  • The Herschel-Heterodyne Instrument for the Far-Infrared (HIFI)

    • Aims: This paper describes the Heterodyne Instrument for the Far-Infrared (HIFI) that was launched onboard ESA's Herschel Space Observatory in May 2009. Methods: The instrument is a set of 7 heterodyne receivers that are electronically tuneable, covering 480-1250 GHz with SIS mixers and the 1410-1910 GHz range with hot electron bolometer (HEB) mixers. The local oscillator (LO) subsystem comprises a Ka-band synthesizer followed by 14 chains of frequency multipliers and 2 chains for each frequency band. A pair of auto-correlators and a pair of acousto-optical spectrometers process the two IF signals from the dual-polarization, single-pixel front-ends to provide instantaneous frequency coverage of 2 × 4 GHz, with a set of resolutions (125 kHz to 1 MHz) that are better than 0.1 km s-1. Results: After a successful qualification and a pre-launch TB/TV test program, the flight instrument is now in-orbit and completed successfully the commissioning and performance verification phase. The in-orbit performance of the receivers matches the pre-launch sensitivities. We also report on the in-orbit performance of the receivers and some first results of HIFI's operations. Herschel is an ESA space observatory with science instruments provided by European-led Principal Investigator consortia and with important participation from NASA.

  • Haslea ostrearia-like Diatoms: Biodiversity out of the Blue

    • Diatoms are usually referred to as golden-brown microalgae, due to the colour of their plastids and to their pigment composition, mainly carotenoids (fucoxanthin, diadinoxanthin, diatoxanthin), which mask chlorophylls a and c. The species Haslea ostrearia Gaillon/Bory (Simonsen) appears unique because of its extraplastidial bluish colour, a consequence of the presence of a water-soluble blue pigment at cell apices, marennine. When released in seawater, marenbine can be fixed on gills of oysters and other bivalves, which turn green. This greening phenomenon is economically exploited in Southwestern France, as it gives an added value to oysters. For decades, this singularity ascribed a worldwide distribution to H. ostrearia, first as Vibrio ostrearius, then Navicula ostrearia, last as H. ostrearia, when the genus Haslea was proposed by R. Simonsen (1974). Indeed, this 'birthmark' (presence of blue apices) made H. ostrearia easily recognisable without further scrutiny and identification of the microalga as well as its presence easily deduced from the greening of bivalves. Consequently, the widely admitted cosmopolitan character of H. ostrearia has only been questioned recently, following the discovery in 2008, of a new species of blue diatom in the Black Sea, Haslea karadagensis. The biodiversity of blue diatoms suddenly increased with the finding of other blue species in the Mediterranean Sea, the Canary Islands, etc., the taxonomic characterization of which is in progress. This review thus focuses on the unsuspected biodiversity of blue diatoms within the genus Haslea. Methods for species determination (morphometrics, chemotaxonomy, genomics), as well as a new species, are presented and discussed.

  • Hydro-economic modelling of cost-effective transboundary water quality management in the Baltic Sea

    • Helcom's recently revised Baltic Sea Action Plan (BSAP) aims to reduce eutrophication in the, Baltic Sea by reducing nutrient loads from all discharging drainage basins. The BALTCOST costminimisation, model, featuring abatement cost and effect functions which utilise spatial data down to, 10×10km2 grid cell resolution, is used to identify a cost-effective distribution of nutrient abatement, measures between drainage basins whilst attempting to achieve specified nutrient load reductions for, separate Baltic Sea regions. Results indicate that the 2013 BSAP load reduction targets for nitrogen and phosphorus can be achieved in most Baltic Sea regions, except for phosphorus into the Baltic Proper, and the Gulf of Finland. The estimated minimum total cost of delivering these reductions is 4.17 billion, Euros annually, with substantial differences in the cost burden between countries and measures.Results are strongly influenced by differences in nutrient retention between drainage basins. Detailed, retention modelling and high spatial resolution source data are major novel features of this research.

  • Dissociation of Bone Resorption and Bone Formation in Adult Mice with a Non-Functional V-ATPase in Osteoclasts Leads to Increased Bone Strength

    • Osteopetrosis caused by defective acid secretion by the osteoclast, is characterized by defective bone resorption, increased osteoclast numbers, while bone formation is normal or increased. In contrast the bones are of poor quality, despite this uncoupling of formation from resorption. To shed light on the effect of uncoupling in adult mice with respect to bone strength, we transplanted irradiated three-month old normal mice with hematopoietic stem cells from control or oc/oc mice, which have defective acid secretion, and followed them for 12 to 28 weeks. Engraftment levels were assessed by flow cytometry of peripheral blood. Serum samples were collected every six weeks for measurement of bone turnover markers. At termination bones were collected for mu CT and mechanical testing. An engraftment level of 98% was obtained. From week 6 until termination bone resorption was significantly reduced, while the osteoclast number was increased when comparing oc/oc to controls. Bone formation was elevated at week 6, normalized at week 12, and reduced onwards. mu CT and mechanical analyses of femurs and vertebrae showed increased bone volume and bone strength of cortical and trabecular bone. In conclusion, these data show that attenuation of acid secretion in adult mice leads to uncoupling and improves bone strength.

  • Inheritance of Mitochondrial DNA in the Pennate Diatom Haslea ostrearia (Naviculaceae) during Auxosporulation Suggests a Uniparental Transmission

    • We present the first study examining mtDNA transmission in diatoms, using sexual progeny of the pennate species Has lea ostrearia (Naviculaceae). A fragment of the cytochrome oxidase subunit I gene (cox1) with 7 nucleic substitutions between parental clones was used as a parental tracer in 16 F1 clones obtained from two pairs of mating crosses. Each cross involved a parental clone isolated from France (Bay of Bourgneuf) and Sweden (Kattegat Bay). We determined that all progeny possessed only one cox1 parental haplotype. These results suggest that the mitochondrial DNA transmission in H. ostrearia is uniparental. Implications and new topics of investigation are discussed. (C) 2013 Elsevier GmbH. All rights reserved.

  • Time-resolved investigation of nanometer scale deformations induced by a high flux x-ray beam

    • We present results of a time-resolved pump-probe experiment where a Si sample was exposed to an intense 15 keV beam and its surface monitored by measuring the wavefront deformation of a reflected optical laser probe beam. By reconstructing and back propagating the wavefront, the deformed surface can be retrieved for each time step. The dynamics of the heat bump, build-up and relaxation, is followed with a spatial resolution in the nanometer range. The results are interpreted taking into account results of finite element method simulations. Due to its robustness and simplicity this method should find further developments at new x-ray light sources (FEL) or be used to gain understanding on thermo-dynamical behavior of highly excited materials.   • Picosecond time-resolved x-ray refectivity of a laser-heated amorphous carbon film

    • We demonstrate thin film x-ray reflectivity measurements with picosecond time resolution. Amorphous carbon films with a thickness of 46 nm were excited with laser pulses characterized by 100 fs duration, a wavelength of 800 nm, and a fluence of 70 mJ/cm(2). The laser-induced stress caused a rapid expansion of the thin film followed by a relaxation of the film thickness as heat diffused into the silicon substrate. We were able to measure changes in film thickness as small as 0.2 nm. The relaxation dynamics are consistent with a model which accounts for carrier-enhanced substrate heat diffusivity.     • Thioredoxin and thioredoxin reductase can regulate cell metabolism through redox regulation of disulfide bridges or through removal of H(2)O(2). These two enzymatic functions are combined in NADPH-dependent thioredoxin reductase C (NTRC), which contains an N-terminal thioredoxin reductase domain fused with a C-terminal thioredoxin domain. Rice NTRC exists in different oligomeric states, depending on the absence or presence of its NADPH cofactor. It has been suggested that the different oligomeric states may have diverse activity. Thus, the redox status of the chloroplast could influence the oligomeric state of NTRC and thereby its activity. We have characterized the oligomeric states of NTRC from barley (Hordeum vulgare L.). This also includes a structural model of the tetrameric NTRC derived from cryo-electron microscopy and single-particle reconstruction. We conclude that the tetrameric NTRC is a dimeric arrangement of two NTRC homodimers. Unlike that of rice NTRC, the quaternary structure of barley NTRC complexes is unaffected by addition of NADPH. The activity of NTRC was tested with two different enzyme assays. The N-terminal part of NTRC was tested in a thioredoxin reductase assay. A peroxide sensitive Mg-protoporphyrin IX monomethyl ester (MPE) cyclase enzyme system of the chlorophyll biosynthetic pathway was used to test the catalytic ability of both the N- and C-terminal parts of NTRC. The different oligomeric assembly states do not exhibit significantly different activities. Thus, it appears that the activities are independent of the oligomeric state of barley NTRC.

  • Light-Induced Structural Changes in a Photosynthetic Reaction Center Caught by Laue Diffraction

    • Photosynthetic reaction centers convert the energy content of light into a transmembrane potential difference and so provide the major pathway for energy input into the biosphere. We applied time-resolved Laue diffraction to study light-induced conformational changes in the photosynthetic reaction center complex of Blastochloris viridis. The side chain of TyrL162, which lies adjacent to the special pair of bacteriochlorophyll molecules that are photooxidized in the primary light conversion event of photosynthesis, was observed to move 1.3 angstroms closer to the special pair after photoactivation. Free energy calculations suggest that this movement results from the deprotonation of this conserved tyrosine residue and provides a mechanism for stabilizing the primary charge separation reactions of photosynthesis.
