    • Minimal residual disease (MRD) assessment is a powerful prognostic factor for determining the risk of relapse in childhood acute lymphoblastic leukaemia (ALL). In this Swedish multi-centre study of childhood ALL diagnosed between 2002 and 2006, the MRD levels were analysed in 726 follow-up samples in 228 children using real-time quantitative polymerase chain reaction (RQ-PCR) of rearranged immunoglobulin/T-cell receptor genes and multicolour flow cytometry (FCM). Using an MRD threshold of 0·1%, which was the sensitivity level reached in all analyses, the concordance between RQ-PCR and FCM MRD values at day 29 was 84%. In B-cell precursor ALL, an MRD level of ≥0·1% at day 29 predicted a higher risk of bone marrow relapse (BMR) with both methods, although FCM was a better discriminator. However, considering the higher median MRD values achieved with RQ-PCR, a higher MRD cut-off (≥0·2%) improved the predictive capacity of RQ-PCR. In T-ALL, RQ-PCR was notably superior to FCM in predicting risk of BMR. That notwithstanding, MRD levels of ≥0·1%, detected by either method at day 29, could not predict isolated extramedullary relapse. In conclusion, the concordance between RQ-PCR and FCM was high and hence both methods are valuable clinical tools for identifying childhood ALL cases with increased risk of BMR.

  • Folding and interaction studies of subunits in protein complexes

    • Proteins function as worker molecules in the cell and their natural environment is crowded. How they fold in a cell-like environment and how they recognize their interacting partners in such conditions, are questions that underlie the work of this thesis.Two distinct subjects were investigated using a combination of biochemical- and biophysical methods. First, the unfolding/dissociation of a heptameric protein (cpn10) in the presence of the crowding agent Ficoll 70. Ficoll 70 was used to mimic the crowded environment in the cell and it has been used previously to study macromolecular crowding effects, or excluded volume effects, in protein folding studies. Second, the conformational changes upon interaction between the Mediator subunit Med25 and the transcription factor Dreb2a from Arabidopsis thaliana. Mediator is a transcriptional co-regulator complex which is conserved from yeast to humans. The molecular mechanisms of its action are however not entirely understood. It has been proposed that the Mediator complex conveys regulatory signals from promoter-bound transcription factors (activators/repressors) to the RNA polymerase II machinery through conformational rearrangements.The results from the folding study showed that cpn10 was stabilized in the presence of Ficoll 70 during thermal- and chemical induced unfolding (GuHCl). The thermal transition midpoint increased by 4°C, and the chemical midpoint by 0.5 M GuHCl as compared to buffer conditions. Also the heptamer-monomer dissociation was affected in the presence of Ficoll 70, the transition midpoint was lower in Ficoll 70 (3.1 μM) compared to in buffer (8.1 μM) thus indicating tighter binding in crowded conditions. The coupled unfolding/dissociation free energy for the heptamer increased by about 36 kJ/mol in Ficoll. Altogether, the results revealed that the stability effect on cpn10 due to macromolecular crowding was larger in the individual monomers (33%) than at the monomer-monomer interfaces (8%).The results from the interaction study indicated conformational changes upon interaction between the A. thaliana Med25 ACtivator Interaction Domain (ACID) and Dreb2a. Structural changes were probed to originate from unstructured Dreb2a and not from the Med25-ACID. Human Med25-ACID was also found to interact with the plant-specific Dreb2a, even though the ACIDs from human and A. thaliana share low sequence homology. Moreover, the human Med25-interacting transcription factor VP16 was found to interact with A. thaliana Med25. Finally, NMR, ITC and pull-down experiments showed that the unrelated transcription factors Dreb2a andVP16 interact with overlapping regions in the ACIDs of A. thaliana and human Med25.The results presented in this thesis contribute to previous reports in two different aspects. Firstly, they lend support to the findings that the intracellular environment affects the biophysical properties of proteins. It will therefore be important to continue comparing results between in vitro and cell-like conditions to measure the magnitude of such effects and to improve the understanding of protein folding and thereby misfolding of proteins in cells. Better knowledge of protein misfolding mechanisms is critical since they are associated to several neurodegenerative diseases such as Alzheimer’s and Parkinson's. Secondly, our results substantiate the notion that transcription factors are able to bind multiple targets and that they gain structure upon binding. They also show that subunits of the conserved Mediator complex, despite low sequence homologies, retain a conserved structure and function when comparing evolutionary diverged species.

    • Mediator is an evolutionary conserved multi-protein complex present in all eukaryotes. It functions as a transcriptional coregulator by conveying signals from activators and repressors to the RNA polymerase II transcription machinery. The Arabidopsis thaliana Med25 (aMed25) ACtivation Interaction Domain (ACID) interacts with the Dreb2a activator which is involved in plant stress response pathways, while Human Med25-ACID (hMed25) interacts with the herpes simplex virus VP16 activator. Despite low sequence similarity, hMed25-ACID also interacts with the plant-specific Dreb2a transcriptional activator protein. We have used GST pull-down-, surface plasmon resonance-, isothermal titration calorimetry and NMR chemical shift experiments to characterize interactions between Dreb2a and VP16, with the hMed25 and aMed25-ACIDs. We found that VP16 interacts with aMed25-ACID with similar affinity as with hMed25-ACID and that the binding surface on aMed25-ACID overlaps with the binding site for Dreb2a. We also show that the Dreb2a interaction region in hMed25-ACID overlaps with the earlier reported VP16 binding site. In addition, we show that hMed25-ACID/Dreb2a and aMed25-ACID/Dreb2a display similar binding affinities but different binding energetics. Our results therefore indicate that interaction between transcriptional regulators and their target proteins in Mediator are less dependent on the primary sequences in the interaction domains but that these domains fold into similar structures upon interaction.

  • Operationalizing Industrial Ecology in the Waste Sector : Roles and tactics for circular value innovation

    • The take-make-waste approach to resource management in human production and consumption systems is contributing to a variety of environmental and social problems worldwide. Additionally, as the world’s population and affluence increase, so do the negative impacts of poor resource management. Lifting the waste management (WM) sector into a new phase of development, which takes its lead from the ideals of Industrial Ecology and circular economy, is seen by many scholars and practitioners as one potential to assist in alleviating these impacts. While there are many studies on how more efficient inter-organizational resource management is (or could be) constructed, there are relatively few business development studies which have explored novel approaches (from roles to tactics) that WM organizations might operationalize toward more efficient resource management.The aim of this thesis is to contribute to the development of knowledge and understanding of how the waste management sector can operationalize more effective and efficient resource management. In approaching this aim, two research questions guided the exploration of: 1) novel roles for WM and 2) support tactics for such roles. Grounded in the broader context of Industrial Ecology (IE) and Business Development, five studies were performed. Two studies, focused on the novel roles of inter-organizational resource management and high value secondary resource extraction, were performed through literature review and interviews, and market driver analysis respectively. In exploring support tactics, two design and proof of concept studies were carried out to investigate data analysis tools for inter-organizational resource management, and one long-term action research engagement project was coordinated to study hands-on inter-organizational collaboration tactics.The studies highlighted that the Swedish WM sector holds some key capacities for operationalizing (and in some cases, is already developing) the novel resource management roles identified: industrial symbiosis facilitator, eco-industrial park manager, holistic facility management, and high value resource extractor. However, depending on the portfolio of services to be performed in such roles, several capacities may need to be developed or strengthened. Main opportunities seen for these roles were – staying ahead of market developments, and aligning activities with organizational goals. The main general risk related to these roles was insufficient returns on investment. Looking forward, the main enablers identified were policy leadership for more balanced market mechanisms, increasing use of external knowledge, developing long term partnerships, lobbying, stockpiling resources, and carefully crafting new business models.The tools developed for strategically applying external information toward the identification of opportunities within new roles showed tactical potential. However, their implementation in broader development processes has yet to be fully validated. The hands-on exploration of change oriented collaboration, highlighted collective system framing and goal setting and face-to-face interaction as key activities for inter-organizational approaches within roles such as industrial symbiosis facilitator.Throughout the studies, several novel roles were investigated. Each of these roles will need to be individually evaluated by directing bodies of WM organizations, and evaluated from the organization’s vision and strategy. If certain roles are chosen to be explored in more detail, they will need to be developed within full business models - addressing issues such as income structure, internal processes and capacities to be developed, and key customers. Through applying IE and business development concepts and findings, WM organizations have possibilities to translate ambitious visions into novel offerings.

  • Churning losses and efficiency in gearboxes

    • Efficient transmissions systems are key to producing competitive motor vehicles that have a smaller environmental impact. Gears are the main components in vehicle transmissions and although they are already highly efficient, there is still room for improvement. In this study, the focus falls on the lubricant used to create separating films between gears and todissipate heat. When driving, the gears churn this lubricant, giving rise to power losses that are related to the amount and properties of the lubricant. However, any attempt to reduce these losses must not compromise the required lubrication and heat dissipation. Paper A reports on the use of an FZG gear test rig to investigate power losses and heat generation for different gear immersion depths, surface roughness and coatings. The results show that lower gear roughness reduces gear mesh losses and heat generation. A polishing affect was obtained when a non-coated gear ran against a coated gear.The aim of the research reported in paper B was to increase the accuracy of efficiency testing. It investigated how and whether repeated disassembly and re-assembly of the same test equipment, as well as test performance and rig conditions, affect the measured torque loss in an FZG gear test rig. It was shown that the measured torque loss changes between one assembly and another. Repeatability between tests is crucial for accurate conclusions.The aim of the research reported in paper C was to study whether gear efficiency could be increased by a running-in procedure, which would reduce the need for a coolant. A back-to-back gear test rig was used to test two running-in loads. Higher gear mesh efficiency was seen when a higher running-in load was used.

  • Influence of real surface topography on the contact area ratio in differently manufactured spur gears

    • Surface topography measurements from spur gears produced using four different manufacturing methods were used as input to a contact analysis programme. All test gears were case-hardened, two gears were machined in the hardened state using honing and grinding respectively, and two gears were machined in the non-hardened state using hobbing and hobbing followed by green-shaving respectively. The results show that the surface topography caused by the manufacturing methods has a large influence on the real contact area in the early life of the gear. The green-shaved gear surfaces and the honed gear surfaces have the highest contact area ratio after manufacturing (as-manufactured), which could be advantageous for future gear life with respect to e.g. the running-in process.

  • Logarithmical Crowning for Spur Gears

    • Gear tooth modifications, such as lead crowning, are often recommended to compensate for misalignment (e.g. assembly deviations). Lead crowning means that the tooth centre is slightly thicker than the tooth edges and is usually described as a circular arc profile. The use of crowning shifts the peak load from the tooth flank edges and, therefore, reduces the risk for high contact pressures at the edges, which can otherwise result in a shortened service life. In this study a logarithmical lead profile was compared with traditional lead profile modifications for gears. The profiles were applied on a spur gear pair and a numerical method for contact analysis was used to calculate the contact pressure distribution at the pitch diameter. All lead profiles were optimised with respect to low contact pressure at a specific normal load, a specified maximum misalignment in the plane of action and a tooth flank edge contact criteria. The results show that the logarithmical profile responds differently to misalignments compared to the traditional lead profile modifications. The logarithmical profile resulted in lower maximum contact pressures for small misalignments and is, therefore, of further interest in terms of achieving a robust gear design.     • Several highly potent BACE-1 protease inhibitors have been developed from an inhibitor series containing a novel hydroxyethylene (HE) core structure displaying aryloxymethyl or benzyloxymethyl P1 side chains and a methoxy P1’ side chain. The target molecules were readily synthesized from chiral carbohydrate starting materials, furnishing the inhibitor compounds in good overall yields. The inhibitors show both high BACE-1 potency and good selectivity against cathepsin D, where the most potent inhibitor furnish a BACE-1 IC50 value of 0.32 nM and displays > 3000 fold selectivity over cathepsin D.

  • Postoperative mortality 90 days after robot-assisted laparoscopic prostatectomy and retropubic radical prostatectomy : a nationwide population-based study

    • Objective To assess 90-day postoperative mortality after robot-assisted laparoscopic radical prostatectomy (RARP) and retropubic radical prostatectomy (RRP) using nationwide population-based registry data. Patients and Methods We conducted a cohort study using the National Prostate Cancer Register of Sweden, including 22 344 men with localized prostate cancer of clinical stage T1-T3, whose prostate-specific antigen levels were <50 mu g/mL and who had undergone primary radical prostatectomy in the period 1998-2012. Vital status was ascertained through the Total Population Register. The rates for 90-day postoperative mortality were analysed using logistic regression analysis, and comparisons of 90-day mortality with the background population were made using standardized mortality ratios (SMRs). Results Of the 14 820 men who underwent RRP, 29 (0.20%) died, and of the 7 524 men who underwent RARP, 10 (0.13%) died. Mortality in the cohort during the 90-day postoperative period was lower than in an age-matched background population: SMR 0.57 (95% confidence interval [CI] 0.39-0.75). There was no statistically significant difference in 90-day mortality according to surgical method: RARP vs RRP odds ratio (OR) 1.14; 95% CI 0.46-2.81. Postoperative 90-day mortality decreased over time: 2008-2012 vs 1998-2007 OR 0.44; 95% CI 0.21-0.95, mainly because of lower mortality after RARP. Conclusion The 90-day postoperative mortality rates were low after RARP and RRP and there was no statistically significant difference between the methods. Given the long life expectancy among men with low-and intermediate-risk prostate cancer, very low postoperative mortality is a prerequisite for RP, which was fulfilled by both RRP and RARP. The selection of healthy men for RP is highlighted by the lower 90-day mortality after RP compared with the background population.

    • The attractive properties achieved by Suspension Plasma Spraying (SPS), combined with the availability of high throughput capable plasma spray systems that permit axial feeding, provide encouragement to explore use of suspensions for next generation functional applications. This paper deals with realization of coatings with various pre-determined function-dependent architectures by employing a hybrid powder-suspension feedstock. Some illustrative application-relevant coating architecture designs are discussed, along with the specific benefits that can accrue by deploying a multi-scale powder-suspension feedstock combination. An elegant feedstock delivery arrangement to enable either simultaneous or sequential feeding of powders and suspensions to enable convenient processing of coatings with desired architectures is presented. As proof-of-concept, deposition of layered, composite and functionally graded coatings using the above system is also demonstrated using appropriate case studies

    • Mediator is a multiprotein coregulatory complex that conveys signals from DNA-bound transcriptional regulators to the RNA polymerase II transcription machinery in eukaryotes. The molecular mechanisms for how these signals are transmitted are still elusive. By using purified transcription factor Dreb2a, mediator subunit Med25 from Arabidopsis thaliana, and a combination of biochemical and biophysical methods, we show that binding of Dreb2a to its canonical DNA sequence leads to an increase in secondary structure of the transcription factor. Similarly, interaction between the Dreb2a and Med25 in the absence of DNA results in conformational changes. However, the presence of the canonical Dreb2a DNA-binding site reduces the affinity between Dreb2a and Med25. We conclude that transcription regulation is facilitated by small but distinct changes in energetic and structural parameters of the involved proteins.

    • Yeast cell subjected to many different stresses elicit an acute transcriptional stress response mediated by the Msn2 transcription factor, which alters expression of both a stress specific-cohort of genes as well as a common cohort of genes that changes expression in a stereotypic fashion upon exposure to any of a wide variety of stresses. We have shown by dynamic single cell analysis that stresses regulate Msn2 activity through cytoplasm to nuclear relocalization but do so in an unusual way: stresses induce increased frequency of bursts of short-lived, recurrent periods of Msn2 nuclear localization with different stresses eliciting different patterns of bursts. Moreover, genetically identical cells subject to an identical stress can behave quite differently, with some cells mounting a robust nuclear occupancy of Msn2 while others show no nuclear localization at all. We have proposed that this idiosyncratic behavior allows populations of cells to “hedge their bet” as to what will be the optimum strategy for surviving the ensuing stress. We have used computational modeling and single cell analysis to determine that bursting is a consequence of noise in the stress signaling pathways amplified by the small number of Msn2 molecules in the cell. Moreover, we have applied genome wide chromatin immunoprecipitation and nucleosome profiling to address how different stresses determine where Msn2 binds under a particular stressful conditions, and thus what genes are regulated by that stress, and how that binding affects, and is affected by, nucleosome positioning and other transcription factor binding. These results provide in vivo validation of Widon's model of indirect cooperativity of transcription factor binding, mediated by partial unwinding of nucleosomes by one transcription factor to allow access for a second transcription factor to a previously occluded binding site. Finally, we have addressed the “bet hedging” hypothesis by showing that persistence of the Msn2-mediated stress response yields cell growth arrest and have identified the targets responsible for that growth arrest. We have applied experimental evolution paradigms to address the relative fitness of cells exhibiting stochastic stress responses versus those with a uniform response. In short, our results indicate that the stress response is complex and that complexity is critical for cell survival.

    • Mediator is a multi-unit molecular complex that plays a key role in transferring signals from transcriptional regulators to RNA polymerase II in eukaryotes. We have combined biochemical purification of the Sac-charomyces cerevisiae Mediator from chromatin with chromatin immunoprecipitation in order to reveal Mediator occupancy on DNA genome-wide, and to identify proteins interacting specifically with Mediator on the chromatin template. Tandem mass spectrometry of proteins in immunoprecipitates of mediator complexes revealed specific interactions between Mediator and the RSC, Arp2/Arp3, CPF, CF 1A and Lsm complexes in chromatin. These factors are primarily involved in chromatin remodeling, actin assembly, mRNA 3'-end processing, gene looping and mRNA decay, but they have also been shown to enter the nucleus and participate in Pol II transcription. Moreover, we have found that Mediator, in addition to binding Pol II promoters, occupies chromosomal interacting domain (CID) boundaries and that Mediator in chromatin associates with proteins that have been shown to interact with CID boundaries, such as Sth1, Ssu72 and histone H4. This suggests that Mediator plays a significant role in higher-order genome organization.

    • Suspension plasma spraying has become an emerging technology for the production of thermal barrier coatings for the gas turbine industry. Presently, though commercial systems for coating production are available, coatings remain in the development stage. Suitable suspension parameters for coating production remain an outstanding question and the influence of suspension properties on the final coatings is not well known. For this study, a number of suspensions were produced with varied solid loadings, powder size distributions and solvents. Suspensions were sprayed onto superalloy substrates coated with high velocity air fuel (HVAF) -sprayed bond coats. Plasma spray parameters were selected to generate columnar structures based on previous experiments and were maintained at constant to discover the influence of the suspension behavior on coating microstructures. Testing of the produced thermal barrier coating (TBC) systems has included thermal cyclic fatigue testing and thermal conductivity analysis. Pore size distribution has been characterized by mercury infiltration porosimetry. Results show a strong influence of suspension viscosity and surface tension on the microstructure of the produced coatings.

It conveys signals from promoter-bound transcriptional regulators to RNA polymerase II and thus plays an essential role in eukaryotic gene regulation. We describe subunit localization and activities of Mediator in Arabidopsis through metabolome and transcriptome analyses from a set of Mediator mutants. Functional metabolomic analysis based on the metabolite profiles of Mediator mutants using multivariate statistical analysis and heat-map visualization shows that different subunit mutants display distinct metabolite profiles, which cluster according to the reported localization of the corresponding subunits in yeast. Based on these results, we suggest localization of previously unassigned plant Mediator subunits to specific modules. We also describe novel roles for individual subunits in development, and demonstrate changes in gene expression patterns and specific metabolite levels in med18 and med25, which can explain their phenotypes. We find that med18 displays levels of phytoalexins normally found in wild type plants only after exposure to pathogens. Our results indicate that different Mediator subunits are involved in specific signaling pathways that control developmental processes and tolerance to pathogen infections.

    • Axial suspension plasma spraying (ASPS) is a relatively new, innovative technique with which microstructures have been produced that are similar to the ones produced by electron beam physical vapor deposition. They have a columnar structure and consist of nm- and µm-sized pores. However, so far the formation of the microstructure is not fully understood because fragmentation and vaporisation of the liquid significantly affects the deposition process. Analysis of single splats can provide important information on the phenomena controlling the coating formation process and the final coating properties. Therefore, the present study aims at providing first results of 8 wt-% yttria-stabilised zirconia single splats sprayed onto a steel substrate by use of ASPS. Scanning electron microscopy and atomic force microscopy have been used to characterise the splats with respect to appearance, shape, and size distribution.

    • The transcription factor Msn2 mediates a significant proportion of the environmental stress response, in which a common cohort of genes changes expression in a stereotypic fashion upon exposure to any of a wide variety of stresses. We have applied genome-wide chromatin immunoprecipitation and nucleosome profiling to determine where Msn2 binds under stressful conditions and how that binding affects, and is affected by, nucleosome positioning. We concurrently determined the effect of Msn2 activity on gene expression following stress and demonstrated that Msn2 stimulates both activation and repression. We found that some genes responded to both intermittent and continuous Msn2 nuclear occupancy while others responded only to continuous occupancy. Finally, these studies document a dynamic interplay between nucleosomes and Msn2 such that nucleosomes can restrict access of Msn2 to its canonical binding sites while Msn2 can promote reposition, expulsion and recruitment of nucleosomes to alter gene expression. This interplay may allow the cell to discriminate between different types of stress signaling.

    • Mediator has been shown to be essential for regulation of RNA Polymerase II mediated transcription. Mediator functions as an interface between the general transcriptional machinery and a multitude of DNA binding transcriptional regulators, although the molecular mechanism for the process is elusive. Mediator is a large complex of over twenty subunits, most of which are conserved from yeast to plants to mammals. Many of these subunits are essential for viability in yeast, and mutations in the corresponding genes have global effects on transcription. Mediator was originally identified in Saccharomyces cerevisiae, but has since been described in most eukaryotes. However, until recently the Mediator complex was not identified in plants. This thesis describes the first successful identification and isolation of the Mediator complex from the plant Arabidopsis thaliana. By raising antibodies against candidate A. thaliana Mediator subunits, we were able to purify a multisubunit protein complex. Mass spectrometry and bioinformatics analysis allowed us to identify 21 of these subunits as conserved Mediator components and six as A. thaliana specific subunits. Some of the genes that encode the identified Mediator subunits had earlier been described as components of specific regulatory pathways controlling for example cell proliferation and flowering time. Subsequent genetic analysis confirmed that the A. thaliana Mediator complex is important for several plant signaling pathways, including flowering and stress pathways. This thesis also describes identification of regulators that interact with the A. thaliana Mediator subunit Med25, previously identified as PFT1 (Phytochrome and Flowering Time 1) and implicated in regulation of flowering time in response to light quality. Finally, we describe the function of Mediator in S. cerevisiae using genome-wide approaches. We have carried out a transcriptional switch where half of the genome changes expression and determined Mediator occupancy across the genome before and after such a switch, using ChIP-SEQ on tagged subunits from different Mediator domains. Unexpectedly, we find that Mediator occupancy is limited at most promoters. However, at the highly occupied promoters, we see different modes of changes in occupancy as a result of the transcriptional switch. These highly occupied promoters control genes involved in different stress response pathways. Thus, our results suggest that Mediator function and composition differ considerably between different promoters.

    • Development in plants is controlled by abiotic environmental cues such as day length, light quality, temperature, drought, and salinity. These signals are sensed by a variety of systems and transmitted by different signal transduction pathways. Ultimately, these pathways are integrated to control expression of specific target genes, which encode proteins that regulate development and differentiation. The molecular mechanisms for such integration have remained elusive. We here show that a linear 130-amino-acids-long sequence in the Med25 subunit of the Arabidopsis thaliana Mediator is a common target for the drought response element binding protein 2A, zinc finger homeodomain 1, and Myb-like transcription factors which are involved in different stress response pathways. In addition, our results show that Med25 together with drought response element binding protein 2A also function in repression of PhyB-mediated light signaling and thus integrate signals from different regulatory pathways.

    • Children’s early mathematics learning and development have become a topic of increasing interest over the past decade since early mathematical knowledge and skills have been shown to be a strong predictor of later mathematics performance. Understanding how children develop mathematical knowledge and skills and how they can be supported in their early learning could thus prove to be a vital component in promoting learning of more formal mathematics.In light of the above, with this thesis I sought to contribute to an increased understanding of children’s early mathematics learning and development by examining effects of playing different number games on children’s number knowledge and skills, and by investigating children’s representations of numbers on number line tasks.Two number game intervention studies were performed, and effects of three different number game conditions (linear number, circular number and nonlinear number) were investigated by examining 5- and 6-year-old children’s pre- and posttest performance on different numerical tasks. The findings indicate that playing number games in general support children’s development of number knowledge and skills, where the specific learning outcomes are affected differently depending on the type of number game utilized.To elucidate children’s representations of numbers, their performance on two different  umber line tasks have been analyzed using a latent class modeling approach. The results reveal that there is a heterogeneity in 5- and 6-year-old children’s number line estimations and subgroups of children showing different estimation patterns were distinguished. In addition, it is shown that children’s number line estimations can be associated to their number knowledge as well as to task specific aspects.The findings presented in this thesis contribute to the discussion of the value of selecting game activities in a conscious way to support children’s early mathematics learning and development. They also add to the discussion regarding the number line task and how children’s number line estimations can be analyzed and interpreted.

    • Rolling contacts are utilized in many technical applications, both in bearings and in the contact between gear teeth. These components are often highly loaded, which makes them susceptible to suffer from rolling contact fatigue. This work focuses on the rolling contact fatigue mechanism of pitting. In order to attain a better understanding of why pitting initiates and grows, detailed simulations of rolling contacts have been performed. In particular the contact between two gears in a truck retarder was here used as a case study. The investigated contact experienced elastohydrodynamic lubrication conditions since the load was high enough to causes the surfaces in contact to deform and the viscosity of the lubricant to increase significantly.In Paper A it was investigated if surface irregularities in the size of the surface roughness are large enough to cause surface initiated fatigue. The investigation focused on the pitch line since small surface initiated pits were found here even though there was no slip present. Since there were pits present at the pitch line, it is important that the theories of pitting can explain the development of pits also in the absence of slip. The conclusion of the work was that surface irregularities of the size of normal surface roughness are enough to cause surface initiated fatigue at the pitch line.In Paper B it was investigated why pits are more likely to initiate in the dedendum of pinion gears than in the addendum. In both areas slip is present but in different directions. In the dedendum the friction from slip is against the rolling direction which enhances the risk for pitting. The investigation was performed by studying the effect of the temperature rise in the contact caused by the slip. The conclusion drawn was that the temperature rise in the contact explained why pitting was more common in the dedendum than in the addendum.

As a result, Mediator subunits play multiple roles in regulating developmental as well as abiotic and biotic stress pathways. In this report we identify the head domain subunits MEDIATOR18 and MEDIATOR20 as important susceptibility factors for Fusarium oxysporum infection in Arabidopsis thaliana. Mutants of MED18 and MED20 display down-regulation of genes associated with jasmonate signaling and biosynthesis while up-regulation of salicylic acid associated pathogenesis related genes and reactive oxygen producing and scavenging genes. We propose that MED18 and MED20 form a sub-domain within Mediator that controls the balance of salicylic acid and jasmonate associated defense pathways.

  • A Facile Approach to Deposit Graphenaceous Composite Coatings by Suspension Plasma Spraying

    • This paper demonstrates, for the first time ever, the deposition of graphenaceous composite coatings using an easy, yet robust, suspension plasma spraying (SPS) process. As a case study, a composite coating comprising 8 wt.% of yttria-stabilized-zirconia (8YSZ) and reinforced with graphene oxide (GO) was deposited on a steel substrate. The coatings were sprayed using an 8YSZ-GO mixed suspension with varied plasma spray parameters. Establishing the possibility of retaining the graphene in a ceramic matrix using SPS was of specific interest. Electron microscopy and Raman spectroscopy confirmed the presence of graphenaceous material distributed throughout the coating in the 8YSZ matrix. The experimental results discussed in this work confirm that SPS is an immensely attractive pathway to incorporate a graphenaceous material into virtually any matrix material and can potentially have major implications in enabling the deposition of large-area graphene-containing coatings for diverse functional applications.

    • The paper aims at demonstrating various microstructures which can be obtained using the suspension spraying technique and their respective significance in enhancing the thermal insulation property of a thermal barrier coating. Three different types of coating microstructures are discussed which were produced by the Axial Suspension Plasma Spraying. Detailed characterization of coatings was then performed. Optical and scanning electron microscopy were utilized for microstructure evaluations; x-ray diffraction for phase analysis; water impregnation, image analysis, and mercury intrusion porosimetry for porosity analysis, and laser flash analysis for thermal diffusivity measurements were used. The results showed that Axial Suspension Plasma Spraying can generate vertically cracked, porous, and feathery columnar-type microstructures. Pore size distribution was found in micron, submicron, and nanometer range. Higher overall porosity, the lower density of vertical cracks or inter-column spacing, and higher inter-pass porosity favored thermal insulation property of the coating. Significant increase in thermal diffusivity and conductivity was found at higher temperature, which is believed to be due to the pore rearrangement (sintering and pore coarsening). Thermal conductivity values for these coatings were also compared with electron beam physical vapor deposition (EBPVD) thermal barrier coatings from the literature and found to be much lower. © 2015 ASM International

    • Suspension plasma spray is a relatively new thermal spray technique which enables feeding of fine powder to produce advanced coatings for varied applications. This work investigates the difference in structure and performance of Al2O3 coatings manufactured using conventional micron-sized powder feedstock and a suspension of sub-micron to few micron sized powder. Axial injection was implemented for deposition in both cases. The effect of feedstock size and processing on the tribological performance of the two coatings was of specific interest. The coatings were characterized by Optical and Scanning Electron Microscopy, micro-hardness and scratch resistance testing, and their dry sliding wear performance evaluated. The suspension sprayed coatings yielded significantly higher scratch resistance, lower friction coefficient and reduced wear rate compared to conventional coatings. The improved tribological behaviour of the former is attributable to finer porosity, smaller splat sizes, and improved interlamellar bonding.

    • Suspension Plasma Spray (SPS) is an emerging technique which overcomes the difficulties typically associated with feeding of fine (submicron or nano-sized) powders in conventional atmospheric plasma spraying (APS) to obtain superior coating properties for various engineering applications. The advent of plasma spray systems that allow axial injection of feedstock, which considerably improves thermal exchange between the plasma plume and the injected feedstock, has enabled substantial enhancement in deposition rates/efficiencies to make SPS techno-commercially exciting. The present study utilizes both the above advances in plasma spraying to demonstrate the ability to deposit novel coating architectures by sequential/simultaneous axial injection of both powder and a suspension feedstock. The results reveal that composite coatings uniquely combining the micron-size features arising from the spray-grade powder and the submicron or nano-sized features attributable to the suspension, can be conveniently realized using the above approach. Three different kinds of coating architectures were generated, namely layered, composite, and functionally graded. The sprayed coatings were extensively characterized for attributes such as surface morphology, microstructure, and composition particularly in the case of composite coatings.

    • The ability of suspension plasma spraying (SPS) to overcome difficulties associated with feeding of fine (submicron or nano-sized) powders and achieve more refined microstructures than possible in atmospheric plasma spraying (APS) is well established. In recent times, the use of axial injection plasma spray systems has yielded substantial enhancement in deposition rates/efficiencies due to improved thermal exchange between the plasma plume and injected feedstock. The present paper describes utilization of both the above advances in plasma spraying to create various function-dependent coating architectures through simultaneous and/or sequential spraying of hybrid powder-suspension feedstock. A specific variant of such hybrid axial plasma spraying that enables deposition of composite coatings by simultaneous injection of a powder and a suspension is discussed in particular detail. Results obtained using an Al2O3-ZrO2 material system as a case study reveal that composite coatings combining the micron-size features arising from the spray-grade Al2O3 powder and submicron or nano-sized features attributable to the ZrO2 suspension can be conveniently realized. The surface morphology, microstructure, and composition of these coatings, as well as their tribological behaviour determined using scratch and ball-on-disc tests, are presented herein. The utility of this method to develop a wide array of composite coatings is also discussed.

    • Latex coatings are environmentally friendly i.e., they are formed from aqueous polymer dispersions, are cheap to produce and provide exceptional mechanical properties. Therefore, they are ubiquitous and can be found in a wide range of different applications such as paints and varnishes, pressure-sensitive adhesives, textiles, construction materials, paper coatings and inks. However, they also have weaknesses and their surfactant content is among them. Surfactants are often needed to stabilize polymer particles in the aqueous latex dispersions. These surfactants also form part of the coatings formed from these dispersions, and it is well-known that they can lower their performance. This work further explores this aspect and focuses on the role that embedded surfactant domains play in the response of latex coatings to humid environments. For this purpose, we made use of several experimental techniques where humidity control was implemented: quartz crystal microbalance with dissipation, atomic force microscopy and differential scanning calorimetry. By means of this multimethodological approach, we report that surfactants embedded in latex coatings can undergo humidity-induced transitions towards more hydrated and softer phases, and that this results in a drastic decrease of the mechanical and water barrier properties of the whole coatings. Subsequently, this work highlights the potential of taking into account the phase behavior of surfactants when choosing which ones to use in the synthesis of latex dispersions as this would help in predicting their performance under different environmental conditions.

    • Dissolution of atmospheric plasma sprayed (APS) hydroxyapatite (HAp) coatings on Ti-6Al-4 V medical implants have always been a challenge to overcome in the field of biomedical industry. In the present work, an attempt has been made to develop a HAp coating using a novel thermal spray process called axial suspension plasma spraying (SPS), which leads to thin adherent coatings. Two HAp coatings fabricated by APS (P1 and P2) and four SPS HAp coatings (S1, S2, S3 and S4) produced with varying spraying parameters were characterized in terms of (1) microstructure, porosity, hardness, adhesion strength, contact angle and phase purity; (2) corrosion resistance in 10% Fetal bovine serum (FBS); (3) in-vitro cell adherence and cell viability using human umbilical cord blood-derived mesenchymal stem cells (hMSCs). Amongst different APS and SPS coatings, P1 and S3 exhibited superior properties. S3 coating developed using SPS exhibited 1.3 times higher adhesion strength when compared to APS coating (P1) and 9.5 times higher corrosion resistance than P1. In addition, both S3 and P1 exhibited comparatively higher biocompatibility as evidenced by the presence of more than 92% viable hMSCs. © 2018 Elsevier B.V.

    • The strong market trend toward lower fuel consumption for heavy road transports requires more frequent gear shifting and increased gear shift performance, i.e. shorter shift time. Increased shift performance means higher loads for the synchronizer which brings component and shifting process optimization more into focus.Traditionally, synchronizer development has relied on physical testing of complete synchronizers in general gearbox test rigs or in specialized synchronization test rigs leaving much of the causes of the observed effects unclear. This paper presents a generalized FE-based thermomechanical simulation model to be used for model-based synchronizer analysis and design. The model is targeted for studies of how different external loads and the values of different synchronizer design parameters affect the temperature transient in the friction lining. Recommendations of how major modeling complications should be treated are presented. The developed simulation model is verified and validated with a combination of analytical means and transient temperature measurements of bulk and surface temperatures. The applicability of the presented model, as well as its limitations, are discussed and exemplified with different design cases.

    • Molybdenum coated gearbox synchronizers are tested in a mu-comp test rig under varying loading conditions until failure. Four different parameters used to describe the thermomechanical load are evaluated just before failure to compare their ability to predict failure. The parameters evaluated are the synchronized kinetic energy, the synchronization power, and the focal as well as the average surface temperature increase. The focal surface temperature increase as well as the average surface temperature increase is found to predict failure with relatively good accuracy. It is shown that there exists a threshold which divides the synchronizer into either a very long or a very short service life. Additionally, a method to determine the average surface temperature in the gearbox management system is proposed.

    • Gear shifts are becoming more and more important as engines are adapted to low speed high torque working conditions. Synchronizers are key components for successful gear shifts. To adapt the synchronizers to new working conditions due to adaptations to new engines, improved development tools are needed. The presented thesis describes the development of two types of numerical models for the synchronization process, namely fluid-structure interaction to simulate the pre-synchronization phase and thermomechanical FE models to simulate the main synchronization phase. A methodology for developing friction models based on a combination of physical testing and numerical simulations is presented. Additionally, a comprehensive gear shift and synchronizer frame of reference section is presented.In paper A, two numerical fluid-structure interaction simulation models for assessing the pre-synchronization phase are presented. Simulations show that the synchronizer functionality is highly dependent on the gear shift maneuvering system, and that grooves in the synchronizer surface have a positive effect on the oil evacuation during the pre-synchronization phase.Paper B describes the development of a numerical thermomechanical model for simulating the main synchronization phase. Two parameter studies were performed, one based on external loads and one based on synchronizer geometry. The effect on the temperature increase from differences in thermal properties between molybdenum and carbon friction linings are presented.In paper C, a verification and validation methodology for highly transient thermomechanical processes was presented. Numerical verification, bulk temperature measurement, surface temperature measurement and qualitative visual inspection were combined to verify and validate the simulation model presented in paper B.In paper D, a methodology combining physical testing with an thermomechanical simulation model to develop a friction model was exemplified by a molybdenum coated synchronizer. A simplified thermal model was developed to remove the dependence of full finite element thermal models. The friction models shows good agreement with measured data.

    • The coefficient of friction in synchronizers is important from both a performance and a functional point of view. The synchronization process is highly transient, and the parameters affecting the coefficient of friction have strong mutual dependences, making analysis highly complex. A new friction model for a lubricated molybdenum-steel contact has been developed based on integrating results from physical rig tests and FEM simulations. A simplified thermal model has also been developed, with the purpose to quickly assess the coefficient of friction based on transient force and synchronizer dimensions. The results show good correlation with measured data except at very low sliding speed.

    • Transmission performance directly influences vehicle behaviour, energy efficiency, and perceived quality. A modern transmission is an aggregate of interacting sub-systems, e. g. gears, bearings, synchronisers and other shifting elements. New transmission trends ask for increased shift performance with higher loads for the synchronising components. Synchroniser development has traditionally required exhaustive physical testing and few analytical methods exist. A numerical method for assessing pre-synchronisation performance, using a multi-physics FEM software, is presented. Simulations show that grooves in the synchroniser surface have a positive effect on pre-synchronisation, and that the presence of grooves seems to be more important than the groove design.

    • Transmission performance is crucial for heavy trucks and connected vehicles in general and for platooning of trucks in particular. Gearbox synchronizers are highly loaded conical friction brakes used during gear shifts. Service life and, thus, the gear shifting reliability, of the synchronizer depend on the local thermomechanical loading of the contact surface. To achieve a robust and cost-efficient system, more knowledge is needed of how manufacturing tolerances affect the local thermomechanical loading and therefore service life and reliability of a synchronizer. The effects from angle deviations between the mating cones and cone out-of-roundness on focal maximum temperature during a synchronization sequence have been studied with transient thermomechanical simulations. It is shown that thermomechanical effects will significantly magnify the nominal effects on synchronization performance caused by shape deviations given by the specified manufacturing tolerances.

    • This study investigates the corrosion resistance of double layer Gd2Zr2O7/YSZ, triple layer dense Gd2Zr2O7 / Gd2Zr2O7/YSZ and a reference single layer YSZ coating with a similar overall top coat thickness of 300-320 ÎŒm. All the coatings were manufactured by suspension plasma spraying (SPS), resulting in a columnar structure. Corrosion tests were conducted at 900°C for 8 hours using vanadium pentoxide and sodium sulphate as corrosive salts at a concentration of 4 mg/cm2. SEM investigations after the corrosion tests show that Gd2Zr2O7 coatings exhibited lower corrosion resistance than the reference material, YSZ. Reaction between the corrosive salts and Gd2Zr2O7 results in the formation of gadolinium vanadate ( GdVO4) along the top surface and between the columns. While the stresses due to phase transformation of zirconia can be relieved to some extent by realigning of the columns in the top coat, it is believed that GdVO4 formation between the columns, along with low fracture toughness of Gd2Zr2O7 had resulted in lower corrosion resistance. Furthermore, the presence of a relatively dense layer of Gd2Zr2O7 on the top, as a preventive layer for salt infiltration, did not improve the corrosion resistance.

It is believed that the GdVO4 formation between the columns reduced the strain tolerance of the coating and also due to the fact that Gd2Zr2O7 has a lower fracture toughness value made it more susceptible to corrosion-induced damage. Furthermore, the presence of a relatively dense layer of Gd2Zr2O7 on the top did not improve in reducing the corrosion-induced damage. For the reference YSZ coating, the observed corrosion-induced damage was lower probably due to combination of more limited salt penetration, the SPS microstructure and superior fracture toughness of YSZ.

    • Background: It is unclear whether preoperative staging using Magnetic Resonance Imaging (MRI) reduces the risk of positive margins in prostate cancer. We aimed to assess the effect on surgical margins and degree of nerve sparing of a pelvic MRI presented at a preoperative MRI conference. Methods: Single institution, observational cohort study including 1037 men that underwent robot assisted radical prostatectomy between October 2013 and June 2015. Of these, 557 underwent a preoperative MRI combined with a preoperative MRI conference and 410 did not. With whole-mount prostate specimen histopathology as gold standard we assessed the ability of MRI in finding the index tumor and the sensitivity and specificity for extra prostatic extension. We calculated relative risks for positive surgical margins and non-nerve sparing procedure, adjusting for preoperative risk factors using stabilized inverse-probability weighting. Results: MRI detected the index tumor in 80% of the cases. Non-organ confined disease (pT3) at histology was present in the MRI and the non-MRI group in 42% and 24%, respectively. Rate of positive surgical margins comparing the MRI and non-MRI groups was 26.7% and 33.7%, respectively, relative risk 0.79 [95% CI 0.65-0.96], weighted relative risk (wRR) 0.69 [95% CI 0.55-0.86]. The wRR of extensive positive surgical margins was 0.45 [95% CI 0.31-0.67]. Undergoing MRI was also associated with an increased risk of being operated with a non-nerve sparing technique (wRR, 1.84 [95% CI 1.11-3.03]). Conclusions: Our study suggests that preoperative prostate MRI in combination with a preoperative MRI conference affects the degree of nerve-sparing surgery and reduces positive surgical margins.

    • In order to predict and optimize energy efficiency, fuel consumption and service life, friction and wear need to be predetermined with higher accuracy than what is possible today. This prediction and optimization is crucial for the development of sustainable mechanical components and systems with excellent environmental performance.Better and more reliable models for predicting friction, wear and scuffing risk in boundary lubricated contacts will be developed in this project. This includes a model for asperity-asperity collision with components of contact mechanics, thermodynamics and physics.In the boundary lubricated contact, loads are mostly carried by asperities. This makes the real area of contact is so different from the nominal contact area, a small fraction of the nominal contact area supporting the load will cause high contact stress and large deformation. Surfaces of machine components operating under high stress in long period can easily cause damage. Therefore, an elastoplastic analysis of asperity collision was conducted with the Finite Element Method. The contact area and contact stress were studied based on the change of parameters as adhesive friction coefficient, interference and collision velocity. The plastically deformed area and residual stress after collision were also depicted in figures.Friction will generate heat in the sliding contact, and eventually cause a temperature rise. Due to the heat is generated at asperities, heat flux is not continuous and the temperature both increase to a relatively high value and decrease to a small value in very short time. This kind of temperature is often called flahtemperature, and it is important to study because it can affect the viscosity of the lubricant, the formation of tribolayer and in turn it will affect the mechanical properites of the surface. The flash temperature was analyzed based on the previous study of the elastoplastic asperity collision, the times for flash temperature to reach maximum value were given and thermal expansion was also included.The FEM model can conduct a study regardless of the geometry and material properties of the surface asperity, but due to the very fine mesh required at the interface, it is not suitable to carry out an analysis of the rough surface contact. Therefore the Boundary Element Method was adopted to have a thorough study of the rough surface contact. The features of the analysis coudcuted in the FEM model, such as strain hardening and friction, should be replicable in the BEM model. In the end, an Engineering tool for the rough surface contact will be developed.

  • Gear Rolling for Production of High Gears

    • Gears are used to transmit mechanical workfrom one point to another. They are widely used in different mechanisms and they are the most important components of a transmission system. Thus, it is important that they are manufactured with high precision to deliver the work with highest possible efficiency. The dominant gear production method is metal cutting, like hobbing. The gear manufacturing industry aims to replace their traditional production lines with greener processes and thereby urge engineers to think about using metal forming methods instead of the traditional metal cutting solutions when possible. Gear rolling is an interesting metal forming method that can be an alternative method to fabricate gear wheels.Research on gear rolling firstly came into interest around 2000. Very few papers are published that covers the development of the method and its limitations and advantages. Almost all of these publications considered rolling of gear wheels with small modules. The focus of this study will be on application of gear rolling for gear wheels with large module (over 3 mm) where the amount of deformation is much larger than found in previous studies.In this thesis the Finite Element Method has been used to simulate and predict the results of rolling of high gears. In addition to that experiments were performed to validate the numerical results and develop the modelling technique for further investigations. The main topic of discussion is about the gear quality as a measure of the success for the process. Extra attention has been paid to the effects of friction and process planning in the result of process loads and also on the gear quality.The thesis structure is based on four published papers, and some additional results from the experiments which have not yet been published. According to the results of these papers, the effect of friction and process parameters are recognized in the final product quality. It is shown that FEM has a great potential in order to model and analyze the gear rolling process. A new approach to combine numerical methods with quality measurement to predict the process outcomes is also presented. The results show that gear wheels with a module of 4mm reach an ISO quality level between 11 and 12. This is substantially lower quality than can be obtained with metal cutting operations. The results of this thesis can be used as the starting point for future research to optimize the quality of gear rolling for high gears.

    • Chromium oxide (Cr2O3) is commonly used as an atmospheric plasma-sprayed (APS) coating from powder feedstock in applications requiring resistance to sliding wear and corrosion, as well as amenability to texturing, e.g., in anilox rolls. Recently, high-velocity oxy-fuel spray methods involving suspension feedstock have been considered an extremely promising alternative to produce denser and more homogeneous chromium oxide coatings with lower as-sprayed surface roughness, higher hardness and potentially superior wear performance compared to conventional APS-sprayed coatings. In this study, the impact of process parameters namely auxiliary air cleaning nozzles and a transverse air curtain on suspension high-velocity oxy-fuel-sprayed Cr2O3 suspensions is presented. The produced coatings are characterized for their microstructure, mechanical properties and wear resistance by cavitation erosion. The results reveal the importance of optimized air nozzles and air curtain to achieve a vastly improved coating structure and performance.

    • Abstract The use of specimen loss factor as fatigue damage indicator of Hastelloy-X substrates with different surface treatments was investigated together with other fatigue damage indicators, namely resonance frequency and crack mouth length. The tested surface treatments included grit-blasting and plasma spraying of NiCoCrAlY bond coat and yttria stabilized zirconia (YSZ) top coat. The loss factors of fatigue test specimens were measured repeatedly during the resonance bending fatigue test using the conventional free decay method. The analysis of the damping spectra, i.e. the model describing the relation of loss factor to maximum macroscopic specimen strain εyy was drafted. The model is based on the combination of defect models developed by Göken and Riehemann [1] and classical dislocation model of Granato and LÃŒcke [2]. It appears, that the damping spectra can be well approximated as a combination of two defect peaks (C1 and C2) and one dislocation peak (D1). The low strain defect peak (peak C1) is sensitive to the presence of fatigue cracks. The second defect peak (peak C2) can be attributed to the remaining substrate and coating defects such as embedded grit particles, coating porosity, surface roughness and sliding in the sample clamping area. The fatigue damage detection using the C1 peak magnitude was performed and its results were related to the crack length obtained by digital image correlation (DIC) method. In the crack initiation stage I., the C1 peak height shows different behavior than the resonance frequency and therefore provides new information. The underlying processes causing C1 peak changes need to be found yet, however. In the crack growth stage II., both resonance frequency and peak height C1 correlate with the measured fatigue crack size.

    • Many applications of thermally sprayed coatings call for increased fatigue resistance of coated parts. Despite the intensive research in this area, the influence of coating on fatigue is still not completely understood. In this paper, the localization of crack initiation sites and the dynamics of crack propagation are studied. The resonance bending fatigue test was employed to test flat specimens with both sides coated. Hastelloy-X substrates coated with classical thermal barrier coating consisting of yttria stabilized zirconia and NiCoCrAlY layers. The strain distribution on the coating surface was evaluated by the Digital Image Correlation method through the whole duration of the fatigue test. Localization of crack initiation sites and the mode of crack propagation in the coated specimen are related to the observed resonance frequency. The individual phases of specimen degradation, i.e., the changes of material properties, crack initiation, and crack propagation, were identified. The tested coatings strongly influenced the first two phases, and the influence on the crack propagation was less significant. In general, the presented crack detection method can be used as a sensitive nondestructive testing method well suited for coated parts. © 2015 ASM International
