  • Interspecific competition affects spore germination and gametophore development of mosses

    • Background: Interactions between moss species in their earliest growth stages have received little attention. To what extent interspecific competition or priority effects influence spore germination, protonemal development and gametophore emergence is unknown. We evaluated such effects in pairwise interaction between six common bryophyte species: Atrichum undulatum, Bryum argenteum, Ceratodon purpureus, Funaria hygrometrica, Hypnum cupressiforme, Leptobryum pyriforme Methods: Interspecific interactions were assessed in vitro. Spores were sterilized and sown on agar plates in three treatments: 1) as single species cultures (controls), 2) as pairwise species cultures inoculated simultaneously, and 3) with a time lag of 20 days between species. Data on the time needed for spore germination, germination rate, the time needed for gametophore differentiation, number of gametophores per germinated spore and average diameter of colonies were collected. We also performed spore germination tests in single-species cultures at the start and end of the study, as well as tests for density-dependency at spore germination and gametophore formation. Results: We observed strong pairwise interactive effects when sowing spores of different species simultaneously or with a delay of 20 days. The results indicate that spore germination is often inhibited by interspecific competition. The first species has an advantage as compared to the later colonizing species, i.e., an apparent priority effect. Interspecific interactions were also evident during gametophore development and included both inhibition and facilitation. Conclusion: We found pronounced differences in the relative performance of species in interaction with other species during spore germination and gametophore formation. Allelopathic effects are the most probable explanation for these observations. Our results under sterile lab conditions are likely to reflect processes that occur in the wild, governing biotic filtering and bryophyte community assembly during primary and secondary colonization.

  • Creating personas for exposome research: the experience from the HEAP project

    • The exposome is a complex scientific field that has enjoyed consistent growth over the last two decades, defined as the composite of every exposure to which an individual is subjected from conception to death. The study of the exposome requires consideration of both the nature of those exposures and their changes over time, and as such necessitates high quality data and software solutions. As the exposome is both a broad and a recent concept, it is challenging to define or to introduce in a structured way. Thus, an approach to assist with clear definitions and a structured framework is needed for the wider scientific and public communication. Results: A set of 14 personas were developed through three focus groups and a series of 14 semi-structured interviews. The focus groups defined the broad themes specific to exposome research, while the sub-themes emerged to saturation via the interviews process. Personas are imaginary individuals that represent segments/groups of real people within a population. Within the context of the HEAP project, the created personas represented both exposome data generators and users. Conclusion: Personas have been implemented successfully in computer science, improving the understanding of human-computer interaction. The creation of personas specific to exposome research adds a useful tool supporting education and outreach activities for a complex scientific field.

  • Quasi-static and dynamic mechanical properties of a low-modulus bone cement for spinal applications

    • Background:Polymethylmethacrylate (PMMA) bone cement is extensively used in spinal procedures such as vertebroplasty and kyphoplasty, while its use in percutaneous cement discoplasty (PCD) is not yet widely spread. A main issue for both application sites, vertebra and disc, is the mismatch in stiffness between cement and bone, potentially resulting in adjacent vertebral fractures and adjacent segment disease. Tailoring the cement modulus using additives is hence an interesting strategy. However, there is a lack of data on the tensile and tension-compression fatigue properties of these cements, relevant to the newly researched indication of PCD. Method:A commercial PMMA cement (VS) was modified with 12%vol of linoleic acid (VSLA) and tested for quasi-static tensile properties. Additionally, tension-compression fatigue testing with amplitudes ranging from +/-5MPa to +/-7MPa and +/-9MPa was performed, and a Weibull three-parameter curve fit was used to calculate the fatigue parameters. Results:Quasi-static testing revealed a significant reduction in VSLA’s Young’s Modulus (E=581.1±126.4MPa) compared to the original cement (E=1478.1±202.9MPa). Similarly, the ultimate tensile stress decreased from 36.6±1.5MPa to 11.6±0.8MPa. Thus, VSLA offers improved compatibility with trabecular bone properties. Fatigue testing of VSLA revealed that as the stress amplitude increased the Weibull mean number decreased from 3591 to 272 and 91 cycles, respectively. In contrast, the base VS cement reached run-out at the highest stress amplitude. However, the lowest stress amplitude used exceeds the pressures recorded in the disc in vivo, and VSLA displayed a similar fatigue life range to that of the annulus fibrosis tissue.Conclusions:While the relevance of fully reversed tension-compression fatigue testing can be debated for predicting cement performance in certain spinal applications, the results of this study can serve as a benchmark for comparison of low-modulus cements for the spine. Further investigations are necessary to assess the clinical feasibility and effectiveness of these cements.

  • Comparing energy system optimization models and integrated assessment models: Relevance for energy policy advice

    •  BackgroundThe transition to a climate neutral society such as that envisaged in the European Union Green Deal requires careful and comprehensive planning. Integrated assessment models (IAMs) and energy system optimisation models (ESOMs) are both commonly used for policy advice and in the process of policy design. In Europe, a vast landscape of these models has emerged and both kinds of models have been part of numerous model comparison and model linking exercises. However, IAMs and ESOMs have rarely been compared or linked with one another.MethodsThis study conducts an explorative comparison and identifies possible flows of information between 11 of the integrated assessment and energy system models in the European Climate and Energy Modelling Forum. The study identifies and compares regional aggregations and commonly reported variables. We define harmonised regions and a subset of shared result variables that enable the comparison of scenario results across the models.ResultsThe results highlight how power generation and demand development are related and driven by regional and sectoral drivers. They also show that demand developments like for hydrogen can be linked with power generation potentials such as onshore wind power. Lastly, the results show that the role of nuclear power is related to the availability of wind resources.ConclusionsThis comparison and analysis of modelling results across model type boundaries provides modellers and policymakers with a better understanding of how to interpret both IAM and ESOM results. It also highlights the need for community standards for region definitions and information about reported variables to facilitate future comparisons of this kind. The comparison shows that regional aggregations might conceal differences within regions that are potentially of interest for national policy makers thereby indicating a need for national-level analysis.

We present the activities conducted within the Multi3Generation COST Action (CA18231), as well as current trends and future perspectives for multitask, multilingual and multimodal language generation.

    • Verification and validation (V&V) are complex processes combining different approaches and incorporating many different methods including many activities. System engineers regularly face the question if their V&V activities lead to better products, and having appropriate criteria at hand for evaluation of safety and cybersecurity of the systems would help to answer such a question. Additionally, when there is a demand to improve the quality of an already managed V&V process, there is a struggle over what criteria to use in order to measure the improvement. This paper presents an extensive set of criteria suitable for safety and cybersecurity evaluation of cyberphysical systems. The evaluation criteria are agreed upon by 60 researchers from 32 academic and industrial organizations jointly working in a large-scale European research project on 13 real-world use cases from the domains of automotive, railway, aerospace, agriculture, healthcare, and industrial robotics.

  • Ergolide mediates anti-cancer effects on metastatic uveal melanoma cells and modulates their cellular and extracellular vesicle proteomes

    • Background: Uveal melanoma is a poor prognosis cancer. Ergolide, a sesquiterpene lactone isolated from Inula Brittanica, exerts anti-cancer properties. The objective of this study was to 1) evaluate whether ergolide reduced metastatic uveal melanoma (MUM) cell survival/viability in vitro and in vivo; and 2) to understand the molecular mechanism of ergolide action. Methods: Ergolide bioactivity was screened via long-term proliferation assay in UM/MUM cells and in zebrafish MUM xenograft models. Mass spectrometry profiled proteins modulated by ergolide within whole cell or extracellular vesicle (EVs) lysates of the OMM2.5 MUM cell line. Protein expression was analyzed by immunoblots and correlation analyses to UM patient survival used The Cancer Genome Atlas (TCGA) data. Results: Ergolide treatment resulted in significant, dose-dependent reductions (48.5 to 99.9%; p<0.0001) in OMM2.5 cell survival in vitro and of normalized primary zebrafish xenograft fluorescence (56%; p<0.0001) in vivo, compared to vehicle controls. Proteome-profiling of ergolide-treated OMM2.5 cells, identified 5023 proteins, with 52 and 55 proteins significantly altered at 4 and 24 hours, respectively (p<0.05; fold-change >1.2). Immunoblotting of heme oxygenase 1 (HMOX1) and growth/differentiation factor 15 (GDF15) corroborated the proteomic data. Additional proteomics of EVs isolated from OMM2.5 cells treated with ergolide, detected 2931 proteins. There was a large overlap with EV proteins annotated within the Vesiclepedia compendium. Within the differentially expressed proteins, the proteasomal pathway was primarily altered. Interestingly, BRCA2 and CDKN1A Interacting Protein (BCCIP) and Chitinase Domain Containing 1 (CHID1), were the only proteins significantly differentially expressed by ergolide in both the OMM2.5 cellular and EV isolates and they displayed inverse differential expression in the cells versus the EVs. Conclusions: Ergolide is a novel, promising anti-proliferative agent for UM/MUM. Proteomic profiling of OMM2.5 cellular/EV lysates identified candidate pathways elucidating the action of ergolide and putative biomarkers of UM, that require further examination.   • Global sensitivity analysis to enhance the transparency and rigour of energy system optimisation modelling

    • Background: Energy system optimisation models (ESOMs) are commonly used to support long-term planning at national, regional, or continental scales. The importance of recognising uncertainty in energy system modelling is regularly commented on but there is little practical guidance on how to best incorporate existing techniques, such as global sensitivity analysis, despite some good applications in the literature.Methods: In this paper, we provide comprehensive guidelines for conducting a global sensitivity analysis of an ESOM, aiming to remove barriers to adopting this approach. With a pedagogical intent, we begin by exploring why you should conduct a global sensitivity analysis. We then describe how to implement a global sensitivity analysis using the Morris method in an ESOM using a sequence of simple illustrative models built using the Open Source energy Modelling System (OSeMOSYS) framework, followed by a realistic example.Results: Results show that the global sensitivity analysis identifies influential parameters that drive results in the simple and realistic models, and identifies uninfluential parameters which can be ignored or fixed. We show that global sensitivity analysis can be applied to ESOMs with relative ease using freely available open-source tools. The results replicate the findings of best-practice studies from the field demonstrating the importance of including all parameters in the analysis and avoiding a narrow focus on particular parameters such as technology costs.Conclusions: The results highlight the benefits of performing a global sensitivity analysis for the design of energy system optimisation scenarios. We discuss how the results can be interpreted and used to enhance the transparency and rigour of energy system modelling studies.
