  • DIII-D research advancing the physics basis for optimizing the tokamak approach to fusion energy
  • 2022
  • Ingår i: Nuclear Fusion. - : IOP Publishing. - 0029-5515 .- 1741-4326. ; 62:4
  • Tidskriftsartikel (refereegranskat)abstract
    • DIII-D physics research addresses critical challenges for the operation of ITER and the next generation of fusion energy devices. This is done through a focus on innovations to provide solutions for high performance long pulse operation, coupled with fundamental plasma physics understanding and model validation, to drive scenario development by integrating high performance core and boundary plasmas. Substantial increases in off-axis current drive efficiency from an innovative top launch system for EC power, and in pressure broadening for Alfven eigenmode control from a co-/counter-I p steerable off-axis neutral beam, all improve the prospects for optimization of future long pulse/steady state high performance tokamak operation. Fundamental studies into the modes that drive the evolution of the pedestal pressure profile and electron vs ion heat flux validate predictive models of pedestal recovery after ELMs. Understanding the physics mechanisms of ELM control and density pumpout by 3D magnetic perturbation fields leads to confident predictions for ITER and future devices. Validated modeling of high-Z shattered pellet injection for disruption mitigation, runaway electron dissipation, and techniques for disruption prediction and avoidance including machine learning, give confidence in handling disruptivity for future devices. For the non-nuclear phase of ITER, two actuators are identified to lower the L-H threshold power in hydrogen plasmas. With this physics understanding and suite of capabilities, a high poloidal beta optimized-core scenario with an internal transport barrier that projects nearly to Q = 10 in ITER at ∼8 MA was coupled to a detached divertor, and a near super H-mode optimized-pedestal scenario with co-I p beam injection was coupled to a radiative divertor. The hybrid core scenario was achieved directly, without the need for anomalous current diffusion, using off-axis current drive actuators. Also, a controller to assess proximity to stability limits and regulate β N in the ITER baseline scenario, based on plasma response to probing 3D fields, was demonstrated. Finally, innovative tokamak operation using a negative triangularity shape showed many attractive features for future pilot plant operation.

  • The Eighth Visual Object Tracking VOT2020 Challenge Results
  • 2020
  • Ingår i: Computer Vision. - Cham : Springer International Publishing. - 9783030682378 ; , s. 547-601
  • Konferensbidrag (refereegranskat)abstract
    • The Visual Object Tracking challenge VOT2020 is the eighth annual tracker benchmarking activity organized by the VOT initiative. Results of 58 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The VOT2020 challenge was composed of five sub-challenges focusing on different tracking domains: (i) VOT-ST2020 challenge focused on short-term tracking in RGB, (ii) VOT-RT2020 challenge focused on “real-time” short-term tracking in RGB, (iii) VOT-LT2020 focused on long-term tracking namely coping with target disappearance and reappearance, (iv) VOT-RGBT2020 challenge focused on short-term tracking in RGB and thermal imagery and (v) VOT-RGBD2020 challenge focused on long-term tracking in RGB and depth imagery. Only the VOT-ST2020 datasets were refreshed. A significant novelty is introduction of a new VOT short-term tracking evaluation methodology, and introduction of segmentation ground truth in the VOT-ST2020 challenge – bounding boxes will no longer be used in the VOT-ST challenges. A new VOT Python toolkit that implements all these novelites was introduced. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website (http://votchallenge.net ).

  • Examining the generalizability of research findings from archival data
  • 2022
  • Ingår i: Proceedings of the National Academy of Sciences of the United States of America. - : Proceedings of the National Academy of Sciences. - 0027-8424 .- 1091-6490. ; 119:30
  • Tidskriftsartikel (refereegranskat)abstract
    • This initiative examined systematically the extent to which a large set of archival research findings generalizes across contexts. We repeated the key analyses for 29 original strategic management effects in the same context (direct reproduction) as well as in 52 novel time periods and geographies; 45% of the reproductions returned results matching the original reports together with 55% of tests in different spans of years and 40% of tests in novel geographies. Some original findings were associated with multiple new tests. Reproducibility was the best predictor of generalizability-for the findings that proved directly reproducible, 84% emerged in other available time periods and 57% emerged in other geographies. Overall, only limited empirical evidence emerged for context sensitivity. In a forecasting survey, independent scientists were able to anticipate which effects would find support in tests in new samples.

  • Vertical Sandwich Gate-All-Around Field-Effect Transistors with Self-Aligned High-k Metal Gates and Small Effective-Gate-Length Variation
  • 2020
  • Ingår i: IEEE Electron Device Letters. - : Institute of Electrical and Electronics Engineers Inc.. - 0741-3106 .- 1558-0563. ; 41:1, s. 8-11
  • Tidskriftsartikel (refereegranskat)abstract
    • A new type of vertical nanowire (NW)/nanosheet (NS) field-effect transistors (FETs), termed vertical sandwich gate-all-around (GAA) FETs (VSAFETs), is presented in this work. Moreover, an integration flow that is compatible with processes used in the mainstream industry is proposed for the VSAFETs. Si/SiGe epitaxy, isotropic quasi-atomic-layer etching (qALE), and gate replacement were used to fabricate pVSAFETs for the first time. Vertical GAA FETs with self-aligned high-k metal gates and a small effective-gate-length variation were obtained. Isotropic qALE, including Si-selective etching of SiGe, was developed to control the diameter/thickness of the NW/NS channels. NWs with a diameter of 10 nm and NSs with a thickness of 20 nm were successfully fabricated, and good device characteristics were obtained. Finally, the device performance was investigated and is discussed in this work. © 2019 IEEE.

  • Cell transcriptomic atlas of the non-human primate Macaca fascicularis
  • 2022
  • Ingår i: Nature. - : Springer Nature. - 0028-0836 .- 1476-4687. ; 604:7907, s. 723-731
  • Tidskriftsartikel (refereegranskat)abstract
    • Studying tissue composition and function in non-human primates (NHPs) is crucial to understand the nature of our own species. Here we present a large-scale cell transcriptomic atlas that encompasses over 1 million cells from 45 tissues of the adult NHP Macaca fascicularis. This dataset provides a vast annotated resource to study a species phylogenetically close to humans. To demonstrate the utility of the atlas, we have reconstructed the cell–cell interaction networks that drive Wnt signalling across the body, mapped the distribution of receptors and co-receptors for viruses causing human infectious diseases, and intersected our data with human genetic disease orthologues to establish potential clinical associations. Our M. fascicularis cell atlas constitutes an essential reference for future studies in humans and NHPs.

  • Triboelectric backgrounds to radio-based polar ultra-high energy neutrino (UHEN) experiments
  • 2023
  • Ingår i: Astroparticle physics. - : Elsevier. - 0927-6505 .- 1873-2852. ; 145
  • Tidskriftsartikel (refereegranskat)abstract
    • In the hopes of observing the highest-energy neutrinos (E> 1 EeV) populating the Universe, both past (RICE, AURA, ANITA) and current (RNO-G, ARIANNA, ARA and TAROGE-M) polar-sited experiments exploit the impulsive radio emission produced by neutrino interactions. In such experiments, rare single event candidates must be unambiguously identified above backgrounds. Background rejection strategies to date primarily target thermal noise fluctuations and also impulsive radio-frequency signals of anthropogenic origin. In this paper, we consider the possibility that 'fake' neutrino signals may also be generated naturally via the `triboelectric effect' This broadly describes any process in which force applied at a boundary layer results in displacement of surface charge, leading to the production of an electrostatic potential difference AV. Wind blowing over granular surfaces such as snow can induce such a potential difference, with subsequent coronal discharge. Discharges over timescales as short as nanoseconds can then lead to radio-frequency emissions at characteristic MHz-GHz frequencies. Using data from various past (RICE, AURA, SATRA, ANITA) and current (RNO G, ARIANNA and ARA) neutrino experiments, we find evidence for such backgrounds, which are generally characterized by: (a) a threshold wind velocity which likely depends on the experimental trigger criteria and layout; for the experiments considered herein, this value is typically O(10 m/s), (b) frequency spectra generally shifted to the low-end of the frequency regime to which current radio experiments are typically sensitive (100-200 MHz), (c) for the strongest background signals, an apparent preference for discharges from above-surface structures, although the presence of more isotropic, lower amplitude triboelectric discharges cannot be excluded.

  • Physiological responses to salinity change and diel-cycling hypoxia in gills of Hong Kong oyster Crassostrea hongkongensis
  • 2023
  • Ingår i: Aquaculture. - : Elsevier BV. - 0044-8486. ; 570
  • Tidskriftsartikel (refereegranskat)abstract
    • Global climate change is a frequent cause of salinity fluctuation in seawater, especially in aquaculture sites. Moreover, anthropologic activities often cause seawater eutrophication with the consequence that hypoxia ap-pears often during nighttime. The Hong Kong oyster Crassostrea hongkongensis, as a species that inhabits estuarine and coastal waters, is faced with such challenges. In this study, oyster physiological changes were considered to be closely related to hypoxia and salinity changes. Physiological indices were examined in Hong Kong oysters by employing six treatments to shed light into the effects of diel-cycling hypoxia (periodical hypoxia) and salinity change. Three salinities (10%o, low salinity; 25%o, normal salinity; and 35%o, high salinity) and two types of dissolved oxygen (normoxia, 6 mg/L throughout the day) and periodical hypoxic condition (6 mg/L at daytime for 12 h and 2 mg/L at nighttime for 12 h) were set. After 14-and 28-day exposures, gill tissues were sampled to detect changes in gill ATP production, mitochondrial membrane potential (MMP), mitochondrial reactive oxygen species production (ROS), and gill respiratory metabolic enzymes. Results indicated that periodical hypoxia and salinity change led to increased hexokinase (HK) and pyruvate kinase (PK) (p < 0.05). By contrast, they had no significant effect on mitochondrial number (MN). Adenosine-triphosphate (ATP) production only increased in the early exposure. In addition, low salinity with periodical hypoxia resulted in decreased MMP, lactate dehy-drogenase (LDH), and succinate dehydrogenase (SDH, p < 0.05). On the contrary, periodical hypoxia with high salinity led to increases in ATP and ROS and decreases in SDH, MMP, and LDH (p < 0.05). These results revealed that when diel-cycling hypoxia occurs with salinity change, the gill metabolism of Hong Kong oysters are gradually dominated by glycolysis while aerobic respiration decreases. Moreover, gill functions could be affected although energy accumulation exists during early exposure. Therefore, long-term exposure to periodical hypoxia with salinity change poses risk to the health and growth of Hong Kong oysters, impairing oyster aquaculture and coastal ecosystem health.

  • An optimal charging scheduling model and algorithm for electric buses
  • 2023
  • Ingår i: Applied Energy. - : Elsevier Ltd. - 0306-2619 .- 1872-9118. ; 332
  • Tidskriftsartikel (refereegranskat)abstract
    • Electrification poses a promising low-carbon or even zero-carbon transportation solution, serving as a strategic approach to reducing carbon emissions and promoting carbon neutrality in the transportation sector. Along the transportation electrification pathway, the goal of carbon neutrality can be further accelerated with an increasing amount of electricity being generated from renewable energies. The past decade observed the rapid development of battery technologies and deployment of electricity infrastructure worldwide, fostering transportation electrification to expand from railways to light and then heavy vehicles on roadways. In China, a massive number of electric buses have been employed and operated in dozens of metropolises. An important daily operations issue with these urban electric buses is how to coordinate their charging activities in a cost-effective manner, considering various physical, financial, institutional, and managerial constraints. This paper addresses a general charging scheduling problem for an electric bus fleet operated across multiple bus lines and charging depots and terminals, aiming at finding an optimal set of charging location and time decisions given the available charging windows. The charging windows for each bus are predetermined in terms of its layovers at depots and terminals and each of them is discretized into a number of charging slots with the same time duration. A mixed linear integer programming model with binary charging slot choice and continuous state-of-charge (SOC) variables is constructed for minimizing the total charging cost of the bus fleet subject to individual electricity consumption rates, electricity charging rates, time-based charging windows, battery SOC bounds, time-of-use (TOU) charging tariffs, and station-specific electricity load capacities. A Lagrangian relaxation framework is employed to decouple the joint charging schedule of a bus fleet into a number of independent single-bus charging schedules, which can be efficiently addressed by a bi-criterion dynamic programming algorithm. A real-world regional electric bus fleet of 122 buses in Shanghai, China is selected for validating the effectiveness and practicability of the proposed charging scheduling model and algorithm. The optimization results numerically reveal the impacts of TOU tariffs, station load capacities, charging infrastructure configurations, and battery capacities on the bus system performance as well as individual recharging behaviors, and justify the superior solution efficiency of our algorithm against a state-of-the-art commercial solver.

  • A multimodal cell census and atlas of the mammalian primary motor cortex
  • 2021
  • Ingår i: Nature. - : Springer Science and Business Media LLC. - 1476-4687 .- 0028-0836. ; 598:7879, s. 86-102
  • Tidskriftsartikel (refereegranskat)abstract
    • Here we report the generation of a multimodal cell census and atlas of the mammalian primary motor cortex as the initial product of the BRAIN Initiative Cell Census Network (BICCN). This was achieved by coordinated large-scale analyses of single-cell transcriptomes, chromatin accessibility, DNA methylomes, spatially resolved single-cell transcriptomes, morphological and electrophysiological properties and cellular resolution input–output mapping, integrated through cross-modal computational analysis. Our results advance the collective knowledge and understanding of brain cell-type organization1–5. First, our study reveals a unified molecular genetic landscape of cortical cell types that integrates their transcriptome, open chromatin and DNA methylation maps. Second, cross-species analysis achieves a consensus taxonomy of transcriptomic types and their hierarchical organization that is conserved from mouse to marmoset and human. Third, in situ single-cell transcriptomics provides a spatially resolved cell-type atlas of the motor cortex. Fourth, cross-modal analysis provides compelling evidence for the transcriptomic, epigenomic and gene regulatory basis of neuronal phenotypes such as their physiological and anatomical properties, demonstrating the biological validity and genomic underpinning of neuron types. We further present an extensive genetic toolset for targeting glutamatergic neuron types towards linking their molecular and developmental identity to their circuit function. Together, our results establish a unifying and mechanistic framework of neuronal cell-type organization that integrates multi-layered molecular genetic and spatial information with multi-faceted phenotypic properties.

  • Improving the genome and proteome annotations of the marine model diatom Thalassiosira pseudonana using a proteogenomics strategy
  • 2023
  • Ingår i: Marine Life Science & Technology. - : Springer Science and Business Media LLC. - 2096-6490 .- 2662-1746. ; 5:1, s. 102-115
  • Tidskriftsartikel (refereegranskat)abstract
    • Diatoms are unicellular eukaryotic phytoplankton that account for approximately 20% of global carbon fixation and 40% of marine primary productivity; thus, they are essential for global carbon biogeochemical cycling and climate. The availability of ten diatom genome sequences has facilitated evolutionary, biological and ecological research over the past decade; however, a complimentary map of the diatom proteome with direct measurements of proteins and peptides is still lacking. Here, we present a proteome map of the model marine diatom Thalassiosira pseudonana using high-resolution mass spectrometry combined with a proteogenomic strategy. In-depth proteomic profiling of three different growth phases and three nutrient-deficient samples identified 9526 proteins, accounting for similar to 81% of the predicted protein-coding genes. Proteogenomic analysis identified 1235 novel genes, 975 revised genes, 104 splice variants and 234 single amino acid variants. Furthermore, our quantitative proteomic analysis experimentally demonstrated that a considerable number of novel genes were differentially translated under different nutrient conditions. These findings substantially improve the genome annotation of T. pseudonana and provide insights into new biological functions of diatoms. This relatively comprehensive diatom proteome catalog will complement available diatom genome and transcriptome data to advance biological and ecological research of marine diatoms.

  • Non-invasive early detection of cancer four years before conventional diagnosis using a blood test
  • 2020
  • Ingår i: Nature communications. - : Springer Science and Business Media LLC. - 2041-1723. ; 11:1, s. 3475-
  • Tidskriftsartikel (refereegranskat)abstract
    • Early detection has the potential to reduce cancer mortality, but an effective screening test must demonstrate asymptomatic cancer detection years before conventional diagnosis in a longitudinal study. In the Taizhou Longitudinal Study (TZL), 123,115 healthy subjects provided plasma samples for long-term storage and were then monitored for cancer occurrence. Here we report the preliminary results of PanSeer, a noninvasive blood test based on circulating tumor DNA methylation, on TZL plasma samples from 605 asymptomatic individuals, 191 of whom were later diagnosed with stomach, esophageal, colorectal, lung or liver cancer within four years of blood draw. We also assay plasma samples from an additional 223 cancer patients, plus 200 primary tumor and normal tissues. We show that PanSeer detects five common types of cancer in 88% (95% CI: 80–93%) of post-diagnosis patients with a specificity of 96% (95% CI: 93–98%), We also demonstrate that PanSeer detects cancer in 95% (95% CI: 89–98%) of asymptomatic individuals who were later diagnosed, though future longitudinal studies are required to confirm this result. These results demonstrate that cancer can be non-invasively detected up to four years before current standard of care.
