    • This paper considers approximations of marginalization sums thatarise in Bayesian inference problems. Optimal approximations ofsuch marginalization sums, using a fixed number of terms, are analyzedfor a simple model. The model under study is motivated byrecent studies of linear regression problems with sparse parametervectors, and of the problem of discriminating signal-plus-noise samplesfrom noise-only samples. It is shown that for the model understudy, if only one term is retained in the marginalization sum, thenthis term should be the one with the largest a posteriori probability.By contrast, if more than one (but not all) terms are to be retained,then these should generally not be the ones corresponding tothe components with largest a posteriori probabilities.

    • This paper deals with the problem of discriminating samples that contain only noise from samples that contain a signal embedded in noise. The focus is on the case when the variance of the noise is unknown. We derive the optimal soft decision detector using a Bayesian approach. The complexity of this optimal detector grows exponentially with the number of observations and as a remedy, we propose a number of approximations to it. The problem under study is a fundamental one and it has applications in signal denoising, anomaly detection, and spectrum sensing for cognitive radio. We illustrate the results in the context of the latter.

  • Capacity Considerations for Uncoordinated Communication in Geographical Spectrum Holes

    • Cognitive radio is a new concept of reusing a licensed spectrum in an unlicensed manner. The motivation for cognitive radio is various measurements of spectrum utilization, that generally show unused resources in frequency, time and space. These "spectrum holes" could be exploited by cognitive radios. Some studies suggest that the spectrum is extremely underutilized, and that these spectrum holes could provide ten times the capacity of all existing wireless devices together. The spectrum could be reused either during time periods where the primary system is not active, or in geographical positions where the primary system is not operating. In this paper, we deal primarily with the concept of geographical reuse, in a frequency-planned primary network. We perform an analysis of the potential for communication in a geographical spectrum hole, and in particular the achievable sum-rate for a secondary network, to some order of magnitude. Simulation results show that a substantial sum-rate could be achieved if the secondary users communicate over small distances. For a small number of secondary links, the sum-rate increases linearly with the number of links. However, the spectrum hole gets saturated quite fast, due to interference caused by the secondary users. A spectrum hole may look large, but it disappears as soon as someone starts using it.

    • Aim: Hypertension is a common disease in the industrialized world and approximately 5% of all cases are secondary to kidney malfunction. We have recently shown that hydronephrosis due to partial unilateral ureteral obstruction (PUUO) causes salt-sensitive hypertension in rats. The mechanisms are still unclear, but appear to be intrarenal and primarily located to the diseased kidney. In the present study, we have developed a model for PUUO to study if hydronephrotic mice develop salt-sensitive hypertension. Methods: PUUO was created in 3-week-old mice (C57bl/6J). Blood pressure and heart rate were measured telemetrically in adult animals on normal and high salt diets. Metabolism cages were used to study the renal excretion of electrolytes and water. Plasma samples for renin analysis were collected and renal histological changes were evaluated. Results: All hydronephrotic animals developed salt-sensitive hypertension that correlated to the degree of hydronephrosis. In hydronephrotic animals, blood pressure increased from 114 ± 1 mmHg on normal salt diet to 120 ± 2 mmHg on high salt diet, compared with 103 ± 1 to 104 ± 1 in controls. Hydronephrotic animals showed increased diuresis and reduced ability to regulate electrolyte concentration. No differences in plasma renin concentration were found between the groups. The parenchymal weight and glomerular area of contralateral kidneys were significantly increased in the hydronephrotic animals. Histopathology of the hydronephrotic kidneys displayed areas with fibrosis, inflammation and glomerular changes. Conclusion: This study provides a model for PUUO in mice and demonstrates the presence of salt-sensitive hypertension and an impaired renal concentrating ability in mice which has not been described before.

  • Role of nitric oxide deficiency in the development of hypertension in hydronephrotic animals

    • Hydronephrotic animals develop renal injury and hypertension, which is associated with an abnormal tubuloglomerular feedback (TGF). The TGF sensitivity is coupled to nitric oxide (NO) in the macula densa. The involvement of reduced NO availability in the development of hypertension in hydronephrosis was investigated. Hydronephrosis was induced by ureteral obstruction in young rats. Blood pressure and renal excretion were measured in adulthood, under different sodium conditions, and before and after chronic administration of either N-G- nitro-L-arginine methyl ester (L-NAME) or L-arginine. Blood samples for ADMA, SDMA, and L-arginine analysis were taken and the renal tissue was used for histology and determination of NO synthase (NOS) proteins. TGF characteristics were determined by stop-flow pressure technique before and after administration of 7-nitroindazole (7-NI) or L-arginine. Hydronephrotic animals developed salt-sensitive hypertension, which was associated with pressure natriuresis and diuresis. The blood pressure response to L-NAME was attenuated and L-arginine supplementation decreased blood pressure in hydronephrotic animals, but not in the controls. Under control conditions, reactivity and sensitivity of the TGF response were greater in the hydronephrotic group. 7-NI administration increased TGF reactivity and sensitivity in control animals, whereas, in hydronephrotic animals, neuronal NOS (nNOS) inhibition had no effect. L-Arginine attenuated TGF response more in hydronephrotic kidneys than in controls. The hydronephrotic animals displayed various degrees of histopathological changes. ADMA and SDMA levels were higher and the renal expressions of nNOS and endothelial NOS proteins were lower in animals with hydronephrosis. Reduced NO availability in the diseased kidney in hydronephrosis, and subsequent resetting of the TGF mechanism, plays an important role in the development of hypertension.

    • Background: Hydronephrosis causes renal dysfunction and salt-sensitive hypertension, which is associated with NO-deficiency and abnormal tubuloglomerular feedback (TGF) response. We investigated the role of oxidative stress for salt-sensitivity and for hypertension in hydronephrosis. Methods: Hydronephrosis was induced in SOD1-transgenic (SOD1-tg), SOD1-deficient (SOD1-ko) and wild-type mice and in rats. In mice, telemetric measurements were performed during normal (0.7% NaCl) and high sodium (4% NaCl) diets and with chronic Tempol supplementation. 8-iso-prostaglandin-F2alpha (F2-IsoPs) and protein excretion profiles and histology were investigated. The acute effects of Tempol on blood pressure and TGF were studied in rats. Results: In hydronephrosis, wild-type mice developed salt-sensitive hypertension (114+/-1 to 120+/-2 mmHg) which was augmented in SOD1-ko (125+/-3 to 135+/-4 mmHg), but abolished in SOD1-tg (109+/-3 to 108+/-3 mmHg). SOD1-ko controls displayed salt-sensitive blood pressure (108+/-1 to 115+/-2 mmHg), which was not found in wild-types or SOD1-tg. Chronic Tempol treatment reduced blood pressure in SOD1-ko controls (-7 mmHg) and in hydronephrotic wild-types (-8 mmHg) and SOD1-ko mice (-16 mmHg), but had no effect on blood pressure in wild-type or SOD1-tg controls. SOD1-ko controls and hydronephrotic wild-type and SOD1-ko mice exhibited increased fluid excretion associated with increased F2-IsoPs and protein excretion. The renal histopathological changes found in hydronephrotic wild-types were augmented in SOD1-ko and diminished in SOD-tg mice. Tempol attenuated blood pressure and normalized TGF response in hydronephrosis (DeltaPSF: 15.2+/-1.2 to 9.1+/-0.6 mmHg, TP: 14.3+/-0.8 to 19.7+/-1.4 nl/min). Conclusion: Oxidative stress due to SOD1-deficiency causes salt-sensitivity and plays a pivotal role for the development of hypertension in hydronephrosis. Increased superoxide formation may enhance TGF response and thereby contribute to hypertension.

  • Uninephrectomy in Young Age or Chronic Salt Loading Causes Salt-Sensitive Hypertension in Adult Rats

    • The importance of nephron endowment and salt intake for the development of hypertension is under debate. The present study was designed to investigate whether reduced nephron number, after completion of nephrogenesis, or chronic salt loading causes renal injury and salt-sensitive hypertension in adulthood. Rats were operated at 3 weeks of age (after completed nephrogenesis) and then subjected to either normal or high-salt diets for 6 to 8 weeks. Four different experimental groups were used: sham-operated animals raised with normal-salt diet (controls) or high-salt diet (HS) and uninephrectomized animals raised with normal-salt diet (UNX) or high-salt diet (UNX+HS). In the adult animals, renal and cardiovascular functions were evaluated and blood pressure recorded telemetrically under different sodium conditions (normal, high, and low). Hypertension was present in UNX+HS (122±9 mm Hg), UNX (101±3 mm Hg), and HS (96± 1 mm Hg) groups on normal-salt diets compared with the controls (84±2 mm Hg), and the blood pressure was salt sensitive (high- versus normal-salt diet; 23±3, 9±2, 7±2, and 1±1 mm Hg, respectively). The hypertensive groups (UNX+HS, UNX, and HS) had increased diuresis and reduced ability to concentrate urine. The glomerular filtration rate (milliliters per minute) in anesthetized rats was reduced in the UNX+HS (2.36±0.30) and UNX animals (2.00±0.31) compared with both HS animals (3.55±0.45) and controls (3.01±0.35). Hypertensive groups displayed reduced plasma renin concentrations during high sodium conditions and hypertrophic kidneys and hearts with various degrees of histopathologic changes. In conclusion, at a young age after completed nephrogenesis, uninephrectomy or chronic salt loading causes renal and cardiovascular injury with salt-sensitive hypertension.

    • Recent meta-analyses of the methylenetetrahydrofolate reductase gene (MTHFR) have suggested association between two of its functional single gene polymorphisms (SNPs, C677T and A1298C) and schizophrenia. Studies have also suggested association between MTHFR C677T and A1298C variation and bipolar disorder. In a replication attempt the MTHFR C677T and A1298C SNPs were analyzed in three Scandinavian schizophrenia case-control samples. In addition, Norwegian patients with bipolar disorder were investigated. There were no statistically significant allele or genotype case-control differences. The present Scandinavian results do not verify previous associations between the putative functional MTHFR gene polymorphisms and schizophrenia or bipolar disorder. However, when combined with previous studies in meta-analyses there is still evidence for association between the MTHFR C677T polymorphism. and schizophrenia. Additional studies are warranted to shed further light on these relationships. (c) 2007 Wiley-Liss, Inc.

    • In this paper, we present a computationally efficient sliding window time updating of the Capon and amplitude and phase,estimation (APES) matched filterbank spectral estimators based on the time-variant displacement structure of the data covariance matrix. The presented algorithm forms a natural extension of the most computationally efficient algorithm to date, and offers a significant computational gain as compared to the computational complexity associated with the batch re-evaluation of the spectral estimates for each time-update. Furthermore, via simulations, the algorithm is found to be numerically superior to the time-updated spectral estimate formed from directly updating the data covariance matrix.

  • Early-Decision Decoding of LDPC Codes

    • Since their rediscovery in 1995, low-density parity-check (LDPC) codes have received wide-spread attention as practical capacity-approaching code candidates. It has been shown that the class of codes can perform arbitrarily close to the channel capacity, and LDPC codes are also used or suggested for a number of important current and future communication standards. However, the problem of implementing an energy-efficient decoder has not yet been solved. Whereas the decoding algorithm is computationally simple, withuncomplicated arithmetic operations and low accuracy requirements, the random structure and irregularity of a theoretically well-defined code does not easily allow efficient VLSI implementations. Thus the LDPC decoding algorithm can be said to be communication-bound rather than computation-bound.In this thesis, a modification to the sum-product decoding algorithm called early-decision decoding is suggested. The modification is based on the idea that the values of the bits in a block can be decided individually during decoding. As the sum-product decoding algorithm is a soft-decision decoder, a reliability can be defined for each bit. When the reliability of a bit is above a certain threshold, the bit can be removed from the rest of the decoding process, and thus the internal communication associated with the bit can be removed in subsequent iterations. However, with the early decision modification, an increased error probability is associated. Thus, bounds on the achievable performance as well as methods to detect graph inconsistencies resulting from erroneous decisions are presented. Also, a hybrid decoder achieving a negligible performance penalty compared to the sum-product decoder is presented. With the hybrid decoder, the internal communication is reduced with up to 40% for a rate-1/2 code with a length of 1152 bits, whereas increasing the rate allows significantly higher gains.The algorithms have been implemented in a Xilinx Virtex 5 FPGA, and the resulting slice utilization andenergy dissipation have been estimated. However, due to increased logic overhead of the early decision decoder, the slice utilization increases from 14.5% to 21.0%, whereas the logic energy dissipation reduction from 499 pJ to 291 pJ per iteration and bit is offset by the clock distribution power, increased from 141 pJ to 191 pJ per iteration and bit. Still, the early decision decoder shows a net 16% estimated decrease of energy dissipation.

    • There is a need for potent invertebrate systems for assessing the impacts of environmental contaminants on marine ecosystems. Balanus improvisus, a marine athropod, has a number of promising characteristics that make it a good candidate in such efforts. We have conducted sequencing of a cDNA library from the cyprid larval stage and identified several detoxification systems as well as novel B. improvisus specific genes. To investigate the toxicological gene expression response in this organism, we performed a short-term exposure experiment of the cyprid larvae to two different biofouling substances. From a natural population of B. improvisus, 300 - 1000 cyprids were treated for 23 hours with 390nM CuO or with two different concentrations (10nM or 10μM) of meditomidine. Protein expression changes were studied by 2D-PAGE analysis after DIGE labelling. For gene expression analysis by DNA miroarrays total RNA was extracted and used for cDNA and cRNA/aRNA templates. Roughly 2000 B. improvisus genes were studied represented by 3000 different probes on the arrays (each in duplicates). Candidate genes were confirmed by qPCR. A number of protein expression changes were detected on the 2D gels as a result of the different treatments. Interestingly, the response to the different treatments clearly formed distinct groups in principle component analysis. The DNA microarray analysis revealed several genes as toxicity indicators, e.g. various heat shock proteins, some proteases and factors involved in regulatory processes (transcription factors). Our data indicate that B. improvisus may serve as a tool to evaluate the impacts of marine pollution, and thus to fill the niche as an important invertebrate marine model organism for ecotoxicology and environmental genomics.

    • Game theory is a branch of mathematics aimed at the modeling and understanding of resource conflict problems. Essentially, the theory splits into two branches: noncooperative and cooperative game theory. The distinction between the two is whether or not the players in the game can make joint decisions regarding the choice of strategy. Noncooperative game theory is closely connected to minimax optimization and typically results in the study of various equilibria, most notably the Nash equilibrium. Cooperative game theory examines how strictly rational (selfish) actors can benefit from voluntary cooperation by reaching bargaining agreements. Another distinction is between static and dynamic game theory, where the latter can be viewed as a combination of game theory and optimal control. In general, the theory provides a structured approach to many important problems arising in signal processing and communications, notably resource allocation and robust transceiver optimization. Recent applications also occur in other emerging fields, such as cognitive radio, spectrum sharing, and in multihop-sensor and adhoc networks.

    • This paper studies the potential performance im- provement from cooperative diversity transmission in a cellular network. We consider a simpli ed coopera- tive relaying system which allows the mobile terminals to relay data packets from their partners. We obtain numerical results for the outage probability, taking into account log-distance path loss, spatially corre- lated shadowing and small-scale fading. A compari- son with conventional macrodiversity is performed for various scenarios of interest. The simulation results demonstrate the superiority of cooperative diversity transmission.

    • We consider soft MIMO detection for the case of block fading. That is, the transmitted codeword spans over several independent channel realizations and several instances of the detection problem must be solved for each such realization. We develop methods that adaptively allocate the computational resources to the detection problems of each channel realization, under a total per-codeword complexity constraint. Our main results are a formulation of the problem as a mathematical optimization problem and a greedy algorithm to approximate it in a computationally feasible fashion.

    • Different antenna placements in nonuniform linear arrays for the joint estimation of the range and direction-of-arrival (DOA) of one source are considered. It is shown that the Cramér-Rao bound (CRB) for the range and DOA estimates can be expressed as a function of the angles at which the source sees the antennas. This new formulation of the CRB easily reveals the geometrical properties that the array should have in order to minimise the CRB. Based on the new representation of the CRB, a result found in the literature (which was concerned with the special case of a number of antennas that is divisible by four) is generalised to include the case of an arbitrary number of antennas. Furthermore, it is shown that symmetric arrays are close to uniformly optimal for different DOAs when the range is much larger than the aperture of the arrays.

    • We design an optimum modulator for the Costa (dirty-paper) precoding problem under the constraint of a binary signaling alphabet, and assuming the interference symbols belong to a binary constellation. We evaluate the performance of our technique in terms of the mutual information between the channel input and output, and compare it to that of Tomlinson-Harashima precoding (THP) with optimized parameters. We show that our optimal modulator is always better than THP. In many relevant scenarios, the performance difference is significant.

  • On Coding of Scheduling Information in OFDM

    • Control signaling strategies for scheduling information in cellular OFDM systems are studied. A single-cell multiuser system model is formulated that provides system capacity estimates accounting for the signaling overhead. Different scheduling granularities are considered, including the one used in the specifications for the 3G Long Term Evolution (LTE). A greedy scheduling method is assumed, where each resource is assigned to the user for which it can support the highest number of bits. The simulation results indicate that the cost of control signaling does not outweigh the scheduling gain, when compared with a simple round-robin scheme that does not need signaling of scheduling information. Furthermore, in the studied scenario, joint coding and signaling of scheduling information over all selected users is found to be superior to separate coding and signaling for each user. The results also indicate that the scheduling granularity used for LTE provides better performance than the full granularity.

    • Pharmaceuticals are typically found in very low concentrations in the aquatic environment. Accordingly, environmental effects clearly assigned to residual drugs are consistent with high affinity interactions with conserved targets in affected wildlife species rather than with a general toxic effect. Thus, evolutionarily well-conserved targets in a given species are associated with an increased risk. In this study orthologs for 1318 human drug targets were predicted in 16 species of which several are relevant for ecotoxicity testing. The conservation of different functional categories of targets was also analyzed. Zebrafish had orthologs to 86% of the drug targets while only 61% were conserved in Daphnia and 35% in green alga. The predicted presence and absence of orthologs agrees well with published experimental data on the potential for specific drug target interaction in various species. Based on the conservation of targets we propose that aquatic environmental risk assessments for human drugs should always include comprehensive studies on aquatic vertebrates. Furthermore, individual targets, especially enzymes, are well conserved suggesting that tests on evolutionarily distant organisms would be highly relevant for certain drugs. We propose that the results can guide environmental risk assessments by improving the possibilities to identify species sensitive to certain types of pharmaceuticals or to other contaminants that act through well defined mechanisms of action. Moreover, we suggest that the results can be used to interpret the relevance of existing ecotoxicity data.

    • Patancheru, near Hyderabad, India, is a major production site for the global bulk drug market. Approximately 90 manufacturers send their wastewater to a common treatment plant in Patancheru. Extraordinary high levels of a wide range of pharmaceuticals have recently been demonstrated in the treated effluent. As little as 0.2% of this effluent can strongly reduce the growth rate of tadpoles, but the underlying mechanisms of toxicity are not known. To begin addressing how the effluent affects aquatic vertebrates, rainbow trout (Oncorhynchus mykiss) were exposed to 0.2% effluent for five days. Several physiological endpoints, together with effects on global hepatic gene expression patterns, were analyzed. The exposed fish showed both an induction of hepatic cytochrome P450 1A (CYP1A) gene expression, as well as enzyme activity. Clinical blood chemistry analyses revealed an increase in plasma phosphate levels, which in humans indicates impaired kidney function. Several oxidative stress-related genes were induced in the livers; however, no significant changes in antioxidant enzyme activities or in the hepatic glutathione levels were found. Furthermore, estrogen-regulated genes were slightly up-regulated following exposure, and moderate levels of estriol were detected in the effluent. The present study identifies changes in gene expression triggered by exposure to a high dilution of the effluent, supporting the hypothesis that these fish are responding to chemical exposure. The pattern of regulated genes may contribute to the identification of mechanisms of sub-lethal toxicity, as well as illuminate possible causative agents.

    • Background Vitellogenin is a well established biomarker for estrogenic exposure in fish. However, effects on gonadal differentiation at concentrations of estrogen not sufficient to give rise to a measurable vitellogenin response suggest that more sensitive biomarkers would be useful. Induction of zona pellucida genes may be more sensitive but their specificities are not as clear. The objective of this study was to find additional sensitive and robust candidate biomarkers of estrogenic exposure. Results Hepatic mRNA expression profiles were characterized in juvenile rainbow trout exposed to a measured concentration of 0.87 and 10 ng ethinylestradiol/L using a salmonid cDNA microarray. The higher concentration was used to guide the subsequent identification of generally more subtle responses at the low concentration not sufficient to induce vitellogenin. A meta-analysis was performed with data from the present study and three similar microarray studies using different fish species and platforms. Within the generated list of presumably robust responses, several well-known estrogen-regulated genes were identified. Two genes, confirmed by quantitative RT-PCR (qPCR), fulfilled both the criteria of high sensitivity and robustness; the induction of the genes encoding zona pellucida protein 3 and a nucleoside diphosphate kinase (nm23). Conclusion The cross-species, cross-platform meta-analysis correctly identified several robust responses. This adds confidence to our approach used for identifying candidate biomarkers. Specifically, we propose that analyses of an nm23 gene together with zona pellucida genes may increase the possibilities to detect an exposure to low levels of estrogenic compounds in fish.
