  • A control oriented strategy of disruption prediction to avoid the configuration collapse of tokamak reactors

    • The objective of thermonuclear fusion consists of producing electricity from the coalescence of light nuclei in high temperature plasmas. The most promising route to fusion envisages the confinement of such plasmas with magnetic fields, whose most studied configuration is the tokamak. Disruptions are catastrophic collapses affecting all tokamak devices and one of the main potential showstoppers on the route to a commercial reactor. In this work we report how, deploying innovative analysis methods on thousands of JET experiments covering the isotopic compositions from hydrogen to full tritium and including the major D-T campaign, the nature of the various forms of collapse is investigated in all phases of the discharges. An original approach to proximity detection has been developed, which allows determining both the probability of and the time interval remaining before an incoming disruption, with adaptive, from scratch, real time compatible techniques. The results indicate that physics based prediction and control tools can be developed, to deploy realistic strategies of disruption avoidance and prevention, meeting the requirements of the next generation of devices.

  • Overview of IFMIF-DONES diagnostics : Requirements and techniques

    • The IFMIF-DONES Facility is a unique first-class scientific infrastructure whose construction is foreseen in Granada, Spain, in the coming years. Strong integration efforts are being made at the current project phase aiming at harmonizing the ongoing design of the different and complex Systems of the facility. The consolidation of the Diagnostics and Instrumentation, transversal across many of them, is a key element of this purpose. A top-down strategy is proposed for a systematic Diagnostics Review and Requirement definition, putting emphasis in the one-of-a-kind instruments necessary by the operational particularities of some of the Systems, as well as to the harsh environment that they shall survive. In addition, other transversal aspects such as the ones related to Safety and Machine Protection and their respective requirements shall be also considered. The goal is therefore to advance further and solidly in the respective designs, identify problems in advance, and steer the Diagnostics development and validation campaigns that will be required. The present work provides an overview of this integration strategy as well as a description of some of the most challenging Diagnostics and Instruments within the facility, including several proposed techniques currently under study.

  • Fusion product measurements by nuclear diagnostics in the Joint European Torus deuterium-tritium 2 campaign (invited)

    • A new deuterium-tritium experimental, DTE2, campaign has been conducted at the Joint European Torus (JET) between August 2021 and late December 2021. Motivated by significant enhancements in the past decade at JET, such as the ITER-like wall and enhanced auxiliary heating power, the campaign achieved a new fusion energy world record and performed a broad range of fundamental experiments to inform ITER physics scenarios and operations. New capabilities in the area of fusion product measurements by nuclear diagnostics were available as a result of a decade long enhancement program. These have been tested for the first time in DTE2 and a concise overview is provided here. Confined alpha particle measurements by gamma-ray spectroscopy were successfully demonstrated, albeit with limitations at neutron rates higher than some 10(17) n/s. High resolution neutron spectroscopy measurements with the magnetic proton recoil instrument were complemented by novel data from a set of synthetic diamond detectors, which enabled studies of the supra-thermal contributions to the neutron emission. In the area of escaping fast ion diagnostics, a lost fast ion detector and a set of Faraday cups made it possible to determine information on the velocity space and poloidal distribution of the lost alpha particles for the first time. This extensive set of data provides unique information for fundamental physics studies and validation of the numerical models, which are key to inform the physics and scenarios of ITER.     • This study explores how Swedish manufacturing companies integrate sustainability into their product development process. We study the driving forces behind the decision to develop sustainability strategies and the factors that influence the integration of sustainability into product development. The primary conclusion is that the concept of sustainability has entered the Swedish business culture and become a normative part of the decision ecology to an extent greater than what is mandated by legal requirements. All the actors that we investigate in this study (owners, top managers, employees, suppliers, customers) appear to be pushing for enhanced sustainability. Nonetheless, the main, and most powerful, impetus is very clearly from top management or owners rather than the other actors. This is consistent with an evolving notion that sustainability in the environmental sense is now felt to be related to financial sustainability. Thus, the elite sectors of business (top managers and owners) appear to be imbuing their planning with additional aspects, such as sustainability, as the societal awareness of the connections between forms of sustainability develop. Additionally, we reveal differences based on organization size in terms of how they attempt to implement sustainable new product designs. Smaller companies tend to use more intuitive approaches while larger companies use more "standardized" and globally known "methods," in part because they may be more tightly connected with the global business knowledge ecosystems which tend to evolve and propagate "methods" or "tools."

  • Costs of diabetes complications : hospital based care and production loss for 392,200 people with type 2 diabetes and matched controls in Sweden

    • Background and aims: The prevalence of diabetes has increased rapidly over the last decades worldwide. The risk of complications and medical consequences is well known and identified as key driver of costs. Less evidence on the impact of individual diabetic complications on the societal burden is available. The objective was to analyse costs of hospital-based health care and work absence related to individual macrovascular and microvascular complications of type 2 diabetes in Sweden in 2016.Materials and methods: The study used data from a Swedish retrospective observational database cross-linking 20 years of individual-level data (1997-2016) from national population-based health, social insurance and socio-economic registers for 392,200 people with type 2 diabetes and matched controls (5:1). Diabetes status and presence of 19 types of complications were derived from years 1997-2016 while the costs of hospital-based care and of production loss due to diabetes complications were estimated for 2016. Regression analysis was used for comparison to controls, to attribute production loss to individual complications, and to account for joint presence of complications.Results: Complications are prevalent and patterns complex in type 2 diabetes (Fig. 1). Use of hospital care for complications was higher compared to controls: 86,104 vs 24,608 outpatient visits per 100,000 persons and 9,894 vs 2,546 inpatient admissions per 100,000 persons (p<0.001) in 2016. 26% vs 12% had ≥1 hospital contact. The corresponding total costs of hospital-based care fo rcomplications were EUR 91,875 vs EUR 23,222 per 100 persons (p<0.001) and 75% were directly attributed to diabetes (EUR 689/person). Regression analyses distributed the costs of days absent from work across diabetes complications, basic type 2 diabetes effect and unattributed causes: diabetes complications amounted to EUR 2,165/person in 2016. Key drivers of costs of production loss were macrovascular complications angina pectoris, heart failure and stroke, and microvascular complications eye disease including retinopathy, kidney disease and neuropathy. Early mortality in working ages cost additional EUR 579/person and medications used in risk-factor treatment amounted to EUR 418/person.Conclusion: The economic burden of complications in type 2 diabetes is substantial. Costs of productivity loss in this study were found to be greater than those of hospital-based care highlighting the need for considering treatment consequences in a societal perspective in research and policy.

    • Spherical tokamaks are key to the successful design of operating scenarios of future fusion reactors in the areas of divertor physics, neutral beam current drive and fast ion physics. MAST Upgrade, which has successfully concluded its first experimental campaign, was specifically designed to address the role of the radial gradient of the fast ion distribution in driving the excitation of magneto-hydrodynamic (MHD) instabilities, such as toroidal Alfven eigenmodes, fish-bones and long-lived mode, thanks to its two tangential neutral beam injection systems, one on the equatorial plane and one that is vertically shifted 65 cm above the equatorial plane. To study the fast ion dynamics in the presence of such instabilities, as well as of sawteeth and neo-classical tearing modes, several fast ion diagnostics were upgraded and new ones added. Among them, the MAST prototype neutron camera (NC) has been upgraded to six, equatorial sight-lines. The first observations of the confined fast ion behavior with the upgraded NC in a wide range of plasma scenarios characterized by on-axis and/or off-axis heating and different MHD instabilities are presented here. The observations presented in this study confirm previous results on MAST but with a higher level of detail and highlight new physics observations unique to the MAST Upgrade. The results presented here confirm the improved performance of the NC Upgrade, which thus becomes one of the key elements, in combination with the rich set of fast ion diagnostics available on the MAST Upgrade, for a more constrained modeling of the fast ion dynamics in fusion reactor relevant scenarios.

  • A Contoured-Beam Reflector Satellite Antenna Using Two Doubly Curved Circular Polarization Selective Surfaces

    • We present a reflector antenna system using a circular polarization selective surface (CPSS) to realize a circular polarization (CP) equivalent to the widely used linear polarization (LP) gridded or dual-gridded reflector (DGR). The CPSS is realized by a multilayer stack of non-resonant anisotropic meander line surfaces, achieving a very wide bandwidth. The CPSS reflection and transmission dyadics are computed using full-wave simulations for all angles of incidence and design frequencies applicable, and the doubly curved reflector surface shapes are optimized in the TICRA POS software. The data are input to the TICRA Tools software framework for antenna-system level RF simulations. A planar CPSS sample was first manufactured and tested to demonstrate the adequacy of the selected multilayer design. A doubly curved reflector demonstrator based on the same technology and with a projected aperture of 750 mm was then designed, manufactured, and tested at Ku-band. While promising, the experimental results emphasize the difficulty of producing an electrically large multilayer doubly curved surface with RF performance equivalent to the original flat design and recommendations are provided to improve further the implementation of the proposed design.

    • Background: The putative involvement of chromatin states in multiple sclerosis (MS) is thus far unclear. Here we determined the association of chromatin-accessibility with concurrent genetic, epigenetic and transcriptional events. Material & methods: We generated paired assay for transposase-accessible chromatin sequencing and RNA-sequencing profiles from sorted blood immune CD4+ and CD8+ T cells, CD14+ monocytes and CD19+ B cells from healthy controls (HCs) and MS patients. Results: We identified differentially accessible regions between MS patients and HCs, primarily in CD4+ and CD19+. CD4+ regions were enriched for MS-associated single nucleotide polymorphisms and differentially methylated loci. In the vicinity of differentially accessible regions of CD4+ cells, 42 differentially expressed genes were identified. The top two dysregulated genes identified in this multilayer analysis were CCDC114 and SERTAD1. Conclusion: These findings provide new insight into the primary role of CD4+ and CD19+ cells in MS.

  • Overview on the progress of the conceptual studies of a gamma ray spectrometer instrument for DEMO

    • The future DEMO tokamak will be equipped with a suite of diagnostics which will operate as sensors to monitor and control the position and operation parameters of DT plasmas. Among the suite of sensors, an integrated neutron and gamma-ray diagnostic system is also studied to verify its capability and performance in detecting possible DEMO plasma position variations and contribute to the feedback system in maintaining DEMO DT plasma in stable conditions. This work describes the progress of the conceptual study of the gamma-ray diagnostic for DEMO reactor performed during the first Work-Package contract 2015-2020. The reaction of interest for this Gamma-Ray Spectrometer Instrument (GRSI) consists of D(T, gamma)He-5 with the emission of 16.63 MeV gamma rays. Due to DEMO tokamak design constraints, the gamma and neutron diagnostics are integrated, both featuring multi-line of sight (camera type), viewing DEMO plasma radially with vertical (12) and horizontal (13) viewing lines to diagnose the. and neutron emission from the DT plasma poloidal section. The GRSI design is based on the investigation of the reaction cross sections, on the calculations performed with GENESIS and MCNP simulation codes and on the physics and geometry constrains of the integrated instrument. GRSI features long collimators which diameters are constrained by the neutron flux at the neutron detectors of the Radial Neutron Camera (RNC) system placed in front, which are key to control DEMO DT plasma position. For these reasons, only few GRSI parameters can be independently selected to optimize its performance. Among these, the choice of the collimator diameters at the back side of the neutron detector box up to the GRSI detector, the use of LiH neutron attenuators in front of the GRSI detectors, the GRSI detector material and shielding. The GRSI detector is based on commercial LaBr3(Ce) inorganic scintillating crystal coupled with a photomultiplier tube or a silicon photomultiplier. They are designed to operate at high count rate although GRSI geometry constraints severely impact on this feature. The GRSI can also provide an independent assessment of DEMO DT fusion power and T burning.

    • Embedded systems are today often self-sufficient with limited and predefined communication. However, this traditional view of embedded systems is changing through advancements in technologies such as, communication, cloud technologies, and advanced analytics including machine learning. These advancements have increased the benefits of building Systems of Systems (SoS) that can provide a functionality with unique capabilities that none of the included subsystems can accomplish separately. By this gain of functionality the embedded system is evolving towards a 'dual use' purpose11In this paper we define dual usage as a control system having two purposes. In other contexts such as politics, diplomacy and export control, the term 'dual-use' refers to technology that can be used for both peaceful and military aims, e.g., nuclear power technology., The use is dual in the sense that the system still needs to handle its original task, e.g., control and protect of an asset, and it must provide information for creating the SoS. Larger installations, e.g., industry plants, power systems and generation, have in most cases a long expected life-cycle, some up to 30-40 years without significant updates, compared to analytical functions that evolve and change much faster, i.e., requiring new types of data sets from the subsystems, not know at its first deployment. This difference in development cycles calls for new solutions supporting updates related to new requirements inherent in analytical functions. In this paper, within the context of 'dual usage' of systems and subsystems, we analyze the impact on an embedded system, new or legacy, when it is required to provide analytic data with high quality. We compare a reference system, implementing all functions in one CPU core, to three other alternative solutions: a) a multi-core system where we are using a separate core for analytics, b) using a separate analytics CPU and c) analytics functionality located in a separate subsystem. Our conclusion is that the choice of analytics information collection method should to be based on intended usage, along with resulting complexity and cost of updates compared to hardware cost.

  • Evaluating the hip-flask defence using analytical data from ethanol and ethyl glucuronide. A comparison of two models

    • Aim: Claimed intake of alcohol after a traffic incident, called the hip-flask defence, can be objectively assessed by different methods. One of them is the use of two consecutive ethanol concentrations in urine and the ratio between ethanol concentrations in urine and blood. Another one is the concentrations of ethyl glucuronide (EtG) and ethyl sulphate (EtS) in blood and their ratio to ethanol. The experimental basis for both these models is from single dose studies only. The aim of this study was therefore to describe the kinetics of ethanol, EtG and EtS after ingestion of two repeated doses of ethanol and to investigate the usefulness of the different models for the assessment of the hip-flask defence. Methods: Thirty-five subjects ingested a first dose of 0.51 g of ethanol per kilo body weight, and two hours later a second dose (the hip-flask drink) of 0.25, 0.51 or 0.85 g of ethanol per kilo body weight. Ten urine and 17 blood samples were collected and analysed for ethanol, EtG and EtS using fully validated methods. It was investigated if all subjects fulfilled the criteria for recent drinking, according to the two different models, when using the samples collected 180-240 minutes after start of first dose drinking. According to the first model, increase in urinary ethanol concentrations and a ratio UAC/BAC below 1.3 indicated recent drinking. According to the second model, increase in blood EtG concentrations and a ratio ethanol (g/kg)/EtG (mg/L) above 1 indicated recent drinking. Results: All subjects in the high dose group fulfilled all criteria for recent drinking. One subject in the medium dose group and nine subjects in the low dose group failed to show increasing UAC and/or a UAC/ BAC ratio below 1.3. One subject in the low dose group failed to show increasing concentrations of blood EtG, but all subjects showed a ratio ethanol/EtG above 1. Conclusions: The present study showed, by the use of experimental data, that both two models used to investigate the hip-flask defence can be used, but only when the hip-flask dose is sufficiently high.   • Quantifying Process Quality : The Role of Effective Organizational Learning in Software Evolution

    • Real-world software applications must constantly evolve to remain relevant. This evolution occurs when developing new applications or adapting existing ones to meet new requirements, make corrections, or incorporate future functionality. Traditional methods of software quality control involve software quality models and continuous code inspection tools. These measures focus on directly assessing the quality of the software. However, there is a strong correlation and causation between the quality of the development process and the resulting software product. Therefore, improving the development process indirectly improves the software product, too. To achieve this, effective learning from past processes is necessary, often embraced through post mortem organizational learning. While qualitative evaluation of large artifacts is common, smaller quantitative changes captured by application lifecycle management are often overlooked. In addition to software metrics, these smaller changes can reveal complex phenomena related to project culture and management. Leveraging these changes can help detect and address such complex issues.Software evolution was previously measured by the size of changes, but the lack of consensus on a reliable and versatile quantification method prevents its use as a dependable metric. Different size classifications fail to reliably describe the nature of evolution. While application lifecycle management data is rich, identifying which artifacts can model detrimental managerial practices remains uncertain. Approaches such as simulation modeling, discrete events simulation, or Bayesian networks have only limited ability to exploit continuous-time process models of such phenomena. Even worse, the accessibility and mechanistic insight into such gray- or black-box models are typically very low. To address these challenges, we suggest leveraging objectively captured digital artifacts from application lifecycle management, combined with qualitative analysis, for efficient organizational learning. A new language-independent metric is proposed to robustly capture the size of changes, significantly improving the accuracy of change nature determination. The classified changes are then used to explore, visualize, and suggest maintenance activities, enabling solid prediction of malpractice presence and -severity, even with limited data. Finally, parts of the automatic quantitative analysis are made accessible, potentially replacing expert-based qualitative analysis in parts.

  • Characterization of Selective and Potent JAK1 Inhibitors Intended for the Inhaled Treatment of Asthma

    • Purpose: Janus kinase 1 (JAK1) is implicated in multiple inflammatory pathways that are critical for the pathogenesis of asthma, including the interleukin (IL)-4, IL-5, IL-13, and thymic stromal lymphopoietin cytokine signaling pathways, which have previously been targeted to treat allergic asthma. Here, we describe the development of AZD0449 and AZD4604, two novel and highly selective JAK1 inhibitors with promising properties for inhalation.Methods: The effects of AZD0449 and AZD4604 in JAK1 signaling pathways were assessed by measuring phosphorylation of signal transducer and activator of transcription (STAT) proteins and chemokine release using immunoassays of whole blood from healthy human volunteers and rats. Pharmacokinetic studies performed on rats evaluated AZD0449 at a lung deposited dose of 52 mu g/kg and AZD4604 at 30 mu g/kg. The efficacy of AZD0449 and AZD4604 was assessed by evaluating lung inflammation (cell count and cytokine levels) and the late asthmatic response (average enhanced pause [Penh]).Results: Both compounds inhibited JAK1-dependent cytokine signaling pathways in a dose-dependent manner in human and rat leukocytes. After intratracheal administration in rats, both compounds exhibited low systemic exposures and medium-to-long terminal lung half-lives (AZD0449, 34 hours; AZD4604, 5 hours). Both compounds inhibited STAT3 and STAT5 phosphorylation in lung tissue from ovalbumin (OVA)-challenged rats. AZD0449 and AZD4604 also inhibited eosinophilia in the lung and reduced the late asthmatic response, measured as Penh in the OVA rat model.Conclusion: AZD0449 and AZD4604 show potential as inhibitors of signaling pathways involved in asthmatic immune responses, with target engagement demonstrated locally in the lung. These findings support the clinical development of AZD0449 and AZD4604 for the treatment of patients with asthma.

    • The Joint European Torus (JET) has recently conducted its second deuterium-tritium (DT) experimental campaign DTE2, providing unique opportunity for studying both physics and engineering aspects of nuclear fusion plasmas. This also allowed the exploitation of new diagnostics and technologies that were not available during the first JET DT campaign held in 1997. Among these new instruments, the enhancement projects of the JET nuclear diagnostics lead to the development and installation of synthetic single crystal diamond detectors along different collimated line of sights. This paper describes the single crystal diamond-based diagnostic suite of the JET tokamak and the enhanced 14 MeV neutron diagnostic capabilities in terms of neutron yield and high resolution neutron spectroscopy. The diamond characterization measurements and the calibration procedure at JET are shown, together with performance of the diamond based neutron spectrometer as 14 MeV neutron yield monitor which allows the separation of 2.5 MeV and 14 MeV neutrons in trace tritium plasmas. The first high-resolution 14 MeV neutron spectroscopy measurements in neutral beam injection-heated DT plasmas are presented, allowing thermal and non-thermal neutron component separation. Prospects for the diagnose of DT burning plasmas such as ITER and SPARC will be presented.

    • We aimed to determine whether the sodium/glucose cotransporter family member SGLT3, a proposed glucose sensor, is expressed in the intestine and/or kidney, and if its expression is altered in mouse models of obesity and in humans before and after weight-loss surgery.We used in-situ hybridization and quantitative PCR to determine whether the Sglt3 isoforms 3a and 3b were expressed in the intestine and kidney of C57, leptin-deficient ob/ob, and diabetic BTBR ob/ob mice. Western blotting and immunohistochemistry were also used to assess SGLT3 protein levels in jejunal biopsies from obese patients before and after weight-loss Roux-en-Y gastric bypass surgery (RYGB), and in lean healthy controls.Sglt3a/3b mRNA was detected in the small intestine (duodenum, jejunum and ileum), but not in the large intestine or kidneys of mice. Both isoforms were detected in epithelial cells (confirmed using intestinal organoids). Expression of Sglt3a/3b mRNA in duodenum and jejunum was significantly lower in ob/ob and BTBR ob/ob mice than in normal-weight littermates. Jejunal SGLT3 protein levels in aged obese patients before Roux-en-Y gastric bypass (RYGB) were lower than in lean individuals, but substantially upregulated 6months post-RYGB.Our study shows that Sglt3a/3b is expressed primarily in epithelial cells of the small intestine in mice. Furthermore, we observed an association between intestinal mRNA Sglt3a/3b expression and obesity in mice, and between jejunal SGLT3 protein levels and obesity in humans. Further studies are required to determine the possible role of SGLT3 in obesity.

  • High rate neutron and gamma ray spectroscopy of magnetic confinement fusion plasmas

    • An important instrumental development work has been done in the last two decades in the field of neutron and gamma ray spectroscopic measurements of magnetic confinement plasmas. Starting from the present state of the art instrumentation installed at JET, this paper reviews the recent development that has been carried out within the EUROFUSION programme for the forthcoming high power JET D and DT campaign. This development was dedicated to the realization of new compact neutron and gamma-ray spectrometers which combine very high energy resolution (typically better than 5%) and MHz counting rate capabilities allowing for time resolution in the 10 ms time scale. One of the advantages offered by the compact dimensions of these spectrometers is to make possible their use in multiple sight-line camera configurations, such as for future burning plasma reactors (ITER and DEMO). New compact neutron spectrometers based on single crystal diamond detectors have been developed and installed at JET for measurements of the 14MeV neutron spectrum. Measurements on a portable DT neutron generator have shown that neutron spectroscopy of the accelerated beam ions at unprecedented energy resolution (similar to 1% at 14 MeV) is possible, which opens up new opportunities for diagnosing DT plasmas. For what concerns gamma ray measurements, the JET gamma ray camera has been recently upgraded with new compact spectrometers based on a LaBr3 scintillator coupled to Silicon Photomultiplier with the dual aim to improve the spectroscopic and rate capabilities of the detectors. The upgrade camera system will reconstruct the spatial gamma ray emissivity from the plasma in the MeV energy range at MHz counting rates and energy resolution in the 2-4% range. This will allow physics studies of gamma rays produced by the interaction of fast ions with impurities in the plasma and bremsstrahlung emission from runaway electrons.
