  • Evidence for octupole collectivity in 172Pt

    • Excited states in the extremely neutron-deficient nucleus 172Pt were populated via 96Ru(78Kr,2p) and 92Mo(83Kr,3n) reactions. The level scheme has been extended up to an excitation energy of  ~ 5 MeV and tentative spin-parity assignments up to Iπ = 18+. Linear polarization and angular distribution measurements were used to determine the electromagnetic E1 character of the dipole transitions connecting the positive-parity ground-state band with an excited side-band, firmly establishing it as a negative-parity band. The lowest member of this negative-parity structure was firmly assigned spin-parity 3-. In addition, we observed an E3 transition from this 3- state to the ground state, providing direct evidence for octupole collectivity in 172Pt. Large-scale shell model (LSSM) and total Routhian surface (TRS) calculations have been performed, supporting the interpretation of the 3- state as a collective octupole-vibrational state.

  • Lifetime measurements in the even-even 102-108Cd isotopes

    • Background: The heaviest T-z = 0 doubly-magic nucleus, Sn-100, and the neighboring nuclei offer unique opportunities to investigate the properties of nuclear interaction. For instance, the structure of light-Sn nuclei has been shown to be affected by the delicate balance between nuclear-interaction components, such as pairing and quadrupole correlations. From Cd to Te, many common features and phenomena have been observed experimentally along the isotopic chains, leading to theoretical studies devoted to a more general and comprehensive study of the region. In this context, having only two proton holes in the Z = 50 shell, the Cd isotopes are expected to present properties similar to those found in the Sn isotopic chain.Purpose: The aim of this work was to measure lifetimes of excited states in neutron-deficient nuclei in the vicinity of Sn-100.Methods: The neutron-deficient nuclei in the N approximate to Z approximate to 50 region were populated using a multinucleon transfer reaction with a Cd-106 beam and a Mo-92 target. The beamlike products were identified by the VAMOS++ spectrometer, while the gamma rays were detected using the AGATA array. Lifetimes of excited states were determined using the recoil distance Doppler-shift method, employing the Cologne differential plunger.Results: Lifetimes of low-lying states were measured in the even-mass Cd-102-(108) isotopes. In particular, multiple states with excitation energy up to MeV, belonging to various bands, were populated in approximate to 3 Cd-106 via inelastic scattering. The transition strengths corresponding to the measured lifetimes were compared with those resulting from state-of-the-art beyond-mean-field calculations using the symmetry-conserving configuration-mixing approach.Conclusions: Despite the similarities in the electromagnetic properties of the low-lying states, there is a fundamental structural difference between the ground-state bands in the Z = 48 and Z = 50 isotopes. The comparison between experimental and theoretical results revealed a rotational character of the Cd nuclei, which have prolate-deformed ground states with beta(2) approximate to 0.2. At this deformation Z = 48 becomes a closed-shell configuration, which is favored with respect to the spherical one.

  • Single-particle states and parity doublets in odd- Z Ac 221 and Pa 225 from α -decay spectroscopy

    • Low-lying states in the odd-Z isotopes Ac13289221 and Pa13491225 have been studied using α-particle and αγ-coincidence spectroscopy in the Pa225→Ac221→Fr217 decay chain. Ground-state spin and parity assignments of Iπ = 5/2- are proposed for both Ac221 and Pa225, with the odd proton occupying the ω = 5/2 orbital of the quadrupole-octupole deformed shell model in both nuclei. In Ac221, excited states in the bands based on the ω = 5/2 and ω = 3/2 orbitals have been identified, including proposed parity-doublet states. The results suggest that reflection-asymmetric deformation of the ground state persists in the odd-A members of the isotope chains down to N = 132 for Ac and N = 134 for Pa, before reaching the transitional region at N = 130.

  • Pairing-quadrupole interplay in the neutron-deficient tin nuclei : First lifetime measurements of low-lying states in Sn-106,Sn-108

    • The lifetimes of the low-lying excited states 2(+) and 4(+) have been directly measured in the neutron-deficient Sn-106,Sn-108 isotopes. The nuclei were populated via a deep-inelastic reaction and the lifetime measurement was performed employing a differential plunger device. The emitted gamma rays were detected by the AGATA array, while the reaction products were uniquely identified by the VAMOS++ magnetic spectrometer. Large-Scale Shell-Model calculations with realistic forces indicate that, independently of the pairing content of the interaction, the quadrupole force is dominant in the B(E2; 2(1)(+) -> 0(g.s)(+)) values and it describes well the experimental pattern for Sn104-114 ; the B(E2;(+)(4) -> 2(1)(+)) values, measured here for the first time, depend critically on a delicate pairing-quadrupole balance, disclosed by the very precise results in Sn-108.

  • Reassigning the shapes of the 0+ states in the 186Pb nucleus

    • Across the physics disciplines, the 186Pb nucleus is the only known system, where the two first excited states, together with the ground state, form a triplet of zero-spin states assigned with prolate, oblate and spherical shapes. Here we report on a precision measurement where the properties of collective transitions in 186Pb were determined in a simultaneous in-beam γ-ray and electron spectroscopy experiment employing the recoil-decay tagging technique. The feeding of the 02+ state and the interband 22+→21+ transition have been observed. We also present direct measurement of the energies of the electric monopole transitions from the excited 0+ states to the 0+ ground state. In contrast to the earlier understanding, the obtained reduced transition probability B(E2;21+→02+) value of 190(80) W.u., the transitional quadrupole moment ∣Qt(21+→02+)∣=7.7(33) eb and intensity balance arguments provide evidence to reassign the 02+ and 03+ states with predominantly prolate and oblate shape, respectively. Our work demonstrates a step-up in experimental sensitivity and paves the way for systematic studies of electric monopole transitions in this region. These electric monopole transitions probe the nuclear volume in a unique manner and provide unexploited input for development of the next-generation energy density functional models.

    • BackgroundGenesis of acquired epilepsy includes transformations spanning genetic-to- network-level modifications, disrupting the regional excitatory/inhibitory balance. Methodology concurrently tracking changes at multiple levels is lacking. Here, viral vectors are used to differentially express two opsin proteins in neuronal populations within dentate gyrus (DG) of hippocampus. When activated, these opsins induced excitatory or inhibitory neural output that differentially affected neural networks and epileptogenesis. In vivo measures included behavioral observation coupled to real-time measures of regional glutamate flux using ceramic-based amperometric microelectrode arrays (MEAs).

  • In-situ temperatures and thermal properties of the East Siberian Arctic shelf sediments : Key input for understanding the dynamics of subsea permafrost

    • Significant reserves of methane (CH4) are held in the Arctic shelf, but the release of CH4 to the overlying ocean and, subsequently, to the atmosphere has been believed to be restricted by impermeable subsea permafrost, which has sealed the upper sediment layers for thousands of years. Our studies demonstrate progressive degradation of subsea permafrost which controls the scales of CH4 release from the sediment into the water-atmospheric system. Thus, new knowledge about the thermal state of subsea permafrost is crucial for better understanding of the permafrost -hydrate system and associated CH4 release from the East Siberian Arctic Shelf (ESAS) – the broadest and shallowest shelf in the World Ocean, which contains about 80% of subsea permafrost and giant pools of hydrates. Meanwhile, the ESAS, still presents large knowledge gaps in many aspects, especially with respect to subsea permafrost distribution and physical properties of bottom sediments. New field data show that the ESAS has an unfrozen (ice-free) upper sediment layer, which in-situ temperature is −1.0 to −1.8 °C and 0.6оС above the freezing point. On one hand, these cold temperature patterns may be related to the presence of subsea permafrost, which currently primarily occurs in the part of the ESAS that is shallower than 100 m, while ice-bearing sediments may also exist locally under deeper water in the Laptev Sea. On the other hand, the negative bottom sediment temperatures of −1.8 °C measured on the Laptev Sea continental slope sediments underlying water columns as deep as down to 330 m may result from dissociation of gas hydrates or possibly from dense water cascading down from the shelf. In contrast, data collected on recent expeditions in the northern Laptev shelf, zones of warmer bottom temperatures are coinciding with methane seeps, likely induced by seismic and tectonic activity in the area. These warm temperatures are not seen in the East Siberian Sea area, not even in areas of methane seeps, yet with little seismic activity.The thermal conductivity and heat capacity of bottom sediments recorded in the database of thermal parameters for the ESAS areas mainly depend on their lithification degree (density or porosity), moisture content, and particle size distribution. The thermal conductivity and heat capacity average about 1.0 W/(m·K) and 2900 kJ/(m3·K), with ±20% and ±10% variance, respectively, in all sampled Arctic sediments to a sub-bottom interval of 0–0.5 m.

    • We reconstructed Holocene paleoceanography of the Sherard Osborn Fjord (SOF), N Greenland, and Lincoln Sea in the eastern Arctic Ocean using sediment properties and micropaleontology from cores obtained during the Ryder 2019 Expedition. Our aims were to better understand faunal indicators of water mass influence on Ryder Glacier and the Lincoln Sea at water depths >500 m. Benthic microfaunal reflect glacio-marine interval during late deglaciation ~10.5 to 8.5 ka (kiloannum) during the Holocene Thermal Maximum (HTM) with dominant benthic foraminiferal species Cassidulina neoteretis, Cassidulina reniforme, and the ostracode Rabilimis mirabilis. Casssidulina neoteretis is considered an indicator of Atlantic Water (AW) throughout the Arctic Ocean and Nordic Seas; C. reniforme reflects glacio-marine conditions from the retreating Ryder Glacier. Deglaciation was followed by a period of elevated productivity and diverse ostracode faunal assemblages that suggest AW influence from 8.5 to 6 ka in the Lincoln Sea and inside SOF. The Holocene occurrence of the ostracode species Acetabulastoma arcticum, that appears in low numbers in the Lincoln Sea and briefly (~ 4–3 ka) in SOF, reflects the presence of variable sea ice in this region. Based on the similarities of the Lincoln Sea and fjord ostracodes to modern and glacial-deglacial faunas from the central Arctic Ocean, the AW influence likely originates from recirculation of AW water from the central Arctic Basin. In general, our results suggest a strong but temporally varying influence of AW during the entire 10.5 kyr record of the Lincoln Sea and SOF.

    • Hypopituitarism means a partial or complete inability to produce pituitary hormones, including those regulating gonadal function. The condition therefore leads to hormone deficiencies that may affect sexuality in various ways. This study aimed to explore patients' own experiences of hypopituitarism in relation to sexuality, and it is based on interviews with 19 men and women. A qualitative methodology inspired by Gadamer's philosophical hermeneutics was used. Analysis revealed that current healthcare practices, which focus primarily on sexual functioning, fail to address patients' existential concerns about loneliness, loving relationships and selfhood. Another important finding was that women felt that their sexual functioning and wellbeing tended to go unnoticed, while men said that healthcare professionals focused mainly on erectile function rather than problems with sexual wellbeing. Cultural and gender norms were apparent in these healthcare interactions, and proactive efforts to improve awareness is required in order to improve the quality of care provided to these patients. Healthcare professionals need time, space and support for reflection so that they may enhance their knowledge about the complexity of sexuality and discuss the results of research into hormone replacement in men and women. They also need support to develop critical awareness of how gendered norms about sexuality may negatively affect the care provided to persons with hypopituitarism.

  • The International Bathymetric Chart of the Southern Ocean Version 2

    • The Southern Ocean surrounding Antarctica is a region that is key to a range of climatic and oceanographic processes with worldwide effects, and is characterised by high biological productivity and biodiversity. Since 2013, the International Bathymetric Chart of the Southern Ocean (IBCSO) has represented the most comprehensive compilation of bathymetry for the Southern Ocean south of 60 degrees S. Recently, the IBCSO Project has combined its efforts with the Nippon Foundation - GEBCO Seabed 2030 Project supporting the goal of mapping the world's oceans by 2030. New datasets initiated a second version of IBCSO (IBCSO v2). This version extends to 50 degrees S (covering approximately 2.4 times the area of seafloor of the previous version) including the gateways of the Antarctic Circumpolar Current and the Antarctic circumpolar frontal systems. Due to increased (multibeam) data coverage, IBCSO v2 significantly improves the overall representation of the Southern Ocean seafloor and resolves many submarine landforms in more detail. This makes IBCSO v2 the most authoritative seafloor map of the area south of 50 degrees S.

  • Gestational hypertension, preeclampsia, and gestational diabetes mellitus after high exposure to perfluoroalkyl substances from drinking water in Ronneby, Sweden

    • Background: Leakage of fire-fighting foam from an airfield caused contamination of the drinking water supplied to a third of the population in Ronneby, resulting in very high serum levels of predominantly perfluorooctane sulfonate (PFOS) and perfluorohexane sulfonate (PFHxS). The results of studies investigating the association between exposure to perfluorinated alkyl substances (PFAS) and pregnancy complications are inconsistent, and studies at high exposures of PFOS and PFHxS are lacking. Objectives: To investigate the association between exposure to high levels of PFAS and gestational hypertension and preeclampsia, and gestational diabetes mellitus. Methods: We retrieved data on 27 292 childbirths between 1995 and 2013 from the National Medical Birth Register for women that had a residential address in Blekinge county for at least one year before delivery. Residential history was used as a proxy for exposure by categorizing women into high-, intermediate-, or background exposed based on their residential address during the five-year period before childbirth. Data on confounders were retrieved from administrative registers. The outcomes were defined based on International Classification of Diseases codes. We used logistic regression to estimate odds ratios (OR) for gestational hypertension and preeclampsia, and gestational diabetes mellitus. We also investigated effect modification by fetal sex. Results: We found no evidence of increased risk of gestational hypertension and preeclampsia (OR 0.80; CI 0.63-1.03), nor gestational diabetes (OR 1.03; CI 0.67-1.58) after high PFAS exposure. There was no effect modification by fetal sex. Discussion: This was the first study to investigate the association between high exposure to PFOS and PFHxS and pregnancy complications. The results from this study add important knowledge to public health management as new hotspots with high levels of PFAS are continuously discovered.

  • Lifetimes of core-excited states in semi-magic Rh-95

    • Lifetimes of negative-parity states have been determined in the neutron deficient semi-magic (N = 50) nucleus Rh-95. The fusion-evaporation reaction Ni-58(Ca-40, 3p) was used to populate high-spin states in Rh-95 at the Grand Accelerateur National d'Ions Lourds (GANIL) accelerator facility. The results were obtained using the Doppler Shift Attenuation Method (DSAM) based on the Doppler broadened line shapes produced during the slowing down process of the residual nuclei in a thick 6 mg/cm(2) metallic target. B(M1) and B(E2) reduced transition strengths are compared with predictions from large-scale shell-model calculations. state-of-the-art theory. Remarkably, the structural features up to moderate angular momentum of nuclei immediately below the N = Z = 50 shell closures can be described with high accuracy in a very simple way by shell-model calculations including only the g(9/2) and p(1/2) subshells. Of special interest is the neutron-proton pair coupling scheme which is expected to appear in the heaviest N=Z nuclei [1,2] and the seniority structure of the N = 50 isotones [3-7]. However, multiple core-excited states have been observed in the semi-magic nuclei of the Sn-100 region [8-10]. The theoretical study of those states is a challenging task, which requires a significantly larger model space for their interpretation. Transition probabilities between nuclear states provide important constraints for theoretical modelling of the structure of the nuclei of interest. Our previous lifetime study of the semimagic (N = 50) nucleus Ru-94 [ 11,12] provided information on the electromagnetic decay properties of neutron-core excited states. We now address lifetime measurements in its closest, more neutron deficient, isotone Rh-95 using the same DSAM technique. The experimental results have been interpreted within the framework of large-scale shell-model (LSSM) calculations.

    • Relative Sea Levels (RSLs) derived primarily from marine bivalves near Petermann Glacier, NW Greenland, constrain past regional ice-mass changes through glacial isostatic adjustment (GIA) modeling. Oxygen isotopes measured on bivalves corrected for shell-depth habitat and document changing meltwater input. Rapid RSL fall of up to 62 m/kyr indicates ice loss at or prior to ∼9 ka. Transition to an RSL stillstand starting at ∼6 ka reflects renewed ice-mass loading followed by further mass loss over the past few millennia. GIA simulations of rapid early RSL fall suggest a low regional upper-mantle viscosity. Early loss of grounded ice tracks atmospheric warming and pre-dates the eventual collapse of Petermann Glacier's floating ice tongue near ∼7 ka, suggesting grounding zone stabilization during early phases of deglaciation. We hypothesize mid-Holocene regrowth of regional ice caps in response to cooling and increased precipitation, following loss of the floating shelf ice. Remnants of these ice caps remain present but are now melting.

    • The spinal locomotor network is frequently used for studies into how neuronal circuitsare formed and how cellular activity shape behavioral patterns. A population of dI6interneurons, marked by the Doublesex and mab-3 related transcription factor 3(Dmrt3), has been shown to participate in the coordination of locomotion and gaitsin horses, mice and zebrafish. Analyses of Dmrt3 neurons based on morphology,functionality and the expression of transcription factors have identified differentsubtypes. Here we analyzed the transcriptomes of individual cells belonging to theDmrt3 lineage from zebrafish and mice to unravel the molecular code that underliestheir subfunctionalization. Indeed, clustering of Dmrt3 neurons based on their geneexpression verified known subtypes and revealed novel populations expressing uniquemarkers. Differences in birth order, differential expression of axon guidance genes,neurotransmitters, and their receptors, as well as genes affecting electrophysiologicalproperties, were identified as factors likely underlying diversity. In addition, thecomparison between fish and mice populations offers insights into the evolutionarydriven subspecialization concomitant with the emergence of limbed locomotion

  • Deciphering neural networks in the somatosensory system using single-cell transcriptomics and rabies tracing

    • Itch has evolved to protect us from malicious parasites keen to suck our blood or lay their eggs in our skin. We can detect both the movement of these parasites and the toxins they introduce with specialized neurons called pruriceptors. When we feel an itch, we get a desire to scratch it. Scratching an itch eases the itch sensation, and this is thought to be regulated by neuronal circuits in the spinal cord. This reactive aspect of itch makes is an interesting system to study as it involves both sensory and motor circuitry. The spinal cord hosts a vast number of different neuronal cell types, and better understanding of these are needed to efficiently delineate the circuitry between them. To find these cell types, we sequence the transcriptome of thousands of individual neurons in the dorsal horn of the spinal cord and identified 15 excitatory and 15 inhibitory neuronal populations (Paper I). Furthermore, we found that cell types expressing neuropeptide Y (NPY) contributed to the inhibition of chemically induced itch via the NPY receptor 2 (Paper II) and inhibition of somatostatin-induced itch via NPY receptor 1 (Paper III). We are currently mapping the neurons presynaptic to the NPY neurons using a Npy-Cre mouse line combined with monosynaptic rabies tracing and find inputs from the dorsal root ganglions, the spinal cord, and the brain (Paper IV). To help decipher circuit connectivity, we developed a method that links cell types expressing matching ligand and receptor pairs in single cell RNA-sequencing (scRNA-seq) datasets (Paper V). We furthermore used scRNA-seq to identify differences and similarities of locomotor circuitry related cells expressing doublesex and mab-3 related transcription factor 3 in zebrafish and mouse (Paper VI).In this thesis, we used a combination of powerful and novel tools to investigate questions that were previously difficult to address. It is my belief that spatial transcriptomics, now poised with the knowledge gained from scRNA-seq, will transform how we think about cell types in the central nervous system, since the location of a neuron is critical for its role in a circuit.

Such complex data structures demand novel analysis methods and tools for exploratory analysis.ResultsWe propose a method to predict the putative ligand–receptor interactions between cell types from single-cell RNA-sequencing data. This is achieved by inferring and incorporating interactions in a multi-directional graph, thereby enabling contextual exploratory analysis. We demonstrate that our approach can detect common and specific interactions between cell types in mouse brain and human tumors, and that these interactions fit with expected outcomes. These interactions also include predictions made with molecular ligands integrating information from several types of genes necessary for ligand production and transport. Our implementation is general and can be appended to any transcriptome analysis pipeline to provide unbiased hypothesis generation regarding ligand to receptor interactions between cell populations or for network analysis in silico.Availability and implementationscConnect is open source and available as a Python package at https://github.com/JonETJakobsson/scConnect. scConnect is directly compatible with Scanpy scRNA-sequencing pipelines.

    • The time dependence of ice-nucleating particle (INP) activity is known to exist, yet for simplicity it is often omitted in atmospheric models as an approximation. Hitherto, only limited experimental work has been done to quantify this time dependency, for which published data are especially scarce regarding ambient aerosol samples and longer timescales. In this study, the time dependence of INP activity is quantified experimentally for six ambient environmental samples. The experimental approach includes a series of hybrid experiments with alternating constant cooling and isothermal experiments using a recently developed cold-stage setup called the Lund University Cold-Stage (LUCS). This approach of observing ambient aerosol samples provides the optimum realism for representing their time dependence in any model. Six ambient aerosol samples were collected at a station in rural Sweden representing aerosol conditions likely influenced by various types of INPs: marine, mineral dust, continental pristine, continental-polluted, combustion-related and rural continental aerosol. Active INP concentrations were seen to be augmented by about 40% to 100% (or 70% to 200%), depending on the sample, over 2h (or 10h). Mineral dust and rural continental samples displayed the most time dependence. This degree of time dependence observed was comparable to, but weaker than, that seen in previous published works. A general tendency was observed for the natural timescale of the freezing to dilate increasingly with time. The fractional freezing rate was observed to decline steadily with the time since the start of isothermal conditions following a power law. A representation of time dependence for incorporation into schemes of heterogeneous ice nucleation that currently omit it is proposed. Our measurements are inconsistent with the simplest purely stochastic model of INP activity, which assumes that the fractional freezing rate of all unfrozen drops is somehow constant and would eventually overpredict active INPs. In reality, the variability of efficiencies among INPs must be treated with any stochastic theory.
