    • This paper addresses a built-in-self-test (BiST) to characterize IP3 linearity of a RF receiver front-end. A two-tone stimulus is generated by a phase-lock loop (PLL) in GHz frequency range. The PLL is designed to keep the frequency difference between the two tones under control and in this way to avoid a possible injection-locking. One of the oscillation frequencies and the difference (beat) frequency can be externally controlled. According to the test requirements the phase noise and nonlinear distortion of the two-tone generator are considered as a merit for the VCO and analog adder design. A highly linear analog adder with output referred IP3 of more than +15 dBm is used to generate the RF stimulus. The two-tone power across 50 Ω receiver input impedance can be more than -25 dBm with very low intermodulation distortion of PIM3 = -75 dBc. The receiver performance is not affected significantly by the test set-up. Simulations for linearity and noise performance of the PLL designed in 65nm CMOS show sufficient potential for on-chip IP3 measurements in the GHz frequency range.

    • Recent research has highlighted that positive biodiversity-ecosystem functioning relationships hold for all groups of organisms, including microbes. Yet, we still lack understanding regarding the drivers of microbial diversity, in particular, whether diversity of microbial communities is a matter of local factors, or whether metacommunities are of similar importance to what is known from higher organisms. Here, we explore the driving forces behind spatial variability in lake phytoplankton diversity in Fennoscandia. While phytoplankton biovolume is best predicted by local phosphorus concentrations, phytoplankton diversity (measured as genus richness, G) only showed weak correlations with local concentrations of total phosphorus. By estimating spatial averages of total phosphorus concentrations on various scales from an independent, spatially representative lake survey, we found that close to 70 per cent of the variability in local phytoplankton diversity can be explained by regionally averaged phosphorus concentrations on a scale between 100 and 400 km. Thus, the data strongly indicate the existence of metacommunities on this scale. Furthermore, we show a strong dependency between lake productivity and spatial community turnover. Thus, regional productivity affects beta-diversity by controlling spatial community turnover, resulting in scale-dependent productivity-diversity relationships. As an illustration of the interaction between local and regional processes in shaping microbial diversity, our results offer both empirical support and a plausible mechanism for the existence of common scaling rules in both the macrobial and the microbial worlds. We argue that awareness of regional species pools in phytoplankton and other unicellular organisms may critically improve our understanding of ecosystems and their susceptibility to anthropogenic stressors.

    • Na+/H+ exchangers (NHEs) are ancient membrane-bound nanoma- chines that work to regulate intracellular pH, sodium levels and cell volume. NHE activities contribute to the control of the cell cycle, cell proliferation, cell migration and vesicle trafficking. NHE dysfunction has been linked to many diseases, and they are targets of pharma- ceutical drugs. Despite their fundamental importance to cell home- ostasis and human physiology, structural information for the mammalian NHEs was lacking. Here, we report the cryogenic elec- tron microscopy structure of NHE isoform 9 (SLC9A9) from Equus caballus at 3.2 Å resolution, an endosomal isoform highly expressed in the brain and associated with autism spectrum (ASD) and atten- tion deficit hyperactivity (ADHD) disorders. Despite low sequence identity, the NHE9 architecture and ion-binding site are remarkably most similar to distantly related bacterial Na+/H+ antiporters with 13 transmembrane segments. Collectively, we reveal the conserved architecture of the NHE ion-binding site, their elevator-like structural transitions, the functional implications of autism disease mutations and the role of phosphoinositide lipids to promote homodimerization that, together, have important physiological ramifications.

    • We present the results of an online, video-based experimental study investigating the impact of robot agency on perceptions of a socially assistive robot (SAR) shown supporting in-home care. We consider two key participant groups: care givers and care receivers. We did not find significant results regarding the impact of agency on overall participant perceptions of the SAR, but we did identify some differences in what these two participant groups might perceive as being best for themselves versus each other. Firstly, care givers perceived more potential benefit from the robot than care receivers did, challenging possible assumptions about who is set to gain most from deployment of these systems. Secondly, care receivers generally perceived the lower agency robot as being more beneficial for themselves, even as they ascribed the higher agency robot more potential to benefit care receivers.

  • Development and psychometric evaluation of the Thirst Distress Scale for patients with heart failure

    • Background:Patients with heart failure can experience thirst distress. However, there is no instrument to measure this in patients with heart failure. The aim of the present study was to develop the Thirst Distress Scale for patients with Heart Failure (TDS-HF) and to evaluate psychometric properties of the scale.Methods and results:The TDS-HF was developed to measure thirst distress in patients with heart failure. Face and content validity was confirmed using expert panels including patients and healthcare professionals. Data on the TDS-HF was collected from patients with heart failure at outpatient heart failure clinics and hospitals in Sweden, the Netherlands and Japan. Psychometric properties were evaluated using data from 256 heart failure patients (age 72±11 years). Concurrent validity of the scale was assessed using a thirst intensity visual analogue scale. Patients did not have any difficulties answering the questions, and time taken to answer the questions was about five minutes. Factor analysis of the scale showed one factor. After psychometric testing, one item was deleted. For the eight item TDS-HF, a single factor explained 61% of the variance and Cronbach’s alpha was 0.90. The eight item TDS-HF was significantly associated with the thirst intensity score (r=0.55, p<0.001). Regarding test-retest reliability, the intraclass correlation coefficient was 0.88, and the weighted kappa values ranged from 0.29–0.60.Conclusion:The eight-item TDS-HF is valid and reliable for measuring thirst distress in patients with heart failure.

    • Objectives: To investigate the effect of prolonged neutralisation of tumour necrosis factor alpha (TNFalpha) on the radiological course in rheumatoid arthritis (RA). To assess whether the radiological course can be predicted by clinical variables or biological markers of cartilage and synovium turnover and of endothelial activation. Patients and methods: Forty seven patients with active RA enrolled at our centre in monotherapy trials with adalimumab (D2E7), a fully human anti-TNFalpha monoclonal antibody, were studied for two years. Radiographs of hands and feet obtained at baseline and after one and two years were scored in chronological order by a single, blinded observer using the modified Sharp method. Radiological course was classified as stable or progressive using the smallest detectable difference as cut off point. The relation between radiological course and serum markers of cartilage and synovium turnover (metalloproteinases (MMP-1 and MMP-3), cartilage oligomeric matrix protein (COMP), human cartilage glycoprotein-39 (HC gp-39)), endothelial activation (soluble E-selectin and intercellular adhesion molecule (ICAM-1)), and integrated measures of disease activity were assessed using univariate and multivariate analysis. Results: Radiological evaluation was performed in 36 patients with paired sets of radiographs at baseline and two years. After two years a total of 15/36 (42%) presented no radiological progression. More patients with stable radiological course were still receiving anti-TNFalpha treatment after two years (13/15 (87%) v 11/21 (52%); p=0.03) and had lower baseline COMP and sICAM-1 levels (p=0.01 and 0.04, respectively) than those in the group with progressive disease. In a logistic regression model the combination of sustained TNF neutralisation and baseline COMP and sICAM-1 levels was predictive for radiological outcome (p=0.03). C reactive protein and disease activity score area under the curve were significantly correlated with changes in radiological scores after two years (r=0.40 and 0.37, p<0.05). Long term TNFalpha neutralisation decreased the levels of COMP, sICAM, MMPs, and HC gp-39, but not sE-selectin. Conclusion: The results suggest that long term monotherapy with anti-TNFalpha has a positive effect on radiological outcome and modulates cartilage and synovium turnover as measured by biological markers. Baseline serum sICAM-1 levels and COMP levels may be helpful to identify patients with progressive or non-progressive radiological outcome.

    • Föreliggande utredning över ädellövskogarna i Kungälvs kommun utgör den åttonde sammanställningen i en planerad utgivning för Göteborgs och Bohus län. De ädla lövskogarna utgör i södra och mellersta Bohusläns kusttrakter ett naturligt växtgeografiskt inslag i landskapsbilden. Lövskogarna utgör genom sin biologiska mångfald en av de rikaste och mest skyddsvärda biotoperna i länet. Sedan 1984 gäller en särskild ädellövskogslag (SFS 1984:119) vars syfte är att för framtiden bevara landets ädellövskogar. Detta skall ske genom ett aktivt, produktionsinriktat lövskogsbruk. Detta innebär bl a att befintlig ädellövskog endast får föryngras med ädla lövträd om inte särskilda skäl talar emot detta. Rådgivning och tillsyn sker genom skogsvårdsstyrelsens ordinarie skogliga verksamhet. Frågor rörande överföring av ädellövskog till annat trädslag prövas av länsstyrelsen. Följande inventering och utvärdering är tänkt att tjäna som underlag dels vid myndigheternas prövning och rådgivning enligt gällande skogslagstiftning dels i samband med den naturvårdsplanering som bedrivs i statlig och kommunal regi.

    • Background: Pathological aggregation of tar DNA-binding protein 43 (TDP-43) in the brain is the primary cause of many cases of frontotemporal lobar degeneration (FTLD), amyotrophic lateral sclerosis (ALS) and limbic-predominant age-related TDP-43 encephalopathy (LATE). It is therefore imperative to establish empirical staging systems to characterize and distinguish stereotypical patterns and commonplace deviations of different TDP-43 proteinopathies. Method: We use ordinal ratings of TDP-43 burden from 19 brain regions to perform data-driven disease progression modeling (SuStaIn) to find the most likely trajectories for FTLD-TDP (n = 108), ALS (n = 137) and LATE (n = 283) from the CNDR Brain Bank at the University of Pennsylvania. Subtype number was defined using cross-validated information criterion. Each individual was assigned a subtype and stage. Multivariate OLS models tested differences between subtypes. Stages were compared to age and existing staging schemes. Cross-validated logistic regression was used for 3-way classification using SuStaIn information only. Result: SuStaIn provided data-driven staging of TDP-43 proteinopathies complementing previously described human-defined staging schema, further providing additional detail (Fig1A-C; Fig3A-C). SuStaIn also identified two distinct subtypes within FTLD-TDP and a further two within ALS (Fig1D). FTLD-TDP subtypes differed in TDP-43 type and Alzheimer’s disease pathology (Table1); ALS subtypes were differentiated by age (Table 2) and by antemortem clinical characteristics. No subtypes were observed for the LATE group. Progression along data-driven stages was positively associated with age in LATE individuals, but negatively associated with age in individuals with FTLD-TDP (Fig2). Using only regional TDP-43 severity, our data driven model could distinguish individuals diagnosed with ALS, FTD or LATE with a cross-validated balanced precision of 0.93 and balanced recall of 0.92, and these metrics improved to 0.95 and 0.96 when combined with a logistic regression model (Fig3). Very little stage overlap was found between FTLD-TDP and LATE, but stages that did overlap showed subtly different patterns (Fig4). Conclusion: We provide an empirical pathological staging system for ALS, FTLD-TDP and LATE, which is sufficient for staging and accurate classification. We demonstrate that there is substantial heterogeneity amongst ALS and FTLD-TDP progression patterns, whilst LATE exhibits a homogeneous progression pattern.
