  • Ethics rounds in the ambulance service: a qualitative evaluation

    • BackgroundIt is a common ethical challenge for ambulance clinicians to care for patients with impaired decision-making capacities while assessing and determining the degree of decision-making ability and considering ethical values. Ambulance clinicians’ ethical competence seems to be increasingly important in coping with such varied ethical dilemmas. Ethics rounds is a model designed to promote the development of ethical competence among clinicians. While standard in other contexts, to the best of our knowledge, it has not been applied within the ambulance service context. Thus, the aim of this study was to describe ambulance clinicians’ experiences of participating in ethics rounds.MethodsThis was a qualitative descriptive study, evaluating an intervention. Data were collected through sixteen interviews with ambulance clinicians who had participated in an intervention involving ethics rounds. The analysis was performed by use of content analysis.ResultsTwo themes describe the participants’ experiences: (1) Reflecting freely within a given framework, and (2) Being surprised by new insights. The following categories form the basis of the themes; 1a) Gentle guidance by the facilitator, 1b) A comprehensible structure, 2a) New awareness in the face of ethical problems, and 2b) Shared learning through dialogue.ConclusionIncorporating structured ethics rounds seems to create a continuous development in ethical competence that may improve the quality of care in the ambulance service. Structured guidance and facilitated group reflections offer ambulance clinicians opportunities for both personal and professional development. An important prerequisite for the development of ethical competence is a well-educated facilitator. Consequently, this type of ethics rounds may be considered a useful pedagogical model for the development of ethical competence in the ambulance service.

  • Characteristics of finasteride users in comparison with non-users: a Nordic nationwide study based on individual-level data from Denmark, Finland, and Sweden

    • Purpose: Published epidemiological studies on the association between finasterideuse and the risk of male breast cancer have been inconclusive due to methodologicallimitations including a few male breast cancer cases included. Determinants of malebreast cancer have been studied, but it remains unexplored whether these are alsorelated to finasteride use and thereby constitute potential confounders. This studyaimed to assess whether there are differences between finasteride users andnonusers with regard to numerous potential confounders.Methods: In total, 246 508 finasteride users (≥35 years) were identified in the pre-scription registries of Denmark (1995-2014), Finland (1997-2013), and Sweden(2005-2014). An equal number of nonusers were sampled. The directed acyclic graph(DAG) methodology was used to identify potential confounders for the associationbetween finasteride and male breast cancer. A logistic regression model comparedfinasteride users and nonusers with regard to potential confounders that were mea-surable in registries and population surveys.Results: Finasteride users had higher odds of testicular abnormalities (odds ratio[OR] 1.40; 95% confidence interval [CI] 1.36-1.44), obesity (1.31; 1.23-1.39), exoge-nous testosterone (1.61; 1.48-1.74), radiation exposure (1.22; 1.18-1.27), and diabe-tes (1.07; 1.04-1.10) and lower odds of occupational exposure in perfume industry orin high temperature environments (0.93; 0.87-0.99), living alone (0.89; 0.88-0.91), liv-ing in urban/suburban areas (0.97; 0.95-0.99), and physical inactivity (0.70;0.50-0.99) compared to nonusers.Conclusions: Systematic differences between finasteride users and nonusers werefound emphasizing the importance of confounder adjustment of associationsbetween finasteride and male breast cancer.

  • Pharmacological targeting of MTHFD2 suppresses acute myeloid leukemia by inducing thymidine depletion and replication stress

    • The folate metabolism enzyme MTHFD2 (methylenetetrahydrofolate dehydrogenase/cyclohydrolase) is consistently overexpressed in cancer but its roles are not fully characterized, and current candidate inhibitors have limited potency for clinical development. In the present study, we demonstrate a role for MTHFD2 in DNA replication and genomic stability in cancer cells, and perform a drug screen to identify potent and selective nanomolar MTHFD2 inhibitors; protein cocrystal structures demonstrated binding to the active site of MTHFD2 and target engagement. MTHFD2 inhibitors reduced replication fork speed and induced replication stress followed by S-phase arrest and apoptosis of acute myeloid leukemia cells in vitro and in vivo, with a therapeutic window spanning four orders of magnitude compared with nontumorigenic cells. Mechanistically, MTHFD2 inhibitors prevented thymidine production leading to misincorporation of uracil into DNA and replication stress. Overall, these results demonstrate a functional link between MTHFD2-dependent cancer metabolism and replication stress that can be exploited therapeutically with this new class of inhibitors. Helleday and colleagues describe a nanomolar MTHFD2 inhibitor that causes replication stress and DNA damage accumulation in cancer cells via thymidine depletion, demonstrating a potential therapeutic strategy in AML tumors in vivo.

  • GWAS Meta-Analysis of Suicide Attempt: Identification of 12 Genome-Wide Significant Loci and Implication of Genetic Risks for Specific Health Factors.

    • Suicidal behavior is heritable and is a major cause of death worldwide. Two large-scale genome-wide association studies (GWASs) recently discovered and cross-validated genome-wide significant (GWS) loci for suicide attempt (SA). The present study leveraged the genetic cohorts from both studies to conduct the largest GWAS meta-analysis of SA to date. Multi-ancestry and admixture-specific meta-analyses were conducted within groups of significant African, East Asian, and European ancestry admixtures.This study comprised 22 cohorts, including 43,871 SA cases and 915,025 ancestry-matched controls. Analytical methods across multi-ancestry and individual ancestry admixtures included inverse variance-weighted fixed-effects meta-analyses, followed by gene, gene-set, tissue-set, and drug-target enrichment, as well as summary-data-based Mendelian randomization with brain expression quantitative trait loci data, phenome-wide genetic correlation, and genetic causal proportion analyses.Multi-ancestry and European ancestry admixture GWAS meta-analyses identified 12 risk loci at p values <5×10-8. These loci were mostly intergenic and implicated DRD2, SLC6A9, FURIN, NLGN1, SOX5, PDE4B, and CACNG2. The multi-ancestry SNP-based heritability estimate of SA was 5.7% on the liability scale (SE=0.003, p=5.7×10-80). Significant brain tissue gene expression and drug set enrichment were observed. There was shared genetic variation of SA with attention deficit hyperactivity disorder, smoking, and risk tolerance after conditioning SA on both major depressive disorder and posttraumatic stress disorder. Genetic causal proportion analyses implicated shared genetic risk for specific health factors.This multi-ancestry analysis of suicide attempt identified several loci contributing to risk and establishes significant shared genetic covariation with clinical phenotypes. These findings provide insight into genetic factors associated with suicide attempt across ancestry admixture populations, in veteran and civilian populations, and in attempt versus death.

  • Risk of Juvenile Idiopathic Arthritis and Rheumatoid Arthritis in Patients With Celiac Disease : A Population-Based Cohort Study

    • INTRODUCTION: Celiac disease (CD) is associated with many immune-mediated conditions, but a definitive epidemiological association between CD and juvenile idiopathic arthritis (JIA) or rheumatoid arthritis (RA) has not been established. We quantified the risk of JIA and RA among patients with CD using a population-based cohort.METHODS: We identified patients diagnosed with biopsy-proven CD between 2004 and 2017 using data from a national histopathology cohort in Sweden. Each patient was matched by age, sex, calendar year, and geographic region to reference individuals in the general population. We calculated the incidence and estimated the relative risk, through Cox proportional hazards models, of JIA in individuals with CD aged ≥18.RESULTS: We identified 24,014 individuals with CD who were matched to 117,397 reference individuals from the general population. Among individuals aged <18, the incidence rate of JIA was 5.9 per 10,000 person-years in patients with CD and 2.2 per 10,000 person-years in the general population (n events = 40 and 73, respectively; hazard ratio [HR] 2.68, 95% confidence interval 1.82-3.95) over a follow-up of 7.0 years. Among individuals aged >= 18, the incidence of RA was 8.4 per 10,000 person-years in CD and 5.1 per 10,000 person-years in matched comparators (n events = 110 and 322, respectively; HR 1.70, 95% confidence interval 1.36-2.12) over a follow-up of 8.8 years.DISCUSSION: Among children with CD, JIA develops nearly 3 times as often as it does in the general population, and among adults with CD, RA occurs nearly 2 times as often. Clinicians caring for patients with CD with joint symptoms should have a low threshold to evaluate for JIA or RA.

    • Ett ständigt aktuellt tema är studenter som upplever att undervisningsformerna karakteriseras av leda (se Daschman et al 2013) och lärare som tycker sig se ett bristande engagemang (se Foley 2013). Intrycket från en programmeringskurs med fokus på mobilappar och tjänster var projekten som studenterna skapade brast i konceptuell och teknisk höjd.  Vare sig studenter eller lärare tyckte kursen var särskilt intressant.  Två samverkande omständigheter identifierades: Lärarna förmådde inte att relevantgöra innehållet på ett engagerade och inspirerande sätt.  Studenterna tyckte innehållet var för svårt och vågade inte chansa. Foley (2013) diskuterar hur lärare kör på i trygga banor (complacency) och inte engagerar sig, något som leder till studenter som inte gör mer än vad som krävs. Faktorer som undervisningens upplägg, över- eller understimulering leder till alltså till brist på engagemang och avhopp.  Det som krävs är en mer varierad, engagerande och individanpassad undervisning.  För att bryta detta har pedagogiken ändrats på kursen för att skapa engagemang på olika sätt.   Kraven ställs på ett annat sätt:  genom att använda noggrant kvalificerade värdeladdade ord för att beskriva kraven på studenternas projekt och laborationer. Exempelvis ska tekniska laborationer ska vara ”avancerade” och tjänsterna/koncepten ”spännande” eller ”unika”.  Vi är dock tydliga med att utmaningarna i sig är något som studenterna får misslyckas med. Istället för att ställa omöjliga krav uppmanar vi studenterna utforska gränserna (”hur spännande kan det bli?”).   Praktikbaserade gemenskaper (efter Wenger, Farnsworth et al 2016):  redovisning av laborationer och projekt genomförs i ett interaktivt muntligt format där studenterna själva väljer vad de vill fokusera på inom ramen för en kritisk diskussion. Studenterna sätter därmed sin egen agenda inom kursens tematiska ram där de kan fokusera på teknologi eller på tjänsteutveckling. Genom att de specificerar sina egna tekniska laborationer direkt i dialog med läraren får de omedelbar återkoppling. Studenterna får också till uppgift att designa en lösning som representerar ett för dem verkligt problem – något som kräver engagemang och en gemensam orientering mot kursinnehållet. Kursinnehållet och fokus kommer alltså inte längre från lärarna utan från studenterna som investerat sitt eget skapande (och tänkande).  Lärarna engageras sig för att inte framstå som okunniga inför studenterna.   En mycket försiktig tendens i kursutvärderingar av två omgångar av kursen genomförd nya sättet är att studenter fortfarande tycker laborationerna är för svåra, men samtidigt att anser de att kursen är givande.   Referenser  Daschmann, E. På väg dit ser vi två steg som kan adresseras separat och tillsammans ökad delning av data och en samordnad digital materialsammanställning över en byggnads livscykel. Nya aktörer behöver knytas till ett digitalt informationsekosystem kring miljödata för byggmaterial för att stödja denna utveckling.

  • Celiac disease and upper secondary school achievement in Sweden : A retrospective cohort study

    • BACKGROUND: Both undiagnosed celiac disease and some chronic childhood diseases are associated with lower academic achievement. However, there is little knowledge of achievements in those diagnosed with celiac disease. Our aim was to investigate school achievements in upper secondary school among Swedish adolescents with celiac disease.METHODS: We performed a retrospective cohort study using register data. We analyzed choice of upper secondary school program, completion of upper secondary school including achievements of basic eligibility for college/university, and final grade in individuals with celiac disease diagnosed before 15 years of age, born 1991-97. We compared with the Swedish population of the same birth years. Analyses were adjusted for sex, year of birth, living region at 17 years of age, and parental education as well as income.RESULTS: The cohort included 734 074 individuals, whereof 3 257 (62% females) with celiac disease. There was no significant difference in choice of upper secondary school program. No significant difference was found in completion or achieving basic eligibility for college/university in adjusted analyses. The mean final grade in the celiac disease group was 13.34 (standard deviation 4.85) compared to 12.78 (standard deviation 5.01) in the reference population (p < 0.001), out of a maximum of 20. The effect of celiac disease on final grade remained in adjusted analyses (p = 0.012).CONCLUSIONS: We found that diagnosed celiac disease does not negatively affect school achievements in upper secondary school. This finding suggests the diagnosis, treatment and follow-up programs of celiac disease could reverse potential deleterious academic processes.

    • Plant traits-the morphological, anatomical, physiological, biochemical and phenological characteristics of plants-determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait-based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits-almost complete coverage for 'plant growth form'. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait-environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives.

  • Dissecting the Shared Genetic Architecture of Suicide Attempt, Psychiatric Disorders, and Known Risk Factors

    • BACKGROUND: Suicide is a leading cause of death worldwide, and nonfatal suicide attempts, which occur far more frequently, are a major source of disability and social and economic burden. Both have substantial genetic etiology, which is partially shared and partially distinct from that of related psychiatric disorders.METHODS: We conducted a genome-wide association study (GWAS) of 29,782 suicide attempt (SA) cases and 519,961 controls in the International Suicide Genetics Consortium (ISGC). The GWAS of SA was conditioned on psychiatric disorders using GWAS summary statistics via multitrait-based conditional and joint analysis, to remove genetic effects on SA mediated by psychiatric disorders. We investigated the shared and divergent genetic architectures of SA, psychiatric disorders, and other known risk factors.RESULTS: Two loci reached genome-wide significance for SA: the major histocompatibility complex and an intergenic locus on chromosome 7, the latter of which remained associated with SA after conditioning on psychiatric disorders and replicated in an independent cohort from the Million Veteran Program. This locus has been implicated in risk-taking behavior, smoking, and insomnia. SA showed strong genetic correlation with psychiatric disorders, particularly major depression, and also with smoking, pain, risk-taking behavior, sleep disturbances, lower educational attainment, reproductive traits, lower socioeconomic status, and poorer general health. After conditioning on psychiatric disorders, the genetic correlations between SA and psychiatric disorders decreased, whereas those with nonpsychiatric traits remained largely unchanged.CONCLUSIONS: Our results identify a risk locus that contributes more strongly to SA than other phenotypes and suggest a shared underlying biology between SA and known risk factors that is not mediated by psychiatric disorders.

  • The Palomar Transient Factory Core-collapse Supernova Host-galaxy Sample. I. Host-galaxy Distribution Functions and Environment Dependence of Core-collapse Supernovae

    • Several thousand core-collapse supernovae (CCSNe) of different flavors have been discovered so far. However, identifying their progenitors has remained an outstanding open question in astrophysics. Studies of SN host galaxies have proven to be powerful in providing constraints on the progenitor populations. In this paper, we present all CCSNe detected between 2009 and 2017 by the Palomar Transient Factory. This sample includes 888 SNe of 12 distinct classes out to redshift z approximate to 1. We present the photometric properties of their host galaxies from the far-ultraviolet to the mid-infrared and model the host-galaxy spectral energy distributions to derive physical properties. The galaxy mass function of Type Ic, Ib, IIb, II, and IIn SNe ranges from 10(5) to 10(11.5) M (circle dot), probing the entire mass range of star-forming galaxies down to the least-massive star-forming galaxies known. Moreover, the galaxy mass distributions are consistent with models of star-formation-weighted mass functions. Regular CCSNe are hence direct tracers of star formation. Small but notable differences exist between some of the SN classes. Type Ib/c SNe prefer galaxies with slightly higher masses (i.e., higher metallicities) and star formation rates than Type IIb and II SNe. These differences are less pronounced than previously thought. H-poor superluminous supernovae (SLSNe) and SNe Ic-BL are scarce in galaxies above 10(10) M (circle dot). Their progenitors require environments with metallicities of < 0.4 and < 1 solar, respectively. In addition, the hosts of H-poor SLSNe are dominated by a younger stellar population than all other classes of CCSNe. Our findings corroborate the notion that low metallicity and young age play an important role in the formation of SLSN progenitors.

    • IntroductionThis study employed an integrative system and causal inference approach to explore molecular signatures in blood and CSF, the amyloid/tau/neurodegeneration [AT(N)] framework, mild cognitive impairment (MCI) conversion to Alzheimer's disease (AD), and genetic risk for AD. MethodsUsing the European Medical Information Framework (EMIF)-AD cohort, we measured 696 proteins in cerebrospinal fluid (n = 371), 4001 proteins in plasma (n = 972), 611 metabolites in plasma (n = 696), and genotyped whole-blood (7,778,465 autosomal single nucleotide epolymorphisms, n = 936). We investigated associations: molecular modules to AT(N), module hubs with AD Polygenic Risk scores and APOE4 genotypes, molecular hubs to MCI conversion and probed for causality with AD using Mendelian randomization (MR). ResultsAT(N) framework associated with protein and lipid hubs. In plasma, Proprotein Convertase Subtilisin/Kexin Type 7 showed evidence for causal associations with AD. AD was causally associated with Reticulocalbin 2 and sphingomyelins, an association driven by the APOE isoform. DiscussionThis study reveals multi-omics networks associated with AT(N) and causal AD molecular candidates.

  • Toward productive complicity: Applying 'traditional ecological knowledge' in environmental science

    • Culture and tradition have long been the domains of social science, particularly social/cultural anthropology and various forms of heritage studies. However, many environmental scientists whose research addresses environmental management, conservation, and restoration are also interested in traditional ecological knowledge, indigenous and local knowledge, and local environmental knowledge (hereafter TEK), not least because policymakers and international institutions promote the incorporation of TEK in environmental work. In this article, we examine TEK usage in peer-reviewed articles by environmental scientists published in 2020. This snapshot of environmental science scholarship includes both critical discussions of how to incorporate TEK in research and management and efforts to do so for various scholarly and applied purposes. Drawing on anthropological discussions of culture, we identify two related patterns within this literature: a tendency toward essentialism and a tendency to minimize power relationships. We argue that scientists whose work reflects these trends might productively engage with knowledge from the scientific fields that study culture and tradition. We suggest productive complicity as a reflexive mode of partnering, and a set of questions that facilitate natural scientists adopting this approach: What and/or who is this TEK for? Who and what will benefit from this TEK deployment? How is compensation/credit shared? Does this work give back and/or forward to all those involved?

    • Yield assessments (YA) and Long-Term Yield Predictions (LTYP) are a prerequisite for business decisions on long term investments into photovoltaic (PV) power plants. Together with cost data (CAPEX, OPEX and discount rate), the output of a YA and LTYP (utilisation rate, performance loss rate and lifetime) provides to the financial investors the parameters needed for the calculation of the Levelised Cost of Electricity (LCOE) and to assess the cash flow model of an investment with relative Internal Rate of Return (IRR) and Net Present Value (NPV).YA and LTYP outputs should be provided with a related exceedance probability. This gives the right tool to stakeholders involved in PV projects to take the best decision in terms of riskaversion. A reduction in the uncertainty of the energy yield can lead to higher values for a given exceedance probability and hence a stronger business case. Various efforts in the literature show the importance of having a common framework that can assess the impact of technical risks on the economic performance of a PV project.The most important parameter influencing the energy yield assessment is the site-specific insolation. Several aspects need to be considered: reliability of the database, interannual variability, long term trends.Site adaptation techniques combine short-term measured data and long-term satellite estimates. Short periods of measured data but with site-specific seasonal and diurnal characteristics are combined with satellite-derived data having a long period of record with not necessarily site-specific characteristics. Upon completion of the measurement campaign, which is typically around one-year, different methodologies can be applied between the measured data at the target site, spanning a relatively short period, and the satellite data, spanning a much longer period. The complete record of satellite data is then used in this relationship to predict the long-term solar resource at the target site. Assuming a strong correlation, the strengths of both data sets are captured and the uncertainty in the long-term estimate can be reduced.  In Müller et al [1] an analysis on long-term trends for measured in-plane irradiance, Performance Ratio and energy yield for 44 rooftop installations in Germany was performed showing an average increase of in-plane irradiance of 1.1 %/year or about 11 %/decade over the period 2008 to 2018 for these systems. The increase in irradiance was especially higher than the observed Performance Loss Rate so that the energy yields of the systems analysed increased over the years with an average trend of 0.3 %/year.  The typical output of Yield Assessments should report the contribution to each derating factor, starting from the Global Horizontal Irradiation to the energy injected in the grid. The starting point of PR = 100 is considered after applying the horizon shading as this become the annual insolation seen by the PV modules. The following table shows a best practice in providing an overview of gains/losses along each modelling step and the related uncertainty. The uncertainty related to each modelling step can be provided already referred to the irradiation/yield value or to the parameter that is modelled. The value in the table for the specific yield (including its uncertainty) is to be understood as an average value over the entire operating period. The possible deviations between the yields for individual recorded years and the specific yield calculated can be assessed by including interannual variability. For example, for temperature-dependent losses, the value of uncertainty could be referred to the temperature variability of the profile used in the assessment or to the temperature model used in the assessment. The ambient temperature variability and the various temperature models will lead to a different contribution in terms of yield loss and in terms of uncertainty.An emerging challenge in YAs is also due to the deployment of novel technologies (e.g. bifacial PV modules) with a contribution in terms of uncertainty that needs to be properly assessed.Building upon the knowledge available in the literature and the previous IEA PVPS Task 13 report [2], in this report we have moved forward from the uncertainty framework in yield assessment to two real implementations of it and the impact that uncertainties can have on lifetime yield predictions, on the LCOE and on the cash-flow.One of the most relevant question that we have tried to answer is also the following: How reliable are YA’s?This is an apparently simple question; however, the answer is not equally simple. Typically, investors require one YA. In some cases, more YAs might be requested if results are unclear. The various YAs can be averaged to assign a purchase value to a given project. In any case the question remains unanswered: why different assessors obtain different answers? Is one YA more reliable than others?

  • The Seventeenth Data Release of the Sloan Digital Sky Surveys : Complete Release of MaNGA, MaStar, and APOGEE-2 Data

    • This paper documents the seventeenth data release (DR17) from the Sloan Digital Sky Surveys; the fifth and final release from the fourth phase (SDSS-IV). DR17 contains the complete release of the Mapping Nearby Galaxies at Apache Point Observatory (MaNGA) survey, which reached its goal of surveying over 10,000 nearby galaxies. The complete release of the MaNGA Stellar Library accompanies this data, providing observations of almost 30,000 stars through the MaNGA instrument during bright time. DR17 also contains the complete release of the Apache Point Observatory Galactic Evolution Experiment 2 survey that publicly releases infrared spectra of over 650,000 stars. The main sample from the Extended Baryon Oscillation Spectroscopic Survey (eBOSS), as well as the subsurvey Time Domain Spectroscopic Survey data were fully released in DR16. New single-fiber optical spectroscopy released in DR17 is from the SPectroscipic IDentification of ERosita Survey subsurvey and the eBOSS-RM program. Along with the primary data sets, DR17 includes 25 new or updated value-added catalogs. This paper concludes the release of SDSS-IV survey data. SDSS continues into its fifth phase with observations already underway for the Milky Way Mapper, Local Volume Mapper, and Black Hole Mapper surveys.

    • Bygg- och fastighetssektorn står för nästan en femtedel av Sveriges klimatpåverkan, varav hälften kommer från byggverksamheten. Projektet Klimatkrav till rimlig kostnad har därför tagit fram en vägledning för att underlätta för bostadsföretag att ställa klimatkrav för byggprojekt. Förutsättningen för att ställa klimatkrav vid upphandling är att det finns ett enhetligt sätt att beräkna klimatpåverkan så att till exempel beräkningar från olika anbudsgivare är möjliga att jämföra.Därför har projektet tagit fram anvisningar för LCA-beräkning av byggprojekt som bland annat anger att vedertagen LCA-metodik skall användas, vilka delar av byggnaden som skall inkluderas och så vidare. Anvisningarna är en tolkning av Boverkets lagförslag på klimatdeklaration samt erfarenheter från praktiska beräkningar inom detta och andra projekt.Vägledningen presenterar förslag på konkreta informations-, förbättrings-, prestanda- och tilldelningskrav vilka kan kompletterats med negativa eller positiva ekonomiska konsekvenser. Entreprenörens uppföljning mot verkligt utförande beskrivs samt hur en beställare utvärderar och granskar beräkningar.Vägledningen kan användas av både privata och offentliga beställare och den kan användas i framtagande av upphandlingsdokument, löpande arbete under ett byggprojekt och i slutskedet av ett projekt. Den är särskilt användbar för nybyggnationer, men går att anpassa till ombyggnation.

    • BACKGROUND: physiological differences between males and females could contribute to the development of Alzheimer's Disease (AD). Here, we examined metabolic pathways that may lead to precision medicine initiatives.METHODS: We explored whether sex modifies the association of 540 plasma metabolites with AD endophenotypes including diagnosis, cerebrospinal fluid (CSF) biomarkers, brain imaging, and cognition using regression analyses for 695 participants (377 females), followed by sex-specific pathway overrepresentation analyses, APOE ε4 stratification and assessment of metabolites' discriminatory performance in AD.RESULTS: In females with AD, vanillylmandelate (tyrosine pathway) was increased and tryptophan betaine (tryptophan pathway) was decreased. The inclusion of these two metabolites (area under curve (AUC) = 0.83, standard error (SE) = 0.029) to a baseline model (covariates + CSF biomarkers, AUC = 0.92, SE = 0.019) resulted in a significantly higher AUC of 0.96 (SE = 0.012). Kynurenate was decreased in males with AD (AUC = 0.679, SE = 0.046).CONCLUSIONS: metabolic sex-specific differences were reported, covering neurotransmission and inflammation pathways with AD endophenotypes. Two metabolites, in pathways related to dopamine and serotonin, were associated to females, paving the way to personalised treatment.
