  • Increased concentrations of Serum amyloid A in dogs with sepsis caused by pyometra

    • Background: Sepsis is a serious and potentially life-threatening condition and early diagnosis and appropriate treatment is crucial for survival. Pyometra is one of the most common diseases in intact female dogs. The disease often leads to sepsis (systemic inflammatory response syndrome, SIRS, caused by infection). Diagnostic markers for detecting sepsis are gaining increasing interest in veterinary medicine. Acute phase proteins (APPs) such as C-reactive protein (CRP) are useful for detecting systemic inflammation in dogs. Serum amyloid A (SAA) is another major APP in dogs that is not yet as widely used. Albumin is regarded as a negative APP and has earlier been evaluated for prediction of prognosis in septic dogs. The aim of the present study was to determine SAA, CRP and albumin concentrations in dogs with sepsis and pyometra and to evaluate whether these inflammatory markers are associated with length of postoperative hospitalization.Results: Thirty-one surgically treated bitches with pyometra were included, whereof 23 septic (SIRS-positive) and eight non-septic (SIRS-negative). Albumin concentrations were analyzed by routine automated methods. SAA and CRP analyses were performed with previously validated commercially available assays (ELISA and immunoturbidimetric). Mean (+/- SE) serum concentrations of SAA were significantly higher in septic (130.8 +/- 8.0 mg/L) compared to non-septic bitches (88.5 +/- 12.5 mg/L). Using a cut-off value for SAA of 109.07 mg/L (n = 31 bitches), the sensitivity and specificity for detecting sepsis was 74% and 50%, respectively. Serum albumin concentrations were not significantly different in septic compared to non-septic bitches (mean +/- SE, 25 +/- 1 g/L and 26 +/- 1 g/L, respectively). CRP concentrations were also not significantly different in septic (mean +/- SE 225.6 +/- 16.0 mg/L) compared to non-septic bitches (mean +/- SE, 176.0 +/- 27.1 mg/L). None of these inflammatory markers were associated with the outcome as measured by length of hospitalization.Conclusions: SAA concentrations were increased in dogs with sepsis induced by pyometra and may be useful as an adjunctive diagnostic marker for sepsis. To evaluate the full potential of SAA as a marker for sepsis also in other diseases, further studies are warranted.

    • Osteochondral lesions in the joints of the distal tarsal region of young Icelandic horses provide a natural model for the early stages of osteoarthritis (OA) in low-motion joints. We describe and characterise mineralised and non-mineralised osteochondral lesions in left distal tarsal region joint specimens from twenty-two 30 ±1 month-old Icelandic horses. Combinations of confocal scanning light microscopy, backscattered electron scanning electron microscopy (including, importantly, iodine staining) and three-dimensional microcomputed tomography were used on specimens obtained with guidance from clinical imaging. Lesion-types were described and classified into groups according to morphological features. Their locations in the hyaline articular cartilage (HAC), articular calcified cartilage (ACC), subchondral bone (SCB) and the joint margin tissues were identified and their frequency in the joints recorded. Associations and correlations between lesion-types were investigated for centrodistal joints only. In centrodistal joints the lesion-types HAC chondrocyte loss, HAC fibrillation, HAC central chondrocyte clusters, ACC arrest and ACC advance had significant associations and strong correlations. These lesion-types had moderate to high frequency in centrodistal joints but low frequencies in tarsometatarsal and talocalcaneal-centroquartal joints. Joint margin lesion-types had no significant associations with other lesion-types in the centrodistal joints but high frequency in both the centrodistal and tarsometatarsal joints. The frequency of SCB lesion-types in all joints was low. Hypermineralised infill phase lesion-types were detected. Our results emphasise close associations between HAC and ACC lesions in equine centrodistal joints and the importance of ACC lesions in the development of OA in low-motion compression-loaded equine joints.

  • Diabetes Mellitus in Elkhounds Is Associated with Diestrus and Pregnancy

    • Background: Female Elkhounds are shown to be at increased risk for diabetes mellitus, and occurrence of diabetes during pregnancy has been described in several cases. Hypothesis: Onset of diabetes mellitus in Elkhounds is associated with diestrus. Animals: Sixty-three Elkhounds with diabetes mellitus and 26 healthy controls. Methods: Medical records from 63 Elkhounds with diabetes were reviewed and owners were contacted for follow-up information. Blood samples from the day of diagnosis were available for 26 dogs. Glucose, fructosamine, C-peptide, growth hormone (GH), insulin-like growth factor-1, progesterone, and glutamate decarboxylase isoform 65-autoantibodies were analyzed and compared with 26 healthy dogs. Logistic models were used to evaluate the association of clinical variables with the probability of diabetes and with permanent diabetes mellitus after ovariohysterectomy (OHE). Results: All dogs in the study were intact females and 7 dogs (11%) were pregnant at diagnosis. The 1st clinical signs of diabetes mellitus occurred at a median of 30 days (interquartile range [IQR], 3-45) after estrus, and diagnosis was made at a median of 46 days (IQR, 27-62) after estrus. Diabetes was associated with higher concentrations of GH and lower concentrations of progesterone compared with controls matched for time after estrus. Forty-six percent of dogs that underwent OHE recovered from diabetes with a lower probability of remission in dogs with higher glucose concentrations (odds ratio [OR], 1.2; P = .03) at diagnosis and longer time (weeks) from diagnosis to surgery (OR, 1.5; P = .05). Conclusions: Diabetes mellitus in Elkhounds develops mainly during diestrus and pregnancy. Immediate OHE improves the prognosis for remission of diabetes.

    • Objective: Describe the local morphological response of the articular-epiphyseal cartilage complex to surgical stab incision in the distal femur of foals, with emphasis on the relationship between growth cartilage injury, enchondral ossification, and repair. Design: Nine foals were induced into general anesthesia at the age of 13 to 15 days. Four full-thickness stab incision defects were created in the cartilage on the lateral aspect of the lateral trochlear ridge of the left distal femur. Follow-up examination was carried out from 1 to 49 days postoperatively, including examination of intact bones, sawed slabs, and histological sections. Results: Incision defects filled with cells displaying fibroblast-, chondrocyte-, and osteoblast-like characteristics, potentially validating the rationale behind the drilling of stable juvenile osteochondritis dissecans lesions in children. Incisions induced necrosis within the cartilage on the margins at all depths of the defects. Sharp dissection may therefore be contraindicated in cartilage repair in young individuals. Incisions caused a focal delay in enchondral ossification in 2 foals, apparently related to the orientation of the incision defect relative to the direction of ossification. Defects became progressively surrounded by subchondral bone, in which granulation tissue containing clasts and foci of osteoblast-like cells was observed. Continued enchondral ossification was therefore likely to result in healing of uncomplicated defects to morphologically normal bone. Conclusions: Epiphyseal growth cartilage injury had the potential to exert a negative effect on enchondral ossification. Enchondral ossification exerted a beneficial effect on repair. This relationship warrants consideration in future studies of cartilage injury and repair within the articular-epiphyseal cartilage complex of all species.

    • Freemartinism is one of the most commonly found intersex conditions in cattle, although it may also occur in small ruminants. The freemartin phenotype appears in a dizygotic twin pregnancy where one twin is a male and the other is a female. Due to precocious anastomoses between the placental vascular systems of the two fetuses, masculinising molecules reach the female twin and disrupt the normal sexual differentiation, whilst in the male the effects of this association are usually minimal. In cattle, this condition is observed in 90 to 97% of twin pregnancies. A freemartin is, by definition, a genetically female fetus masculinised in the presence of a male co-twin, giving rise to a sterile heifer. Genital tract defects with varying severity can be observed in freemartin animals, which often present suppression and disorganization of the ovary, originating a rudimentary or a testis-like gonad depleted of germ cells. The uterine horns may be hypoplastic or instead may be reduced to a cord-like structure suspended in the broad ligament. Anatomic continuity between the uterus and the vagina is frequently absent, and the existence of rudimentary vesicular glands is typical. The external genitalia commonly presents enlarged clitoris, small vulva and a prominent, male-like tuft of hair. As a rule, heifers born twin to a bull have to be considered sterile and should be identified as early as possible to cull them from replacement stock. Despite its limitations, freemartinism is currently diagnosed by physical examination, as karyotyping or blood typing is often considered an unnecessary expense. In cattle, twinning trend has a genetic background that has been associated to hormonal regulation in favor of double ovulations. However, the genetic determinant on the basis of twinning seems to have small importance when compared to environmental or management-associated factors, particularly in dairy cows. In fact, in dairy animals, in particular in high milk producing cows, it has long been proven that the increase of twin calvings occurs due to the hormonal and metabolic disturbances in the energy balance early in the post-partum period. With increased incidence of twin births in cow it is reasonably expectable a small increase in the occurrence of freemartins at the farm levels. In this paper it is the intent to describe the gross and histopathological findings of freemartinism in cattle, using data gathered from a study at an abattoir (17 cases) and from 3 cases diagnosed in living animals, supported by a review of the pathophysiology of the process, and to discuss the available methods for identification of freemartin animals at farm level.

  • Markers of Borna disease virus infection in cats with staggering disease

    • Borna disease virus (BDV) is a RNA-virus causing neurological disorders in a wide range of mammals. In cats, BDV infection may cause staggering disease. Presently, staggering disease is a tentative clinical diagnosis, only confirmed at necropsy. In this study, cats with staggering disease were investigated to study markers of BDV infection aiming for improvement of current diagnostics. Nineteen cats fulfilled the inclusion criteria based on neurological signs and pathological findings. In 17/19 cats, BDV infection markers (BDV-specific antibodies and/or BDV-RNA) were found, and antibodies in serum (13/16, 81%) were the most common marker. BDV-RNA was found in 11/19 cats (58%). In a reference population without neurological signs, 4/25 cats were seropositive (16%). The clinical history and neurological signs in combination with presence of BDV infection markers, where serology and rRT-PCR on blood can be helpful tools, improve the diagnostic accuracy in the living cat.

  • Carriage of methicillin-resistant Staphylococcus pseudintermedius in dogs–a longitudinal study

    • Background: Methicillin-resistant S. pseudintermedius strains (MRSP) are reported with increasing frequency in bacterial cultures from dogs. The objectives of this study were to determine whether MRSP could be found in dogs several months after a clinically apparent infection and whether the length of carriage varied depending on systemic antimicrobial treatment, diagnosis at time of the first positive MRSP culture and the presence of skin disease or wounds. Thirty-one dogs previously diagnosed with a clinical infection were sampled repeatedly for a minimum of eight months or, with the exception of two dogs, until two consecutive negative results were obtained. Five specified locations were sampled, and the results were evaluated to determine future recommendations concerning sample strategies when screening for MRSP carriage. Information was collected from medical records and questionnaires to evaluate factors that may influence length of carriage.Results: The overall median length of MRSP carriage was 11 months (48 weeks). The presence of wounds and signs of dermatitis did not influence length of carriage. Systemic treatment for three weeks or longer with antimicrobial agents to which the bacterium was resistant was associated with prolonged carriage compared to dogs treated for a shorter period of time. Three of five dogs treated with an antimicrobial to which their MRSP-isolates were susceptible (tetracycline) were found to still be MRSP-positive when sampled after the end of treatment. Wound samples had the highest positive MRSP yield (81%) for the positive sample sites, compared to less than 70% for each of the other four sample sites. Cultures from the nostrils were less likely to detect MRSP carriage relative to the pharynx, perineum, wounds and the corner of the mouth.Conclusions: Dogs can carry MRSP for more than a year after a clinically apparent infection. Systemic antimicrobial treatment of infections with antimicrobial agents to which the MRSP-bacteria are resistant should be avoided when possible in dogs with possible or confirmed MRSP carriage or infection, since it may prolong time of MRSP carriage. Simultaneous sampling of pharynx, perineum, and the corner of the mouth as well as wounds when present is recommended when screening for MRSP. Cultures from nostrils were shown to be less likely to detect MRSP carriage.

    • Pyometra is a common and life-threatening disease of intact bitches caused by hormonal influence of the uterus in combination with bacterial infection. The treatment of choice is ovariohysterectomy, but several purely medical options are available. Common drugs used for medical treatment in combination with antimicrobials are progesterone receptor blockers, prostaglandins, and dopamine agonists. The aim of this study was to evaluate long-term recovery and fertility after treatment with the progesterone receptor blocker aglepristone in bitches with pyometra. Data from 28 bitches with pyometra, admitted to the University Animal Hospital, Swedish University of Agricultural Sciences, during a 9-year period, were studied retrospectively and followed up by telephone interviews with the owners. The bitches had been treated with aglepristone at a dose of 10 mg/kg on a median of four occasions. All bitches had also been treated with antimicrobial drugs for a mean duration of 23 days, the most frequently used being enrofloxacin. Escherichia coli were the most commonly isolated bacteria from cranial vagina. The outcome was assessed for up to 6 years after treatment. The success rate, determined as restored clinically healthy status, was 75% (21/28 bitches), and the recurrence rate of disease was 48% (10/21 bitches). The mean time until recurrence was 10.5 months after the end of treatment. After treatment, 69% (9/13) of the mated bitches produced puppies. Of the seven bitches that did not have a successful treatment, six were ovariohysterectomized and one was euthanized. In conclusion, medical treatment with aglepristone in combination with antimicrobial therapy was successful in 75% of the bitches studied and the recurrence rate was 48%. (C) 2014 The Authors.   • Human-Human and Human-Animal Interaction : Some Common Physiological and Psychological Effects

    • The aim of the present thesis was to investigate hormonal and physiological effects in mothers during a breastfeeding session and in dogs and their owners in response to short-term interaction. In study one, sixty-six mothers receiving either exogenous oxytocin infusion and/or epidural analgesia (EDA) during labor or intramuscular oxytocin injection post partum were studied. Oxytocin, prolactin, adrenocorticotrophic hormone (ACTH) and cortisol levels, as well as blood pressure were measured during a breastfeeding session two days after birth. In response to breastfeeding two days after birth, the mothers displayed a pulsatile release of oxytocin and increasing prolactin levels. In addition, the activity in the HPA-axis was reduced and maternal blood pressure decreased. The results also show that EDA administration in combination with oxytocin during labor resulted insignificantly lower oxytocin levels and higher cortisol levels, as well as higher bloodpressure in response to breastfeeding two days after birth, compared to EDA administration alone. In addition, oxytocin infusions dose-dependently lowered the mothers’ endogenous oxytocin levels two days after birth. In study two, ten female dog owners and their male Labrador dogs participated, together with ten controls. Their levels of oxytocin, cortisol and insulin, as well as their heart rate, were measured. The connection between the quality of the dogowner relationship and hormone levels was also explored. Short-term interaction between dogs and their owners resulted in oxytocin release in both species and their cortisol levels and heart rate were also affected. Oxytocin levels and positive attitudes regarding the dog-owner relationship were positively correlated. In conclusion, both human-human and human-animal interactions induce oxytocin release and promote oxytocin mediated effects, such as decreasing cortisol levels and blood pressure. In addition, social interaction and oxytocin levels arepositively related.

    • Conclusions: Several clinically useful indicators were identified. Leucopenia was the most important marker, associated with 18-fold increased risk for peritonitis and an over three-point-five increased risk for prolonged hospitalization. Fever/hypothermia, depression and pale mucous membranes were associated with increased risk for peritonitis and/or prolonged hospitalization. The results of the present study may be valuable for identifying peritonitis and predicting increased morbidity in surgically treated bitches with pyometra.

    • Introduction: Histologic examination with light microscopy is considered the reference standard to detect osteoarthritis (OA) in its earliest stages1, 2. In early OA, articular cartilage changes develop in focal and specific regions of the affected joint3 but the locations of these regions in specific joints and species are uncertain and are likely to vary among individuals. When osteochondral samples are collected from predetermined anatomic sites for histologic examination of joints early in the disease process, then it is possible that OA will fail to be detected because the associated lesions may not be included in the collection site. This study evaluates a method for computed tomography (CT) and magnetic resonance imaging (MRI) guided osteochondral sample collection for the detection of early stage OA lesions in joints from young Icelandic horses. Materials and Methods: CT and MRI were semiquanitatively graded to evaluate the extent of suspected OA changes in right centrodistal joints from the cadavers of 24 Icelandic horses aged 29 to 31 months. The anatomic regions with the highest grade of change were identified, and osteochondral samples were obtained from these regions. Samples were also obtained from the same centrodistal joints at predetermined sites. Histologic examination of all samples was performed, with samples classified as negative or positive for OA, and results were compared between sample collection method. Results: Histologic examination revealed OA lesions in 29% (7/24) of centrodistal joints with predetermined method and in 62% (15/24) with the image-guided method. Significant associations were detected between histologic OA and the summed image-guided sample collection site image grades, central osteophytes, articular cartilage thickness abnormalities, grade-2 articular mineralisation front defects, and grade-2 marginal osteophytes. Discussion/Conclusion:An image-guided sample collection method is recommended when histologic examination will be used as the reference standard for the detection of early-stage OA in centrodistal joints of horses. CT and MRI aided the detection of focal changes suggestive of early stage OA in the centrodistal joints of equine cadavers and may be useful for detection of similar disease in live horses. The first morphological changes of centrodistal joint OA are suspected to be in the articular cartilage and the articular mineralisation front regions.

Plasma vasopressin and urinary noradrenalin and creatinine concentrations were analyzed.Dogs were premedicated with acepromazine, carprofen, and methadone andflurane. Blood pressure, heart rate, and end‐tidalflurane concentration were measured every minute. The effects of various events Results: greater than for the 2nd ovary because of an elevation in baseline. Similarly, the heart rate increased at the removal of the 1st ovary but not at removal of the 2nd ovary. Plasma vasopressin concentration increased at removal of both ovaries. Urinary noradrenalin/ creatinine ratio increased at anesthesia, removal of both ovaries, and was elevated at closure of the abdomen. End pressure and vasopressin concentrations changed in parallel using z comparison.The magnitude of blood pressure increase at removal of the 1st ovary was‐tidal isoflurane concentration did not change. Blood‐scores for Conclusions: concentration, and urinary noradrenalin/creatinine ratio did not differ between removals of the ovaries. Relative changes differed between repeated noxious stimuli, which should be considered in evaluation of methods at ovary removal.Peak values for blood pressure, heart rate, plasma vasopressin

    • Bornaviruses are known to cause neurological disorders in a number of animal species. Avian Bornavirus (ABV) causes proventricular dilatation disease (PDD) in birds and Borna disease virus (BDV) causes Borna disease in horses and sheep. BDV also causes staggering disease in cats, characterised by ataxia, behavioural changes and loss of postural reactions. BDV-infection markers in cats have been reported throughout the world. This review summarizes the current knowledge of Borna disease viruses in cats, including etiological agent, clinical signs, pathogenesis, epidemiology and diagnostics, with comparisons to Bornavirus infections in other species.

  • Treponema spp. in porcine skin ulcers : clinical aspects

    • The hypothesis tested in this work is that bacteria of genus Treponema play a main role when shoulder ulcers and ear necrosis occur in an infectious or severe form, and perhaps also in other skin conditions in the pig. Samples were collected from pigs in 19 Swedish herds 2010-2011. The sampled skin lesions included 52 shoulder ulcers, 57 ear necroses, 4 facial necroses and 5 other skin ulcers. Occurrence of spirochetes was detected by phase contrast microscopy, Warthin-Starry silver staining, PCR and Fluorescent In Situ Hybridization (FISH). Treponemal diversity was investigated by sequencing of 16S-23S rRNA intergenic spacer region 2 (ISR2) and high-throughput sequencing (HTS) of a part of the 16S rRNA gene. Culturing and characterization of treponemes by biochemical analyses, testing of antimicrobial susceptibility and fingerprinting by random amplified polymorphic DNA (RAPD) were carried out. A challenge study was performed to test if Treponema pedis induced skin lesions. Serological response towards TPE0673, a T. pedis protein, was tested with ELISA. Spirochetes were found in all types of skin ulcers and in all herds. The occurrence of Treponema spp. detected by PCR was 52% in shoulder ulcers, 46% in ear necrosis and 9.7% in gingiva. Treponemes were identified in 69% of the shoulder ulcers and in 59% of the ear necroses by FISH. A phylogenetic tree revealed a great variability of treponemes. Three main phylotypes were identified; T. pedis, Treponema parvum and one phylotype without designation. Twelve isolates of T. pedis, T. parvum, and one phylotype most similar to Treponema sp. OMZ 840 were obtained. All except two had unique RAPD fingerprints. Biochemical tests could not differentiate between the isolates and they were generally susceptible to tested antimicrobials. By FISH, treponemes were visualized deep in the ulcers and a predominance of T. pedis was noted, and confirmed by HTS. Challenged pigs did not develop any lesions or IgG response towards the T. pedis protein. Most sows with shoulder ulcers showed a strong, and most cases of ear necrosis a weak IgG response towards TPE0673. In conclusion, Treponema spp. are frequently abundant in ear necroses and shoulder ulcers in pigs. Identical phylotypes and ISR2 sequences from ulcers and gingiva indicate spreading from mouth to ulcer. A broad diversity of phylotypes was revealed, but the predominance of T. pedis suggests specific importance of this species. Our results point towards an important role of treponemes in chronic and severe skin ulcers in pigs.

    • Summary High yielding dairy cows are housed for most of the year, most of their production cycle, or most of their life. When dairy cows are confined, as in a free stall system, their feet (when standing) and legs (when lying) are influenced in several ways: wear, trauma, loading, and by microorganisms especially if the hygiene is poor. Moreover, exposure for environmental risk factors are also important and therefore cow traffic is of concern in the planning. Even if the lying area is of outmost importance it cannot compensate for poor flooring because animals still have to move around to socialize, to feed and to be milked. The ability for each specific function such as resting, standing and walking must, therefore, be optimized. That means a soft yielding floor with, enough friction to reduce claw over growth yet preventing over wear and avoiding poor hygiene by ensuring as dry and clean floors as possible. Although optional, some kind of foot wash is beneficial.

    • The aim of the present study was to explore possible correlations between dog owners' relationships with their dogs, as measured with the Monash Dog Owner Relationship Scale (MDORS), and oxytocin and cortisol levels in both the owners and their dogs. Ten female owners of male Labrador Retrievers completed the MDORS. The scores obtained from the single items, subscales, and total score of the MDORS were calculated. Ten blood samples were collected from each dog owner and her dog during a 60-minute interaction. Blood samples were analyzed for oxytocin and cortisol by Enzyme Immuno Assay (EIA) and mean values of oxytocin and cortisol were calculated in both owners and dogs. The MDORS scores obtained were correlated with basal and mean oxytocin and cortisol levels. The correlation analysis revealed some relationships between the scores of items in the MDORS that reflect the character of the dog-owner-relationship and the owners' hormone levels. For example, higher oxytocin levels in the owners were associated with greater frequency in kissing their dogs (rs = 0.864, p = 0.001). Lower cortisol levels in the owners were associated with their perception that it will be more traumatic when their dog dies (rs = -0.730, p = 0.025). The correlation analysis also revealed some relationships between the scores of items in the MDORS and the dogs' hormone levels. For example, greater frequency in owners kissing their dogs was associated with higher oxytocin levels in the dogs (rs = 0.753, p = 0.029). Six items in the subscale Perceived Costs, as well as the subscale itself, correlated significantly with the dogs' oxytocin levels (rs = 0.820, p = 0.007), that is, the lower the perceived cost, the higher the dogs' oxytocin levels. In addition, significant correlations between the oxytocin levels of the owners and the dogs were demonstrated. Possible mechanisms behind these correlations are discussed. In conclusion, the scores of some items and the subscales of the MDORS correlated with oxytocin, and to a lesser extent cortisol, levels in both the owners and dogs.

    • The aim of this exploratory study was to determine heart rate and the levels of oxytocin, cortisol, and insulin in dogs and their owners in response to a short-term interaction. In addition, the dogs' behavior was studied. The owners' responses were compared with those obtained from a control group. Ten female volunteers and their own male Labrador dogs participated in an experiment during which the owner stroked, petted, and talked with her dog during the first 3 minutes. Blood samples were collected from both dog and owner before (0) and at 1, 3, 5, 15, 30, and 60 minutes after the start of the interaction. Blood samples were analyzed by EIA. Heart rate was monitored telemetrically. The data were analyzed using linear mixed models and paired t-tests. The dogs' oxytocin levels were significantly increased 3 minutes after the start of the interaction (p = 0.027). Cortisol levels were significantly increased after 15 and 30 minutes (p = 0.004 and p = 0.022, respectively), and heart rate was significantly decreased after 55 minutes (p = 0.008). The dogs displayed normal behaviors during the experiment. The owners' oxylocin levels peaked between 1 and 5 minutes after interaction (p = 0.026). No such effect was seen in the controls. Cortisol levels displayed a significant decrease at 15 or 30 minutes in both owners and controls, and insulin levels did so at 60 minutes (p = 0.030, p = 0.002 and p = 0.002, p < 0.0001, respectively). Heart rate decreased significantly in the owners at 55 and 60 minutes (p = 0.0008) but not in the controls. In conclusion, short-term sensory interaction between dogs and their owners influences hormonal levels and heart rate. However, further studies need to be performed in order to better understand the effects of interaction between dogs and their owners.

    • Following a request from the Commission, the Panel on Animal Health and Welfare was asked to deliver a Scientific Opinion on: 1) the expected prevalence (design prevalence) under different circumstances, and, 2) an updated scientific assessment of the size of the relevant geographical area for the purpose of monitoring and surveillance programmes for bluetongue. A systematic literature review and a review of monitoring and surveillance data from European Union Member States was performed in order to estimate the prevalences observed in the Member States. The prevalences observed in areas that have been infected for several years were slightly lower than the design prevalence of 2 % currently used for monthly testing of sentinel animals, but much lower than the design prevalences of 20 % and 10 % for annual surveys in populations of unvaccinated and vaccinated ruminants, respectively. Currently there is no scientific evidence that suggests an optimal size of the relevant geographic unit for BTV monitoring and surveillance, since it depends on many factors, including the goal of the surveillance programmes. Early warning based on passive surveillance will take place irrespective of the size of the geographical unit but, when based on active surveillance, it is best targeted at regions considered at risk for introduction, using small geographical units, a high sampling frequency and sample size. For estimating the impact of interventions on the prevalence of infected animals, smaller areas result in more precise estimates of the prevalence and also take better account of local differences. For establishing freedom from infection, smaller areas result in lower design prevalence for a region as a whole and take better account of local differences in infection dynamics.

    • To answer a question from the European Commission on the potential special characteristics of bluetongue virus (BTV) serotype 8 (BTV-8) compared to other serotypes and their possible impact on the epidemiology of the disease, a systematic literature review was carried out by a working group established by the Animal Health and Welfare Panel. Currently, three special features can be assigned to BTV-8, which are the ability to cause serious disease in cattle and goats, the ability to be transmitted transplacentally, and the ability to contaminate semen. The transplacental transmission and the contamination of semen are also observed for several serotypes of modified live virus (MLV) vaccines and for some cell culture/egg passaged strains. These two features may have an impact on the epidemiology of the disease, since they may increase the ability of BTV-8 to survive the winter period, for example, when pregnant cows are infected in late autumn and give birth to viraemic offspring in the next vector season, or, through infecting the recipient dam via artificial insemination (AI) with frozen contaminated semen. Furthermore, the chance of BTV-8 spread may be increased either through movement of seropositive but virus negative pregnant animals, which may give birth to viraemic calves, or through natural mating or AI using BTV-8 contaminated semen by transmission from semen to receiving dam. The current legislation provides effective measures to ensure that all dams are immune to BTV before insemination or mating, so there is no subsequent risk of transplacental infection of their offspring. Furthermore, pregnant animals are effectively restricted in their movement. More research is needed to determine whether oral transmission and/or transmission through embryo transfer are more likely to occur for BTV-8 than for other BTV serotypes.

    • The AHAW panel conducted an assessment of the risk of entry, release, spread and possible impact of epizootic ulcerative syndrome (EUS) in Europe. The risk of entry is linked to the probability of infection in the source region and the frequency of imports. A greater risk of entry was associated with the importation of ornamental fish into closed ornamental facilities (COF). COF, as defined by EU legislation, are pet shops, garden centres, garden ponds, commercial aquaria or wholesalers keeping ornamental aquatic animals without any direct contact with natural waters; or equipped with an effluent treatment system. The release of pathogens from COF could occur through the sale of live fish to hobbyists, insufficient effluent treatment or unintentional contact with natural waters. Assuming regular entry and release the long-term lack of reporting of EUS is a apparent contradiction. The following scenarios could explain it: i) Fish in European waters have never been exposed to the pathogen because the survival of released zoospores is too short, ii) EUS does not establish, but the pathogen fades out after entry into the EU, due to unfavourable conditions, iii) Ecological or epidemiological circumstances are not favourable for disease expression, iv) EUS is endemic and regularly misdiagnosed. The insufficient data from monitoring of aquaculture fish or wild stocks in Europe preclude confirmation or estimation of the likelihoods of these scenarios. The impact of EUS on EU aquaculture, would range from no impact, if there was no spread, to low impact if there was spread without disease, low incidence or weak disease patterns. However, if endemic the impact may change in the future if conditions such as climate, water quality or availability of susceptible species, either in aquaculture or in natural water bodies, become favourable for initiating epidemics and disease expression.

    • Following the emergence in 2009 of the new pandemic H1N1 influenza virus, which contained gene segments from pig, bird and human influenza viruses, it was apparent that a better scientific understanding is required of influenza viruses to protect public and animal health. The latest scientific data on biological properties of the virus, transmissibility, host susceptibility and epidemiology has been evaluated in order to identify factors that could be monitored in animals and that would suggest a risk of emergence of a new pandemic influenza strains. Virological studies and animal models have highlighted the importance of individual virus proteins but virulence and transmissibility are polygenic effects and no single genetic marker can be reliably associated with increased pathogenicity or transmissibility. It was concluded that current monitoring of the influenza gene pool in humans has been able to provide an alert for the emergence of new human influenza strains of public health significance. In contrast, there is an incomplete view of the influenza virus strains circulating among pigs and birds at the global level. Interpretation of the origins and pandemic potential of influenza viruses do require knowledge of the influenza gene pools in both pigs and birds, as well as other animal species. It is recommended that there should be long term support for a passive monitoring network in pigs and birds in order to promote greater understanding of the evolution of influenza viruses at the global level. Maximum benefit can only be obtained by applying an integrated approach involving the medical and veterinary networks including development of harmonised tools and approaches, exchange of virus strains and sequence data and enhancing the coordination and dissemination of the findings from the human, swine and avian networks.

    • Following a request from the European Commission, a scientific opinion was prepared by EFSA's Animal Health and Welfare Panel to determine the magnitude, distribution, impact and significance of infection and disease in domestic ruminants and humans, risk factors for the maintenance (in domestic ruminant populations) and spillover (from these populations to humans) of Coxiella burnetii (the causative agent of Q fever), and control options in domestic ruminant populations. A range of approaches were used, including an assessment of monitoring/surveillance data, the development of a simple conceptual model, a critical review of available literature, and several country case studies. Control options for C. burnetii infection in small ruminants were qualitatively assessed. Infection is endemic in domestic ruminants in most, if not all, EU member states, however, disease is rare and impact is limited. In the EU, Q fever is a zoonotic disease with limited public health impact, except under certain epidemiological circumstances and for particular risk groups. Human cases are often associated with proximity to small ruminants (particularly at parturition or during abortions) and dry, windy weather. Currently, there is no clear evidence of an association between bacterial genotypes/isolates and virulence. A number of longer-term options to control C. burnetii infection in domestic ruminants were identified; these should be considered in those situations where the public health risk is considered unacceptable. Some additional options were not considered sustainable for long-term control, but may have a role in the face of an outbreak. Persistent environmental contamination may confound animal-based control efforts. Vaccination should be considered a long-term control option, noting that effectiveness may not be observed in the short-term. Antibiotic treatment of animals is not recommended. There is no conclusive evidence that the consumption of milk and milk products containing C. burnetii has resulted in clinical Q fever in humans.

    • Embryo transfer (ET) in the donkey resulted in a very low recipient pregnancy rates. The aim of these studies was to investigate if nonsurgical transfer techniques or donkey embryo quality affect donkey recipient pregnancy failure. In Study 1, the impact of transfer technique was investigated by evaluating if cervical catheterization is associated with prostaglandin release and suppression of luteal function and if donkey recipients would become pregnant after nonsurgical transfer of horse embryos. Four jennies, from 5 to 8 d after ovulation, were submitted to a sham transcervical ET and to evaluation of POEM and progesterone plasma concentrations. Five 8 d horse embryos were nonsurgically transferred into synchronized donkey recipients (HD). Cervical stimulation caused a transient PGF(2 alpha) release in two of four jennies in the absence of a significant decrease in progesterone plasma concentration. All transferred horse embryos resulted in pregnancies in the jenny recipients. In Study 2, donkey embryo viability was investigated by 1.2 meters, 6-diamidino-2-phenylindole (DAPI) staining of 10 embryos and by the transfer of 6 and 12 donkey embryos in synchronized mare (DH) and donkey (DD) recipients, respectively, of known fertility. The estimated proportion of dead cells in DAPI stained embryos was 0.9% (range 0-3.9%) and below what is considered normal (20%) for horse embryos. Three of six and six of 12 of the DH and DD ETs, respectively resulted in pregnancies at 14 and 25 d (50%), a higher pregnancy rate than previously reported after DD ET. The overall results of this study suggest that the transcervical technique for ET and donkey embryo viability are not the reasons for the low pregnancy rates that have previously been described in donkey recipients, and that nonsurgical ET in donkeys can result in acceptable results.     • Background Strangles is a contagious equine-specific disease caused by Streptococcus equi subsp. equi. Unfortunately, detection of S. equi can fail in up to 40% of horses with strangles. Whereas recent molecular biologic methods and sampling techniques have improved recovery of S. equi optimal sampling methods and laboratory analyses remain ill-defined. Objectives To determine the yield of S. equi from horses with acute strangles in confirmed outbreaks by field-sampling methods subjected to culture and biochemical identification, and real-time PCR directly and after culture. Animals Fifty-seven horses of varying breeds and ages from 8 strangles outbreaks. Methods Prospective study. Culture with biochemical identification and real-time PCR directly, and from culture, were performed on nasal swabs, nasopharyngeal swabs, and nasopharyngeal lavages. Results Real-time PCR directly from samples identified the highest number of infected horses, with 45/57 nasal swabs, 41/57 nasopharyngeal swabs, and 48/57 nasopharyngeal lavages S. equi positive. Biochemical identification (highest positives 22/57) was inferior to real-time PCR for S. equi recovery regardless of sampling method. Real-time PCR of nasopharyngeal lavage directly and after culture yielded 52/57 positives whereas direct real-time PCR of nasopharyngeal lavage combined with either nasopharyngeal swabs or nasal swabs yielded 53/57 positives. Three horses were negative on all samples. Conclusions and Clinical Importance Nasopharyngeal lavage analyzed by a combination of real-time PCR directly and after culture or, alternatively, real-time PCR directly on a nasopharyngeal lavage and a nasal/nasopharyngeal swab can identify S. equi in over 90% of acute strangles cases.

    • The objective of this study was to study factors affecting a reproductive performance indicator at the cow level adjusted for herd management strategy. Associations between the outcome variable, pregnant or not at the herd voluntary waiting period (VWP) plus 30 d (pregnant at VWP+30), and the predictor variables were analyzed using a multivariable, generalized estimation equations model that adjusted for clustering of the data at the herd level. The statistical analysis was stratified on parity. In total, 132,721 cows were retained for analyses, of which 29,113 (22%) were pregnant at VWP+30 d. Of the nonpregnant cows, 81,483 cows had records of artificial inseminations (AI) and 22,125 cows had no records of AI. The chance of pregnancy was higher for cows of the Swedish Red and for other/crossbreeds compared with Swedish Holstein, for cows from herds with high heat detection efficiency compared with cows from herds with medium and low heat detection efficiency, for cows from herds with long VWP (i.e., >51 d) compared with cows from herds with short VWP (<51 d), and for cows in freestalls compared with cows in tiestalls. The chance for pregnancy was lower for cows with severe problems at claw trimming compared with cows with no problems at trimming (only for second- and higher-parity cows), for cows that had a record of reproduction-related disease, for cows that had a record of any other disease compared with cows without record, for second- and higher-parity cows with records of dystocia compared with cows with no record of dystocia, for first-parity cows in the group with the highest milk yield compared with first-parity cows in the group with the lowest milk yield, for cows of third and higher parity in the group with the lowest milk yield compared with cows in higher yielding groups, for cows bred in summer compared with those bred in winter-spring (not significant for first-parity cows), and for cows with a twin birth had compared with cows with a single birth. We observed associations of the dose-response type, such that when the milk fat-to-protein ratio increased, the chance for pregnancy decreased, and as the somatic cell count increased, the chance for pregnancy decreased. In conclusion, factors that are known to affect reproductive efficiency also affect the chance of cows being pregnant at the herd VWP plus 30 d.

    • The objective of this study was to investigate if and how cow factors and intramammary infection (IMI) are associated with 4 different udder-health indicators in dairy cows as a first step in investigating whether the diagnostic performance of these indicators can be improved. The investigated indicators were somatic cell count (SCC), lactate dehydrogenase (LDH), N-acetyl-beta-D-glucosaminidase (NAGase), and alkaline phosphatase (AP) measured in milk. In this cross-sectional study, approximately 1,000 cows from 25 dairy herds were sampled for bacteriology (quarter milk samples) during 3 consecutive days: the day before test milking, at the day of test milking, and at the day after test milking. The whole-udder test milking sample was analyzed for milk composition, SCC, LDH, NAGase, and AP. Cow data (parity, breed, milk yield, percentage of milk fat and protein, milk urea concentration, and days in milk from the sampled test milking) were collected from the Swedish milk-recording scheme. Of the sampled cows 485 were considered IMI negative and were used in multivariable mixed-effect linear regression models to investigate associations between cow factors and the udder-health indicators. A second modeling including all cows, both IMI negative and IMI positive (256 cows), was also performed. The results showed that all udder-health indicators were affected by cow factors but that different cow factors were associated with different indicators. Intramammary-infection status was significantly associated with all udder-health indicators except AP. Parity and milk urea concentration were the only cow factors associated with all indicators in all models. The significant cow factors explained 23% of the variation in SCC and >30% of the variation in LDH, NAGase, and AP in IMI-negative cows, showing that LDH, NAGase, and AP are more affected than SCC by cow factors. The IMI status explained 23% of the variation in SCC in the model with all cows but only 7% of the variation in LDH and 2% of the variation in NAGase, indicating that SCC has the best potential as a diagnostic tool in finding cows with IMI. However, further studies are needed to investigate whether the diagnostic properties of these udder-health indicators will improve with adjustment according to their associations with different cow factors when used as a diagnostic tool for finding cows with IMI.

    • The objective of this study was to examine the dynamics of bovine corona virus (BCV) and bovine respiratory syncytial virus (BRSV) infections in dairy herds over a 3-year period. The status of 79 dairy herds located in two Northern and two Southern Regions of Sweden were surveyed by measuring antibody concentrations to BCV and BRSV in pooled milk samples from primiparous cows, and in bulk-tank milk twice annually. In the Southern Regions the percentage of antibody-positive herds remained persistently high (75-100%), whereas in herds based in the Northern Region, the percentage of positive herds for BCV and BRSV was 38-80% and 0-80%, respectively, with antibody levels to BRSV decreasing over time. Pooled milk samples of 'home-bred' primiparous animals were found to be most useful in terms of monitoring herd status but could gradually be replaced by bulk-tank sampling once freedom from infection was established.     • Methicillin-resistant Staphylococcus pseudintermedius (MRSP) strains have been isolated from dogs with increasing frequency; prolonged or excessive use of antimicrobials is associated with the selection of MRSP, and misuse of antimicrobials is frequent in breeding kennels. This study was carried out in two breeding kennels (A and B) in which we had isolated MRSP in 2008: the aim was to assess colonization of previously positive bitches and of other bitches sharing the same environment and to assess the genetic profile of both the old and the new strains [spa typing, staphylococcal cassette chromosome mec (SCCmec) typing and Pulsed-field Gel Electrophoresis (PFGE)]. Six animals from Kennel A (two from 2008) and eight from Kennel B (one from 2008) were tested: 16 MRSP strains were isolated only from bitches housed in Kennel B. Old and new isolates were mecA positive, resulted spa type t02 and carried SSCmec II-III. PGFE showed that all isolates were related and belonged to the main clone lineage dominating in Europe, ST71-J-t02-II-III. Kennels A and B differ in the use of antimicrobials, which has been reduced over time in Kennel A, while has remained excessive in Kennel B, where many agents belonging to different classes (third-generation cephalosporins, fluoroquinolones, macrolids) are administered to dogs, without veterinary supervision, especially around parturition. Misuse of antimicrobials is the key factor for the selection of MRSP strains in healthy dogs and for their persistence over time. Dog breeders should be aware that infections caused by multiresistant bacteria have very limited therapeutical options and represent a huge challenge for animal health.
