    • This article documents the addition of 411 microsatellite marker loci and 15 pairs of Single Nucleotide Polymorphism (SNP) sequencing primers to the Molecular Ecology Resources Database. Loci were developed for the following species: Acanthopagrus schlegeli, Anopheles lesteri, Aspergillus clavatus, Aspergillus flavus, Aspergillus fumigatus, Aspergillus oryzae, Aspergillus terreus, Branchiostoma japonicum, Branchiostoma belcheri, Colias behrii, Coryphopterus personatus, Cynogolssus semilaevis, Cynoglossus semilaevis, Dendrobium officinale, Dendrobium officinale, Dysoxylum malabaricum, Metrioptera roeselii, Myrmeciza exsul, Ochotona thibetana, Neosartorya fischeri, Nothofagus pumilio, Onychodactylus fischeri, Phoenicopterus roseus, Salvia officinalis L., Scylla paramamosain, Silene latifo, Sula sula, and Vulpes vulpes. These loci were cross-tested on the following species: Aspergillus giganteus, Colias pelidne, Colias interior, Colias meadii, Colias eurytheme, Coryphopterus lipernes, Coryphopterus glaucofrenum, Coryphopterus eidolon, Gnatholepis thompsoni, Elacatinus evelynae, Dendrobium loddigesii Dendrobium devonianum, Dysoxylum binectariferum, Nothofagus antarctica, Nothofagus dombeyii, Nothofagus nervosa, Nothofagus obliqua, Sula nebouxii, and Sula variegata. This article also documents the addition of 39 sequencing primer pairs and 15 allele specific primers or probes for Paralithodes camtschaticus.

  • Visible Light Driven Water Splitting in a Molecular Device with Unprecedentedly High Photocurrent Density

    • A molecular water oxidation catalyst (2) has been synthesized and immobilized together with a molecular photosensitizer (1) on nanostructured TiO2 particles on FTO conducting glass, forming a photoactive anode (TiO2(1+2)). By using the TiO2(1+2) as working electrode in a three-electrode photoelectrochemical cell (PEC), visible light driven water splitting has been successfully demonstrated in a phosphate buffer solution (pH 6.8), with oxygen and hydrogen bubbles evolved respectively from the working electrode and counter electrode. By applying 0.2 V external bias vs NHE, a high photocurrent density of more than 1.7 rnA.cm(-2) has been achieved. This value is higher than any PEC devices with molecular components reported in literature.

  • Combination of Diane-35 and Metformin to Treat Early Endometrial Carcinoma in PCOS Women with Insulin Resistance

    • Background: Young women with polycystic ovary syndrome (PCOS) have a high risk of developing endometrial carcinoma. There is a need for the development of new medical therapies that can reduce the need for surgical intervention so as to preserve the fertility of these patients. The aim of the study was to describe and discuss cases of PCOS and insulin resistance (IR) women with early endometrial carcinoma while being co-treated with Diane-35 and metformin. Methods: Five PCOS-IR women who were scheduled for diagnosis and therapy for early endometrial carcinoma were recruited. The hospital records and endometrial pathology reports were reviewed. All patients were co-treated with Diane-35 and metformin for 6 months to reverse the endometrial carcinoma and preserve their fertility. Before, during, and after treatment, endometrial biopsies and blood samples were obtained and oral glucose tolerance tests were performed. Endometrial pathology was evaluated. Body weight (BW), body mass index (BMI), follicle-stimulating hormone (FSH), luteinizing hormone (LH), total testosterone (TT), sex hormone-binding globulin (SHBG), free androgen index (FAI), insulin area under curve (IAUC), and homeostasis model assessment of insulin resistance (HOMA-IR) were determined. Results: Clinical stage 1a, low grade endometrial carcinoma was confirmed before treatment. After 6 months of co-treatment, all patients showed normal epithelia. No evidence of atypical hyperplasia or endometrial carcinoma was found. Co-treatment resulted in significant decreases in BW, BMI, TT, FAI, IAUC, and HOMA-IR in parallel with a significant increase in SHBG. There were no differences in the FSH and LH levels after co-treatment. Conclusions: Combined treatment with Diane-35 and metformin has the potential to revert the endometrial carcinoma into normal endometrial cells in PCOS-IR women. The cellular and molecular mechanisms behind this effect merit further investigation.

    • Abstract Polycystic ovary syndrome (PCOS) is a state of altered steroid hormone production and activity. Chronic estrogen exposure or lack of progesterone due to ovarian dysfunction can result in endometrial hyperplasia and carcinoma. A key contributor to our understanding of progesterone as a critical regulator for normal uterine function has been the elucidation of progesterone receptor (PR) expression, regulation, and signaling pathways. Several human studies indicate that PR-mediated signaling pathways in the nucleus are associated with progesterone resistance in women with PCOS. The aim of this review is to provide an overview of endometrial progesterone resistance in women with PCOS; to present the PR structure, its different isoforms, and their expression in the endometrium; to illustrate the possible regulation of PR and PR-mediated signaling in progesterone resistance in women with PCOS; and to discuss current clinical treatments for atypical endometrial hyperplasia and endometrial carcinoma in women with PCOS and accompanying progesterone resistance.

    • We applied the master equation method to investigate the anisotropic transport property of the 4,4'-bis((E)-2-(naphthalen-2-yl)vinyl)-1,1'-biphenyl molecular crystal based on first-principles calculation. It is found that the hole mobility has the largest value along the [100] direction, while electrons have the best transport property along the [010] direction. The anisotropic transport property was found to have close relationship with the charge transfer integral which is determined by the molecular stacking network in the crystals as well as the intermolecular frontier orbital overlap. In addition, the effect of the charge carrier density and the electronic field on the charge transport was also studied, and little effect was found except that the density is larger than 0.01 and the electronic field is increased to 1.0 x 106 V/cm. The kinetic Monte Carlo simulation method has also been used to study the anisotropic charge transport property, and consistent results were obtained as with the master equation method.

    • In this paper, we study an aspiration-induced migration model, in which each individual plays the prisoner’s dilemma game with those being within a circle of radius r centered on himself/herself. An individual will migrate to a randomly chosen place with the velocity v if his/her payoff is below the aspiration level. We report that cooperative behavior is favored when the aspiration level and interaction radius are moderate, and the migration velocity is slow.

  • Evaluation of terrestrial carbon cycle models for their response to climate variability and to CO2 trends

    • The purpose of this study was to evaluate 10 process-based terrestrial biosphere models that were used for the IPCC fifth Assessment Report. The simulated gross primary productivity (GPP) is compared with flux-tower-based estimates by Jung etal. [Journal of Geophysical Research 116 (2011) G00J07] (JU11). The net primary productivity (NPP) apparent sensitivity to climate variability and atmospheric CO2 trends is diagnosed from each model output, using statistical functions. The temperature sensitivity is compared against ecosystem field warming experiments results. The CO2 sensitivity of NPP is compared to the results from four Free-Air CO2 Enrichment (FACE) experiments. The simulated global net biome productivity (NBP) is compared with the residual land sink (RLS) of the global carbon budget from Friedlingstein etal. [Nature Geoscience 3 (2010) 811] (FR10). We found that models produce a higher GPP (133 +/- 15Pg Cyr-1) than JU11 (118 +/- 6Pg Cyr-1). In response to rising atmospheric CO2 concentration, modeled NPP increases on average by 16% (5-20%) per 100ppm, a slightly larger apparent sensitivity of NPP to CO2 than that measured at the FACE experiment locations (13% per 100ppm). Global NBP differs markedly among individual models, although the mean value of 2.0 +/- 0.8Pg Cyr-1 is remarkably close to the mean value of RLS (2.1 +/- 1.2 Pg Cyr-1). The interannual variability in modeled NBP is significantly correlated with that of RLS for the period 1980-2009. Both model-to-model and interannual variation in model GPP is larger than that in model NBP due to the strong coupling causing a positive correlation between ecosystem respiration and GPP in the model. The average linear regression slope of global NBP vs. temperature across the 10 models is -3.0 +/- 1.5Pg Cyr-1 degrees C-1, within the uncertainty of what derived from RLS (-3.9 +/- 1.1Pg Cyr-1 degrees C-1). However, 9 of 10 models overestimate the regression slope of NBP vs. precipitation, compared with the slope of the observed RLS vs. precipitation. With most models lacking processes that control GPP and NBP in addition to CO2 and climate, the agreement between modeled and observation-based GPP and NBP can be fortuitous. Carbon-nitrogen interactions (only separable in one model) significantly influence the simulated response of carbon cycle to temperature and atmospheric CO2 concentration, suggesting that nutrients limitations should be included in the next generation of terrestrial biosphere models.

    • Digital LLRF technology has been widely used in new generation particle accelerators. IF quadrature sampling is a common method for amplitude and phase detection. Many strategies, which obey the same rule of f(sample) = (M/N)f(IF) (M/N is a rational number), have been proposed to reduce the effects of spectrum aliasing. However, we found that M/N does not need to be a rational number according to Shannon's theorem. Therefore, we propose a new IQ detection method in this paper. This method is based on a special IIR filter which is derived from an RLC circuit. The unique characteristic of the method is that the value of f(IF) is independent of the value of, f(sample). We have set up an experimental platform to verify our method. A 122.88 MHz sampling clock is used to sample a 3 MHz IF signal. The DOS and PI control techniques are used to realize the closed-loop control. Results show that the stability of the system is within +/- 0.05% (peak to peak) for the amplitude, and with +/- 0.03 degrees (peak to peak) for the phase in 5 h. (C) 2012 Elsevier B.V.     • To gain insight into how genomic information is translated into cellular and developmental programs, the Drosophila model organism Encyclopedia of DNA Elements (modENCODE) project is comprehensively mapping transcripts, histone modifications, chromosomal proteins, transcription factors, replication proteins and intermediates, and nucleosome properties across a developmental time course and in multiple cell lines. We have generated more than 700 data sets and discovered protein-coding, noncoding, RNA regulatory, replication, and chromatin elements, more than tripling the annotated portion of the Drosophila genome. Correlated activity patterns of these elements reveal a functional regulatory network, which predicts putative new functions for genes, reveals stage- and tissue-specific regulators, and enables gene-expression prediction. Our results provide a foundation for directed experimental and computational studies in Drosophila and related species and also a model for systematic data integration toward comprehensive genomic and functional annotation.

    • Abstract Although a number of in vitro studies have demonstrated the antiproliferative, anti-invasive, and antimetastatic effects of metformin in multiple cancer cell types, its cellular and molecular mechanisms of anti-cancer action in the endometrium of women with polycystic ovary syndrome (PCOS) have not yet been fully elucidated. Organic cation transporters (OCTs) and multidrug and toxin extrusion proteins (MATEs) are known to be involved in metformin uptake and excretion in cells. In this article, we discuss the novel therapeutic possibilities for early-stage endometrial carcinoma (EC) in women with PCOS focusing on metformin, which might have a direct effect in the endometrium through the OCTs and MATEs. We then review the molecular mechanism(s) of the action of metformin in the endometrium and highlight possible mechanistic insights into the inhibition of cell proliferation and tumor growth and, ultimately, the reversal of early-stage EC into normal endometria in women with PCOS.

    • GLP-1 (glucagon-like peptide I) and its mimetic exendin-4 are used against Type 2 diabetes. C-peptide has also proven promising to enhance insulin action. Since insulin secretion in vivo can be rapidly tuned by changes in islet microcirculation, we evaluated the influence of GLP-1, exendin-4 and C-peptide on pancreatic IBF (islet blood flow), and dynamic changes in insulin secretion and glycaemia in the rat. Adult male Wistar rats were divided into four groups given intravenous saline, GLP-1, exendin-4 or C-peptide respectively and administered either saline or 30% glucose. Furthermore, we investigated the effect of intravenous infusion of different doses of exendin-4 into either the femoral vein or the portal vein on islet microcirculation. A non-radioactive microsphere technique was adopted to measure the regional blood flow. Both GLP-1 and exendin-4 prevented the glucose-induced PBF (pancreatic blood flow) redistribution into the islets. Infusion of exendin-4 into the portal vein did not alter pancreatic islet microcirculation, while infusion of exendin-4 into femoral vein significantly decreased basal IBF. C-peptide increased basal IBF and the proportion of IBF out of total PBF, but did not affect the islet microcirculation after glucose administration. GLP-1, exendin-4 and C-peptide stimulated insulin secretion and significantly decreased glycaemia. Blocking NO formation did not prevent the decreased IBF and post-load glycaemia evoked by exendin-4, but further decreased IBF and KBF (kidney blood flow) and increased basal glycaemia. Blocking the vagus nerve enhanced pancreatic IBF and further decreased post-load glycaemia and KBF and increased basal glycaemia. The vascular modulatory effect on pancreatic islet microcirculation described herein, with subsequent effects on in vivo insulin secretion and glycaemia, might be one of the mechanisms underlying the anti-diabetic actions of GLP-1 and its long acting mimetic exendin-4, as well as that of C-peptide.

    • Two new near-infrared chemodosimeters for cyanide anion based on 5,10-dihexyl-5,10-dihydrophenazine were designed and synthesized. With dicyano-vinyl groups as the recognition site and electron-withdrawing groups on both sides, probe 1 exhibited an intramolecular charge transfer (ICT) absorption band at 545 nm and emission band at 730 nm, respectively, and thus showed an ICT block process and realized an 'on-off" response after bilateral reaction with cyanide anions in CH3CN. Probe 2 utilized an unreactive formyl group instead of one of the two reactive dicyano-vinyl groups as the electron-withdrawing component. Due to the unilateral recognition process the ICT of probe 2 was redirected and lead to a remarkably colorimetric and ratiometric near-infrared (NIR) fluorescent response for cyanine. Both probes provided high sensitivity and selectivity with apparent response signals which can be observed by naked eyes, even in the copresence of various other interference anions. Optical spectroscopic techniques, NMR titration measurements, and density functional theory calculations were conducted to rationalize the sensing mechanisms of these two probes.

    • Two chemodosimeters PDMI and PMI for cyanide detection were designed and synthesized based onphenazinecyanine dyes with N-methyl indolium group as receptor unit. According to the specific reactivity of indolium C-N+ bond against cyanide anion, both of them featured high sensitivity with detectionlimit of 1.4 mu M and 200 nM, respectively, and high selectivity against other anions. The quenching effecton phenazine-cyanine fluorophore by strong intramolecular charge transfer (ICT) from phenazine donorto indolium receptor made both PDMI and PMI non-emissive at the original state. After addition ofcyanide, the ICT effect decreased and vanished leading to dramatic " off-on" fluorescence enhancement. PDMI which proceeded bilateral electrophilic reaction toward cyanide anion provided an emission signal at 580 nm in HEPES buffer with naked-eye detectable color change. Probe PMI utilized an unreactiveformyl group instead of one reactive N-methyl indolium group as the electron-withdrawing component. Due to the unilateral recognition process for cyanide the ICT orientation of PMI was redirectedthus exhibited fluorescence enhancement with maximum emission at 630 nm. Meanwhile, PMI wasapplied for monitoring intracellular cyanide in Hela cells and proved to achieve "off-on" fluorescentsignal confirmed by confocal laser scanning microscopic imaging.

    • A novel super-macroporous monolithic composite cryogel was prepared by embedding macroporous cellulose beads into poly(hydroxyethyl methacrylate) cryogel. The cellulose beads were fabricated by using a microchannel liquid-flow focusing and cryopolymerization method, while the composite cryogel was prepared by cryogenic radical polymerization of the hydroxyethyl methacrylate monomer with poly(ethylene glycol) diacrylate as cross-linker together with the cellulose beads. After graft polymerization with (vinylbenzyl)trimethylammonium chloride, the composite cryogel was applied to separate immunoglobulin-G and albumin from human serum. Immunoglobulin-G with a mean purity of 83.2% and albumin with a purity of 98% were obtained, indicating the composite cryogel as a promising chromatographic medium in bioseparation for the isolation of important bioactive proteins like immunoglobulins and albumins.

  • Oxidation and Corrosion of New MCrAlX Coatings : Modelling and Experiments

    • MCrAlY coatings (“M” for Ni and/or Co) are widely used for the protection of superalloy components operated at high temperatures such as in the hot sections of gas turbines. The exposure to high temperature can cause coating degradation due to oxidation or hot corrosion at the coating surface. Microstructures in the coating and the coating life are affected also by the diffusion of alloying elements through the coating-superalloy interface. This PhD project, by applying thermodynamic modelling and experimental tests, investigates the oxidation and hot corrosion behavior of new MCrAlX coatings, in which X, referring to minor elements, is used to highlight the functions of such elements.In order to understand and predict the coating degradation progress during thermal exposure, an oxidation-diffusion model has been established for MCrAlX coating-superalloy systems, which integrates the oxidation of aluminum at coating surface, diffusion of alloying elements, and the diffusion-blocking effect in the materials. The predicted chemical composition profile and microstructure agreed well with experimental results in a CoNiCrAlYSiTa-Inconel 792 system. The model was further applied in several coating-superalloy systems to study the influence of coating composition, superalloy composition and temperature on the evolution of microstructure in the coating and the coating life. The results have demonstrated the potential of the model in designing new durable MCrAlX coatings. In addition to the applications in coating-superalloy systems, the model was also adapted for studying the microstructural development in a superalloy in which internal oxidation and nitridation occurred in an oxidation process.The oxidation behavior of some HVOF MCrAlX coatings was studied by thermal exposure at different temperatures (900, 1000, 1100 °C). Different spinels formed above the alumina scale, depending on the oxidation temperature. The minor alloying elements, Ru and Ir, had no direct influence on the oxidation behavior but may affect the phase stability in the coating.MCrAlX coatings were also tested in 48-hour cycles at 900 °C in different hot corrosion environments containing sulphates and/or SO2. The results showed that the coating performance was dependent on coating quality, concentration of Al and Cr in the coating, and the hot corrosion condition. It was also found that the addition of SO2 in the environment may not necessarily be bad for hot corrosion resistance of some MCrAlY coatings.

  • Thermal and Mechanical Behaviors of High Temperature Coatings

    • With superior oxidation and corrosion resistance, metallic coatings (i.e. diffusion coatings and MCrAlX coatings) are widely used to protect hot components made of superalloys in turbine engines. Two issues are critically important for the coating at high temperatures: thermal property related to oxidation/corrosion behavior and microstructure stability, and mechanical properties (e.g. creep and fatigue). The aim of this project is to develop better understanding of the thermal and mechanical behaviors of metallic coatings on superalloys and to improve the accuracy of prediction of their lifetime by thermodynamic modeling. The present work includes an investigation on the oxidation behavior of MCrAlX coating with a new lifetimeprediction model and a study on the influence of diffusion coatings on creep and fatigue behaviors of the superalloy IN792.Experiments on isothermal and thermal cycling oxidation were designed to investigate the oxidation behavior of a HVOF CoNiCrAlYSi coating on superalloy IN792. It is found that the oxidation behaviors of the coating are related to its thermodynamic property. A diffusion model has been established using the homogenization models in the DICTRA software and taking into consideration of the influence of surface oxidation, coating-substrate interdiffusion and diffusion blocking effect caused by internal voids and oxides. The simulation results show an improved accuracy of lifetime prediction by introducing the diffusion blocking effect.Microstructural evolution during creep process at high temperatures was studied in different diffusion coatings (NiAl and PtAl). It is found that the inward diffusion of aluminum controls the thickening rate of the diffusion coatings. The developed coatings displayed two types of mechanical behavior - being easily plasticized or cracked - dependent on temperature and type of coating, and therefore could be considered as non-load carrying material during creep test. The influence of cracking of PtAl coating on the high-cycle fatigue (HCF) behavior of the IN792 was also investigated. The results show that precracking of the coating prior to the fatigue test has little influence on the fatigue limit of specimens with thin coating (50 μm) but lowers the fatigue limit of specimens with thick coating (70 μm). The through-coating crack has enough mobility to penetrate into the substrate and causes fatigue failure only when the driving force for crack propagation is increased above a critical value due to a higher applied stress or a larger crack length (thicker coating).
