    • Plant cells exposed to photo-inhibitory conditions respond by accumulation of the early light-induced proteins (Elips) with a potential photoprotective function. Here we studied the expression of Elip in various pea cultivars grown under agricultural or climate-chamber conditions. We demonstrated that the expression of Elip in all cultivars was developmentally regulated and its level decreased during flowering and post-flowering periods. Surprisingly, significant amounts of Elip transcripts, but not proteins, accumulated in senescing leaves already under low light conditions and the exposure to light stress resulted in a 10-times higher induction of Elip transcripts. Furthermore, the expression pattern of Elip transcript and protein significantly differed under field and growth-chamber conditions. First, the expression level of Elip was much higher in field-grown than in chamber-grown cultivars. Second, substantial amounts of Elip transcripts and protein were detected during the night in field-grown plants in contrast to chamber-grown cultivars due to a synergistic effect of light stress occurring during the day and low temperature present during the following night. The expression of the PsbS protein related to Elips and involved in the photoprotection of the photosystem II was relatively constant under all conditions tested.

  • Inhibitory effects of N-acetylcysteine on scavenger receptor class A expression in human macrophages.

    • OBJECTIVE: The formation of foam cells from monocyte-derived macrophages involves the uptake of modified lipoproteins by scavenger receptors. Antioxidants inhibit lipoprotein oxidation and may also modulate gene expression. We investigated the effect of the antioxidant N-acetylcysteine on the expression of the class A scavenger receptor (SR-A) types I and II in human macrophages. DESIGN: Monocytes and macrophages from healthy blood donors and plaque-derived macrophages from patients undergoing carotid endarterectomy were used for experiments. SR-A mRNA was analysed with quantitative and semiquantitative reverse transcription-polymerase chain reaction, and ligand binding and uptake were assessed with 125I-labelled acetylated low-density lipoprotein (LDL). RESULTS: Incubation of monocytes and monocyte-derived macrophages with N-acetylcysteine decreased both SR-A I and II mRNA expression. N-Acetylcysteine also reduced SR-A mRNA in lesion-derived cells. Binding and uptake of 125I-acetylated LDL was decreased after brief incubation with N-acetylcysteine. After longer periods of incubation with N-acetylcysteine we observed an increased degradation of lipoproteins. CONCLUSIONS: Our results imply that N-acetylcysteine leads to a decrease in SR-A mRNA and initially also to an attenuated uptake of modified lipoproteins. This adds more to the knowledge about the cellular actions of this drug.

    • Many dinoflagellate species form dormant resting cysts as a part of their life cycle, and in some freshwater species, hatching of these cysts can be delayed by the presence of water-borne signals from grazing zooplankton. Some marine dinoflagellates can form temporary cysts, which may function to resist unfavourable short-term environmental conditions. We investigated whether the marine dinoflagellate Alexandrium ostenfeldii is able to induce an increased resistance to the parasitic flagellate Parvilucifera infectans by forming temporary cysts. We performed several laboratory experiments where dinoflagellates were exposed either to direct contact with parasites or to filtered water from cultures of parasite-infected conspecifics (parasite-derived signals). Infection by P. infectans is lethal to motile A. ostenfeldii cells, but temporary cysts were more resistant to parasite infection. Furthermore, A. ostenfeldii induced a shift in life-history stage (from motile cells to temporary cysts) when exposed to parasite-derived water-borne signals. The response was relaxed within a couple of hours, indicating that A. ostenfeldii may use this behaviour as a short-term escape mechanism to avoid parasite infection. The results suggest that intraspecies chemical communication evoked by biotic interactions can be an important mechanism controlling life-history shifts in marine dinoflagellates, which may have implications for the development of toxic algal blooms.
