt. The reason is that adaptive change not only depends on genetic variation, but also on the extent to which this genetic variation can be realized as adaptive phenotypic variation. This requires attention to developmental systems and how plasticity influences evolutionary potential. Finally, we discuss how a better understanding of the different factors that contribute to evolvability can be exploited in conservation practice.

    • The maritime domain is under pressure from changing economic, political and environmental factors. Technological advancements facilitate increased monitoring and control from land. By viewing the maritime domain as a complex socio-technical system, the importance of understanding the role of the on board and shore-side operator in maintaining safety and efficiency of navigation becomes apparent, particularly when introducing new technology. This paper looks at the success factors for navigational assistance, as currently performed by maritime pilots and Vessel Traffic Service (VTS) operators, aiming to identify issues worth consideration in future navigational assistance services. One focus group and one combined workshop/focus group were held with three pilots and two VTS operators respectively. The first looked at the prerequisites for successful navigational assistance from the perspective of the pilot. Using a grounded theory-style approach, a proposition was created that the main indicator of success is “no incidents”, that success depends on the integration of local knowledge, preparation and foresight into the ship-shore system and that good communication is vital to achieving this. Testing this, the second study considered the role of communication in enabling the VTS operator to support the pilot; it confirmed the results of the first study, emphasising the importance of communication when working both with on board and shore-based pilots.

  • The elemental compositions of lower marine organisms

    • A study of 33 new and 87 previous analyses of marine biological matter shows that the proportions of the minor and trace elements (Ti, Fe, Al, Mn, Cr, Cu, Ni, Cd, B, V, Zr and Mo) in lower marine organisms scatter much less than the major elements Na, K, Sr and SiO2 do. Particularly small variations are found for Ti, Fe, Al, Mn, Cr, Cu, Cd and V. Scatter ranges between these extremes are found for Ca, Mg, Ba, Pb and Zn. Biological matter from the Pacific and the Atlantic Oceans show almost identical trace- and minor-element compositions with significant discrepancies only for Al and Zn. Pacific samples are rich in Zn and Atlantic samples are enriched in Al.

    • A peptide derived from the N-terminus of the unprocessed bovine prion protein (bPrPp), incorporating the hydrophobic signal sequence (residues 1–24) and a basic domain (KKRPKP, residues 25–30), internalizes into mammalian cells, even when coupled to a sizeable cargo, and therefore functions as a cell-penetrating peptide (CPP). Confocal microscopy and co-localization studies indicate that the internalization of bPrPp is mainly through macropinocytosis, a fluid-phase endocytosis process, initiated by binding to cell-surface proteoglycans. Electron microscopy studies show internalized bPrPp–DNA–gold complexes residing in endosomal vesicles. bPrPp induces expression of a complexed luciferase-encoding DNA plasmid, demonstrating the peptide’s ability to transport the cargo across the endosomal membrane and into the cytosol and nucleus. The novel CPP activity of the unprocessed N-terminal domain of PrP could be important for the retrotranslocation of partly processed PrP and for PrP trafficking inside or between cells, with implications for the infectivity associated with prion diseases.

  • Collateral development in limb ischemia : aspects of endogenous and stimulated arteriogenesis

    • Peripheral arterial disease (PAD) is common among elderly and affects about 20% of the population above the age of 60 years. PAD is a strong marker for future cardiovascular events and increased mortality due to widespread atherosclerotic lesions in the coronary and cerebral arteries. Intermittent claudication (IC) with lower extremity pain on walking has a prevalence of 3-6 % in men at the age of 60-70 years according to large population studies. Among these patients the majority remain stable without progression of symptoms. The symptoms progress in about 25% of the patients and distal ulceration or gangrene and ischemic rest pain, i.e critical limb ischemia (CLI) develops. Despite liberal indications for surgical or endovascular revascularizations, major amputation is required in as much as one third of the patients with CLI and for these patients there is a great need for alternative treatment strategies. Collateral development, bridging stenoses or occlusions, is one of the reparative processes that reduce the consequences of coronary artery disease and PAD. The growth of these collaterals from pre-existing arteriole is called arteriogenesis. Angiogenesis, the sprouting of new capillaries, may also be of importance in the pathophysiology of PAD. There is little data establishing any impact of angiogenesis on PAD but it is likely that an angiogenic process occurs in ischemic tissues as a response to acute ischemia. Accordingly, hemodynamic forces, cytokines and inflammatory processes appear to be of importance in arteriogenesis, but the exact role of these processes is not firmly established. The arteriogenic mechanisms studied in animal models of limb ischemia and in vitro models aiming to stimulate arteriogenesis by supplying vascular growth factors have shown promising results. The aim of this thesis was to assess the role of vascular growth factors in the arteriogenic process in PAD patients, and furthermore to evaluate gene therapy of FGF-4 in an animal model and humans as a method for therapeutic arteriogenesis. First we explored the expression of growth factors in skin and muscle samples from 25 patients with CLI. There were increased levels of FGF-2 but not VEGF in distal muscle compared to proximal. Furthermore the expression of growth factors and inflammatory cytokines was investigated in 20 patients with IC after walking provocation. The proinflammatory cytokine IL-6 was significantly elevated one hour after exercise. VEGF mRNA also increased significantly in ischemic muscle samples after exercise. Taken together these results indicate that VEGF expression rises locally in response to maximal exercise to the absolute limit of pain in patients with IC but not in patients with CLI at rest. FGF-2 seems to be upregulated in distal muscle in CLI patients which could be due to local inflammatory processes. Gene therapy with adenovirus mediated FGF-4 was assessed in a rat model of severe limb ischemia and in a phase I/II multicenter clinical trial in CLI patients. No signs of stimulated arteriogenesis were detected in the animal model 4 weeks after gene transfer. Gene therapy in 13 patients seemed safe, but no sign of stimulated arteriogenesis was able to be detected in this small patient cohort.

  • On the relationship between blend state and dispersibility of adhesive mixtures containing active pharmaceutical ingredients

    • The objectives of this investigation were to study the evolution in blend state of adhesive mixtures containing the active pharmaceutical ingredients (APIs) salbutamol, budesonide and AZD5423 and to study the relationship between blend state and dispersibility of the mixtures, as assessed by the fine particle fraction (FPF). A series of adhesive mixtures of varied fines concentration were prepared for each API using the same type of carrier. Based on visual examination and powder mechanics, blend states were identified and summarized as blend state maps for each API. The dispersibility of the mixtures was studied using a Fast Screening Impactor (FSI) equipped with a ScreenHaler. The evolution in blend state differed between the APIs in terms of the width of the blend states. The structure of the adhesion layer also differed between the APIs, from relatively uniform to a heterogeneous layer with small agglomerates dispersed on the carrier surface. All three APIs expressed a similar type of bended relationship between FPF and fines concentration. However, the initial rate of increase and the fines concentration of the plateau differed between the APIs. The adhesive mixtures of all APIs followed the three main states in terms of structural evolution and the overall shape of the FPF-fines concentration profiles could be explained by the evolution in blend state. It is proposed that the structure of the adhesion layer is an important factor explaining the differences in blend state - blend dispersibility relationships between the APIs.

  • New C-C coupling Reactions Enabled by Main-group Organometallics

    • The carbon-carbon bond has always been at the very core of chemical research. Strategies for the creation of C−C bonds are one of the keys to the construction game that organic chemists play with the building blocks provided by Nature, with the ultimate goal of producing useful molecular structures that will serve society as medicines, materials, imaging tools, catalysts, and ligands (to mention but a few). While very different in their structure, all of these molecules are often prepared by the same methods. However, efficiency could be improved with tailored chemical strategies that would serve an individual purpose. Ideally, these chemical manipulations should be efficient, selective, environmentally friendly and economic, in order to truly fulfill their final objective.However, despite the ever-expanding rule-book of chemical reactions, target molecules of increasing complexity often face chemists with daunting challenges, whose success rely on multi-step synthetic sequences. There is therefore a permanent need for new, specific methods and strategies that are capable of seamlessly creating C−C bonds, evading the synthesis of difficult or expensive substrates. In this regard, common organometallic reagents display a unique behavior as carbon precursors, in particular as powerful nucleophiles. Reagents based on main-group elements such as lithium or magnesium have therefore played a central role in organic synthesis ever since their discovery. The challenge often lies in controlling their high reactivity, as well as their basic character. Tuning and taming these properties provides chemists with a wide range of unique strategies for the selective synthesis of countless molecular targets.In the first part of this thesis, a scalable and stereoselective [3+3] homocoupling of imines in which two C−C bonds are formed in a single step is reported. This reaction relies on an unusual combination of visible-light irradiation and aluminum organometallics. This photochemical process enables the circumvention of the native [3+2] reactivity of these readily available starting materials, thus enabling rapid access to densely functionalized piperazines. Thanks to the congested environment they provide, these heterocyclic scaffolds can be used as ligands to prevent catalyst deactivation through oligomerization.The next chapter presents a novel Pummerer-type redox-neutral coupling of sulfoxides and Grignard reagents. This reaction is enabled by a unique turbo-magnesium amide base, and allows the use of a wide range of carbon nucleophiles in intermolecular Pummerer C−C coupling for the streamlined preparation of thioethers. Given the central character of sulfur in organic chemistry, these compounds can then be converted to a variety of unrelated functional groups for the streamlined preparation of diverse building blocks.In the final two chapters, the development of a method for the direct conversion of carboxylic acids to ketones with Grignard reagents is described. Using the above-mentioned combination of organometallics, a wide variety of carboxylic acids substrates and Grignard reagents can be coupled in a convenient, scalable and highly selective method that suppresses the need for activation and offers a straightforward approach to ketones from readily available starting materials.

  • Phenotypic responses of zooplankton to variable conditions

    • Organisms are continually challenged by multiple threats in the environment, and such threats are seldom constant in either time or space. Therefore, organisms must maintain physiological, behavioural, morphological and life- history adaptations across environments to prevent reductions in fitness. Freshwater ecosystems are particularly variable environments, and so organisms inhabiting lakes and ponds exhibit a range of different adaptations in order to survive and propagate. Despite this, the energetic constraints and potentially divergent responses required towards multiple threats creates the necessity to make trade-offs in their phenotypic repertoire. The outcomes of such phenotypic compromises are often difficult to predict and requires both a mechanistic understanding of the threats and responses involved, as well as insights into the resulting fitness consequences. Although this is relatively well understood with regards to some threats like predation for example, other biotic and abiotic stressors are less well studied. In this thesis I identify phenotypic compromises in behavioural, morphological and life-history traits of zooplankton with regards to variable environments.Behaviour is often considered one of the most labile traits and consequently it may provide a quick and inexpensive response to infrequent or rapidly changing contexts. Using 3D tracking, I focused on swimming behaviour of Daphnia magna in response to social context and found that females avoid males in a way that resembles a predation event. In a separate experiment I investigated how the presence of a high-density food patch altered the response to the abiotic threat ultraviolet radiation (UVR). Here I found that despite the potential fitness consequences of UVR the foraging opportunity reduced the avoidance response that has been classically described in Daphnia.UVR is a substantial threat in aquatic systems with documented physiological, behavioural, morphological and life- history responses. It is also a particularly variable stressor as it is absent during night and varies in intensity over the seasons. Despite its variable nature, studies have typically only addressed the presence or absence and not fluctuating UVR stress. I addressed this gap in the knowledge by tracking the survival, reproduction and behavioural response to both fluctuating and stable exposure of UVR. Simply by varying the scheduling but not the quantity of UVR stress, I identified fitness costs that appeared to be linked to the cost of the behavioural avoidance.In high-latitude environments, where phenotypic plasticity is promoted due to seasonal variability, copepods increase pigmentation in response to increased UVR but only in the absence of fish. I tested whether this response is ubiquitous at lower latitudes that experience less seasonality and have evolved with different predation regimes. Copepods from fishless environments had higher pigmentation than those with visually hunting predators. I also found that plasticity towards UVR removal was minor, but plasticity towards predation was mostly idiosyncratic. This suggests that plasticity does exist for the threats that are most variable and constitutive responses may have evolved towards ever present danger.

  • A Raman scattering study of the interactions of DNA with its water of hydration

    • Raman spectroscopy is used to probe the nature of the hydrogen bonds which hold the water of hydration to DNA. The approximate to 3450cm(-1) molecular O-H stretching mode shows that the first six water molecules per base pair of the primary hydration shell are very strongly bound to the DNA. The observed shift in the peak position of this mode permits a determination of the length of the hydrogen bonds for these water molecules. These hydrogen bonds appear to be about 0.3 angstrom shorter than the hydrogen bonds in bulk water. The linewidth of this mode shows no significant changes above water contents of about 15 water molecules per base pair. This technique of using a vibrational spectroscopy to obtain structural information about the hydration shells of DNA could be used to study the hydration shells of other biomolecules.

  • Student and teacher co-agency when combining CT with arts and design in a cross-curricular project

    • The technological development has raised awareness for the importance of digital competence and computational thinking (CT) to understand the digital world and has resulted in revised curricula in many countries. In Finland, a new curriculum for grades 1–9 came into force in 2016 introducing digital competence (including programming) to be integrated in other subjects. Most teachers lack prior experience in programming and there is a need for suitable instructional models. This article presents a cross-curricular teaching sequence and the results from a case study conducted in four Finnish schools. Students in grades 4–6 collaboratively worked on a project combining arts, design and CT with other subjects. The results show that students demonstrated several CT abilities while working on their projects, in particular creativity, tinkering and debugging. The findings also indicate that teachers and students learned together (co-agency) and suggest that models like the teaching sequence can help and encourage teachers to integrate programming and CT in a cross-curricular manner. Still, the teachers’ knowledge, ambition level and understanding of the task at hand, as well as the organizational support appear to play a notable role when planning and carrying out projects of this kind. While CT is commonly seen as developed through programming, the teaching sequence seems to have fostered CT abilities through the project as a whole, with programming playing the role of a tool or a glue depending on the time available, and the students’ skill and ambition level.
