  • Plasma biomarkers for Alzheimer's disease: a field-test in a memory clinic

    • Background The key Alzheimer's disease (AD) biomarkers are traditionally measured with techniques/exams that are either expensive (amyloid-positron emission tomography (PET) and tau-PET), invasive (cerebrospinal fluid A beta 42 and p-tau 181), or poorly specific (atrophy on MRI and hypometabolism on fluorodeoxyglucose-PET). Recently developed plasma biomarkers could significantly enhance the efficiency of the diagnostic pathway in memory clinics and improve patient care. This study aimed to: (1) confirm the correlations between plasma and traditional AD biomarkers, (2) assess the diagnostic accuracy of plasma biomarkers as compared with traditional biomarkers, and (3) estimate the proportion of traditional exams potentially saved thanks to the use of plasma biomarkers. Methods Participants were 200 patients with plasma biomarkers and at least one traditional biomarker collected within 12 months. Results Overall, plasma biomarkers significantly correlated with biomarkers assessed through traditional techniques: up to r=0.50 (p<0.001) among amyloid, r=0.43 (p=0.002) among tau, and r=-0.23 (p=0.001) among neurodegeneration biomarkers. Moreover, plasma biomarkers showed high accuracy in discriminating the biomarker status (normal or abnormal) determined by using traditional biomarkers: up to area under the curve (AUC)=0.87 for amyloid, AUC=0.82 for tau, and AUC=0.63 for neurodegeneration status. The use of plasma as a gateway to traditional biomarkers using cohort-specific thresholds (with 95% sensitivity and 95% specificity) could save up to 49% of amyloid, 38% of tau, and 16% of neurodegeneration biomarkers. Conclusion The implementation of plasma biomarkers could save a remarkable proportion of more expensive traditional exams, making the diagnostic workup more cost-effective and improving patient care.

  • Many-body probes for quantum features of spacetime

    • Many theories of quantum gravity can be understood as imposing a minimum length scale the signatures of which can potentially be seen in precise table top experiments. In this work, we inspect the capacity for correlated many-body systems to probe non-classicalities of spacetime through modifications of the commutation relations. We find an analytic derivation of the dynamics for a single mode light field interacting with a single mechanical oscillator and with coupled oscillators to first order corrections to the commutation relations. Our solution is valid for any coupling function as we work out the full Magnus expansion. We numerically show that it is possible to have superquadratic scaling of a nonstandard phase term, arising from the modification to the commutation relations, with coupled mechanical oscillators.

    • We present spectroscopy of the ejecta of SN 1987A in 2017 and 2018 from the Hubble Space Telescope and the Very Large Telescope, covering the wavelength range between 1150 and 10000 angstrom. At 31 yr, this is the first epoch with coverage over the ultraviolet-to-near-infrared range since 1995. We create velocity maps of the ejecta in the H alpha, Mg II lambda lambda 2796, 2804 and [OI] lambda lambda 6302, 6366 (vacuum) emission lines and study their morphology. All three lines have a similar morphology, but Mg II is blueshifted by similar to 1000 km s(-1) relative to the others and stronger in the north-west. We also study the evolution of the line fluxes, finding a brightening by a factor of similar to 9 since 1999 in Mg II, while the other line fluxes are similar in 1999 and 2018. We discuss implications for the power sources of emission lines at late times: thermal excitation due to heating by the X-rays from the ejecta-ring interaction is found to dominate the ultraviolet Mg II lines, while the infrared Mg II doublet is powered mainly by Ly alpha fluorescence. The X-ray deposition is calculated based on merger models of SN 1987A. Far-ultraviolet emission lines of H-2 are not detected. Finally, we examine the combined spectrum of recently discovered hotspots outside the equatorial ring. Their unresolved Balmer emission lines close to zero velocity are consistent with the interaction of fast ejecta and a clumpy, slowly moving outflow. A clump of emission in this spectrum, south of the equatorial ring at similar to 1500 km s(-1), is likely associated with the reverse shock.

  • A global database of Holocene paleotemperature records

    • A comprehensive database of paleoclimate records is needed to place recent warming into the longer-term context of natural climate variability. We present a global compilation of quality-controlled, published, temperature-sensitive proxy records extending back 12,000 years through the Holocene. Data were compiled from 679 sites where time series cover at least 4000 years, are resolved at sub-millennial scale (median spacing of 400 years or finer) and have at least one age control point every 3000 years, with cut-off values slackened in data-sparse regions. The data derive from lake sediment (51%), marine sediment (31%), peat (11%), glacier ice (3%), and other natural archives. The database contains 1319 records, including 157 from the Southern Hemisphere. The multi-proxy database comprises paleotemperature time series based on ecological assemblages, as well as biophysical and geochemical indicators that reflect mean annual or seasonal temperatures, as encoded in the database. This database can be used to reconstruct the spatiotemporal evolution of Holocene temperature at global to regional scales, and is publicly available in Linked Paleo Data (LiPD) format.

  • The resilience of Amazon tree cover to past and present drying

    • The Amazon forest is increasingly vulnerable to dieback and encroachment of grasslands and agricultural fields. Threats to these forested ecosystems include drying, deforestation, and fire, but feedbacks among these make it difficult to determine their relative importance. Here, we reconstruct the central and western Amazon tree cover response to aridity and fire in the mid-Holocene—a time of less intensive human land use and markedly drier conditions than today—to assess the resilience of tree cover to drying and the strength of vegetation-climate feedbacks. We use pollen, charcoal, and speleothem oxygen isotope proxy data to show that Amazon tree cover in the mid-Holocene was resilient to drying in excess of the driest bias-corrected future precipitation projections. Experiments with a dynamic global vegetation model (LPJ-GUESS) suggest tree cover resilience may be owed to weak feedbacks that act to amplify tree cover loss with drying. We also compare these results to observational data and find that, under limited human interference, modern tree cover is likely similarly resilient to mid-Holocene levels of aridification. Our results suggest human-driven fire and deforestation likely pose a greater threat to the future of Amazon ecosystems than drying alone.

  • A JWST Survey of the Supernova Remnant Cassiopeia A

    • We present initial results from a James Webb Space Telescope (JWST) survey of the youngest Galactic core-collapse supernova remnant, Cassiopeia A (Cas A), made up of NIRCam and MIRI imaging mosaics that map emission from the main shell, interior, and surrounding circumstellar/interstellar material (CSM/ISM). We also present four exploratory positions of MIRI Medium Resolution Spectrograph integral field unit spectroscopy that sample ejecta, CSM, and associated dust from representative shocked and unshocked regions. Surprising discoveries include (1) a weblike network of unshocked ejecta filaments resolved to ∼0.01 pc scales exhibiting an overall morphology consistent with turbulent mixing of cool, low-entropy matter from the progenitor's oxygen layer with hot, high-entropy matter heated by neutrino interactions and radioactivity; (2) a thick sheet of dust-dominated emission from shocked CSM seen in projection toward the remnant's interior pockmarked with small (∼1'') round holes formed by ≲01 knots of high-velocity ejecta that have pierced through the CSM and driven expanding tangential shocks; and (3) dozens of light echoes with angular sizes between ∼01 and 1' reflecting previously unseen fine-scale structure in the ISM. NIRCam observations place new upper limits on infrared emission (≲20 nJy at 3 μm) from the neutron star in Cas A's center and tightly constrain scenarios involving a possible fallback disk. These JWST survey data and initial findings help address unresolved questions about massive star explosions that have broad implications for the formation and evolution of stellar populations, the metal and dust enrichment of galaxies, and the origin of compact remnant objects.
