    • BCR::ABL1-independent pathways contribute to primary resistance to tyrosine kinase inhibitor (TKI) treatment in chronic myeloid leukemia (CML) and play a role in leukemic stem cell persistence. Here, we perform ex vivo drug screening of CML CD34+ + leukemic stem/progenitor cells using 100 single drugs and TKIdrug combinations and identify sensitivities to Wee1, MDM2, and BCL2 inhibitors. These agents effectively inhibit primitive CD34+CD38- + CD38- CML cells and demonstrate potent synergies when combined with TKIs. Flow-cytometry-based drug screening identifies mepacrine to induce differentiation of CD34+CD38- + CD38- cells. We employ genome-wide CRISPR-Cas9 screening for six drugs, and mediator complex, apoptosis, and erythroid-lineage-related genes are identified as key resistance hits for TKIs, whereas the Wee1 inhibitor AZD1775 and mepacrine exhibit distinct resistance profiles. KCTD5, a consistent TKI-resistance-conferring gene, is found to mediate TKI-induced BCR::ABL1 ubiquitination. In summary, we delineate potential mechanisms for primary TKI resistance and non-BCR::ABL1-targeting drugs, offering insights for optimizing CML treatment.

    • We investigated the mechanical properties, thermal behavior, physical state, and colloidal structure of a model gel network formulated with various ratios of sodium dodecyl sulfate polyacrylamide (SDS) and cetyl/stearyl fatty alcohol (FA) mixture in constant amounts of water (89%). The metastable gel, formed by slowly cooling stirred samples from temperatures above the chain melting temperature (Tm) of the long chain FA, has a lamellar structure with a periodic inter bilayer spacing of approximately 30nm. The bilayers remain homogeneous with SDS being immobilized in the FA matrix upon chain freezing. The chain length mismatch leads to an increased structural disorder among the alkyl chains upon SDS incorporation among the FA. As a result, the elastic modulus decreased with increasing SDS content. Conclusions were based on a large number of experiments involving differential scanning calorimetry, rheology, 1H NMR spectroscopy, and small angle x-ray scattering. Results from this work have uncovered the physical nature of these networks and gave insight into the role of anionic surfactants on the assembly, physical state, and mechanical properties of gel networks.

    • Groups of spiritual practitioners (P) (n = 38), long-term residents in a center offering spiritual courses (R) (n = 8), and matched control groups from a general, population sample for P (CP) (n = 45) and for R (CR) (n = 14) were compared. P had high levels of education and tended to be in a committed relationship and belong to the Danish National Church, whereas R had a lower level of education and were unlikely to be in a committed relationship or belong to the Church, suggesting social marginality. All groups completed the Brief Symptom Inventory-53 (BSI-53), a measure of psychological distress. P did not differ from the control groups in the Global Severity scale (GSI) or the other subscales of the BSI-53 except for scoring lower on the phobic anxiety subscale. In contrast, R scored higher in the GSI and most BSI-53 subscales than P and the control groups, and published norms for Danish and US populations. P and R did not differ in the subscales of somatization or interpersonal sensitivity. These two groups also filled out measures of dissociation, general and severe trauma, absorption, and fantasy proneness. R scored higher than P in dissociation, severe trauma, and absorption. The GSI correlated with dissociation, a history of serious trauma and (weakly) with absorption, but not with general trauma or fantasy-proneness. Overall, the results do not support the view that most spiritual practitioners have higher psychological distress or are socially marginal, although there is a subset of more troubled individuals.

  • Heating of the Magnetic Chromosphere : Observational Constraints from Ca II lambda 8542 Spectra

    • The heating of the Sun's chromosphere remains poorly understood. While progress has been made on understanding what drives the quiet-Sun internetwork chromosphere, chromospheric heating in strong magnetic field regions continues to present a difficult challenge, mostly because of a lack of observational constraints. We use high-resolution spectropolarimetric data from the Swedish 1 m Solar Telescope to identify the location and spatio-temporal properties of heating in the magnetic chromosphere. In particular, we report the existence of raised-core spectral line profiles in the Ca II lambda 8542 line. These profiles are characterized by the absence of an absorption line core, showing a quasi-flat profile between. lambda approximate to +/- 0.5 angstrom, and are abundant close to magnetic bright points and plage. Comparison with three-dimensional MHD simulations indicates that such profiles occur when the line of sight goes through an "elevated temperature canopy" associated with the expansion with height of the magnetic field of flux concentrations. This temperature canopy in the simulations is caused by ohmic dissipation where there are strong magnetic field gradients. The raised-core profiles are thus indicators of locations of increased chromospheric heating. We characterize the location and temporal and spatial properties of such profiles in our observations, thus providing much stricter constraints on theoretical models of chromospheric heating mechanisms than before.

    • This paper presents an algorithm to ensure the atomicity of a distributed storage that can be read and written by any number of clients. In failure-free and synchronous situations, and even if there is contention, our algorithm has a high write throughput and a read throughput that grows linearly with the number of available servers. The algorithm is devised with a homogeneous cluster of servers in mind. It organizes servers around a ring and assumes oint-to-point communication. It is resilient to the crash failure of any number of readers and writers as well as to the crash failure of all but one server. We evaluated our algorithm on a cluster of 24 nodes with dual fast ethernet network interfaces (100 Mbps). We achieve 81 Mbps of write throughput and 8*90 Mbps of read throughput (with up to 8 servers) which conveys the linear scalability with the number of servers.

  • Mild Stress Conditions during Laboratory Culture Promote the Proliferation of Mutations That Negatively Affect Sigma B Activity in Listeria monocytogenes

    • In Listeria monocytogenes, the full details of how stress signals are integrated into the σB regulatory pathway are not yet available. To help shed light on this question, we investigated a collection of transposon mutants that were predicted to have compromised activity of the alternative sigma factor B (σB). These mutants were tested for acid tolerance, a trait that is known to be under σB regulation, and they were found to display increased acid sensitivity, similar to a mutant lacking σB (ΔsigB). The transposon insertions were confirmed by whole-genome sequencing, but in each case, the strains were also found to carry a frameshift mutation in the sigB operon. The changes were predicted to result in premature stop codons, with negative consequences for σB activation, independently of the transposon location. Reduced σB activation in these mutants was confirmed. Growth measurements under conditions similar to those used during the construction of the transposon library revealed that the frameshifted sigB operon alleles conferred a growth advantage at higher temperatures, during late exponential phase. Mixed-culture experiments at 42°C demonstrated that the loss of σB activity allowed mutants to take over a population of parental bacteria. Together, our results suggest that mutations affecting σB activity can arise during laboratory culture because of the growth advantage conferred by these mutations under mild stress conditions. The data highlight the significant cost of stress protection in this foodborne pathogen and emphasize the need for whole-genome sequence analysis of newly constructed strains to confirm the expected genotype.IMPORTANCE: In the present study, we investigated a collection of Listeria monocytogenes strains that all carried sigB operon mutations. The mutants all had reduced σB activity and were found to have a growth advantage under conditions of mild heat stress (42°C). In mixed cultures, these mutants outcompeted the wild type when mild heat stress was present but not at an optimal growth temperature. An analysis of 22,340 published L. monocytogenes genome sequences found a high rate of premature stop codons present in genes positively regulating σB activity. Together, these findings suggest that the occurrence of mutations that attenuate σB activity can be favored under conditions of mild stress, probably highlighting the burden on cellular resources that stems from deploying the general stress response.

    • Production of plants and mushrooms in substrate based partly on anaerobic digestate from biogas production (30%) and peat (70%) was studied in experiments performed using oyster mushroom (Pleurotus ostreatus) and basil (Ocimum basilicum). Biogas digestate was included in order to decrease use of peat and fertilizer. In separate experiments, combined greenhouse production of mushrooms and plants in fresh substrate, mushroom production in bags of fresh substrate or spent substrate from plant production, and plant production in spent substrate from mushroom production were studied. In terms of plant yield, positive impacts of combined culture were observed, with significantly higher yield of basil when mushroom spawn was added to fresh substrate at a concentration of 2% (p 0.04). Increasing the concentration to 10%, which was sufficient for fruiting body formation in parallel with plant production, did not increase basil yield compared with the control. When fresh substrate was partly replaced with spent substrate from mushroom production, significantly higher yield of basil was obtained (p 0.001). Mushroom production had an impact on the nutritional composition of the substrate, resulting in changes in nitrogen dynamics, a significant decrease in phosphorus concentration by 14% (p 0.001), and a change in extractable concentrations of five of 10 elements studied. In terms of mushroom yield, the impacts of combined production with plants were generally negative.

  • Aggrecan fragments as biomarkers in osteoarthritis

    • Background: In osteoarthritis (OA) the balance of cartilage matrix synthesis and degradation is disturbed, resulting in a gradual destruction of the articular cartilage. Matrix components released into body fluids by proteolytic cleavage can be used as biomarkers of ongoing processes. This thesis focuses on proteolytic degradation of the proteoglycan aggrecan with the overall aim to study the potential of aggrecan fragments as biomarkers in knee OA. Methodology/Principal findings: Using neoepitope specific antibodies in Western blots, aggrecan fragments were identified in knee cartilage and synovial fluid (SF) pooled from individuals with a wide spectrum of disease. Aggrecanases were found to dominate aggrecan proteolysis in disease, although a contribution of matrix metalloprotease (MMP) activity was noted. Western blot quantification in individual samples showed that the proportion of aggrecan released into SF generated by aggrecanases varied in disease, and was higher in diagnostic groups associated with high disease activity. Quantification by ELISA of SF ARGS showed that SF ARGS better distinguished samples from patients with knee joint disease from samples obtained from knee healthy individuals than aggrecan measures not specific for this neoepitope. In patients meniscectomized 18 years earlier, SF ARGS levels were inversely associated with progression of radiographic OA. Conclusions: Aggrecanase is the dominating protease in human knee OA and its activity toward the aggrecan interglobular domain (IGD) is elevated in disease. SF levels of aggrecan ARGS fragments generated by this IGD activity can be used as biomarkers and has diagnostic as well as prognostic capabilities.

  • Risk Factors for Intracerebral Hemorrhage : Genome-Wide Association Study and Mendelian Randomization Analyses

    • BACKGROUND: The genetic and nongenetic causes of intracerebral hemorrhage (ICH) remain obscure. The present study aimed to uncover the genetic and modifiable risk factors for ICH.METHODS: We meta-analyzed genome-wide association study data from 3 European biobanks, involving 7605 ICH cases and 711 818 noncases, to identify the genomic loci linked to ICH. To uncover the potential causal associations of cardiometabolic and lifestyle factors with ICH, we performed Mendelian randomization analyses using genetic instruments identified in previous genome-wide association studies of the exposures and ICH data from the present genome-wide association study meta-analysis. We performed multivariable Mendelian randomization analyses to examine the independent associations of the identified risk factors with ICH and evaluate potential mediating pathways.RESULTS: We identified 1 ICH risk locus, located at the APOE genomic region. The lead variant in this locus was rs429358 (chr19:45411941), which was associated with an odds ratio of ICH of 1.17 (95% CI, 1.11-1.20; P=6.01x10-11) per C allele. Genetically predicted higher levels of body mass index, visceral adiposity, diastolic blood pressure, systolic blood pressure, and lifetime smoking index, as well as genetic liability to type 2 diabetes, were associated with higher odds of ICH after multiple testing corrections. Additionally, a genetic increase in waist-to-hip ratio and liability to smoking initiation were consistently associated with ICH, albeit at the nominal significance level (P<0.05). Multivariable Mendelian randomization analysis showed that the association between body mass index and ICH was attenuated on adjustment for type 2 diabetes and further that type 2 diabetes may be a mediator of the body mass index-ICH relationship.CONCLUSIONS: Our findings indicate that the APOE locus contributes to ICH genetic susceptibility in European populations. Excess adiposity, elevated blood pressure, type 2 diabetes, and smoking were identified as the chief modifiable cardiometabolic and lifestyle factors for ICH.

  • Emergence of Fractional Statistics for Tracer Particles in a Laughlin Liquid

    • We consider a thought experiment where two distinct species of 2D particles in a perpendicular magnetic field interact via repulsive potentials. If the magnetic field and the interactions are strong enough, one type of particles forms a Laughlin state and the other type couples to Laughlin quasiholes. We show that, in this situation, the motion of the second type of particles is described by an effective Hamiltonian, corresponding to the magnetic gauge picture for noninteracting anyons. The argument is in accord with, but distinct from, the Berry phase calculation of Arovas, Schrieffer, and Wilczek. It suggests possibilities to observe the influence of effective anyon statistics in fractional quantum Hall systems.

    • Certain many-particle Hardy inequalities are derived in a simple and systematic way using the so-called ground state representation for the Laplacian on a subdomain of R^n. This includes geometric extensions of the standard Hardy inequalities to involve volumes of simplices spanned by a subset of points. Clifford/multilinear algebra is employed to simplify geometric computations. These results and the techniques involved are relevant for classes of exactly solvable quantum systems such as the Calogero-Sutherland models and their higher-dimensional generalizations, as well as for membrane matrix models, and models of more complicated particle interactions of geometric character.

    • We consider the many-particle quantum mechanics of anyons, i.e. identical particles in two space dimensions with a continuous statistics parameter α∈[0,1] ranging from bosons (α = 0) to fermions (α = 1). We prove a (magnetic) Hardy inequality for anyons, which in the case that α is an odd numerator fraction implies a local exclusion principle for the kinetic energy of such anyons. From this result, and motivated by Dyson and Lenard’s original approach to the stability of fermionic matter in three dimensions, we prove a Lieb-Thirring inequality for these types of anyons.

  • Organizational approaches to greening : technocentrism and beyond

    • How and why do organizations approach greening? How can we conceptualize approaches and how can we encourage reflexive dialogues on them? These are the main questions addressed in this qualitative study on organizational greening. The study sets off by discussing matters of research philosophy, arguing that our trust in science ought to be revised and that a more postmodern and constructionist philosophy might be a way to go. This is then followed by a theoretical review, showing that organizational studies have a history in environmental issues, but that it is basically technocentric in orientation. A more reflexive organizational approach is suggested. The empirical part of the study is based on qualitative research of five case studies, representing a mix of organizations situated in Sweden, all with an explicit ambition to approach greening. The analyses target the organizations' approaches from practice to assumptions, pointing at the commonalities as well as the tensions. Basically, greening was an issue for all studied organizations, but an increasing pressure to market-orient their operations in line with the business rhetoric dominated their identity construction. The environment was included if there were opportunities of win-win situations between environment and economy in sight. Once embarked upon, the organizations tended to focus on technocratic practices, developing or implementing management systems, product development indexes, life-cycle methodologies and other tools. On a more philosophical level, in the study referred to as the worldview level, the approaches were predominandy characterized by a representative epistemology and a dualistic ontology, that is, they were clearly anthropocentric. With a base in these findings, an alternative approach is discussed as a way out, or as a way of constructing a reflexive dialogue on greening. This is partly based on the tensions within and between the cases, which encouraged reflections on how greening was approached. In the alternative, organizations are seen as actors on a symbolic agora where transparency, participation and self-reflexivity are keys to organizational legitimacy. This view frames organizations in the dominating approach as agoraphobic producers of materialistically dependent satisfiers. The alternative also targets the limits of a preference and materialistically oriented view on die satisfaction of human needs. Instead, it is argued that environmental and cultural sensitivity should be acknowledged as natural parts of organizational greening. This, however, demands more room for reflexive dialogues encouraging ontological awareness and a respect for more ecocentric views.

    • CoCrFeMnNi is a prototype fcc-structured high-entropy alloy. Numerous efforts have been paid to strengthen CoCrFeMnNi, by replacing Mn with other elements for an enhancement of the solid solution strengthening. 4d transition metals, including Zr, Nb, and Mo, are of interest for this purpose, since they have much larger atomic radii than that of Mn. However, Nb and Mo are known to have a low solid solubility in fcc-structured CoCrFeNi. Compared to Nb and Mo, Zr has an even larger atomic radius. The solid solubility of Zr in fcc-structured CoCrFeNi was investigated in this work, combining both experimental studies and thermodynamic calculations. In addition, based on previous results and new results obtained here, methods to predict the solid solubility in CoCrFeNiMx (M = Zr, Nb, and Mo) alloys were developed. Particularly, the average d-orbital energy level, Md, was re-evaluated in the present work, for an improved predictability of the solid solubility in fcc-structured high entropy alloys containing 4d transition metals.

The study uses communicative strategies as an umbrella term for the way the texts fulfil the political assignment through adaptation to suit different receivers in a discursive practice with set guidelines for communication between experts and users, and how these strategies are visible in the studied texts.The material consists of six guides for care personnel and ten brochures for smokers. The study is a text-focused critical discourse analysis combining methodological tools from different linguistic traditions. These are chiefly taken from critical text linguistics, new rhetoric, and sociosemiotics. The main question posed in the study is how the public authority’s assignment to influence people through the texts is combined with ideas about smokers’ empowerment. The investigation problematizes how the content and form of the texts relates to the authority’s assignment to exert influence, and to motivational interviewing as a discursive practice with an empowerment perspective on lifestyle changes.The analyses show that controlling ideas and notions about smokers’ empowerment exist in parallel in the government texts about smoking cessation. Controlling elements are most prominent in the sections of the guides about groups who, in the encounter with care personnel, do not show any interest or desire to quit smoking. In the studied brochures the controlling elements are most prominent in those aimed at operation ­patients, adolescents, and parents-to-be. Both groups of material – the guides and the brochures – nevertheless give the impression of being designed to be compatible with a patient-cent­red discursive practice. This is noticeable, for example, in the occurrence of associative and dissociative argumentation strategies which legitimize or tone down controlling elements.

  • Impurity scattering and size quantization effects in a single graphene nanoflake

    • By using Fourier-transform scanning tunneling spectroscopy we measure the interference patterns produced by the impurity scattering of confined Dirac quasiparticles in epitaxial graphene nanoflakes. Upon comparison of the experimental results with tight-binding calculations of realistic model flakes, we show that the characteristic features observed in the Fourier-transformed local density of states are related to scattering between different transverse modes (subbands) of a graphene nanoflake and allow direct insight into the gapped electronic spectrum of graphene. We also observe a strong reduction of quasiparticle lifetime which is attributed to the interaction with the underlying substrate. In addition, we show that the distribution of the on-site energies at flower defects leads to an effectively broken pseudospin selection rule, where intravalley backscattering is allowed.

    • X-ray magnetic linear dichroism spectra measured in reflection (XMLD-R) on crystalline bcc Fe thin films across the 3 p absorption edges are reported. A series of measurements with varying orientation of the electric field vector of the linear polarized synchrotron radiation with respect to the crystal axes reveals a strong magnetocrystalline anisotropy in the XMLD-R spectra. The spectra agree well with theoretical spectra calculated within the framework of the density-functional theory accounting for the spin-orbital and exchange splitting of the 3 p semicore states on an equal footing.

    • A comparative study of chemical looping combustion (CLC) with paper, plastic, and coal as fuel was carried out. Experiments were performed in a laboratory fluidized-bed reactor by alternating between reduction and oxidation cycles. The results obtained indicated that a higher temperature leads to an increase in the CO yield and carbon conversion for all fuels. Paper had the highest fractional conversion of CO to CO followed by polyvinyl chloride (PVC) and coal. This was due to the higher fraction of volatiles in paper compared to PVC and coal. Scanning electron microscopy (SEM) analysis of the oxygen carrier particle after each of the solid fuel experiment was carried out. For the used ilmenite, there was a slight difference in the morphology for the three different fuels.

    • Chemical Looping Combustion (CLC) is a carbon capture technology that uses an oxygen carrier to transfer the oxidizing agent to the fuel for combustion. This study used different machine learning algorithms, Artificial neural network and Response surface methodology to estimate the surface region process performance and optimize the process condition for the CLC of different solid fuels waste paper, plastic waste, and sugarcane bagasse blends. Based on the combustion efficiency, CO2 yield and CO2 capture efficiency responses, A high performance correlation (R2 > 0.8) was obtained for all the combustion parameters analyzed. The perturbation plot derived from the RSM analysis indicated that the most significant input parameters include the steam to fixed carbon, blend ratio and the fuel reaction temperature. The CLC process was optimized using RSM. For blends of SCB/WP, the best operating conditions were found to be 800 °C, a solid flow rate of 197.7 kg/h, an oxygen carrier to fuel ratio of 1.1, a steam to fixed carbon ratio of 2.16, and a blend ratio of 1. Similarly, for blends of SCB/PW, the optimal operating conditions were 800 °C, a solid flow rate of 199.4 kg/h, an oxygen carrier to fuel ratio of 1.3, steam to fixed carbon ratio of 2, and a blend ratio of 0.3. The optimum combustion performance was found to be 0.98, 0.78, and 0.96 for SCB/WP and 0.99, 0.62, and 0.96 for SCB/PW, respectively.

  • Towards Smart Home Automation Using IoT-Enabled Edge-Computing Paradigm

    • Smart home applications are ubiquitous and have gained popularity due to the overwhelming use of Internet of Things (IoT)-based technology. The revolution in technologies has made homes more convenient, efficient, and even more secure. The need for advancement in smart home technology is necessary due to the scarcity of intelligent home applications that cater to several aspects of the home simultaneously, i.e., automation, security, safety, and reducing energy consumption using less bandwidth, computation, and cost. Our research work provides a solution to these problems by deploying a smart home automation system with the applications mentioned above over a resource-constrained Raspberry Pi (RPI) device. The RPI is used as a central controlling unit, which provides a cost-effective platform for interconnecting a variety of devices and various sensors in a home via the Internet. We propose a cost-effective integrated system for smart home based on IoT and Edge-Computing paradigm. The proposed system provides remote and automatic control to home appliances, ensuring security and safety. Additionally, the proposed solution uses the edge-computing paradigm to store sensitive data in a local cloud to preserve the customer's privacy. Moreover, visual and scalar sensor-generated data are processed and held over edge device (RPI) to reduce bandwidth, computation, and storage cost. In the comparison with state-of-the-art solutions, the proposed system is 5% faster in detecting motion, and 5 ms and 4 ms in switching relay on and off, respectively. It is also 6% more efficient than the existing solutions with respect to energy consumption.

    • In this paper we present a framework of how board chairperson practices may promote innovation in small entrepreneurial firms. A case study approach is employed involving interviews with experienced board members. In our analysis we identify a set of chairperson practices related to issues such as structure, processes and culture, which serves as prerequisites for effective board work. In addition, we distinguish another set of innovation-driving chairperson practices related to the cognitive aspects of the board’s work. Overall, our findings provide a conceptual foundation for the value creating potential of board leadership embedded within the boundaries of the entrepreneurial firm and its operations.

    • The bile acid receptor farnesoid X receptor (FXR) is a key regulator of hepatic defense mechanisms against bile acids. A comprehensive study addressing the role of FXR in the coordinated regulation of adaptive mechanisms including biosynthesis, metabolism, and alternative export together with their functional significance is lacking. We therefore fed FXR knockout (FXR−/−) mice with cholic acid (CA) and ursodeoxycholic acid (UDCA). Bile acid synthesis and hydroxylation were assessed by real-time RT-PCR for cytochrome P-450 (Cyp)7a1, Cyp3a11, and Cyp2b10 and mass spectrometry-gas chromatography for determination of bile acid composition. Expression of the export systems multidrug resistance proteins (Mrp)4–6 in the liver and kidney and the recently identified basoalteral bile acid transporter, organic solute transporter (Ost-α/Ost-β), in the liver, kidney, and intestine was also investigated. CA and UDCA repressed Cyp7a1 in FXR+/+ mice and to lesser extents in FXR−/− mice and induced Cyp3a11 and Cyp2b10 independent of FXR. CA and UDCA were hydroxylated in both genotypes. CA induced Ost-α/Ost-β in the liver, kidney, and ileum in FXR+/+ but not FXR−/− mice, whereas UDCA had only minor effects. Mrp4 induction in the liver and kidney correlated with bile acid levels and was observed in UDCA-fed and CA-fed FXR−/− animals but not in CA-fed FXR+/+ animals. Mrp5/6 remained unaffected by bile acid treatment. In conclusion, we identified Ost-α/Ost-β as a novel FXR target. Absent Ost-α/Ost-β induction in CA-fed FXR−/− animals may contribute to increased liver injury in these animals. The induction of bile acid hydroxylation and Mrp4 was independent of FXR but could not counteract liver toxicity sufficiently. Limited effects of UDCA on Ost-α/Ost-β may jeopardize its therapeutic efficacy.
