    • To understand the biology and evolution of ruminants, the cattle genome was sequenced to about sevenfold coverage. The cattle genome contains a minimum of 22,000 genes, with a core set of 14,345 orthologs shared among seven mammalian species of which 1217 are absent or undetected in noneutherian (marsupial or monotreme) genomes. Cattle-specific evolutionary breakpoint regions in chromosomes have a higher density of segmental duplications, enrichment of repetitive elements, and species-specific variations in genes associated with lactation and immune responsiveness. Genes involved in metabolism are generally highly conserved, although five metabolic genes are deleted or extensively diverged from their human orthologs. The cattle genome sequence thus provides a resource for understanding mammalian evolution and accelerating livestock genetic improvement for milk and meat production.

    • The methylation of inorganic Hg(II) by methylcobalamin in aquatic systems was studied using high-performance liquid chromatography coupled with UV-digestion cold vapor atomic fluorescence spectrometry (HPLC-UV-CV AFS). Monomethylmercury (MMC) could be positively identified as the reaction product in the aqueous solution. The salinity and pH of the aquatic system have great effects on the formation of MMC, because they could change the species of the reactants in the solution. From an electrophile reaction point of view, salinity and pH alter the electron density of the methyl donor and the electrophilicity of metal ion in the reaction system. This methylation of inorganic Hg(II) is shown to be possible even in highly saline solutions, which indicates its importance in aquatic environments. Kinetic experiments showed that the methylation reaction was fast and first-order for Hg(II). The first-order reaction rate was determined to be 0.00612 and 0.000287 min-1 for pH 5.0 and 1.5, respectively. It is suggested that this methylation could occur in the absence of enzymes, in which Hg(II) acts as an electrophile to attack methylcobalamin with a subsequent transfer of carbanion methyl group to the higher oxidized state of Hg(II).

    • The methylation reaction of inorganic arsenic occurring in aquatic systems was studied by HPLC-HGAFS as a method to separate and detect soluble methylarsenic species. Transformation from inorganic arsenic to methylarsenic was essential for major changes in toxicity to organisms. Monomethylarsenic [AsOCH(3)(OH)(2)] was the only product in the methylation reaction of inorganic arsenic(III) with methyl iodide (MeI). This process can be described as an oxidative carbonium-ion transfer, with MeI acting as a methyl donor. From a thermodynamic point of view, the activity of the carbonium ion and pH were the two major influencing factors. The pH dependence of redox potential of As(III) was the reason for the effect of pH on methylation of arsenic. The influences of salinity and concentration of the methyl donor may be explained by their effects on the activity of carbonium. Moreover, kinetics experiments demonstrated that the methylation reaction was first-order for both As(III) and methyl iodide. First-order reaction rates were also calculated at different pH, salinity and MeI, and were found to be in the range 0.0026-0.0123 h(-1). The methylation rate varied largely under different reaction conditions.

  • Re-establishing the regenerative potential of central nervous system axons in postnatal mice.

    • At a certain point in development, axons in the mammalian central nervous system lose their ability to regenerate after injury. Using the optic nerve model, we show that this growth failure coincides with two developmental events: the loss of Bcl-2 expression by neurons and the maturation of astrocytes. Before postnatal day 4, when astrocytes are immature, overexpression of Bcl-2 alone supported robust and rapid optic nerve regeneration over long distances, leading to innervation of brain targets by day 4 in mice. As astrocytes matured after postnatal day 4, axonal regeneration was inhibited in mice overexpressing Bcl-2. Concurrent induction of Bcl-2 and attenuation of reactive gliosis reversed the failure of CNS axonal re-elongation in postnatal mice and led to rapid axonal regeneration over long distances and reinnervation of the brain targets by a majority of severed optic nerve fibers up to 2 weeks of age. These results suggest that an early postnatal downregulation of Bcl-2 and post-traumatic reactive gliosis are two important elements of axon regenerative failure in the CNS.

  • A novel composite electrolyte based on CeO2 for low temperature solid oxide fuel cells

    • A novel composite material based on mixture of samarium-doped ceria (SDC)-carbonate was studied as electrolyte in low temperature solid oxide fuel cells. The phase and microstructures of composite electrolyte were examined by XRD and SEM. The electrical conductivity was investigated by AC impedance spectroscopy at 400-700 degrees C in different atmospheres. An abrupt change in the conductivity at about 500 degrees C indicates that different mechanisms affect transfer in different temperature ranges. The conductivity increases with the carbonate fraction above 500 degrees C. The conductivity in reduce atmosphere is higher than that in oxide atmosphere. An anode-supported fuel cell using SDC-carbonate as electrolyte was fabricated and tested. The result shows that all the composite electrolytes exhibit better performance than pure SDC electrolyte. The electrolyte with 20wt% carbonate can achieve the highest power density of 415mW center dot cm(-2) and an open circuit voltage of 1.00V at 500 degrees C.

    • Leukotriene B-4 (LTB4) is a potent pro-inflammatory mediator that has been implicated in the pathogenesis of multiple diseases, including psoriasis, inflammatory bowel disease, multiple sclerosis and asthma. As a method to decrease the level of LTB4 and possibly identify novel treatments, inhibitors of the LTB4 biosynthetic enzyme, leukotriene A(4) hydrolase (LTA(4)-h), have been explored. Here we describe the discovery of a potent inhibitor of LTA(4)-h, arylamide of glutamic acid 4f, starting from the corresponding glycinamide 2. Analogs of 4f are then described, focusing on compounds that are both active and stable in whole blood. This effort culminated in the identification of amino alcohol 12a and amino ester 6b which meet these criteria.

    • Although the Gd ion in Gd@C-82 has been shown to lie above the C-C bond on the C-2 axis as an anomalous structure from the MEM/Rietveld analysis, the present XANES study reveals that it lies above the hexagon on the C-2 axis as a normal structure, and Gd oscillates around its equilibrium position with an amplitude increasing with temperature increase.

  • Oxygen measurement by multimode diode lasers employing gas correlation spectroscopy

    • Multimode diode laser (MDL)-based correlation spectroscopy (COSPEC) was used to measure oxygen in ambient air, thereby employing a diode laser (DL) having an emission spectrum that overlaps the oxygen absorption lines of the A band. A sensitivity of 700 ppm m was achieved with good accuracy (2%) and linearity (R-2 = 0.999). For comparison, measurements of ambient oxygen were also performed by tunable DL absorption spectroscopy (TDLAS) technique employing a vertical cavity surface emitting laser. We demonstrate that, despite slightly degraded sensitivity, the MDL-based COSPEC-based oxygen sensor has the advantages of high stability, low cost, ease-of-use, and relaxed requirements in component selection and instrument buildup compared with the TDLAS-based instrument.     • Tribolium castaneum is a representative of earth’s most numerous eukaryotic order, a powerful model organism for the study of generalized insect development, and also an important pest of stored agricultural products. We describe its genome sequence here. This omnivorous beetle has evolved an ability to interact with a diverse chemical environment as evidenced by large expansions in odorant and gustatory receptors, as well as p450 and other detoxification enzymes. Developmental patterns in Tribolium are more representative of other arthropods than those found in Drosophila, a fact represented in gene content and function. For one, Tribolium has retained more ancestral genes involved in cell-cell communication than Drosophila, and some are expressed in the growth zone crucial for axial elongation in short germ development. Systemic RNAi in T. castaneum appears to use mechanisms distinct from those found in C. elegans, but nevertheless offers similar power for the elucidation of gene function and identification of targets for selective insect control.

  • Clinicopathological significance of microRNA-31, -143 and -145 expression in colorectal cancer

    • We are just beginning to understand how microRNAs (miRNAs) are involved in tumor-related processes in humans. Applying real-time RT-PCR, we investigated the miR-31, miR-143 and miR-145 expression in 98 primary CRC specimens, along with the corresponding normal mucosa specimens, and analyze the relationship of their expression with clinicopathological features. Our results showed the miR-31 expression was up-regulated in CRC compared to normal mucosa (p = 0.001). Furthermore, miR-31 expression was positively related to advanced TNM stage (p = 0.026) and deeper invasion of tumors (p = 0.024). MiR-145 was down-regulated in both colon (p = 0.001) and rectal (p = 0.012) cancer. MiR-143 was only down-regulated in colon cancer (p = 0.023) but not in rectal cancer (p = 0.351). There was no relationship of miR-143 and miR-145 expression with other clinicopathological features (p > 0.05), except that the miR-145 expression was related to cancer site (p = 0.03). In conclusion, the miR-31 overexpression may be involved in the development and progression of CRC. The miR-143 and miR-145 may play a certain role in the development of colon and/or rectal cancers but not in progression of the disease.

  • Carbon doped MO-SDC material as an SOFC anode

    • Oxide mixtures MO-SDC, M=Cu, Ni, Co, SDC=Ce0.9Sm0.1O1.95 were synthesized by employing a citrate/nitrate combustion technique. Two kinds of Carbon materials, activated carbon (AC) and vapor grown carbon fiber (VGCF) were homogeneously dispersed into the MO-SDC. The materials can be used as anodes to fabricate single cells using a uniaxial die-press method. The sintering temperature was studied to optimize cell performance. Experimental results showed that cells sintered at 700 degrees C had better performance. When the temperature was above 750 degrees C, the cells were severely distorted, and cannot be tested. Compared with the basic MO-SDC anode, AC and VGCF improve the solid oxide fuel cell (SOFC) anode properties, due to a change of the microstructures of the anode materials which enhance their electron conductivity. Single cell performances were evaluated by I-V measurements, and when 1.25 wt.%VGCF was introduced into the MO-SDC by ball-milling, termed: 1.25 wt.%VGCF-MO-SDC, the 1.25 wt.%VGCF-MO-SDC anode material could achieve the highest power density of up to 0.326 W cm(-2) with H-2 as fuel. The calcination temperature of the MO-SDC dry gel also strongly influenced the electrochemical performance of the 1.25 wt.%VGCF-MO-SDC material. XRD spectra for each calcined temperature and the I-V measurement both suggest that calcinations at 550 degrees C for 1 h are suitable. 1.0 wt.%AC-MO-SDC and 1.25 wt.%VGCF-MO-SDC have similar performance when the cell was fed in methanol/3%H2O, and the corresponding power density was up to 0.253 W cm(-2). Traces of carbon were found in the off-gases.

    • The nitrogen-fixing symbiosis between cyanobacteria and the water fern Azolla microphylla is, in contrast to other cyanobacteria-plant symbioses, the only one of a perpetual nature. The cyanobacterium is vertically transmitted between the plant generations, via vegetative fragmentation of the host or sexually within megasporocarps. In the latter process, subsets of the cyanobacterial population living endophytically in the Azolla leaves function as inocula for the new plant generations. Using electron microscopy and immunogold-labeling, the fate of the cyanobacterium during colonization and development of the megasporocarp was revealed. On entering the indusium chamber of the megasporocarps as small-celled motile cyanobacterial filaments (hormogonia), these differentiated into large thick-walled akinetes (spores) in a synchronized manner. This process was accompanied by cytoplasmic reorganizations and the release of numerous membrane vesicles, most of which contained DNA, and the formation of a highly structured biofilm. Taken together the data revealed complex adaptations in the cyanobacterium during its transition between plant generations.
