  • Effects of chemotherapy and hormonal therapy for early breast cancer on recurrence and 15-year survival: an overview of the randomised trials

    • Background Quinquennial overviews (1985-2000) of the randomised trials in early breast cancer have assessed the 5-year and 10-year effects of various systemic adjuvant therapies on breast cancer recurrence and survival. Here, we report the 10-year and 15-year effects. Methods Collaborative meta-analyses were undertaken of 194 unconfounded randomised trials of adjuvant chemotherapy or hormonal therapy that began by 1995. Many trials involved CMF (cyclophosphamide, methotrexate, fluorouracil), anthracycline-based combinations such as FAC (fluorouracil, doxombicin, cyclophosphamide) or FEC (fluorouracil, epirubicin, cyclophosphamide), tamoxifen, or ovarian suppression: none involved taxanes, trastuzumab, raloxifene, or modem aromatase inhibitors. Findings Allocation to about 6 months of anthracycline-based polychemotherapy (eg, with FAC or FEC) reduces the annual breast cancer death rate by about 38% (SE 5) for women younger than 50 years of age when diagnosed and by about 20% (SE 4) for those of age 50-69 years when diagnosed, largely irrespective of the use of tamoxifen and of oestrogen receptor (ER) status, nodal status, or other tumour characteristics. Such regimens are significantly (2p=0 . 0001 for recurrence, 2p<0 . 00001 for breast cancer mortality) more effective than CMF chemotherapy. Few women of age 70 years or older entered these chemotherapy trials. For ER-positive disease only, allocation to about 5 years of adjuvant tamoxifen reduces the annual breast cancer death rate by 31% (SE 3), largely irrespective of the use of chemotherapy and of age (<50, 50-69, &GE; 70 years), progesterone receptor status, or other tumour characteristics. 5 years is significantly (2p<0 . 00001 for recurrence, 2p=0 . 01 for breast cancer mortality) more effective than just 1-2 years of tamoxifen. For ER-positive tumours, the annual breast cancer mortality rates are similar during years 0-4 and 5-14, as are the proportional reductions in them by 5 years of tamoxifen, so the cumulative reduction in mortality is more than twice as big at 15 years as at 5 years after diagnosis. These results combine six meta-analyses: anthracycline-based versus no chemotherapy (8000 women); CMF-based versus no chemotherapy (14 000); anthracycline-based versus CMF-based chemotherapy (14 000); about 5 years of tamoxifen versus none (15 000); about 1-2 years of tamoxifen versus none (33 000); and about 5 years versus 1-2 years of tamoxifen (18 000). Finally, allocation to ovarian ablation or suppression (8000 women) also significantly reduces breast cancer mortality, but appears to do so only in the absence of other systemic treatments. For middle-aged women with ER-positive disease (the commonest type of breast cancer), the breast cancer mortality rate throughout the next 15 years would be approximately halved by 6 months of anthracycline-based chemotherapy (with a combination such as FAC or FEC) followed by 5 years of adjuvant tamoxifen. For, if mortality reductions of 38% (age <50 years) and 20% (age 50-69 years) from such chemotherapy were followed by a further reduction of 31% from tamoxifen in the risks that remain, the final mortality reductions would be 57% and 45%, respectively (and, the trial results could well have been somewhat stronger if there had been full compliance with the allocated treatments). Overall survival would be comparably improved, since these treatments have relatively small effects on mortality from the aggregate of all other causes. Interpretation Some of the widely practicable adjuvant drug treatments that were being tested in the 1980s, which substantially reduced 5-year recurrence rates (but had somewhat less effect on 5-year mortality rates), also substantially reduce 15-year mortality rates. Further improvements in long-term survival could well be available from newer drugs, or better use of older drugs.

    • Excited states in Xe-124 were populated in the reaction Se-82(Ca-48, 6n) Xe-124 and gamma-ray coincidence relationships were measured with the Gammasphere spectrometer. Two new bands are observed and several of the previously known bands are extended in the high-as well as in the low-spin region. Two irregular high-spin structures are also added. The irregularities are a fingerprint of a transition from collective to non-collective behaviour. Configuration assignments to the new structures are proposed on the basis of systematics and by comparing experimental properties with calculations within the framework of the cranking model.

    • Patients with acromegaly are currently treated with surgery, radiation therapy, and drugs to reduce hypersecretion of growth hormone, but the treatments may be ineffective and have adverse effects. Pegvisomant is a genetically engineered growth hormone-receptor antagonist that blocks the action of growth hormone.We conducted a 12-week, randomized, double-blind study of three daily doses of pegvisomant (10 mg, 15 mg, and 20 mg) and placebo, given subcutaneously, in 112 patients with acromegaly.The mean (+/-SD) serum concentration of insulin-like growth factor I (IGF-I) decreased from base line by 4.0+/-16.8 percent in the placebo group, 26.7+/-27.9 percent in the group that received 10 mg of pegvisomant per day, 50.1+/-26.7 percent in the group that received 15 mg of pegvisomant per day, and 62.5+/-21.3 percent in the group that received 20 mg of pegvisomant per day (P<0.001 for the comparison of each pegvisomant group with placebo), and the concentrations became normal in 10 percent, 54 percent, 81 percent, and 89 percent of patients, respectively (P<0.001 for each comparison with placebo). Among patients treated with 15 mg or 20 mg of pegvisomant per day, there were significant decreases in ring size, soft-tissue swelling, the degree of excessive perspiration, and fatigue. The score fortotal symptoms and signs of acromegaly decreased significantly in all groups receiving pegvisomant (P< or =0.05). The incidence of adverse effects was similar in all groups.On the basis of these preliminary results, treatment of patients who have acromegaly with a growth hormone-receptor antagonist results in a reduction in serum IGF-I concentrations and in clinical improvement.

  • Long-term follow-up of the SBG 9401 study comparing tailored FEC-based therapy versus marrow-supported high-dose therapy

    • Background: The purpose was to investigate adjuvant marrow-supportive high-dose chemotherapy compared with an equitoxicity-tailored comparator arm. Patients and methods: Five hundred and twenty-five women below theage of 60 years with operated high-risk primary breast cancer were randomised to nine cycles of granulocyte colony-stimulating factor supported and individually tailored FEC (5-fluorouracil, epirubicin, cyclophosphamide), (n = 251) or standard FEC followed by marrow-supported high-dose therapy with CTCb (cyclophosphamide, thiotepa, carboplatin) therapy (n = 274), followed by locoregional radiotherapy and tamoxifen for 5 years. Results: There were 104 breast cancer relapses in the tailored FEC group versus 139 in the CTCb group (double triangular method by Whitehead, P = 0.046), with a median follow-up of all included patients of 60.8 months. The event-free survival demonstrated 121 and 150 events in the tailored FEC- and CTCb group, respectively [P = 0.074, hazard ratio (HR) 0.804, 95% confidence interval (CI) 0.633-1.022]. Ten patients in the tailored FEC regimen developed acute myeloid leukaemia (AML)/myelodysplasia (MDS). One hundred deaths occurred in the tailored FEC group and 121 in the CTCb group (P = 0.287, HR 0.866, 95% CI 0.665-1.129). Conclusion: The update of this study shows an improved outcome linked to the tailored FEC treatment in relation to breast cancer relapse, but also an increased incidence of AML/MDS. © 2007 Oxford University Press.

    • Volatile compounds were collected from apple branches (Malus domestica) at different developmental stages, and the antennal response of codling moth females (Cydia pomonella) to these compounds was recorded by electroantennography coupled to gas chromatography. Presence of a range of terpenoid compounds, many of which had antennal activity, was characteristic for volatile collections from branches with leaves, and from small green apples. Nine compounds from branches with leaves and green fruit consistently elicited an antennal response: methyl salicylate, (E)-beta -farnesene, fi-caryophyllene, 4,8-dimethyl-1,3(E),7-nonatriene, (Z)3-hexenol, (Z,E)-alpha -farnesene, linalool, germacrene D, and (EE)-alpha -farnesene. The bouquet emitted from flowering branches contained in addition several benzenoid compounds which were not found after bloom. Small green apples, which are the main target of codling moth oviposition during the first seasonal flight period, released very few esters. In comparison, fully grown apples released a large number of esters, but fewer terpenoids. The study of apple volatiles eliciting an antennal response, together with a survey of the seasonal change in the release of these compounds, is the first step toward the identification of volatiles mediating host-finding and oviposition in codling moth females.

    • Single sensillum recordings from Cydia pomonella male antennae showed three different types of receptor neurons. The most abundant type was most sensitive to the main pheromone compound (E,E)-8,10-dodecadienol, while its response to the geometric isomers E,Z, Z,E and Z,Z was comparable to a tenfold lower dose of (E,E)-8,10-dodecadienol. This neuron type also responded to the four behaviorally antagonistic isomers of (Delta,Delta)-8,10-dodecadienyl acetate, among which it was most sensitive to the E,E isomer. Cross-adaptation studies showed that these compounds were all detected by the same receptor neuron type. Receptor neurons specifically tuned to (E,Z) or (Z,Z)-8,10-dodecadienol were not found, although these two compounds are behaviorally active. A second type of receptor neuron responded to all isomers of (Delta,Delta)-8,10-dodecadienyl acetate and was most sensitive to the E,E isomer. This neuron type did not respond to any of the isomers of (Delta,Delta)-8,10-dodecadienol. A third receptor neuron type was highly sensitive to the plant compound alpha-farnesene. The finding that the receptor neuron type tuned to the main pheromone compound responded even to strong behavioral antagonists aids the interpretation of ongoing behavioral studies for the development of the mating disruption technique in codling moth.

    • Control and reduction of soot particle emissions from aeronautic turbines requires a monitoring system suitable for quantification of these emissions. Currently, such emissions are estimated using the technique of smoke number. This is an extractive method, which is not sensitive enough for the low emission levels of modern gas turbines. Within a recent European project, AEROTEST, part of the project aimed at investigating an alternative soot monitoring technique, laser-induced incandescence (LII) as an in-situ optical diagnostic for quantification of soot emissions. For aero-engine applications, especially those involving large-scale turbines, it is necessary to perform the measurements at long distance from the turbine. The LII technique is favourable in this respect as it provides for non-intrusive measurements and, by detecting the isotropic LII signal along the same axis as the incoming laser beam (so called backward LII), both the laser and the detector can be built inside one system located several meters from the turbine. The concept was initiated in the previous European projects, AEROJET I and II. This paper describes the modified version of the system and the procedure developed to achieve reliable and quantitative soot volume fraction measurements in the exhausts of aero-engines. Application of the backward LII technique is demonstrated in the exhaust of a military turbojet engine for different engine speeds.

    • Objective: To investigate possible differences, between generally and abdominally obese men, in activity and regulation of the hypothalamic-pituitary-adrenal (HPA) axis and the sympathetic nervous system.Research Methods and Procedures: Fifty non-diabetic, middle-aged men were selected to obtain two groups with similar body mass index (BMI) but different waist/hip circumference ratio (WHR). Measurements were performed of the activity of the HPA axis and the sympathetic nervous system, as well as metabolic and endocrine variables.Results: Men with a high WHR, in comparisons with men with a low WHR, had higher insulin, glucose, and triglyceride values in the basal state and higher glucose and insulin after an oral glucose tolerance test. Men with high WHR had elevated diurnal adrenocorticotropic hormone (ACTH) values but similar cortisol values, except lower cortisol values in the morning. Diurnal growth hormone concentrations showed reduced peak size. Stimulation of the HPA axis with corticotropin-releasing hormone (CRH) and laboratory stress showed no difference in ACTH values between groups, but cortisol values were lower in-men with high WHR. In comparison with men with a low WHR, men with a high WHR had elevated pulse pressure and heart rate in the basal state and after challenges by CRH and laboratory stress. They also had increased urinary excretion of catecholamine metabolites.Discussion: These results suggest a mild dysregulation of the HPA axis, occurring with elevated WHR independent of the BMI. The results also indicate a central activation of the sympathetic nervous system, such as in the early phases of hypertension, correlating with insulin resistance.

    • Possible decay pathways associated with three candidates for triaxial strongly deformed (TSD) bands in Hf-168 have been investigated. The spin and excitation energy of the strongest band, TSD1, were determined approximately based on gamma-ray coincidence relationships. Discrete links were established for the second band. The overall agreement between the observed properties of the bands and cranking calculations using the ULTIMATE CRANKER code provides strong support for an interpretation where band TSD1 is associated with a TSD minimum, (epsilon(2),gamma)similar to(0.43,20(degrees)), involving the pi(i(13/2))(2) and the nu(j(15/2)) high-j orbitals. This constitutes the first identification of a TSD band in Hf isotopes, which has been long-predicted by theoretical studies. The second band is understood as being associated with a near-prolate shape and a deformation enhanced with respect to the normal deformed bands. It is proposed to be built on the pi(i(13/2)h(9/2))circle times nu(i(13/2))(2) configuration.

    • Objective: The aim of the present study was to clarify the relationship between GH deficiency (GHD) andsome cardiovascular risk factors and to analyse the effect of GH replacement therapy in a large numberof patients over a prolonged period of time.Design: Data for analysis were retrieved from KIMS (Pfizer International Metabolic Database). Serumconcentrations of total cholesterol, high-density lipoprotein (HDL)-cholesterol, low-density lipoprotein(LDL)-cholesterol and triglycerides were obtained from 2589 patients at baseline and from 1206patients after 1 and 2 years of GH replacement therapy. Body mass index (BMI), waist and hip, restingblood pressure and body composition were also measured.Results: At baseline, the unfavourable effects of GHD were most obvious in the lipid profiledemonstrating elevated mean total and LDL-cholesterol, in the increased waist circumference and theelevated BMI. The cholesterol concentration, BMI and body composition were significantly adverselyaffected by a number of factors, including age, sex and the use of anti-epileptic drugs. The therapeuticeffect of GH was essentially uniform across the whole population. GH replacement reduced significantlythe mean total and LDL-cholesterol, the waist circumference and the fat mass and was maintainedduring 2 years.Conclusions: This analysis of a large number of patients confirmed that GHD adults present with anincreased cardiovascular risk. The sustained improvement of the adverse lipid profile and bodycomposition suggests that GH replacement therapy may reduce the risk of cardiovascular disease andthe premature mortality seen in hypopituitary patients with untreated GHD.

    • We present results from measurements of the subthreshold lateral spontaneous emission profile in 1.3-mu m wavelength ridge waveguide InGaNAs quantum-well lasers using a scanning near-field optical microscopy technique. The measurements reveal the presence of significant lateral carrier diffusion which has a profound effect on the temperature dependence of the threshold current. This effect is frequently omitted when the characteristic temperature of the threshold current is considered.

    • We present an experimental and theoretical investigationof the temperature dependence of the threshold current fordouble quantum well GaInNAs–GaAs lasers in the temperaturerange 10 C–110 C. Pulsed measurements of the threshold current have been performed on broad and narrow ridge wave guide(RWG) lasers. The narrow RWG lasers exhibit high characteristic temperatures (T0)of 200 K up to a critical temperature (Tc), above which T0 is reduced by approximately a factor of 2. The T0-values for broad RWG lasers are significantly lower than those for the narrow RWG lasers, with characteristic temperatures on the order of 100 (60) K below (above). Numerical simulations, using a model that accounts for lateral diffusion effects, show good agreement with experimental data and reveal that a weakly temperature dependent lateral diffusion current dominates thethreshold current for narrow RWG lasers.

    • Identification of host volatile compounds attractive to codling moth Cydia pomonella, a most important insect of apple, will contribute to the development of safe control techniques. Synthetic apple volatiles in two doses were tested for antennal and behavioural activity in codling moth. Female antennae strongly responded to (Z)3-hexenol, (Z)3-hexenyl benzoate, (Z)3-hexenyl hexanoate, (+/-)-linalool and E,E-alpha-farnesene. Two other compounds eliciting a strong antennal response were the pear ester, ethyl (E,Z)-2,4-decadienoate, and its corresponding aldehyde, E,E-2,4-decadienal, which is a component of the larval defence secretion of the European apple sawfly. Attraction of codling moth to compounds eliciting a strong antennal response was tested in a wind tunnel. Male moths were best attracted to a blend of (E,E)-alpha-farnesene, (E)-beta-farnesene and ethyl (E,Z)-2,4-decadienoate. The aldehyde E,E-2,4-decadienal had an antagonistic effect when added to the above mixture.

    • Chronic hepatic encephalopathy (HE) is a neuropsychiatric syndrome that arises in liver-impaired subjects. Patients with HE display various neuropsychiatric symptoms including affective disturbances and may therefore likely receive treatment with novel thymoleptics like citalopram (CIT). The simultaneous pharmacokinetic and pharmacodynamic outcome of the commonly used serotonin-selective thymoleptic drugs in liver-impaired subjects with pending chronic HE is far from understood today. We therefore investigated the effects of chronic, body-weight-adjusted (10 mg ╖ kg-1 ╖ day-1), treatment with CIT in rats with and without portacaval shunts (PCS). Open-field activity was monitored. The 5-HT, 5-HIAA, noradrenaline (NA), and dopamine (DA) output were assessed in the frontal neocortex. The racemic levels of CIT and its metabolites DCIT and DDCIT, including the S- and R-enantiomers, were determined in serum, brain parenchyma, and extracellular fluid. The rats with PCS showed higher (2-3-fold) levels of CIT than rats undergoing a sham treatment with CIT in all compartments investigated. The PCS rats also showed elevated levels of DCIT and DDCIT. No major differences in the S/R ratios between PCS rats and control rats could be detected. The CIT treatment resulted in neocortical output differences between PCS rats and control rats mainly within the 5-HT and DA systems but not within the NA system. For the 5-HT system, this change was further evidenced by outspoken elevation in 5-HT output after KCl-depolarizing challenges. Moreover, the CIT treatment to PCS rats was shown to "normalize" the metabolic turnover of 5-HT, measured as a profound lowering of a basal elevation in the 5-HIAA levels. The CIT treatment resulted in an increased or "normalized" behavioral activity in the PCS group. Therefore, a dose-equal chronic treatment with CIT in PCS rats produced pharmacokinetic and pharmacodynamic changes not observed in control rats. The results further support the contention of an altered 5-HT neurotransmission prevailing in the chronic HE condition. However, the tentatively beneficial behavioral response also seen following chronic CIT treatment to PCS rats in this study has to be viewed in relation to both the pharmacokinetic and pharmacodynamic changes observed.

    • The antennal responses of codling moth females, Cydia pomonella, to volatiles from apple branches with green fruits were recorded by electroantennography coupled to gas chromatography. The antennae strongly responded to 4,8-dimethyl-1,3(E),7-nonatriene, linalool, beta -caryophyllene, (E)-beta -farnesene, germacrene D, (Z,E)-alpha -farnesene, (E,E)-alpha -farnesene and methyl salicylate. These compounds were all present in volatile collections on Porapak Q from both living and cut branches. Analysis by the solid phase microextraction technique (SPME) showed that the emission of some electrophysiologically active compounds increased after branches had been cut, especially 4,8-dimethyl-1,3(E),7-nonatriene, linalool and (E,E)-alpha -farnesene. The identification of apple volatiles eliciting antennal responses is the first step towards the identification of compounds mediating host-finding and oviposition in codling moth females.

    • In analogy with other hormonal replacement therapy GH treatment should be commenced with a low starting dose, independent of body weight or body surface area. Hormonal replacement should mimic the normal physiology to minimize the risk of side effects in the life-long replacement of adults. We should, therefore, consider individual responsiveness and also be aware of the difference between pattern of GH under normal condition and during s.c. administration. The safety and monitoring of GH replacement therapy in adults have been addressed in the Growth Hormone Research Society Consensus Guidelines for Diagnosis and Treatment of Adults with GH Deficiency from the Port Stephens Workshop, April 1997. Besides finding better and more accurate biochemical markers for choosing correct GH replacement dose, future research should address the long-term benefits and safety with GH replacement in adults, with special emphasize on incipient risks in terms of cardiovascular disease and of neoplasia, in particular.

    • Aims: Two common reasons to explain slow environmental biodegradation of polycyclic aromatic hydrocarbons (PAHs), namely lack of appropriate carbon sources for microbial growth and limited bioavailability of PAHs, were tested in a laboratory bioassay using a creosote-contaminated soil. Methods and Results: The soil, containing a total of 8 mg g(-1) of 16 PAHs, was sieved and incubated in bottles for 45 days. The first explanation was tested by enrichment with the analogue anthracene and the non-analogue myristic acid, and both failed to stimulate degradation of all PAHs except anthracene. The second explanation was tested by addition of different concentrations of dissolved organic carbon (DOC), with effects depending on the DOC concentration and the molecular size of the PAH. The degradation was enhanced from 10 to 35% for 12 PAHs when the soil was saturated. The degraded amounts of individual PAHs were proportional to their concentration in the soil. Conclusions: The slow in situ degradation of PAHs was enhanced by more than three times by adding water as a solvent. Addition of DOC facilitated the degradation of four- to six-ring PAHs. Significance and Impact of Study: Bioremediation of PAH-contaminated sites may be facilitated by creating water-saturated conditions but retarded by addition of other carbon substrates, such as analogue compounds.

    • We review the effects of human impact on biodiversity of European forests in the light of recent views on disturbances and succession in ecosystems, and discuss recent ideas on how biodiversity affects ecosystem functions such as productivity and ecosystem stability. With this as a background we discuss how to better manage European forests for both production and biodiversity. We argue that the next generation of forestry practices need to understand and mimic natural disturbance dynamics much better than the present ones. Of particular importance is the fact that most species in European forests have evolved in forests that were to a large extent influenced by large grazers, first by megaherbivores and later, in historic times, by domestic animals. We highlight several areas where new knowledge and management tools are urgently needed: (i) How do species survive and adapt to the natural disturbance regimes in different regions and forest types? (ii) How can new and imaginative forest management practices be devised that take natural disturbance regimes into account? (iii) How does forest biodiversity affect ecosystem function and stability in a changing world, in particular in the light of predicted climate changes? (iv) How are ecological processes at different levels and scales related to diversity, and how do different management practices affect biodiversity? (v) How can efficient agroforestry methods be developed to preserve biodiversity? (vi) What is the role of humans and human behaviour for sustainable management of ecosystems?     • The Homogeneous charge compression ignition (HCCI) principle holds promise to increase efficiency and to reduce emissions from internal combustion engines. As HCCI combustion lacks direct ignition timing control and auto-ignition depends on the operating condition, control of auto-ignition is necessary. Since auto-ignition of a homogeneous mixture is very sensitive to operating conditions, a fast combustion phasing control is necessary for reliable operation. To this purpose, HCCI modelling and model-based control with experimental validation were studied. A six-cylinder heavy-duty HCCI engine was controlled on a cycle-to-cycle basis in real time using a variety of sensors, actuators and control structures for control of the HCCI combustion in comparison. Combustion phasing control based on ion current was compared to feedback control based on cylinder pressure. With several actuators for controlling HCCI engines suggested, two actuators were compared, dual fuel and variable valve actuation (VVA). Model-based control synthesis requiring dynamic models of low complexity and HCCI combustion models were estimated by system identification and by physical modelling the physical models aiming at describing the major thermodynamic and chemical interactions in the course of an engine stroke and their influence on combustion phasing. The models identified by system identification were used to design model-predictive control (MPC) with several desirable features and today applicable to relatively fast systems, the MPC control results being compared to PID control results. Both control of the combustion phasing and control of load-torque with simultaneous minimization of the fuel consumption and emissions, while satisfying the constraints on cylinder pressure, were included.

  • Model sophistication in relation to scales in snowmelt runoff modeling

    • Snowmelt induced runoff from river basins is usually successfully simulated using a simple degree-day approach and conceptual rainfall-runoff models. Fluctuations within the day can not be described by such crude approaches. In the present paper, it is investigated which degree of sophistication is required in snow models and runoff models to resolve the basin runoff from basins of different character, and also how snow models and runoff models must adapt to each other. Models of different degree of sophistication are tested on basins ranging from 6,000 km(2) down to less than 1 km(2). It is found that for large basins it is sufficient to use a very simple runoff module and a degree day approach, but that the snow model has to be distributed related to land cover and topography. Also for small forested basins, where most of the stream flow is of groundwater origin, the degree-day method combined with a conceptual runoff model reproduces the snowmelt induced runoff well. Where overland flow takes place, a high resolution snow model is required for resolving the runoff fluctuations at the basin outlet.

    • Remote fluorescence imaging of fungal growth on polymeric high-voltage insulators was performed using a mobile lidar system with a laser wavelength of 355 nm. Insulator areas contaminated by fungal growth could be distinguished from clean surfaces and readily be imaged. The experiments were supported by detailed spectral studies performed in laboratory using a fibre-optic fluorosensor incorporating an optical multi-channel analyser system (OMA) and a nitrogen laser emitting radiation at 33 7 nm.

    • The paper focuses on the fact that, since the flow rate in parallel channels relies strongly on the channel width, the combination may lead to inaccurate results if errors in the fabrication process lead to an uneven distribution of channel widths. Parallel channel enzyme reactors were designed with channel widths distributed normally with different degrees of standard deviation. The reactors were then evaluated with regard to dispersion and to catalytic effect of the immobilised enzyme. It was shown that for lower concentrations the catalytic efficiency decreased significantly even for small variations in the distribution of channel widths and reactors with poor homogeneity in channel widths also diluted the sample more than the others.

    • In-situ measurements of soot volume fraction in the exhausts of jet engines can be carried out using the laser-induced incandescence (LII) technique in backward configuration, in which the signal is detected in the opposite direction of the laser beam propagation. In order to improve backward LII for quantitative measurements, we have in this work made a detailed experimental and theoretical investigation in which backward LII has been compared with the more commonly used right-angle LII technique. Both configurations were used in simultaneous visualization experiments at various pulse energies and gate timings in a stabilized methane diffusion flame. The spatial near-Gaussian laser energy distribution was monitored on-line as well as the time-resolved LII signal. A heat and mass transfer model for soot particles exposed to laser radiation was used to theoretically predict both the temporal and spatial LII signals. Comparison between experimental and theoretical LII signals indicates similar general behaviour, for example the broadening of the spatial LII distribution and the hole-burning effect at centre of the beam due to sublimation for increasing laser pulse energies. However, our comparison also indicates that the current heat and mass transfer model overpredicts signal intensities at higher fluence, and possible reasons for this behaviour are discussed.

    • In recent years global problems such as climatic change, acid rain, and water pollution in surface and subsurface environments dominate discussions of world environmental problems. In this paper, the roles of hydrologic processes and hydrogeochemical processes are investigated through development, modification, and application of mathematical models for addressing point and non-point source water quality modelling of receiving waters: surface water, subsurface water and lake water. The paper describes the use of models to simulate the movements of pollutants and water: subsurface water, surface water and lake sediments. A hydrological model was applied to Northeast Pond River watershed to understand climate change effects in the watershed. Four watershed acidification models were applied to compute hydrogen ion, alkalinity and sulphate concentrations from Turkey Lakes watershed, Canada. The computed hydrogen ion was used to estimate acidic events, magnitude of hydrogen ion, and duration using a stochastic model. There exist uncertainties in environmental models due to imperfect knowledge of processes controlling water quality parameters as well as errors in data. Monte Carlo, first order, and inverse method analyses were used to assess uncertainty in models. SUTRA (saturated-unsaturated transport) and SUTRA(-1) models were applied to Lambton county, Ontario, Canada to locate groundwater discharge areas for St. Clair River, calculate discharge rates, and hydrogeologic parameters. A sediment contamination model was developed and applied to Great Lakes sediment data to estimate transport parameters by Pb-210 data. It was then coupled with fatty acid data and results were compared with observed data. A contaminant transport model was developed and applied to two North American streams to compute stream water concentration. The computed data was compared with observed data using a simple statistical method. A hydrological model was coupled with water quality models and RAISON (regional analysis by intelligent systems on) expert system and applied to Canadian watersheds. Digital satellite data was used to locate groundwater discharge and recharge areas in the watershed. This data is useful as observed data for hydrological modelling and GIS (Geographical Information System) system. (C) 2000 Elsevier Science B.V. All rights reserved.

    • Pure rotational coherent anti-Stokes Raman scattering (CARS) experiments have been performed in acetylene for temperatures ranging from 294 to 582 K, and in mixtures of acetylene and nitrogen in the mole fraction range of 0.06-0.32 for acetylene at room temperature. The experimental spectra are evaluated by a least-square fitting to libraries of theoretically calculated spectra using two different Raman linewidth models, one with and one without dependence on the rotational quantum number J. It is found that a J-dependent model is favourable, both regarding temperature measurements in pure acetylene, and simultaneous acetylene concentration and temperature measurements in different mixtures of acetylene and nitrogen. For the temperature measurements performed in pure acetylene the temperature inaccuracy is generally less than 2% when the J-dependent model for the Raman linewidths is used. It is found that fitting the value of the non-resonant susceptibility significantly improves the quality of the spectral fits and is a requirement for high temperature accuracy with the present model. The evaluated concentrations show a maximum error of 13% on a relative scale. Potential sources of systematical errors both regarding measured temperatures and acetylene concentrations are discussed.
