  • Novel TCF21high pericyte subpopulation promotes colorectal cancer metastasis by remodelling perivascular matrix

    • Haematogenous dissemination is a prevalent route of colorectal cancer (CRC) metastasis. However, as the gatekeeper of vessels, the role of tumour pericytes (TPCs) in haematogenous metastasis remains largely unknown. Here, we aimed to investigate the heterogeneity of TPCs and their effects on CRC metastasis.DesignTPCs were isolated from patients with CRC with or without liver metastases and analysed by single-cell RNA sequencing (scRNA-seq). Clinical CRC specimens were collected to analyse the association between the molecular profiling of TPCs and CRC metastasis. RNA-sequencing, chromatin immunoprecipitation-sequencing and bisulfite-sequencing were performed to investigate the TCF21-regulated genes and mechanisms underlying integrin α5 onTCF21DNA hypermethylation. Pericyte-conditionalTcf21-knockout mice were constructed to investigate the effects of TCF21 in TPCs on CRC metastasis. Masson staining, atomic force microscopy, second-harmonic generation and two-photon fluorescence microscopy were employed to observe perivascular extracellular matrix (ECM) remodelling.ResultsThirteen TPC subpopulations were identified by scRNA-seq. A novel subset of TCF21highTPCs, termed ‘matrix–pericytes’, was associated with liver metastasis in patients with CRC. TCF21 in TPCs increased perivascular ECM stiffness, collagen rearrangement and basement membrane degradation, establishing a perivascular metastatic microenvironment to instigate colorectal cancer liver metastasis (CRCLM).Tcf21depletion in TPCs mitigated perivascular ECM remodelling and CRCLM, whereas the coinjection of TCF21highTPCs and CRC cells markedly promoted CRCLM. Mechanistically, loss of integrin α5 inhibited the FAK/PI3K/AKT/DNMT1 axis to impairTCF21DNA hypermethylation in TCF21highTPCs.ConclusionThis study uncovers a previously unidentified role of TPCs in haematogenous metastasis and provides a potential diagnostic marker and therapeutic target for CRC metastasis.

  • Search for inelastic scattering of WIMP dark matter in XENON1T

    • We report the results of a search for the inelastic scattering of weakly interacting massive particles (WIMPs) in the XENON1T dark matter experiment. Scattering off Xe-129 is the most sensitive probe of inelastic WIMP interactions, with a signature of a 39.6 keV deexcitation photon detected simultaneously with the nuclear recoil. Using an exposure of 0.83 tonne-years, we find no evidence of inelastic WIMP scattering with a significance of more than 2 sigma. A profile-likelihood ratio analysis is used to set upper limits on the cross section of WIMP-nucleus interactions. We exclude new parameter space for WIMPs heavier than 100 GeV/c(2), with the strongest upper limit of 3.3 x 10(-39) cm(2) for 130 GeV/c(2) WIMPs at 90% confidence level.

    • In this paper, which is of programmatic rather than quantitative nature, we aim to further delineate and sharpen the future potential of the LISA mission in the area of fundamental physics. Given the very broad range of topics that might be relevant to LISA,we present here a sample of what we view as particularly promising fundamental physics directions. We organize these directions through a "science-first" approach that allows us to classify how LISA data can inform theoretical physics in a variety of areas. For each of these theoretical physics classes, we identify the sources that are currently expected to provide the principal contribution to our knowledge, and the areas that need further development. The classification presented here should not be thought of as cast in stone, but rather as a fluid framework that is amenable to change with the flow of new insights in theoretical physics.

  • Study of excited Ξ baryons with the P¯ ANDA detector

    • The study of baryon excitation spectra provides insight into the inner structure of baryons. So far, most of the world-wide efforts have been directed towards N∗ and Δ spectroscopy. Nevertheless, the study of the double and triple strange baryon spectrum provides independent information to the N∗ and Δ spectra. The future antiproton experiment P¯ANDA will provide direct access to final states containing a Ξ¯ Ξ pair, for which production cross sections up to μb are expected in p¯p reactions. With a luminosity of L= 10 31 cm- 2 s- 1 in the first phase of the experiment, the expected cross sections correspond to a production rate of ∼106events/day. With a nearly 4 π detector acceptance, P¯ANDA will thus be a hyperon factory. In this study, reactions of the type p¯p → Ξ¯ +Ξ∗ - as well as p¯p → Ξ¯ ∗ +Ξ- with various decay modes are investigated. For the exclusive reconstruction of the signal events a full decay tree fit is used, resulting in reconstruction efficiencies between 3 and 5%. This allows high statistics data to be collected within a few weeks of data taking.

  • The potential of Λ and Ξ- studies with PANDA at FAIR

    • The antiproton experiment PANDA at FAIR is designed to bring hadron physics to a new level in terms of scope, precision and accuracy. In this work, its unique capability for studies of hyperons is outlined. We discuss ground-state hyperons as diagnostic tools to study non-perturbative aspects of the strong interaction, and fundamental symmetries. New simulation studies have been carried out for two benchmark hyperon-antihyperon production channels: p¯ p→ Λ¯ Λ and p¯ p→ Ξ¯ +Ξ-. The results, presented in detail in this paper, show that hyperon-antihyperon pairs from these reactions can be exclusively reconstructed with high efficiency and very low background contamination. In addition, the polarisation and spin correlations have been studied, exploiting the weak, self-analysing decay of hyperons and antihyperons. Two independent approaches to the finite efficiency have been applied and evaluated: one standard multidimensional efficiency correction approach, and one efficiency independent approach. The applicability of the latter was thoroughly evaluated for all channels, beam momenta and observables. The standard method yields good results in all cases, and shows that spin observables can be studied with high precision and accuracy already in the first phase of data taking with PANDA.

  • Examining the generalizability of research findings from archival data

    • This initiative examined systematically the extent to which a large set of archival research findings generalizes across contexts. We repeated the key analyses for 29 original strategic management effects in the same context (direct reproduction) as well as in 52 novel time periods and geographies; 45% of the reproductions returned results matching the original reports together with 55% of tests in different spans of years and 40% of tests in novel geographies. Some original findings were associated with multiple new tests. Reproducibility was the best predictor of generalizability-for the findings that proved directly reproducible, 84% emerged in other available time periods and 57% emerged in other geographies. Overall, only limited empirical evidence emerged for context sensitivity. In a forecasting survey, independent scientists were able to anticipate which effects would find support in tests in new samples.

  • ENSO modulates wildfire activity in China

    • China is a key region for understanding fire activity and the drivers of its variability under strict fire suppression policies. Here, we present a detailed fire occurrence dataset for China, the Wildfire Atlas of China (WFAC; 2005-2018), based on continuous monitoring from multiple satellites and calibrated against field observations. We find that wildfires across China mostly occur in the winter season from January to April and those fire occurrences generally show a decreasing trend after reaching a peak in 2007. Most wildfires (84%) occur in subtropical China, with two distinct clusters in its southwestern and southeastern parts. In southeastern China, wildfires are mainly promoted by low precipitation and high diurnal temperature ranges, the combination of which dries out plant tissue and fuel. In southwestern China, wildfires are mainly promoted by warm conditions that enhance evaporation from litter and dormant plant tissues. We further find a fire occurrence dipole between southwestern and southeastern China that is modulated by the El Nino-Southern Oscillation (ENSO). Fire activity in China and its associations with climate are not well quantified at a local scale. Here, the authors present a detailed fire occurrence dataset for China and find a dipole fire pattern between southwestern and southeastern China that is modulated by the El Nino-Southern Oscillation (ENSO).

  • Insights into the accuracy of social scientists' forecasts of societal change

    • How well can social scientists predict societal change, and what processes underlie their predictions? To answer these questions, we ran two forecasting tournaments testing the accuracy of predictions of societal change in domains commonly studied in the social sciences: ideological preferences, political polarization, life satisfaction, sentiment on social media, and gender-career and racial bias. After we provided them with historical trend data on the relevant domain, social scientists submitted pre-registered monthly forecasts for a year (Tournament 1; N = 86 teams and 359 forecasts), with an opportunity to update forecasts on the basis of new data six months later (Tournament 2; N = 120 teams and 546 forecasts). Benchmarking forecasting accuracy revealed that social scientists' forecasts were on average no more accurate than those of simple statistical models (historical means, random walks or linear regressions) or the aggregate forecasts of a sample from the general public (N = 802). However, scientists were more accurate if they had scientific expertise in a prediction domain, were interdisciplinary, used simpler models and based predictions on prior data. How accurate are social scientists in predicting societal change, and what processes underlie their predictions? Grossmann et al. report the findings of two forecasting tournaments. Social scientists' forecasts were on average no more accurate than those of simple statistical models.

  • Evolution of spin excitations from bulk to monolayer FeSe

    • In ultrathin films of FeSe grown on SrTiO (FeSe/STO), the superconducting transition temperature T is increased by almost an order of magnitude, raising questions on the pairing mechanism. As in other superconductors, antiferromagnetic spin fluctuations have been proposed to mediate SC making it essential to study the evolution of the spin dynamics of FeSe from the bulk to the ultrathin limit. Here, we investigate the spin excitations in bulk and monolayer FeSe/STO using resonant inelastic x-ray scattering (RIXS) and quantum Monte Carlo (QMC) calculations. Despite the absence of long-range magnetic order, bulk FeSe displays dispersive magnetic excitations reminiscent of other Fe-pnictides. Conversely, the spin excitations in FeSe/STO are gapped, dispersionless, and significantly hardened relative to its bulk counterpart. By comparing our RIXS results with simulations of a bilayer Hubbard model, we connect the evolution of the spin excitations to the Fermiology of the two systems revealing a remarkable reconfiguration of spin excitations in FeSe/STO, essential to understand the role of spin fluctuations in the pairing mechanism.

  • Metaheuristic Optimization Algorithms Hybridized With Artificial Intelligence Model for Soil Temperature Prediction : Novel Model

    • An enhanced hybrid articial intelligence model was developed for soil temperature (ST) prediction. Among several soil characteristics, soil temperature is one of the essential elements impacting the biological, physical and chemical processes of the terrestrial ecosystem. Reliable ST prediction is signicant for multiple geo-science and agricultural applications. The proposed model is a hybridization of adaptive neuro-fuzzy inference system with optimization methods using mutation Salp Swarm Algorithm and Grasshopper Optimization Algorithm (ANFIS-mSG). Daily weather and soil temperature data for nine years (1 of January 2010 - 31 of December 2018) from ve meteorological stations (i.e., Baker, Beach, Cando, Crary and Fingal) in North Dakota, USA, were used for modeling. For validation, the proposed ANFIS-mSG model was compared with seven models, including classical ANFIS, hybridized ANFIS model with grasshopper optimization algorithm (ANFIS-GOA), salp swarm algorithm (ANFIS-SSA), grey wolf optimizer (ANFIS-GWO), particle swarm optimization (ANFIS-PSO), genetic algorithm (ANFIS-GA),and Dragon y Algorithm (ANFIS-DA). The ST prediction was conducted based on maximum, mean and minimum air temperature (AT). The modeling results evidenced the capability of optimization algorithms for building ANFIS models for simulating soil temperature. Based on the statistical evaluation; for instance, the root mean square error (RMSE) was reduced by 73%, 74.4%, 71.2%, 76.7% and 80.7% for Baker, Beach, Cando, Crary and Fingal meteorological stations, respectively, throughout the testing phase when ANFIS-mSG was used over the standalone ANFIS models. In conclusion, the ANFIS-mSG model was demonstrated as an effective and simple hybrid articial intelligence model for predicting soil temperature based on univariate air temperature scenario.

    • The acidification of coastal waters is distinguished from the open ocean because of much stronger synergistic effects between anthropogenic forcing and local biogeochemical processes. However, ocean acidification research is still rather limited in polar coastal oceans. Here, we present a 17-year (2002-2019) observational data set in the Chukchi Sea to determine the long-term changes in pH and aragonite saturation state (omega(arag)). We found that pH and omega(arag) declined in different water masses with average rates of -0.0047 +/- 0.0026 years(-1) and -0.017 +/- 0.009 years(-1), respectively, and are similar to 2-3 times faster than those solely due to increasing atmospheric CO2. We attributed the rapid acidification to the increased dissolved inorganic carbon owing to a combination of ice melt-induced increased atmospheric CO2 invasion and subsurface remineralization induced by a stronger surface biological production as a result of the increased inflow of the nutrient-rich Pacific water.

In the future, everything in the physical world would be replicated in the digital space through digital twin technology. As a cutting-edge technology, digital twin has received a lot of attention. However, digital twin is far from realizing their potential, which is a complex system and long-drawn process. Researchers must model all the different parts of the objects or systems. Varied types of data needed to be collected and merged. Many researchers and participators in engineering are not clear which technologies and tools should be used. 5-dimension digital twin model provides reference guidance for understanding and implementing digital twin. From the perspective of 5-dimension digital twin model, this paper tries to investigate and summarize the frequently-used enabling technologies and tools for digital twin to provide technologies and tools references for the applications of digital twin in the future.

  • Genetic insights into resting heart rate and its role in cardiovascular disease

    • The genetics and clinical consequences of resting heart rate (RHR) remain incompletely understood. Here, the authors discover new genetic variants associated with RHR and find that higher genetically predicted RHR decreases risk of atrial fibrillation and ischemic stroke. Resting heart rate is associated with cardiovascular diseases and mortality in observational and Mendelian randomization studies. The aims of this study are to extend the number of resting heart rate associated genetic variants and to obtain further insights in resting heart rate biology and its clinical consequences. A genome-wide meta-analysis of 100 studies in up to 835,465 individuals reveals 493 independent genetic variants in 352 loci, including 68 genetic variants outside previously identified resting heart rate associated loci. We prioritize 670 genes and in silico annotations point to their enrichment in cardiomyocytes and provide insights in their ECG signature. Two-sample Mendelian randomization analyses indicate that higher genetically predicted resting heart rate increases risk of dilated cardiomyopathy, but decreases risk of developing atrial fibrillation, ischemic stroke, and cardio-embolic stroke. We do not find evidence for a linear or non-linear genetic association between resting heart rate and all-cause mortality in contrast to our previous Mendelian randomization study. Systematic alteration of key differences between the current and previous Mendelian randomization study indicates that the most likely cause of the discrepancy between these studies arises from false positive findings in previous one-sample MR analyses caused by weak-instrument bias at lower P-value thresholds. The results extend our understanding of resting heart rate biology and give additional insights in its role in cardiovascular disease development.

    • BackgroundGastric cancer is more common in men than in women, but underlying reasons have not been completely understood. This study aimed to assess patterns of the sex difference in the incidence of gastric cancer in the United States.MethodsUsing data from 13 cancer registries in the Surveillance, Epidemiology, and End Results Program, we analyzed the age-specific sex difference in the incidence of gastric cancer by ethnicity, anatomic site and histological type in the United States during 1992–2014. We assessed the temporal trends in the sex differences in the incidence of gastric cancer during the study period.ResultsThe male-to-female incidence ratio of cardia cancer increased with age until peaking at ages 55–69 years and decreased thereafter, while the ratio for non-cardia gastric cancer increased with age before ages < 60 years and remained stable onwards. The age-specific patterns in the sex difference of gastric cancer incidence varied between intestinal and diffuse histological types. The sex difference in the incidence of cardia cancer remained relatively stable except for that the absolute difference between the sexes in whites decreased on average by 0.8% per year from 1992 to 2014. The absolute incidence difference between the sexes in non-cardia gastric cancer decreased over time in whites, blacks, and Asian and Pacific islanders by approximately 4% per year. The male-to-female incidence ratio of non-cardia gastric cancer decreased over time in whites and blacks, but remained relatively stable in Asian and Pacific islanders.ConclusionsBoth extrinsic and intrinsic factors may have contributed to the sex difference in gastric cancer. Sex hormones may play a role in the development of cardia cancer and intestinal type of gastric cancer.

    • Evaporation, one of the fundamental components of the hydrology cycle, is differently influenced by various meteorological variables in different climatic regions. The accurate prediction of evaporation is essential for multiple water resources engineering applications, particularly in developing countries like Iraq where the meteorological stations are not sustained and operated appropriately for in situ estimations. This is where advanced methodologies such as machine learning (ML) models can make valuable contributions. In this research, evaporation is predicted at two different meteorological stations located in arid and semi-arid regions of Iraq. Four different ML models for the prediction of evaporation – the classification and regression tree (CART), the cascade correlation neural network (CCNNs), gene expression programming (GEP), and the support vector machine (SVM) – were developed and constructed using various input combinations of meteorological variables. The results reveal that the best predictions are achieved by incorporating sunshine hours, wind speed, relative humidity, rainfall, and the minimum, mean, and maximum temperatures. The SVM was found to show the best performance with wind speed, rainfall, and relative humidity as inputs at Station I (R2 = .92), and with all variables as inputs at Station II (R2 = .97). All the ML models performed well in predicting evaporation at the investigated locations.

    • The low-lying states of Eu-146 populated in the decay of the 9(+) isomer were studied. The gamma-ray intensities were reanalyzed employing germanium detectors, and the lifetimes of the 6(1)(-) and 6(2)(-) states were measured using the mirror symmetric centroid difference (MSCD) method with fast-timing LaBr3(Ce) scintillator detectors. The B(M1) values of the 6(1)(-) -> 5(1)(-) and 6(2)(-) -> 5(1)(-) transitions were deduced, and all observed states were interpreted as members of the pi d(5/2)(-1)nu f(7/2) and pi g(7/2)(-1)nu f(7/2) multiplets. In particular, the 5(1)(-) level is shown to be dominated by the pi d(5/2)(-1)nu f(7/2) configuration, solving the discrepancy in its configuration assignment proposed in previous works. These experimental results were compared with the shell model calculations using several different effective interactions. The systematics of low-lying structure in the N = 83 isotones Pr-142, Pm-144, and Eu-146 was established.
