    • After the fall of the Berlin wall and the end of the Cold War, a certain sense of optimism swept across Europe. Some 25 years later, everything seems radically different. With the considerable inroads made into mainstream politics by right-wing populist parties across the continent, there is no shortage of gloom and worry. Nevertheless, despite numerous examples of retrogressive forms of mobilization, there are also many cases of progressive mobilization. To capture this dynamic complexity, we posit politics as a site of struggle that constitutes an arena for the conflicting demands of the two master frames of nostalgia and hope. Following this logic of a polarized political terrain, the volume is divided into three parts that address both right-wing populist politics across Europe (Part I) and politics beyond party politics through either retrogressive mobilization (Part II) or emancipatory initiatives (Part III).

  • Integrated Assessment Modeling of Air Pollution in Four European Countries.

    • The integrated assessment modeling on acid rain has incorporated several related effects and pollutants into a multi-pollutant/multi-effect approach, resulting in complex integrated models and policy assessments. The development and implementation of effects-oriented cost-effective emission reduction strategies in Europe are based on integrated assessment models. The project on national integrated assessment modeling in Finland, Denmark, Spain and Sweden aimed to support the national evaluation of European emission reduction strategies. The tasks covered the comparison of inventories and projections for emissions of sulfur, nitrogen oxides, ammonia and volatile organic compounds, assessment of control techniques and related costs, concentration and deposition scenarios to estimate environmental effects of acidification, eutrophication and ground-level ozone and their temporal aspects, uncertainty analyses on both individual modules and whole integrated models, and dissemination of results to stakeholders. The integrated assessment modeling provided a consistent framework for the harmonization of input data and in-depth scientific research tasks on emissions, pollutant loading and impacts including comprehensive uncertainty analyses, and facilitated the dissemination of knowledge to policy-makers.

  • MCM3 upregulation confers endocrine resistance in breast cancer and is a predictive marker of diminished tamoxifen benefit

    • Resistance to endocrine therapy in estrogen receptor-positive (ER+) breast cancer is a major clinical problem with poorly understood mechanisms. There is an unmet need for prognostic and predictive biomarkers to allow appropriate therapeutic targeting. We evaluated the mechanism by which minichromosome maintenance protein 3 (MCM3) influences endocrine resistance and its predictive/prognostic potential in ER+ breast cancer. We discovered that ER+ breast cancer cells survive tamoxifen and letrozole treatments through upregulation of minichromosome maintenance proteins (MCMs), including MCM3, which are key molecules in the cell cycle and DNA replication. Lowering MCM3 expression in endocrine-resistant cells restored drug sensitivity and altered phosphorylation of cell cycle regulators, including p53(Ser(315,33)), CHK1(Ser(317)), and cdc25b(Ser(323)), suggesting that the interaction of MCM3 with cell cycle proteins is an important mechanism of overcoming replicative stress and anti-proliferative effects of endocrine treatments. Interestingly, the MCM3 levels did not affect the efficacy of growth inhibitory by CDK4/6 inhibitors. Evaluation of MCM3 levels in primary tumors from four independent cohorts of breast cancer patients receiving adjuvant tamoxifen mono-therapy or no adjuvant treatment, including the Stockholm tamoxifen (STO-3) trial, showed MCM3 to be an independent prognostic marker adding information beyond Ki67. In addition, MCM3 was shown to be a predictive marker of response to endocrine treatment. Our study reveals a coordinated signaling network centered around MCM3 that limits response to endocrine therapy in ER+ breast cancer and identifies MCM3 as a clinically useful prognostic and predictive biomarker that allows personalized treatment of ER+ breast cancer patients.

  • BMI in early adulthood is associated with severe COVID-19 later in life: A prospective cohort study of 1.5 million Swedish men

    • Objective: Overweight and obesity have been identified as risk factors for severe COVID-19; however, prospective cohort studies investigating the association between overweight early in life and severity of COVID-19 are lacking. Methods: This study included 1,551,670 Swedish men, born between 1950 and 1987, with BMI registered at age 18 years. They were followed until January 9, 2021. COVID-19 cases and comorbidities were identified through the National Patient, Intensive Care, and Cause of Death registries. Outcomes included the following: 1) hospitalization; 2) intensive care unit admission; and 3) death. Results: The study found 4,315 cases (mean age = 56.4 years [SD 8.8]) of patients hospitalized because of COVID-19, of which 729 were admitted to an intensive care unit, and altogether there were 224 deaths. The risk for hospital admission increased with higher values of BMI at age 18 years, despite adjustment for comorbidities, from an odds ratio (OR) of 1.19 (95% CI: 1.08-1.31) at BMI = 22.5 to 25 to an OR of 1.68 (95% CI: 1.39-2.02) at BMI >= 30, compared with BMI = 18.5 to 20. ORs for intensive care unit admission were 1.44 (95% CI: 1.13-1.84) at BMI = 22.5 to 25 and 2.61 (95% CI: 1.73-3.93) at BMI >= 30. Conclusions: Higher BMI in early adulthood was associated with severe COVID-19 many years later, with a risk increase starting already at BMI >= 22.5. This underlines the necessity of preventive actions against overweight in youth to offer protection against coming viral pandemics.

    • Dalton is a powerful general-purpose program system for the study of molecular electronic structure at the Hartree-Fock, Kohn-Sham, multiconfigurational self-consistent-field, MOller-Plesset, configuration-interaction, and coupled-cluster levels of theory. Apart from the total energy, a wide variety of molecular properties may be calculated using these electronic-structure models. Molecular gradients and Hessians are available for geometry optimizations, molecular dynamics, and vibrational studies, whereas magnetic resonance and optical activity can be studied in a gauge-origin-invariant manner. Frequency-dependent molecular properties can be calculated using linear, quadratic, and cubic response theory. A large number of singlet and triplet perturbation operators are available for the study of one-, two-, and three-photon processes. Environmental effects may be included using various dielectric-medium and quantum-mechanics/molecular-mechanics models. Large molecules may be studied using linear-scaling and massively parallel algorithms. Dalton is distributed at no cost from for a number of UNIX platforms.

    • CDK4/6 inhibitors (CDK4/6i) combined with endocrine therapy have shown impressive efficacy in estrogen receptor-positive advanced breast cancer. However, most patients will eventually experience disease progression on this combination, underscoring the need for effective subsequent treatments or better initial therapies. Here, we show that triple inhibition with fulvestrant, CDK4/6i and AKT inhibitor (AKTi) durably impairs growth of breast cancer cells, prevents progression and reduces metastasis of tumor xenografts resistant to CDK4/6i-fulvestrant combination or fulvestrant alone. Importantly, switching from combined fulvestrant and CDK4/6i upon resistance to dual combination with AKTi and fulvestrant does not prevent tumor progression. Furthermore, triple combination with AKTi significantly inhibits growth of patient-derived xenografts resistant to combined CDK4/6i and fulvestrant. Finally, high phospho-AKT levels in metastasis of breast cancer patients treated with a combination of CDK4/6i and endocrine therapy correlates with shorter progression-free survival. Our findings support the clinical development of ER, CDK4/6 and AKT co-targeting strategies following progression on CDK4/6i and endocrine therapy combination, and in tumors exhibiting high phospho-AKT levels, which are associated with worse clinical outcome.

    • The special issue contributes to the exploration of transversal solidarities counterpoised to anexhausted neoliberalism on the one hand and a xenophobic populism on the other. It trackscontours of a multifarious countermovement, traversing ‘race’, class and gender, driven byreimaginings of the common and the renewal of democracy. The emphasis is on the understandingof contending urban justice movements, welcoming communities and their liaisons in a multiscale(local, national, transnational) perspective. A collection of theoretically informed papers discussescases from urban contexts of Europe and the United States, all riveted by schisms of class, ‘race’/ethnicity and gender, occupied by the ‘migration’ issue and challenged by contending movementsfor social cum environmental sustainability. Exploring examples of social movements and formsof mobilisation in different contexts, the overarching aim is to retrieve options for transversalsolidarities transcending identities while focusing on commonalities.

    • A multi-residue pressurised liquid extraction (PLE) methodology has been established for the determination of the four ionophores: lasalocid, monensin, salinomycin and narasin in solid environmental matrices. The PLE methodology is combined with solid phase extraction as clean-up using liquid chromatography coupled to tandem mass spectrometry applying electrospray ionisation for detection. The samples were freeze-dried prior to extraction. The absolute recoveries for soil and sediment ranged from 71 to 123% (relative standard deviation (RSDs) below 16%) and in the range 94–133% (RSDs 9–35%) for poultry manure. The final method allowed for the detection of four ionophores down to a few hundred ng kg−1 in natural solid matrices with limit of quantifications (LOQs) being 0.96, 0.87, 0.98, and 0.64 μg kg−1 in soil for lasalocid, monensin, salinomycin, and narasin, respectively. Corresponding LOQs in sediment were 1.28, 1.34, 1.39, and 0.78 μg kg−1 for the respective ionophores, while in manure the LOQs were 0.98, 1.01, 1.45, and 1.01 μg kg−1.

    • This study is the first to investigate the pharmaceutical burden from point sources affecting the UNESCO Biosphere Reserve Kristianstads Vattenrike, Sweden. The investigated Biosphere Reserve is a >1000 km(2) wetland system with inflows from lakes, rivers, leachate from landfill, and wastewater-treatment plants (WWTPs). We analysed influent and treated wastewater, leachate water, lake, river, and wetland water alongside sediment for six model pharmaceuticals. The two WWTPs investigated released pharmaceutical residues at levels close to those previously observed in Swedish monitoring exercises. Compound-dependent WWTP removal efficiencies ranging from 12 to 100 % for bendroflumethiazide, oxazepam, atenolol, carbamazepine, and diclofenac were observed. Surface-water concentrations in the most affected lake were ≥100 ng/L for the various pharmaceuticals with atenolol showing the highest levels (>300 ng/L). A small risk assessment showed that adverse single-substance toxicity on aquatic organisms within the UNESCO Biosphere Reserve is unlikely. However, the effects of combinations of a large number of known and unknown pharmaceuticals, metals, and nutrients are still unknown.

    • The protein parkin, encoded by the PARK2 gene, is vital for mitochondrial homeostasis, and although it has been implicated in Parkinson’s disease (PD), the disease mechanisms remain unclear. We have applied mass spectrometry-based proteomics to investigate the effects of parkin dysfunction on the mitochondrial proteome in human isogenic induced pluripotent stem cell-derived neurons with and without PARK2 knockout (KO). The proteomic analysis quantified nearly 60% of all mitochondrial proteins, 119 of which were dysregulated in neurons with PARK2 KO. The protein changes indicated disturbances in oxidative stress defense, mitochondrial respiration and morphology, cell cycle control, and cell viability. Structural and functional analyses revealed an increase in mitochondrial area and the presence of elongated mitochondria as well as impaired glycolysis and lactate-supported respiration, leading to an impaired cell survival in PARK2 KO neurons. This adds valuable insight into the effect of parkin dysfunction in human neurons and provides knowledge of disease-related pathways that can potentially be targeted for therapeutic intervention.

The hydrolysis study was carried out by dissolving the ionophores in solutions of pH 4, 7, and 9, followed by incubation at three temperatures of 6, 22, and 28 °C for maximum 34 days. Using LC–MS/MS for chemical analysis, lasalocid was not found to hydrolyse in any of the tested environments. Monensin, salinomycin, and narasin were all stable in neutral or alkaline solution but hydrolysed in the solution with a pH of 4. Half-lives at 25 °C were calculated to be 13, 0.6, and 0.7 days for monensin, salinomycin, and narasin, respectively. Absorbance spectra from each compound indicated that only lasalocid is degraded by photolysis (half-life below 1 h) due to an absorbance maximum around 303 nm, and monensin, salinomycin, and narasin are resistant to direct photolysis because they absorb light of environmentally irrelevant wavelengths.

    • This contribution examines, in the Central and Eastern European context, the interplay between ideals of national specificity, welfare chauvinist appeals, and emerging politics of migration, for the purpose of providing welfare provision to a narrowly defined ethnic group, as promoted by right-wing populist parties in the region. We suggest a comparative framework to account the various positions that such parties occupy in the mainstream political systems in Central and Eastern Europe. Our study deals with the case of a right-wing populist party becoming the main governing force, such as the Law and Justice Party (Prawo i Sprawiedliwość, PiS) in Poland; the case of a right-wing populist party as key opposition force, such as the Movement for a Better Hungary (Jobbik Magyarországért Mozgalom, Jobbik) in Hungary; and thirdly, the case of an unsuccessful right-wing populist party, such as the United Romania Party (Partidul România Unită, PRU). For our qualitative analysis we are drawing on the official discourses of these parties as articulated from 2015 onwards, since it marks the beginning of what has come to be referred to as the European refugee crisis. The aim of this chapter is to map out the various electoral strategies employed, with varying degrees of success, which juxtapose cultural protectionist appeals to welfare chauvinist proposals, and consequently shed light on the culture and welfare nexus in the Central and Eastern European context.

The volume offers a collection of critical essays on human rights movements, sanctuary spaces, and the emplacement of antiracist conviviality in cities across North and South America, Europe, and Africa. Each intervention proceeds from the idea that cities may accommodate both a humanistic sensibility and a radical potential for social transformation. The figure of the 'migrant' is pivotal. It expounds the prospect of transversal solidarity to capture a plurality of commonalities and to abjure dichotomies between in-group and out-group, the national and the international, or society and institutions.

    • DIII-D physics research addresses critical challenges for the operation of ITER and the next generation of fusion energy devices. This is done through a focus on innovations to provide solutions for high performance long pulse operation, coupled with fundamental plasma physics understanding and model validation, to drive scenario development by integrating high performance core and boundary plasmas. Substantial increases in off-axis current drive efficiency from an innovative top launch system for EC power, and in pressure broadening for Alfven eigenmode control from a co-/counter-I p steerable off-axis neutral beam, all improve the prospects for optimization of future long pulse/steady state high performance tokamak operation. Fundamental studies into the modes that drive the evolution of the pedestal pressure profile and electron vs ion heat flux validate predictive models of pedestal recovery after ELMs. Understanding the physics mechanisms of ELM control and density pumpout by 3D magnetic perturbation fields leads to confident predictions for ITER and future devices. Validated modeling of high-Z shattered pellet injection for disruption mitigation, runaway electron dissipation, and techniques for disruption prediction and avoidance including machine learning, give confidence in handling disruptivity for future devices. For the non-nuclear phase of ITER, two actuators are identified to lower the L-H threshold power in hydrogen plasmas. With this physics understanding and suite of capabilities, a high poloidal beta optimized-core scenario with an internal transport barrier that projects nearly to Q = 10 in ITER at ∼8 MA was coupled to a detached divertor, and a near super H-mode optimized-pedestal scenario with co-I p beam injection was coupled to a radiative divertor. The hybrid core scenario was achieved directly, without the need for anomalous current diffusion, using off-axis current drive actuators. Also, a controller to assess proximity to stability limits and regulate β N in the ITER baseline scenario, based on plasma response to probing 3D fields, was demonstrated. Finally, innovative tokamak operation using a negative triangularity shape showed many attractive features for future pilot plant operation.

    • Background: Prognostic factors in breast cancer are often measured on a continuous scale, but categorized for clinical decision-making. The primary aim of this study was to evaluate if accounting for continuous non-linear effects of the three factors age at diagnosis, tumor size, and number of positive lymph nodes improves prognostication. These factors will most likely be included in the management of breast cancer patients also in the future, after an expected implementation of gene expression profiling for adjuvant treatment decision-making. Methods: Four thousand four hundred forty seven and 1132 women with primary breast cancer constituted the derivation and validation set, respectively. Potential non-linear effects on the log hazard of distant recurrences of the three factors were evaluated during 10 years of follow-up. Cox-models of successively increasing complexity: dichotomized predictors, predictors categorized into three or four groups, and predictors transformed using fractional polynomials (FPs) or restricted cubic splines (RCS), were used. Predictive performance was evaluated by Harrell's C-index. Results: Using FP-transformations, non-linear effects were detected for tumor size and number of positive lymph nodes in univariable analyses. For age, non-linear transformations did, however, not improve the model fit significantly compared to the linear identity transformation. As expected, the C-index increased with increasing model complexity for multivariable models including the three factors. By allowing more than one cut-point per factor, the C-index increased from 0.628 to 0.674. The additional gain, as measured by the C-index, when using FP- or RCS-transformations was modest (0.695 and 0.696, respectively). The corresponding C-indices for these four models in the validation set, based on the same transformations and parameter estimates from the derivation set, were 0.675, 0.700, 0.706, and 0.701. Conclusions: Categorization of each factor into three to four groups was found to improve prognostication compared to dichotomization. The additional gain by allowing continuous non-linear effects modeled by FPs or RCS was modest. However, the continuous nature of these transformations has the advantage of making it possible to form risk groups of any size.

    • As programming is typically a static activity in front of a screen, we perform an initial exploration around the capabilities of block-based programming in the immersive space using Virtual Reality (VR) to make an early charting on how programming could involve moving the programmer's body. We created a block-based programming interface in a VR space called BlocklyVR based on the existing Blockly programming environment. To investigate programmer performance and experience in BlocklyVR, we conducted a controlled lab experiment (N = 20) with eight programming tasks that covered mathematical operations, if-statements, and function creation. Our initial exploration contributes by classifying movement types made by BlocklyVR programmers and reflecting on how these movements are related to the programming tasks. Additionally, our data suggests that participant performance in BlocklyVR was not affected compared to the 2D Blockly, even if participants were physically moving in VR space. We also found that the virtual reality sickness was marginal. Lastly, we identified four types of interaction that can potentially be employed by VR designers and developers aiming to convert a static task, like programming at a desk, into a "mobile"immersive experience. © 2023 Owner/Author.

    • Two of every three persons living with dementia reside in low- and middle-income countries (LMICs). The projected increase in global dementia rates is expected to affect LMICs disproportionately. However, the majority of global dementia care costs occur in high-income countries (HICs), with dementia research predominantly focusing on HICs. This imbalance necessitates LMIC-focused research to ensure that characterization of dementia accurately reflects the involvement and specificities of diverse populations. Development of effective preventive, diagnostic, and therapeutic approaches for dementia in LMICs requires targeted, personalized, and harmonized efforts. Our article represents timely discussions at the 2022 Symposium on Dementia and Brain Aging in LMICs that identified the foremost opportunities to advance dementia research, differential diagnosis, use of neuropsychometric tools, awareness, and treatment options. We highlight key topics discussed at the meeting and provide future recommendations to foster a more equitable landscape for dementia prevention, diagnosis, care, policy, and management in LMICs. HIGHLIGHTS: Two-thirds of persons with dementia live in LMICs, yet research and costs are skewed toward HICs. LMICs expect dementia prevalence to more than double, accompanied by socioeconomic disparities. The 2022 Symposium on Dementia in LMICs addressed advances in research, diagnosis, prevention, and policy. The Nairobi Declaration urges global action to enhance dementia outcomes in LMICs.

    • AimsTo assess the variation in ovarian carcinoma type diagnosis among gynaecological pathologists from Nordic countries, and whether a rationally designed panel of immunohistochemical markers could improve diagnostic reproducibility. Methods and resultsEight pathologists from four countries (Sweden, Denmark, Norway, and Finland) received an educational lecture on the diagnosis of ovarian carcinoma type. All tumour-containing slides from 54 ovarian carcinoma cases were independently reviewed by the participants, who: (i) determined type purely on the basis of histology; (ii) indicated whether they would apply immunohistochemistry in their routine practice; and (iii) determined type after reviewing the staining results. The results for six markers (WT1, p53, p16, HNF-1, ARID1A, and progesterone receptor) were determined for all 54 cases, by staining of a tissue microarray. The median concordance with central review diagnosis was 86%, and significantly improved to 90% with the incorporation of immunostaining results (P=0.0002). The median interobserver agreement was 78%, and significantly improved to 85% with the incorporation of immunostaining results (P=0.0002). ConclusionsUse of the immunostaining results significantly improved both diagnostic accuracy and interobserver agreement. These results indicate that ovarian carcinoma type can be reliably diagnosed by pathologists from different countries, and also demonstrate that immunohistochemistry has an important role in improving diagnostic accuracy and agreement between pathologists.

  • Constructing Workers of Migrant Background : A study of class and nation myths in trade union practice

    • How do trade unions represent workers of migrant background? How are factors that affect the quality of life of workers of migrant background, made sense of and responded to by trade unions trying to reverse declining trade union membership numbers? This thesis takes a qualitative approach and utilises discursive constructions as a semiotic entry point into the social practices of trade unions and their responses to workers of migrant background. Combining semiotic theory of Roland Barthes and Stuart Hall with critical discourse theory of Norman Fairclough, trade union work is conceptualised as a social practice that comprises both semiotic and non-semiotic elements. The study focuses on the discursive work of two unions in Copenhagen, Denmark that represent the bike mechanic and the cleaning trade, respectively. The selection of unions allows for an exploration of similarities and differences within the national setting. The analysis is based on individual and group interviews with 28 union officials and active members, supported and complemented by notes (387 pages) from 56 participant observations (147 observation hours) that were conducted in the two unions over several years. The analysis shows how workers of migrant background are constructed through discourses of class and nation. Analysing the interrelationship between semiotic and non-semiotic elements of union events and practices, a four-cell typology is developed that characterise union responses as either internalisation, externalisation, ignorance or misrepresentation. The type of response is defined according to whether the unions signify their constructions of migrant differences as relevant or irrelevant to union work, and whether they address these differences with uniform or differentiated union action. It is argued that the ways in which constructions of national, ethnic and racial differences are responded to through signification and action are crucial to whether unions work to reinforce or ameliorate social inequalities. Finally, the typology is suggested as a tool to nuance the discussion about the inclusive and/or exclusive character of trade union responses to workers of migrant background.

  • NKL-1915 National Nitrogen Budgets in Scandinavia: consequences for climate change and for eutrophication – synthesis report

    • This report presents the results of a project undertaken by Norway, Finland, Sweden, and Denmark related to the National Nitrogen Budget (NNB) with a focus on the NNB pool Forests and semi-natural vegetation (FS). The NNB involves calculations of reactive nitrogen (Nr) inflows and outflows within the FS pool, and the study compares these calculations to reports of carbon sequestration in climate reporting to the United Nations Framework Convention on Climate Change (UNFCCC).Norway and Finland initiated their work on NNB, concentrating on Forests and semi-natural vegetation (FS). The UNECE LRTAP Task Force on Reactive Nitrogen (TFRN) methodology was also used for compiling the FS pool for Denmark, while Sweden had established its FS pool prior to the project.The FS pool comprises three sub-pools: forests, wetlands, and other lands. Inflows of reactive nitrogen (Nr) to the FS pool are primarily from deposition and biological N-fixation, with major outflows being leaching, harvest, and denitrification.In Finland, Sweden, and Denmark, the inflows and outflows are in balance, while in Norway, there is an accumulation of Nr, indicating a surplus of nitrogen. Wetland sub-pools in all four countries and the forest sub-pool in Norway show nitrogen accumulation.All four countries report carbon sequestration in forests to UNFCCC.There is a link between nitrogen and carbon cycles, since the organic matter forming the carbon stock in forests and wetlands relies on nitrogen. The origin of nitrogen contributing to the estimated C-stock increase needs further investigation.There is a discrepancy between the calculated nitrogen changes in the FS pool and the nitrogen needed to support reported carbon stock changes. Possible explanations for the discrepancies are re-location of nitrogen within pools, changes in the C/N ratio, expansion of forested areas, and uncertainties in N-fixation rates and denitrification.There is a need for better integration of carbon and nitrogen flux and stock change estimates in natural ecosystems. Understanding the interactions between carbon and nitrogen is deemed crucial for predicting future developments in carbon sequestration and impacts of nitrogen deposition, especially for the Nordic countries with their significant forests playing roles as carbon and nitrogen sinks.

    • This open access book shows how the politics of migration affect community building in the 21st century, drawing on both retrogressive and progressive forms of mobilization. It elaborates theoretically and shows empirically how the two master frames of nostalgia and hope are used in local, national and transnational settings, in and outside conventional forms of doing politics. It expands on polarized societal processes and external events relevant for the transformation of European welfare systems and the reproduction of national identities today. It evidences the importance of gender in the narrative use of the master frames of nostalgia and hope, either as an ideological tool for right-wing populist and extreme right retrogressive mobilization or as an essential element of progressive intersectional politics of hope. It uses both comparative and single case studies to address different perspectives, and by means of various methodological approaches, the manner in which the master frames of nostalgia and hope are articulated in the politics of culture, welfare, and migration. The book is organized around three thematic sections whereby the first section deals with right-wing populist party politics across Europe, the second section deals with an articulation of politics beyond party politics by means of retrogressive mobilization, and the third and last section deals with emancipatory initiatives beyond party politics as well.

The methodology was applied to various drug products including tablets, capsules, suspensions, syrups, intravenous and injection solutions as well as ear and eye droplets used as essential medicines in a Sub-Saharan country, Ghana.

    • Neurotropic viruses, including herpes simplex virus (HSV) types 1 and 2, have the capacity to infect neurons and can cause severe diseases. This is associated with neuronal cell death, which may contribute to morbidity or even mortality if the infection is not controlled. However, the mechanistic details of HSV-induced neuronal cell death remain enigmatic. Here, we report that lytic HSV-2 infection of human neuron-like SH-SY5Y cells and primary human and murine brain cells leads to cell death mediated by gasdermin E (GSDME). HSV-2-induced GSDME-mediated cell death occurs downstream of replication-induced endoplasmic reticulum stress driven by inositol-requiring kinase 1 alpha (IRE1 alpha), leading to activation of caspase-2, cleavage of the pro-apoptotic protein BH3interacting domain death agonist (BID), and mitochondriadependent activation of caspase-3. Finally, necrotic neurons released alarmins, which activated inflammatory responses in human iPSC-derived microglia. In conclusion, lytic HSV infection in neurons activates an ER stress-driven pathway to execute GSDMEmediated cell death and promote inflammation.

The background activities carried out by the CATRIN IMs in preparation of this report have been: 1. A survey among IMs, in which the CATRIN IMs have reviewed the current pricing principles and have expressed their positions concerning the adoption of other principles in the formulation and implementation of pricing policies. 2. Interviews with the IMs in order to discuss cost allocation methods in use, for which the review of current practices has led to the identification and discussion of the most relevant issues at stake, problems, uncertainties and achievements. 3. Meetings with IMs in which the CATRIN IMs have provided input to the case studies and discussed the possibility to implement the CATRIN recommendations, as emerged from the CATRIN case studies.

    • The current special issue examines the range and strength of analysing contemporary transformations and struggles through the lens of ‘precarity’. Rather than defining a single precariat, the interest is in exploring ‘varieties of precarity’. These take different forms in different parts of the world, on different scales and in different socio-economic contexts, and yet they share certain characteristics in terms of conditions and capacity for agency. Contributions to this volume testify that precarity may be a political proposition as much as a sociological category that offers an analytical description of current transformations. The selection of articles has the ‘politics of precarity’ as a frame of reference. It describes the political economy of neoliberal globalization producing institutionally embedded precarization of labour, livelihoods and citizenship, but also resistance against the systemic structuration within which it is embedded.

  • Towards Operator-managed P2P Content Delivery with Application Layer Traffic Optimization

    • P2P technology provides a flexible and very popular way of content delivery for various services, including networked media applications. However, P2P-based content delivery generates large amount of backbone traffic. Recently, several approaches have been proposed for guiding P2P services based on operator preferences, in order to reduce the amount of costly backbone traffic generated by P2P applications. Application Layer Traffic Optimization (ALTO) is a one key approach for such managed P2P applications. In short, ALTO is a dedicated service, operated by a network operator or ISP, which can provide useful network layer information to application layer clients for improved peer selection and corresponding content delivery. This paper provides an overview of standardisation and research activities for improving P2P content delivery with ALTO which are carried out within the EU FP7 project NAPA-WINE. In particular, we give an overview on standardisation efforts, present simulation results, explain our prototypical implementation of the ALTO concept, and highlight ongoing large-scale operational trials we are currently conducting within the network of Polish Telecom and among NAPA-WINE partners

    • Background: The aim was to confirm a previously defined prognostic index, combining a proliferation marker, histological grade, and estrogen receptor (ER) in different subsets of primary N0/N1 chemo-naive breast cancer patients. Methods/design: In the present study, including 1,854 patients, Ki67 was used in the index (KiGE), since it is the generally accepted proliferation marker in clinical routine. The low KiGE-group was defined as histological grade 1 patients and grade 2 patients which were ER-positive and had low Ki67 expression. All other patients made up the high KiGE-group. The KiGE-index separated patients into two groups with different prognosis. In multivariate analysis, KiGE was significantly associated with disease-free survival, when adjusted for age at diagnosis, tumor size and adjuvant endocrine treatment (hazard ratio: 3.5, 95% confidence interval: 2.6-4.7, P<0.0001). Discussion: We have confirmed a prognostic index based on a proliferation marker (Ki67), histological grade, and ER for identification of a low-risk group of patients with N0/N1 primary breast cancer. For this low-risk group constituting 57% of the patients, with a five-year distant disease-free survival of 92%, adjuvant chemotherapy will have limited effect and may be avoided.
