  • Imputation and subset-based association analysis across different cancer types identifies multiple independent risk loci in the TERT-CLPTM1L region on chromosome 5p15.33
  • 2014
  • Ingår i: Human Molecular Genetics. - : Oxford University Press (OUP). - 0964-6906 .- 1460-2083. ; 23:24, s. 6616-6633
  • Tidskriftsartikel (refereegranskat)abstract
    • Genome-wide association studies (GWAS) have mapped risk alleles for at least 10 distinct cancers to a small region of 63 000 bp on chromosome 5p15.33. This region harbors the TERT and CLPTM1L genes; the former encodes the catalytic subunit of telomerase reverse transcriptase and the latter may play a role in apoptosis. To investigate further the genetic architecture of common susceptibility alleles in this region, we conducted an agnostic subset-based meta-analysis (association analysis based on subsets) across six distinct cancers in 34 248 cases and 45 036 controls. Based on sequential conditional analysis, we identified as many as six independent risk loci marked by common single-nucleotide polymorphisms: five in the TERT gene (Region 1: rs7726159, P = 2.10 × 10(-39); Region 3: rs2853677, P = 3.30 × 10(-36) and PConditional = 2.36 × 10(-8); Region 4: rs2736098, P = 3.87 × 10(-12) and PConditional = 5.19 × 10(-6), Region 5: rs13172201, P = 0.041 and PConditional = 2.04 × 10(-6); and Region 6: rs10069690, P = 7.49 × 10(-15) and PConditional = 5.35 × 10(-7)) and one in the neighboring CLPTM1L gene (Region 2: rs451360; P = 1.90 × 10(-18) and PConditional = 7.06 × 10(-16)). Between three and five cancers mapped to each independent locus with both risk-enhancing and protective effects. Allele-specific effects on DNA methylation were seen for a subset of risk loci, indicating that methylation and subsequent effects on gene expression may contribute to the biology of risk variants on 5p15.33. Our results provide strong support for extensive pleiotropy across this region of 5p15.33, to an extent not previously observed in other cancer susceptibility loci.

  • Ingår i: European Journal of Clinical Pharmacology. - : Springer Science and Business Media LLC. - 0031-6970 .- 1432-1041. ; 66:12, s. 1217-1227
  • Tidskriftsartikel (refereegranskat)abstract
    • There were great interindividual differences in warfarin maintenance dosage (ranging from 0.6 to 8.4 mg/day) among the 127 patients with mechanical heart valve prostheses. VKORC1-1639G>A, CYP2C9, EPHX1691A>G polymorphism, body weight, and age were found to affect the dose demands. Multiple linear regression models incorporating genetic polymorphisms of VKORC1, CYP2C9, EPHX1691A>G, and the nongenetic factors of age and body weight were developed, and explained up to 76.8% of the total variation (adjusted R (2) of 0.743) in warfarin maintenance doses in southwest Chinese patients with mechanical heart valve prostheses.

  • WT1 promotes cell proliferation in non-small cell lung cancer cell lines through up-regulating cyclin D1 and p-pRb in vitro and in vivo
  • 2013
  • Ingår i: PLOS ONE. - San Francisco : PLoS, Public Library of Science. - 1932-6203. ; 8:8
  • Tidskriftsartikel (refereegranskat)abstract
    • The Wilms' tumor suppressor gene (WT1) has been identified as an oncogene in many malignant diseases such as leukaemia, breast cancer, mesothelioma and lung cancer. However, the role of WT1 in non-small-cell lung cancer (NSCLC) carcinogenesis remains unclear. In this study, we compared WT1 mRNA levels in NSCLC tissues with paired corresponding adjacent tissues and identified significantly higher expression in NSCLC specimens. Cell proliferation of three NSCLC cell lines positively correlated with WT1 expression; moreover, these associations were identified in both cell lines and a xenograft mouse model. Furthermore, we demonstrated that up-regulation of Cyclin D1 and the phosphorylated retinoblastoma protein (p-pRb) was mechanistically related to WT1 accelerating cells to S-phase. In conclusion, our findings demonstrated that WT1 is an oncogene and promotes NSCLC cell proliferation by up-regulating Cyclin D1 and p-pRb expression.

  • Early Failure of Frontline Rituximab-Containing Chemo-immunotherapy in Diffuse Large B Cell Lymphoma Does Not Predict Futility of Autologous Hematopoietic Cell Transplantation
  • 2014
  • Ingår i: Biology of blood and marrow transplantation. - : Elsevier BV. - 1083-8791 .- 1523-6536. ; 20:11, s. 1729-1736
  • Tidskriftsartikel (refereegranskat)abstract
    • The poor prognosis for patients with diffuse large B cell lymphoma (DLBCL) who relapse within 1 year of initial diagnosis after first-line rituximab-based chemo-immunotherapy has created controversy about the role of autologous transplantation (HCT) in this setting. We compared autologous HCT outcomes for chemosensitive DLBCL patients between 2000 and 2011 in 2 cohorts based on time to relapse from diagnosis. The early rituximab failure (ERF) cohort consisted of patients with primary refractory disease or those with first relapse within 1 year of initial diagnosis. The ERF cohort was compared with those relapsing >1 year after initial diagnosis (late rituximab failure [LRF] cohort). ERF and LRF cohorts included 300 and 216 patients, respectively. Nonrelapse mortality (NRM), progression/relapse, progression-free survival (PFS), and overall survival (OS) of ERF versus LRF cohorts at 3 years were 9% (95% confidence interval [CI], 6% to 13%) versus 9% (95% CI, 5% to 13%), 47% (95% CI, 41% to 52%) versus 39% (95% CI, 33% to 46%), 44% (95% CI, 38% to 50%) versus 52% (95% CI, 45% to 59%), and 50% (95% CI, 44% to 56%) versus 67% (95% CI, 60% to 74%), respectively. On multivariate analysis, ERF was not associated with higher NRM (relative risk [RR], 1.31; P = .34). The ERF cohort had a higher risk of treatment failure (progression/relapse or death) (RR, 2.08; P < .001) and overall mortality (RR, 3.75; P < .001) within the first 9 months after autologous HCT. Beyond this period, PFS and OS were not significantly different between the ERF and LRF cohorts. Autologous HCT provides durable disease control to a sizeable subset of DLBCL despite ERF (3-year PFS, 44%) and remains the standard-of-care in chemosensitive DLBCL regardless of the timing of disease relapse.

  • Climate control of terrestrial carbon exchange across biomes and continents
  • 2010
  • Ingår i: Environmental Research Letters. - : IOP Publishing. - 1748-9326. ; 5:3
  • Tidskriftsartikel (refereegranskat)abstract
    • Understanding the relationships between climate and carbon exchange by terrestrial ecosystems is critical to predict future levels of atmospheric carbon dioxide because of the potential accelerating effects of positive climate-carbon cycle feedbacks. However, directly observed relationships between climate and terrestrial CO2 exchange with the atmosphere across biomes and continents are lacking. Here we present data describing the relationships between net ecosystem exchange of carbon (NEE) and climate factors as measured using the eddy covariance method at 125 unique sites in various ecosystems over six continents with a total of 559 site-years. We find that NEE observed at eddy covariance sites is (1) a strong function of mean annual temperature at mid-and high-latitudes, (2) a strong function of dryness at mid-and low-latitudes, and (3) a function of both temperature and dryness around the mid-latitudinal belt (45 degrees N). The sensitivity of NEE to mean annual temperature breaks down at similar to 16 degrees C (a threshold value of mean annual temperature), above which no further increase of CO2 uptake with temperature was observed and dryness influence overrules temperature influence.

  • Particle size : A missing factor in risk assessment of human exposure to toxic chemicals in settled indoor dust
  • 2012
  • Ingår i: Environment International. - : Elsevier. - 0160-4120 .- 1873-6750. ; 49, s. 24-30
  • Tidskriftsartikel (refereegranskat)abstract
    • For researches on toxic chemicals in settled indoor dust, selection of dust fraction is a critical influencing factor to the accuracy of human exposure risk assessment results. However, analysis of the selection of dust fraction in recent studies revealed that there is no consensus. This study classified and presented researches on distribution of toxic chemicals according to dust particle size and on relationship between dust particle size and human exposure possibility. According to the literature, beyond the fact that there were no consistent conclusions on particle size distribution of adherent fraction, dust with particle size less than 100 mu m should be paid more attention and that larger than 250 mu m is neither adherent nor proper for human exposure risk assessment. Calculation results based on literature data show that with different selections of dust fractions, analytical results of toxic chemicals would vary up to 10-fold, which means that selecting dust fractions arbitrarily will lead to large errors in risk assessment of human exposure to toxic chemicals in settled dust. Taking into account the influence of dust particle size on risk assessment of human exposure to toxic chemicals, a new methodology for risk assessment of human exposure to toxic chemicals in settled indoor dust is proposed and human exposure parameter systems to settled indoor dust are advised to be established at national and regional scales all over the world.

  • A novel trigeminal zinc porphyrin and corresponding porphyrin monomers for dye-sensitized solar cells
  • 2014
  • Ingår i: RSC Advances. - : Royal Society of Chemistry (RSC). - 2046-2069. ; 4:21, s. 10439-10449
  • Tidskriftsartikel (refereegranskat)abstract
    • A novel trigeminal zinc porphyrin sensitizer (T) and two zinc porphyrin monomers (M1 and M2) were successfully designed and synthesized. The spectral, electrochemical, and photovoltaic properties of the porphyrin dyes were investigated. Compared with M1, the molecule of M2 has an additional aliphatic n-hexyloxyl chain at the meso-position of the porphyrin framework, and such a structure is favorable for the formation of a compact hydrophobic layer at the TiO2 surface and the retardation of the diffusion of I-3(-) ions into the nanoporous TiO2 electrode, resulting in more effective suppression of the charge recombination process and a higher V-oc. Meanwhile, M2 has larger IPCE values than those of M1, leading to the higher J(sc) value. Thus, the DSSC devices based on M2 demonstrated a relatively high power conversion efficiency of 5.77%, with the J(sc), V-oc and ff values of 13.93 mA cm(-2), 732 mV, and 0.566, respectively. Even though dye T has the highest molar absorption coefficients and multiple binding moieties, the corresponding power conversion efficiency is 2.30%, which is lower than those for M1 and M2. These observations may be ascribed to the low efficiency of the electron injection process caused by the isolation of the LUMOs from the anchoring carboxyl groups in addition to the lowest adsorption amount.

  • Full-colour luminescent compounds based on anthracene and 2,2 '-dipyridylamine
  • 2013
  • Ingår i: Journal of Materials Chemistry C. - : Royal Society of Chemistry (RSC). - 2050-7526 .- 2050-7534. ; 1:44, s. 7409-7417
  • Tidskriftsartikel (refereegranskat)abstract
    • Starting from two simple units of anthracene and 2,2'-dipyridylamine, a series of new luminescent compounds 1-8 were designed and synthesized by a combined strategy of changing the connection mode between the two units, extending the conjugation size, and introducing an additional electron donor. Photophysical properties of 1-8 were investigated and discussed on the basis of solvatochromic behaviour, theoretical calculations, crystal structure, and optimized structures. Interestingly, the emission wavelengths of these compounds could be successfully tuned from violet to red both in solutions and the solid-state, and prominent positive solvatochromism was observed for the compounds with a D-p-A framework. Consider compound 7 as an example, it shows peaks at 526 nm and 627 nm in cyclohexane and DMSO, respectively. Meanwhile, the quantum yield was decreased from 0.80 in cyclohexane to 0.12 in DMSO. The introduction of bulky groups was demonstrated to be effective for suppressing the aggregation effect and thus improving the solid state emission quantum yield. These results indicate that the combined structure modulation strategy offers a powerful tool for tuning the emission behaviour. To demonstrate the possibility of practical applications, 2 was employed as the emitting material for the fabrication of deep-blue organic light-emitting diodes (OLEDs), which showed a maximum external quantum efficiency of 2.2%. The CIE coordinates of (0.15, 0.08) are indicative of excellent blue color purity.

  • TICT based fluorescence "turn-on" hydrazine probes
  • 2014
  • Ingår i: Sensors and actuators. B, Chemical. - : Elsevier BV. - 0925-4005 .- 1873-3077. ; 199, s. 93-100
  • Tidskriftsartikel (refereegranskat)abstract
    • Fluorescence "turn-on" probes PC3 and C3 with weak background emissions were developed for hydrazine sensing. The aldehyde and dicyanovinyl groups were used as the recognition units for PD and C3, respectively. Because of low reactivity of the aldehyde group, the fluorescence of PC3 was enhanced by only ca. 93 folds upon addition of a large amount of 1646 eq. hydrazine. In contrast, 0 exhibited fluorescence enhancement by ca. 239 folds upon addition of only 1.3 eq. hydrazine, and thus it showed high sensitivity towards hydrazine, with the detection limit of 7 ppb. In aqueous systems, it also works well with improved selectivity for hydrazine over CN-. The weak fluorescence of PC3 and 0 can be ascribed to twisted intramolecular charge transfer (TICT) processes by the combination of the bulky diphenylamino and 9-anthryl units, which were well demonstrated by theoretical calculations, viscosity dependent fluorescence, and fluorescence decay behaviour. Addition of hydrazine induced the disappearance of the TICT deactivation pathway, resulting in the observed fluorescence enhancement. It can be concluded that the combination of the bulky diphenylamino and 9-anthryl units is an effective approach for developing fluorescence turn-on hydrazine probes based on the TICT mechanism.

  • 2010
  • Ingår i: International journal of hydrogen energy. - : Elsevier BV. - 0360-3199 .- 1879-3487. ; 35:7, s. 2732-2736
  • Tidskriftsartikel (refereegranskat)abstract
    • Direct carbon fuel cell (DCFC) is a kind of high temperature fuel cell using carbon materials directly as anode. Electrochemical reactivity and surface property of carbon were taken into account in this paper. Four representative carbon samples were selected. The most suitable ratio of the ternary eutectic mixture Li2CO3-K2CO3-Al2O3 was determined at 1.05:1.2:1(mass ration). Conceptual analysis for electrochemical reactivity of carbon anode shows the importance of (1) reactive characteristics including lattice disorder, edge-carbon ratio and the number of short alkyl side chain of carbon material, which builds the prime foundation of the anodic half-cell reaction; (2) surface wetting ability, which assures the efficient contact of anode surface with electrolyte. It indicates that anode reaction rate and DCFC output can be notably improved if carbon are pre-dispersed into electrolyte before acting as anode, due to the straightway shift from cathode to anode for CO32- provided by electrolyte soaked in carbon material.

  • Ceria-carbonate composite for low temperature solid oxide fuel cell : Sintering aid and composite effect
  • 2014
  • Ingår i: International journal of hydrogen energy. - : Elsevier BV. - 0360-3199 .- 1879-3487. ; 39:23, s. 12309-12316
  • Tidskriftsartikel (refereegranskat)abstract
    • In this study, the effect of carbonate content on microstructure, relative density, ionic conductivity and fuel cell performance of Ce0.8Sm0.2O1.9-(Li/Na)(2)CO3 (SDC-carbonate, abbr. SCC) composites is systematically investigated. With the addition of carbonate, the nanoparticles of ceria are well preserved after heat-treatment. The relative densities of SCC pellets increase as the carbonate content increases or sintering temperature rises. Especially, the relative density of SCC2 sintered at 900 degrees C is higher than that of pure SDC sintered at 1350 degrees C. Both the AC conductivity and DC oxygen ionic conductivity are visibly improved compared with the single phase SDC electrolyte. Among the composites, SDC-20 wt% (Li/Na)(2)CO3 (SCC20) presents high dispersion, relative small particle size, and the dense microstructure. The optimized microstructure brings the best ionic conductivity and fuel cell performance. It is hoped that the results can contribute the understanding of the role of carbonate in the composite materials and highlight their prospective application.

  • 2013
  • Ingår i: Clinical Microbiology and Infection. - : Elsevier BV. - 1469-0691 .- 1198-743X. ; 19:9, s. 869-874

    • There is a need for analytical methods capable of monitoring blood antibiotic levels in real time. Here we present a method for quantifying antibiotic levels in whole blood that does not require any sample pretreatment. The tests employ the enzyme penicillinase to assay for penicillin G, penicillin V and ampicillin using a flow-injected biosensor, the Enzyme Thermistor. Optimal flow rates, sample volumes and pH were determined to be 0.5mL/min, 100L and 7.0, respectively. Analysis of the antibiotics diluted in buffer gave a linear range of 0.17-5.0mM. Calibration curves prepared using blood spiked with the antibiotics gave a linear range of 0.17-2.0mM. Linear regression values for all of the calibration curves were 0.998 or higher. Assay cycle time was 5min. The relative standard deviation value for 100 determinations of a mock blood sample spiked with penicillin G was 6.71%. Despite the elimination of sample pretreatment, no detectable clogging or signal drift was observed. The assay provides a fast, simple, reliable analytical method for determining antibiotic concentrations in blood without the need for any sample pretreatment. This is an important first step towards developing a device capable of real-time monitoring of antibiotic levels in whole blood. The technology has the potential to significantly improve the outcomes of patients undergoing critical care.

  • Ingår i: Applied Physics Letters. - : AIP Publishing. - 0003-6951 .- 1077-3118. ; 101:15, s. 154106-

    • The unique electronic properties of graphene are exploited for field-effect sensing in both capacitor and transistor modes when operating the sensor device in electrolyte. The device is fabricated using large-area graphene thin films prepared by means of layer-by-layer stacking. Although essentially the same device, its operation in the capacitor mode is found to yield more information than in the transistor mode. The capacitor sensor can simultaneously detect the variations of surface potential and electrical-double-layer capacitance at the graphene/electrolyte interface when altering the ion concentration. The capacitor-mode operation further facilitates studies of the molecular binding-adsorption kinetics by monitoring the capacitance transient

  • 2011
  • Ingår i: Biosensors & Bioelectronics. - : Elsevier BV. - 1873-4235 .- 0956-5663. ; 29:1, s. 115-118
  • Tidskriftsartikel (refereegranskat)abstract
    • Detection of analytes in complex biological samples, such as milk and blood, normally requires sample pretreatment. These pretreatment regimes reduce assay throughput and increase testing costs. Technologies that make it possible to eliminate sample pretreatment are of great industrial interest. Here we report the development of a dual-signal flow injected analysis device which eliminates the need for sample pretreatment. The device employs thermal traducers to measure the signal from an enzyme and a reference column. This makes it possible to independently monitor and correct for non-specifically generated heat, thereby eliminating the need for sample pretreatment. The ability of the dual-signal device to determine urea and lactate in milk samples without any prior treatment was evaluated. The spiked milk samples, the urea assay had a linear range from 0.1 to 50mM (R=0.996), and the lactate assay had a linear range from 0.025 to 5.0mM (R=0.9998). The linear regression values for urea and lactate for 0.5%, 1.5% and 3.0% fat milk were at least 0.990. The dual-signal design improves assay reproducibility, accuracy and sensitivity. Addition benefits are shorter assay times and lowers costs, as well as reducing equipment and training requirements. The potential application of the technology for multi-analyte analysis in point of care and decentralized diagnostic testing in healthcare, agriculture and environmental areas is discussed.

  • Konferensbidrag (refereegranskat)abstract
    • Composites consisting of Ce0.8Sm0.2O1.9 (SDC)-carbonate were developed as electrolytes for low temperature solid oxide fuel cells (LTSOFC). The SDC power was prepared by sol-gel method. The carbonates were binary eutectics of (Li/Na)2CO3, (Li/K)2CO3 and (K/Na)2CO3. Conductivity measurements showed that the conductivities were depended on the type of carbonates. Discontinuities were found in the Arrhenius plots for both SDC-(Li/Na)2CO3 and SDC-(Li/K)2CO3. For SDC-(Na/K)2CO3 composite electrolyte, the conductivity increased as temperature rose following one slope. Single cells based on various composites were fabricated by a uniaxial die-press method and tested at 450-600 °C. The results showed all cells exhibited improved performances upon that of pure SDC-based cell. The best power density of 532 mW cm -2 at 600 °C was achieved for LTSOFC using composite of SDC and (Li/Na)2CO3. Conductivity mechanism was also discussed.

  • Ingår i: Journal of Power Sources. - : Elsevier BV. - 0378-7753 .- 1873-2755. ; 195:15, s. 4695-4699
  • Tidskriftsartikel (refereegranskat)abstract
    • A composite of samarium doped ceria (SDC) and a binary carbonate eutectic (52 mol% Li2CO3/48 mol% Na2CO3) is investigated with respect to its morphology, conductivity and fuel cell performances. The morphology study shows the composition could prevent SDC particles from agglomeration. The conductivity is measured under air, argon and hydrogen, respectively. A sharp increase in conductivity occurs under all the atmospheres, which relates to the superionic phase transition in the interface phases between SDC and carbonates. Single cells with the composite electrolyte are fabricated by a uniaxial die-press method using NiO/electrolyte as anode and lithiated NiO/electrolyte as cathode. The cell shows a maximum power density of 590 mW cm(-2) at 600 degrees C, using hydrogen as the fuel and air as the oxidant. Unlike that of cells based on pure oxygen ionic conductor or pure protonic conductor, the open circuit voltage of the SDC-carbonate based fuel cell decreases with an increase in water content of either anodic or cathodic inlet gas, indicating the electrolyte is a co-ionic (H+/O2-) conductor. The results also exhibit that oxygen ionic conductivity contributes to the major part of the whole conductivity under fuel cell circumstances. (C) 2010 Elsevier B.V. All rights reserved.

  • Ingår i: Langmuir. - : American Chemical Society (ACS). - 0743-7463 .- 1520-5827. ; 27:17, s. 10677-10682
  • Tidskriftsartikel (refereegranskat)abstract
    • In this article, we experimentally investigate the substrate, wavelength, and time dependence of the plasmon-assisted surface-catalyzed dimerization of 4-nitrobenzenethiol to form p,p'-dimercaptoazobenzene on Au, Ag, and Cu films. We provide direct experimental evidence that surface plasmon resonance plays the most important role in these surface-catalyzed reactions. It is found that the reaction is strongly dependent on the substrate, the wavelength of the laser, and the reaction timescales. Our experimental results revealed that optimal experimental conditions can be rationally chosen to control (accelerate or restrain) this reaction. The experimental results are also confirmed by theoretical calculations.

  • Ingår i: International journal of hydrogen energy. - : Elsevier BV. - 0360-3199 .- 1879-3487. ; 38:26, s. 11398-11405
  • Tidskriftsartikel (refereegranskat)abstract
    • In this work, Lithiated NiCuZnOx (LNCZO) composite is synthesized and evaluated as a potential symmetrical electrode for ceria-carbonate composite electrolyte based low temperature ceramic fuel cells. Its crystal structures, the hydrogen oxidation/oxygen reduction electrochemical activities and fuel cell performances are systematically examined on the symmetrical cell configuration. Nano crystallite particles in the form of composite are observed for these oxides. The LNCZO shows relatively high catalytic activities for hydrogen oxidation and oxygen reduction reaction according to the electrochemical impedance spectroscopy measurements. A remarkable low oxygen reduction activation energy of 42 kJ mol(-1) is obtained on the LNCZO/ceria-carbonate composite, demonstrating excellent electro-catalytic activity. Especially, the catalytic activity can be further improved in the presence of water in the cathode chamber. The results show that the lithiated transition metal oxide composite is a promising symmetrical electrode for ceria-carbonate electrolyte and composite approach might a probable solution to develop super-performance electrodes for reduced temperature ceramic fuel cells.

    • Low temperature solid oxide fuel cells (SOFCs) with metal oxide composite cathode on the ceria–carbonate composite electrolyte have shown promising performance. However, the role of individual elements or compound is seldom investigated. We report here the effect of the ZnO on the physico-chemical and electrochemical properties of lithiated NiO cathode. The materials and single cells are characterized by X-ray diffraction, scanning electron microscopy, DC polarization electrical conductivity, electrochemical impedance spectroscopy and fuel cell performance. The ZnO modified lithiated NiO composite materials exhibit smaller particle size and lower electrical conductivity than lithiated NiO. However, improved electro-catalytic oxygen reduction activity and power output are achieved after the ZnO modification. A maximum power density of 808 mW cm−2 and the corresponding interfacial polarization resistance of 0.22 Ω cm2 are obtained at 550 °C using ZnO modified cathode and 300 μm thick composite electrolyte. The single cell keeps reasonable stability over 300 min at 500 °C. Thus, ZnO modified lithiated NiO is a promising cathode candidate for low temperature SOFCs.

    • Nano-structural components have attracted increasing attention in intermediate/low temperature ceramic fuel cell. We reported here a ceramic fuel cell with a configuration of (Ni/Fe)-NSDC/NSDC/LiNiZnO-NSDC by all nano-composite materials and operated at low temperature range of 500-600°C. The prepared nanocomposite materials are characterized by X-ray diffraction (XRD), Emission scanning electron microscopy (SEM) and Transmission electron microscopy (TEM). Electrochemical performances were studied by current -Voltage, power density characteristics and Ac impedance spectroscopy. The short term stability of fuel cell was also investigated in 100 min. The high fuel cell performance and reasonable stability demonstrated that the all nanocomposite fuel cell concept is feasible and may have great potential in future study.

    • Ceria-carbonate composite materials have been widely investigated as candidate electrolytes for solid oxide fuel cells operated at 300-600 degrees C. However, fundamental studies on the composite electrolytes are still in the early stages and intensive research is demanded to advance their applications. In this study, the crystallite structure, microstructure, chemical activity, thermal expansion behavior and electrochemical properties of the samaria doped ceria-carbonate (SCC) composite have been investigated. Single cells using the SCC composite electrolyte and Ni-based electrodes were assembled and their electrochemical performances were studied. The SCC composite electrolyte exhibits good chemical compatibility and thermal-matching with Ni-based electrodes. Peak power density up to 916 mW cm(-2) was achieved at 550 degrees C, which was attributed to high electrochemical activity of both electrolyte and electrode materials. A stable discharge plateau was obtained under a current density of 1.5 A cm(-2) at 550 degrees C for 120 min. In addition, the ionic conducting property of the SCC composite electrolyte was investigated using electrochemical impedance spectroscopy technique. It was found that the hybrid-ionic conduction improves the total ionic conductivity and fuel cell performance. These results highlight potential low-temperature application of ceria-carbonate composite electrolytes for solid oxide fuel cells.

  • Ingår i: International journal of hydrogen energy. - : Elsevier. - 0360-3199 .- 1879-3487. ; 37:24, s. 19388-19394

    • Pr2NiO4-Ag composite was synthesized and evaluated as cathode component for low temperature solid oxide fuel cells based on ceria-carbonate composite electrolyte. X-ray diffraction analysis reveals that the formation of a single phase K2NiF4-type structure occurs at 1000 °C and Pr2NiO4-Ag composite shows chemically compatible with the composite electrolyte. Symmetrical cells impedance measurements prove that Ag displays acceptable electrocatalytic activity toward oxygen reduction reaction at the temperature range of 500-600 °C. Single cells with Ag active component electrodes present better electrochemical performances than those of Ag-free cells. An improved maximum power density of 695 mW cm-2 was achieved at 600 °C using Pr 2NiO4-Ag composite cathode, with humidified hydrogen as fuel and air as the oxidant. Preliminary results suggest that Pr 2NiO4-Ag composite could be adopted as an alternative cathode for low temperature solid oxide fuel cells.

  • Ingår i: International Journal of Electrochemical Science. - 1452-3981. ; 7:9, s. 8420-8435
  • Tidskriftsartikel (refereegranskat)abstract
    • The impressive ionic conductivity and tunable conduction behaviors have made the ceria-carbonate composite an attractive electrolyte for low temperature ceramic fuel cells. However, the conduction mechanism is not yet well studied. In the present study, both proton and oxygen ion conductivity as well as the transport properties of samaria-doped ceria/ sodium-lithium-carbonate (denoted as SDCLN) composite are investigated by the fuel cell study and the modified Hebb-Wagner polarization measurements. The multi-ionic polarization behaviors and the transfer processes in composite electrolyte under external electrical field are analyzed. A maximum power density of 780 mW cm(-2) and a calculated total ion (proton and oxygen ion) conductivity of 0.153 S cm(-1) are obtained under H-2/air condition at 550 degrees C. The Wagner DC polarization measurements show that the proton conduction dominates the total ionic conductivity. A synergistic effect exists between the charge carriers in the doped ceria-carbonate composite system. An ideal interfacial conduction model is also proposed based on the obtained results.

    • In the last ten years, the research of solid oxide fuel cells (SOFCs) or ceramic fuel cells (CFC) had focused on reducing the working temperature through the development of novel materials, especially the high ionic conductive electrolyte materials. Many progresses on single-phase electrolyte materials with the enhanced ionic conductivity have been made, but they are still far from the criteria of commercialization. The studies of ceria oxide based composite electrolytes give an alternative solution to these problems because of their impressive ionic conductivities and tunable ionic conduction behaviors. Significant advances in the understanding the ceria based composite material and construction of efficient fuel cell systems have been achieved within a short period. This report reviews recent developments of ceria-based composite from different aspects: materials, fundamentals, technologies, fabrication/construction parameters, electrochemistry and theoretical studies. Particular attention is given to ceria-carbonate (nano)composite, including its fuel cell performance, multi-ionic transport properties, advanced applications, corresponding electrode material and stability concerning. Besides, several novel fuel cell (FC) concepts like nanowire FC, all-nanocomposite FC and single-component/electrolyte-free fuel cell (SC-EFFC) are presented. This mini-review emphasizes the promise of ceria-based composites for advanced FC application and highlights the breakthrough of SC-EFFC research for high efficient energy conversion.

    • Composite and nanocomposite samarium doped ceria-carbonates powders were prepared by solidstatereaction, citric acid-nitrate combustion and modified nanocomposite approaches and used aselectrolytes for low temperature solid oxide fuel cells. X-ray Diffraction, Scanning Electron Microscope,low-temperature Nitrogen Adsorption/desorption Experiments, Electrochemical ImpedanceSpectroscopy and fuel cell performance test were employed in characterization of these materials. All powders are nano-size particles with slight aggregation and carbonates are amorphous incomposites. Nanocomposite electrolyte exhibits much lower impedance resistance and higher ionicconductivity than those of the other electrolytes at lower temperature. Fuel cell using the electrolyteprepared by modified nanocomposite approach exhibits the best performance in the whole operationtemperature range and achieves a maximum power density of 839 mW cm−2 at 600 C withH2 as fuel. The excellent physical and electrochemical performances of nanocomposite electrolytemake it a promising candidate for low-temperature solid oxide fuel cells.

    • This paper presents the investigation of the resistive humidity-sensing properties of multi-walled carbon nanotubes (MWCNTs). MWCNTs functionalized by acid treatment (f-MWCNTs) exhibit rather high sensitivity in resistance toward humidity, owing to the presence of carboxylic groups on the nanotube surface. By integrating the f-MWCNTs resistor into a wireless sensor platform, flexible humidity sensors for ultra-high frequency applications are investigated. The operating frequency range of the sensor is dramatically increased from 600 MHz to 2 GHz by adjusting the resistor-electrodes' configuration. This enhancement is predominately attributed to the variation in parasitic capacitance between the resistor-electrodes.

    • IntroductionThe renal clearance of infused crystalloid fluid is very low during anaesthesia and surgery, but experiments in conscious sheep indicate that the renal fluid clearance might approach a normal rate when the adrenergic balance is modified.MethodsSixty females (mean age, 32 years) undergoing laparoscopic gynecological surgery were randomized to control group and received only the conventional anesthetic drugs and 20 ml/kg of lactated Ringer's over 30 mins. The others were also given an infusion of 50 μg/kg/min of esmolol (beta1-receptor blocker) or 0.01 μg/kg/min of phenylephrine (alpha1-adrenergic agonist) over 3 hours. The distribution and elimination of infused fluid were studied by volume kinetic analysis based on urinary excretion and blood hemoglobin level.ResultsBoth drugs significantly increased urinary excretion while heart rate and arterial pressure remained largely unaffected. The urine flows during non-surgery were 43, 147, and 176 ml in the control, esmolol, and phenylephrine groups, respectively (medians, P < 0.03). When surgery had started the corresponding values were 34, 65 and 61 ml (P < 0.04). At 3 hours, averages of 9%, 20%, and 25% of the infused volume had been excreted in the three groups (P < 0.01). The kinetic analyses indicated that both treatments slowed down the distribution of fluid from the plasma to the interstitial fluid space, thereby preventing hypovolemia.ConclusionsEsmolol doubled and phenylephrine almost tripled urinary excretion during anesthesia-induced depression of renal fluid clearance.

  • Ingår i: Applied Geochemistry. - : Elsevier BV. - 0883-2927 .- 1872-9134. ; 47, s. 130-140

    • The interaction of an As-rich natural pyrite (FeS2.08As0.043) with aqueous Se(IV) was investigated as a function of pH, ferrous iron concentration, and reaction time. Arsenic is often the most abundant minor constituent of natural pyrite, and is believed to substitute for S in the pyrite structure. EXAFS measurements confirmed the presence of AsS dianion group, with arsenic in the same local configuration as in the arsenopyrite. Speciation studies indicated that Se(0) was the unique reduction product in the pH range 5.05–8.65 over a reaction period of >1 month, while trace amounts of FeSeO3 might be formed at pH â©Ÿ 6.10. At pH > 6.07, the formation of Fe(III)-(oxyhydr)oxide is kinetically favored, and it consumed nearly all the aqueous iron, including the extra added Fe2+, thereby inhibiting the formation of the thermodynamically most stable product: FeSe2. After oxidation by Se(IV), the occurrence of surface S0, significant aqueous sulfur deficit, and excessive leaching of arsenic in solution, indicate the preferential release of As impurity via arsenopyrite oxidation. The data suggest that the polysulfide-elemental sulfur pathway, which prevails in acid-soluble metal sulfides, is an important pathway in the oxidation of As-rich pyrite, in addition to the thiosulfate pathway for acid-insoluble pyrite. Control experiments on As-free natural pyrite further support this mechanism. This study confirms the potential of reductive precipitation to attenuate the mobility of Se in the environment and demonstrates that minor elements commonly present in natural pyrite can play a significant role on its dissolution pathway.

    • Previous studies from our laboratory identified a number of miRNAs that were aberrantly expressed in colorectal cancer (CRC) tissue. However, their diagnostic and prognostic value in serum has not been fully evaluated. In the present study, we measured the levels of five miRNAs (miR-21, miR-31, miR-92a, miR-18a, and miR-106a) in serum samples from 200 CRC patients, 50 advanced adenoma patients, and 80 healthy controls by real-time quantitative polymerase chain reaction (RT-PCR). In our study, the levels of miR-21 and miR-92a in patients with CRC and advanced adenoma were significantly higher than those in healthy controls (all P  < 0.05). MiR-21 yielded an area under the receiver-operating characteristics (ROC) curve (AUC) of 0.802 and miR-92a yielded an AUC of 0.786 in discriminating CRCs from the controls. Additionally, miR-21 and miR-92a yielded an AUC of 0.709 and 0.701, respectively, in discriminating advanced adenomas from the controls. Combined ROC analyses using both miRNAs, revealed an elevated AUC of 0.847 in discriminating CRCs, and an AUC of 0.722 in discriminating advanced adenomas from the controls. In the multivariate Cox proportional hazards analysis, high miR-92a expression in CRC was independently associated with poor survival (P = 0.03; hazard ratio 4.36; 95 % confidence interval = 1.64–11.57). No significant difference was observed in the levels of miR-18a, miR-31, and miR-106a among CRC, advanced adenoma, and control samples. In summary, our data indicate that miR-21 and miR-92a serum levels have potential value for early detection of CRC. Furthermore, miR-92a has prognostic value in CRC patients.

  • 2011
  • Ingår i: IEEE Electron Device Letters. - 0741-3106 .- 1558-0563. ; 32:9, s. 1299-1301
  • Tidskriftsartikel (refereegranskat)abstract
    • Thin-film field-effect transistors (TFTs) are readily fabricated using a semiconductor composite that is solution processed under ambient conditions for the conduction channel. The composite comprises single-walled carbon nanotubes (SWCNTs) embedded in poly-9,9' dioctyl-fluorene-co-bithiophene. Carrier mobility values approaching 10 cm(2)V(-1)s(-1) are obtained for the composite with relatively high SWCNT concentrations. When the SWCNT concentration is reduced for a large ON/OFF current ratio > 10(6), the mobility remains decent around 0.3 cm(2)V(-1)s(-1). The resultant TFTs display remarkable environmental and operational reliability. Nanotube-based composites are therefore of significance in printed electronics owing to their simplicity in device fabrication and competitiveness in device performance.

  • Tidskriftsartikel (refereegranskat)abstract
    • Significant influence on the thermal stability of polyaniline (PANI) in the presence of multi-walled carbon nanotubes (MWCNTs) is reported. By means of in-situ rapid mixing approach, water-dispersible nanofibrillar PANI and composites, consisting of MWCNTs uniformly coated with PANI in the state of emeraldine salt, with a well-defined core-shell heterogeneous structure, were prepared. The de-protonation process in PANI occurs at a lower temperature under the presence of MWCNTs on the polyaniline composite upon thermal treatment. However, it is found that the presence of MWCNTs significantly enhances the thermal stability of PANI's backbone upon exposure to laser irradiation, which can be ascribed to the core-shell heterogeneous structure of the composite of MWCNTs and PANI, and the high thermal conductivity of MWCNTs.

    • Non-Hodgkin lymphoma (NHL) disproportionately affects older patients, who do not often undergo allogeneic hematopoietic cell transplantation (HCT). We analyzed Center for International Blood and Marrow Transplant Research data on 1248 patients age >= 40 years receiving reduced-intensity conditioning (RIC) or nonmyeloablative (NMA) conditioning HCT for aggressive (n = 668) or indolent (n = 580) NHL Aggressive lymphoma was more frequent in the oldest cohort 49% for age 40 to 54 versus 57% for age 55 to 64 versus 67% for age >= 65; P = .0008). Fewer patients aged >= 65 had previous autografting (26% versus 24% versus 9%; P = .002). Rates of relapse, acute and chronic GVHD, and nonrelapse mortality (NRM) at 1 year post-HCT were similar in the 3 age cohorts (22% [95% confidence interval (CI), 19% to 26%] for age 40 to 54, 27% [95% CI, 23% to 31%] for age 55 to 64, and 34% [95% CI, 24% to 44%] for age >= 65. Progression-free survival (PFS) and overall survival (OS) at 3 years was slightly lower in the older cohorts (OS: 54% [95% CI, 50% to 58%] for age 40 to 54; 40% [95% CI, 36% to 44%] for age 55 to 64, and 39% [95% CI, 28% to 50%] for age >= 65; P < .0001). Multivariate analysis revealed no significant effect of age on the incidence of acute or chronic GVHD or relapse. Age >= 55 years, Karnofsky Performance Status <80, and HLA mismatch adversely affected NRM, PFS, and OS. Disease status at HCT, but not histological subtype, was associated with worse NRM, relapse, PFS, and OS. Even for patients age >= 55 years, OS still approached 40% at 3 years, suggesting that HCT affects long-term remission and remains underused in qualified older patients with NHL.

  • Tidskriftsartikel (refereegranskat)abstract
    • In this paper, two microwave irradiation methods: (i) liquid-phase microwave irradiation (MWI) reduction of graphite oxide suspension dissolved in de-ionized water and N, N-dimethylformamide, respectively, and (ii) solid-phase MWI reduction of graphite oxide powder have been successfully carried out to reduce graphite oxide. The reduced graphene oxide products are thoroughly characterized by scanning electron microscopy, atomic force microscopy, X-ray photoelectron spectroscopy, Fourier transform infrared spectral analysis, Raman spectroscopy, UV-Vis absorption spectral analysis, and four-point probe conductivity measurements. The results show that both methods can efficiently remove the oxygen-containing functional groups attached to the graphite layers, though the solid-phase MWI reduction method can obtain far more efficiently a higher quality-reduced graphene oxide with fewer defects. The I( D)/ I( G) ratio of the solid-phase MWI sample is as low as 0.46, which is only half of that of the liquid-phase MWI samples. The electrical conductivity of the reduced graphene oxide by the solid method reaches 747.9 S/m, which is about 25 times higher than that made by the liquid-phase method.

    • The ever-emerging demands on miniaturization of electronic devices have pushed the development of innovative materials with desired properties. One major endeavor is the development of organic- or organic-inorganic hybrid-based electronics as alternatives or supplements to silicon-based devices. Herein we report the first observation of the coexistence of resistance switching and ferroelectricity in a metal-organic framework (MOF) material, [InC16H11N2O8]center dot 1.5H(2)O, denoted as RSMOF-1. The electrical resistance of RSMOF-1 can be turned on and off repeatedly with a current ratio of 30. A first-principles molecular dynamics simulation suggests that the resistive switching effect is related to the ferroelectric transition of N center dot center dot center dot H-O center dot center dot center dot H-N bridge-structured dipoles of the guest water molecules and the amino-tethered MOF nanochannel. The discovery of the resistive switching effect and ferroelectricity in MOFs offers great potential for the physical implementation of novel electronics for next-generation digital processing and communication.

  • Ingår i: IEEE Transactions on Plasma Science. - 0093-3813. ; 39:8, s. 1689-1694
  • Tidskriftsartikel (refereegranskat)abstract
    • A nonequilibrium Ar/O2 plasma at atmospheric pressure was generated between two parallel square electrodes by a 13.56-MHz power supplier. Experimental results demonstrate that the Ar/O2 plasma more effectively kills Staphylococcus aureus and Escherichia coli than the Ar plasma. When the O2 flow rate increases from 0 to 100 sccm step by step, the discharge voltage and the radio-frequency (RF) current always show sinusoidal waveforms, and the discharges maintain the α mode as well. In addition, the sterilization efficiency values of the RF-powered atmospheric pressure Ar/O2 plasma depend on the oxygen fraction in Ar gas. It is found that the sterilization efficiency of the Ar/O2 plasma is highest with 0.5 vol.% oxygen in argon gas, corresponding to the maximum production of O(3P) radicals. It is considered that the O(3P) radicals produced by the Ar/O2 plasma play a significant role in atmospheric plasma sterilization.

    • Tyrosine kinase inhibitors (TKIs) and reduced intensity conditioning (RIC)/nonmyeloablative (NMA) conditioning hematopoietic cell transplants (HCTs) have changed the therapeutic strategy for chronic myelogenous leukemia (CML) patients. We analyzed post-HCT outcomes of 306 CML patients reported to the Center for International Blood and Marrow Transplant Research aged 40 years and older undergoing RIC/NMA HCT from 2001 to 2007: 117 (38%) aged 40 to 49 years, 119 (39%) 50 to 59 years, and 70 (23%) 60 years or older. The majority (74%) had treatment with imatinib before HCT. At HCT, most patients aged 40 to 49 years were in chronic phase (CP) 1 (74%), compared with 31% aged 60 years or older. Siblings were donors for 56% aged 40 to 49 years; older cohorts had more unrelated donors. The majority received peripheral blood grafts and RIC across all age groups. 3 year overall survival (54%, 52%, and 41%), day + 100 grade II-IV acute GVHD (26%, 32%, and 32%), chronic GVHD (58%, 51%, and 43%), and 1-year treatment-related mortality (18%, 20%, and 13%) were similar across ages. The 3-year relapse incidence (36%, 43%, and 66%) and disease-free survival (35%, 32%, and 16%) were inferior in the oldest cohort. Importantly, for CP1 patients, relapse and disease-free survival were similar across age cohorts. Allogeneic RIC HCT for older patients with CML can control relapse with acceptable toxicity and survival in TKI-exposed CML, especially if still in CP1.

    • Background: Making data available as Linked Data using Resource Description Framework (RDF) promotes integration with other web resources. RDF documents can natively link to related data, and others can link back using Uniform Resource Identifiers (URIs). RDF makes thedata machine-readable and uses extensible vocabularies for additional information, making it easier to scale up inference and data analysis. Results: This paper describes recent developments in an ongoing project converting data from the ChEMBL database into RDF triples. Relative to earlier versions, this updated version of ChEMBL-RDF uses recently introduced ontologies, including CHEMINF and CiTO; exposes more information from the database; and is now available as dereferencable, linked data. To demonstrate these new features, we present novel use cases showing further integration with other web resources, including Bio2RDF, Chem2Bio2RDF, and ChemSpider, and showing the use of standard ontologies for querying. Conclusions: We have illustrated the advantages of using open standards and ontologies to link the ChEMBL database to other databases. Using those links and the knowledge encoded in standards and ontologies, theChEMBL-RDF resource creates a foundation for integrated semantic web cheminformatics applications, such as the presented decision support.

    • A local shear flow field was feasibly generated by pulling the ramie fiber in single fiber reinforced poly(lactic acid) (PLA) composites. This was featured by an ultrahigh shear gradient with a maximum shear rate up to 1500 s(-1), a level comparable to that frequently occurring during the practical polymer processing. To distinguish shear-induced self-nucleation and ramie fiber-induced heterogeneous nucleation, the shear history was classified by pulling the fiber for 5 s (pulled sample) and pulling out the fiber during 10 s (pulled-out sample), while the static fiber-induced crystallization was carried out as the counterpart. As a result of the ultrahigh shear gradient, the combination of primary shear-induced nucleation in the central region and secondary nucleation in the outer layer assembled the unique hierarchical superstructures. By comparing the architectural configurations of interphases formed in the static, pulled, and pulled-out samples, it was shown that the hierarchical cylindrites underwent the process of self-nucleation driven by the applied shear flow, very different from the formation of fiber-induced transcrystallinity (TC) triggered by the heterogeneous nucleating sites at the static fiber surface. The twisting of transcrystallized lamellae may take place due to the spatial hindrance induced by the incredibly dense nuclei under the intense shearing flow, as observed in the synchrotron X-ray diffraction patterns. The influence of chain characteristics on the crystalline morphology was further explored by adding a small amount of poly(ethylene glycol) (PEG) to enhance the molecular mobility of PLA. It was of interest to find that the existence of PEG not only facilitated the growth rates of TC and cylindrites but also improved the preferential orientation of PLA chains and thus expanded the ordered regions. We unearthed lamellar units that were composed of rich fibrillar extended chain crystals (diameter of 50-80 nm). These results are of importance to shed light on tailoring crystalline morphology for natural fibers reinforced green composite materials. Of immense practical significance, too, is the crystalline evolution that has been tracked in the simple model penetrated with an ultrahigh shear gradient, which researchers have so far been unable to replicate during the practical melt processing, such as extrusion and injection molding.

    • Organic bulk heterojunction (BHJ) solar cells based on a dicyanovinyl-substituted oligothiophene as a donor and [6,6]-phenyl C61 butyric acid methyl ester (PCBM) as an acceptor were fabricated and characterized. The oligothiophene derivative can absorb long wavelength photons of the solar radiation, which makes the solar cells with an optimized weight ratio of 1:1.4 have a decent short-circuit current density (12.4 mA/cm(2)) and open-circuit voltage (0.88 V) under AM 1.5G illumination with an intensity of 100 mW/cm(2). A power conversion efficiency (PCE) of 3.7% is achieved, which is among the best PCEs of solution processed small molecule BHJ solar cells.

    • Seafood samples were collected from Dalian, China to study the accumulation and distribution characteristics of short chain chlorinated paraffins (SCCPs) by GC/ECNI-LRMS. Sum of SCCPs (dry weight) were in the range of 77-8 250 ng.g-1, with the lowest value in Scapharca subcrenata and highest concentration in Neptunea cumingi. The concentrations of sum of SCCPs (dry weight) in fish, shrimp/crab and shellfish were in the ranges of 100-3 510, 394-5 440, and 77-8 250 ng.g-1 , respectively. Overall, the C10 and C11 homologues were the most predominant carbon groups of SCCPs in seafood from this area, and a relatively higher proportion of C12-13 was observed in seafood with higher concentrations of sum of SCCPs . With regard to chlorine content, Cl1,, CI8 and CI6 were the major groups. Significant correlations were found among concentrations of different SCCP homologues (except C1, vs. Cl10 ) , which indicated that they might share the same sources and/or have similar accumulation, migration and transformation processes.

  • EHL properties of polyalkylene glycols and their aqueous solutions
  • 2012
  • Ingår i: Tribology letters. - : Springer. - 1023-8883 .- 1573-2711. ; 45:3, s. 379-385
  • Tidskriftsartikel (refereegranskat)abstract
    • Polyalkylene glycols (PAGs) are a type of synthetic lubricants widely used as compressor lubricants, gear oils, hydraulic fluids, and metal working fluids. The PAGs with typical molecular structure can dissolve in water, which makes it a candidate for the base stock of water-based lubricants. Till now, most of the investigations on the water-based lubrication have focussed on the additives. In this work, the potential of PAGs aqueous solutions to replace water as base stocks has been investigated. Four types of PAGs with different molecular weight and their aqueous solutions with different concentrations were studied to reveal their elastohydrodynamic lubrication (EHL) behavior. It has been found that the PAGs solutions can form EHL film like traditional oils. The film-forming capability depends on the viscosity, the pressure-viscosity coefficient, and the molecular weight of PAGs. The results indicate that the PAGs aqueous solution can be employed as base stocks of water-based lubricant

  • Wnt inhibitory factor-1 functions as a tumor suppressor through modulating Wnt/β-catenin signaling in neuroblastoma
  • 2014
  • Ingår i: Cancer Letters. - : Elsevier. - 0304-3835 .- 1872-7980. ; 348:1–2, s. 12-19

    • Neuroblastoma is the most common extracranial solid tumor in childhood and is associated with serious morbidity and mortality. The effective treatment of neuroblastoma remains one of the major challenges in pediatric oncology. The Wnt signaling pathway has been shown to play a significant role in the pathogenesis of adult and pediatric tumors. WIF-1 has been identified as an important Wnt antagonist which inhibits Wnt/β-catenin signaling by directly binding to Wnt proteins. However, the expression and function of WIF-1 in neuroblastoma remains unknown. The present study showed that WIF-1 was downregulated with high level promoter methylation in neuroblastoma cells, and was significantly upregulated after exposure to demethylating agent. This finding suggests that downregulation of WIF-1 was associated with its promoter methylation in neuroblastoma. To further study the potential function of WIF-1 in neuroblastoma, we constructed a plasmid that over-expressed WIF-1 and transfected the plasmid into one neuroblastoma cell line SK-N-SH. We found that restoration of WIF-1 inhibited the growth and proliferation of neuroblastoma cells in vitro. Morever, Wnt/β-catenin signaling activity and target genes expression were reduced by WIF-1 restoration. These results provide support that WIF-1 is downregulated and functions as a tumor suppressor by antagonizing Wnt/β-catenin signaling in neuroblastoma, suggesting a potential role as a therapeutic target in neuroblastoma.

  • Knockdown of MMP-7 inhibits cell proliferation and enhances sensitivity to 5-fluorouracil and X-ray irradiation in colon cancer cells
  • 2014
  • Ingår i: Clinical and Experimental Medicine (Testo stampato). - : Springer Verlag (Germany). - 1591-8890 .- 1591-9528. ; 14:1, s. 99-106
  • Tidskriftsartikel (refereegranskat)abstract
    • The role of matrix metalloproteinase-7 (MMP-7) in the pathogenesis of colon cancer is not understood thoroughly. Previous studies from our group have shown that the expression levels of MMP-7 were highly elevated in colorectal cancer patient specimens and were correlated with Dukes Staging, histological differentiation grade and CEA level. The goal of this study was to investigate the cellular impact of MMP-7 in colon cancer. In this study, we used the SW480 colon cancer cell lines of MMP-7 knockdown by lentivirus-mediated RNA interference as a model system to investigate the impact of MMP-7 on cell proliferation and sensitivity to 5-Fluorouracil (5-FU) and X-ray irradiation (IR). Cell proliferation and sensitivity to 5-FU and IR were measured by MTT assay and colony formation assay. Cell cycle was evaluated by flow cytometry. We showed that the down regulation of MMP-7 inhibits colon cancer cell proliferation and sensitizes tumour cells to 5-FU and IR (P less than 0.05). Decreased MMP-7 expression in SW480 cells by RNA interference triggered cell cycle arrest at G1 phase (P less than 0.05). Down regulation of MMP-7 may inhibit the cell proliferation of colon cancer cells and increase tumour cells sensitivity to radiotherapy and chemotherapy. RNAi-mediated silencing of MMP-7 may represent a powerful therapeutic approach for controlling human colorectal cancer growth.
