    • Conservation scientists, national governments, and international conservation groups seek to devise, and implement, governance strategies that mitigate human impact on the environment. However, few studies to date have systematically investigated the performance of different systems of governance in achieving successful conservation outcomes. Here, we use a newly-developed analytic framework to conduct analyses of a suite of case studies, linking different governance strategies to standardized scores for delivering ecosystem services, achieving sustainable use of natural resources, and conserving biodiversity, at both local and international levels. Our results: (i) confirm the benefits of adaptive management; and (ii) reveal strong associations for the role of leadership. Our work provides a critical step toward implementing empirically justified governance strategies that are capable of improving the management of human-altered environments, with benefits for both biodiversity and people.

  • Alpha Emitter Radium-223 and Survival in Metastatic Prostate Cancer

    • Background Radium-223 dichloride (radium-223), an alpha emitter, selectively targets bone metastases with alpha particles. We assessed the efficacy and safety of radium-223 as compared with placebo, in addition to the best standard of care, in men with castration-resistant prostate cancer and bone metastases. Methods In our phase 3, randomized, double-blind, placebo-controlled study, we randomly assigned 921 patients who had received, were not eligible to receive, or declined docetaxel, in a 2:1 ratio, to receive six injections of radium-223 (at a dose of 50 kBq per kilogram of body weight intravenously) or matching placebo; one injection was administered every 4 weeks. In addition, all patients received the best standard of care. The primary end point was overall survival. The main secondary efficacy end points included time to the first symptomatic skeletal event and various biochemical end points. A prespecified interim analysis, conducted when 314 deaths had occurred, assessed the effect of radium-223 versus placebo on survival. An updated analysis, when 528 deaths had occurred, was performed before crossover from placebo to radium-223. Results At the interim analysis, which involved 809 patients, radium-223, as compared with placebo, significantly improved overall survival (median, 14.0 months vs. 11.2 months; hazard ratio, 0.70; 95% confidence interval [CI], 0.55 to 0.88; two-sided P=0.002). The updated analysis involving 921 patients confirmed the radium-223 survival benefit (median, 14.9 months vs. 11.3 months; hazard ratio, 0.70; 95% CI, 0.58 to 0.83; P<0.001). Assessments of all main secondary efficacy end points also showed a benefit of radium-233 as compared with placebo. Radium-223 was associated with low myelosuppression rates and fewer adverse events. Conclusions In this study, which was terminated for efficacy at the prespecified interim analysis, radium-223 improved overall survival.   • Long-term valacyclovir suppressive treatment after herpes simplex virus type 2 meningitis: a double-blind, randomized controlled trial.

    • Herpes simplex virus type 2 (HSV-2) is a common cause of acute and recurrent aseptic meningitis. Our aim was to determine the impact of antiviral suppression on recurrence of meningitis and to delineate the full spectrum of neurological complications.One hundred and one patients with acute primary or recurrent HSV-2 meningitis were assigned to placebo (n = 51) or 0.5 g of valacyclovir twice daily (n = 50) for 1 year after initial treatment with 1 g of valacyclovir 3 times daily for 1 week in a prospective, placebo-controlled, multicenter trial. The primary outcome was time until recurrence of meningitis. The patients were followed up for 2 years.The first year, no significant difference was found between the valacyclovir and placebo groups. The second year, without study drugs, the risk of recurrence of verified and probable HSV-2 meningitis was significantly higher among patients exposed to valacyclovir (hazard ratio, 3.29 [95% confidence interval, 10.06-10.21]). One-third of the patients experienced 1-4 meningitis episodes during the study period. A considerable morbidity rate, comprising symptoms from the central, peripheral, and autonomous nervous system, was found in both groups.Suppressive treatment with 0.5 g of valacyclovir twice daily was not shown to prohibit recurrent meningitis and cannot be recommended for this purpose after HSV meningitis in general. Protection against mucocutaneous lesions was observed, but the dosage was probably inappropriate for the prevention of HSV activation in the central nervous system. The higher frequency of meningitis, after cessation of active drug, could be interpreted as a rebound phenomenon.

  • The Short Non-Coding Transcriptome of the Protozoan Parasite Trypanosoma cruzi

    • The pathway for RNA interference is widespread in metazoans and participates in numerous cellular tasks, from gene silencing to chromatin remodeling and protection against retrotransposition. The unicellular eukaryote Trypanosoma cruzi is missing the canonical RNAi pathway and is unable to induce RNAi-related processes. To further understand alternative RNA pathways operating in this organism, we have performed deep sequencing and genome-wide analyses of a size-fractioned cDNA library (16-61 nt) from the epimastigote life stage. Deep sequencing generated 582,243 short sequences of which 91% could be aligned with the genome sequence. About 95-98% of the aligned data (depending on the haplotype) corresponded to small RNAs derived from tRNAs, rRNAs, snRNAs and snoRNAs. The largest class consisted of tRNA-derived small RNAs which primarily originated from the 3' end of tRNAs, followed by small RNAs derived from rRNA. The remaining sequences revealed the presence of 92 novel transcribed loci, of which 79 did not show homology to known RNA classes.

  • Genome analysis and comparative genomics of a Giardia intestinalis assemblage E isolate.

    • Background Giardia intestinalis is a protozoan parasite that causes diarrhea in a wide range of mammalian species. To further understand the genetic diversity between the Giardia intestinalis species, we have performed genome sequencing and analysis of a wild-type Giardia intestinalis sample from the assemblage E group, isolated from a pig. Results We identified 5012 protein coding genes, the majority of which are conserved compared to the previously sequenced genomes of the WB and GS strains in terms of microsynteny and sequence identity. Despite this, there is an unexpectedly large number of chromosomal rearrangements and several smaller structural changes that are present in all chromosomes. Novel members of the VSP, NEK Kinase and HCMP gene families were identified, which may reveal possible mechanisms for host specificity and new avenues for antigenic variation. We used comparative genomics of the three diverse Giardia intestinalis isolates P15, GS and WB to define a core proteome for this species complex and to identify lineage-specific genes. Extensive analyses of polymorphisms in the core proteome of Giardia revealed differential rates of divergence among cellular processes. Conclusions Our results indicate that despite a well conserved core of genes there is significant genome variation between Giardia isolates, both in terms of gene content, gene polymorphisms, structural chromosomal variations and surface molecule repertoires. This study improves the annotation of the Giardia genomes and enables the identification of functionally important variation.

  • Drömmen om den nya staden : Stadsförnyelse i det postsovjetiska Riga

    • The aim of this thesis is to shed light on Post-Soviet urban renewal and people’s perceptions of changes that recently occurred in both the city of Riga and Latvian society more generally since the dissolution of the Soviet Union in the early 1990s. More specifically, this study examines how these perceptions are manifest in Riga’s ongoing renewal. Through applying aspects of continuity and discontinuity, I illustrate how the urban renewal of Post-Soviet Riga uncovers imaginary and emotional aspects of the city and how these are interpreted in relation to the past, present and the future. This study introduces the concept “urban postperestroika” and one important difference between this concept and the more common concepts post-socialist, post-communist or post-Soviet is that the former highlights a process while the latter ones largely highlight the state. The starting point is how urban phantasmagorias – contemporary dreams of the future of the city – elucidate urban renewal processes in general and urban postperestroika in particular. In Riga’s ongoing renewal processes I identify three main trajectories in relation to aspects of the past, present and the future: de-Sovietization, globalization and Lettification. Together, these three trajectories constitute an engine that produces urban phantasmagorias.The dissolution of the Soviet Union has given rise to notions that everything is “back to normal” again, and these notions of normalcy have influenced urban renewal processes. The dilemmas, as shown in this thesis, concern the Soviet period and its remaining psychical structure which give the impression of being too conspicuous to eliminate and too contradictory to assimilate. The study illustrates the difficulties of building new urban and societal structures on the remains of pre-existing orders. Such difficulties of course lead to contradictory and ambiguous world views and to new dysfunctional situations that have to be managed in the future.

    • Purpose: To investigate the dose-response relationship and pain-relieving effect of radium-223, a highly bone-targeted alpha-pharmaceutical. Methods: One hundred patients with castration-resistant prostate cancer (CRPC) and painful bone metastases were randomized to a single intravenous dose of 5, 25, 50 or 100 kBq/kg radium-223. The primary end-point was pain index (visual analogue scale [VAS] and analgesic use), also used to classify patients as responders or non-responders. Results: A significant dose response for pain index was seen at week 2 (P = .035). At week 8 there were 40%, 63%, 56% and 71% pain responders (reduced pain and stable analgesic consumption) in the 5, 25, 50 and 100 kBq/kg groups, respectively. On the daily VAS, at week 8, pain decreased by a mean of -30, -31, -27 and -28 mm, respectively (P = .008, P = .0005, P = .002, and P < .0001) in these responders (post-hoc analysis). There was also a significant improvement in the brief pain inventory functional index for all dose-groups (P = .04, .01, .002 and .02, Wilcoxon signed rank test). Furthermore, a decrease in bone alkaline phosphatase in the highest dose-group was demonstrated (P = .0067). All doses were safe and well tolerated. Conclusion: Pain response was seen in up to 71% of the patients with a dose response observed 2 weeks after administration. The highly tolerable side-effect profile of radium-223 previously reported was confirmed. (C) 2012 Elsevier Ltd. All rights reserved.

  • Genetic Evaluation of Mastistis Liability and Recovery through Longitudinal Models of SCC

    • Genetic evaluation of mastitis is performed either with cross-sectional or longitudinal models. In this study we aim to develop better longitudinal models using simulated SCC (Somatic Cell Count) which usually is used as a proxy to label clinical mastitis. Data was simulated for mastitis liability and recovery for two scenarios (28% and 95% mastitis cases/lactation) and two daughter groups of 60 and 150 per sire in 1200 herds. Weekly observations for SCC were simulated assuming a baseline curve for non-mastitic cows and deviations in case of a mastitis event. Binary data was created to define presence or absence of mastitis as 1 if the simulated SCC was above pre-specified boundary and 0 otherwise. The boundary was allowed to vary along the lactation curve modeled by a spline function with a multiple of 10 or 15. The dynamic nature of the SCC was taken in to consideration with the longitudinal approach and the patterns were captured by modelling transition probability of moving across the boundary. Thus, a transition from below to above the boundary is an indicator of the probability to contract mastitis, and a transition from above to below the boundary is an indicator of the recovery process. Estimated breeding values for mastitis liabilities and recovery were calculated in DMU. Our preliminary results showed the correlation between true and estimated breeding value for the simulated mastitis liability was 0.72 which is as good as the estimations based on clinical mastitis. Though the estimation accuracy for recovery (0.42) was not as high as for mastitis liability the transition probability model enables us to generate breeding values for mastitis recovery process.

    • In the search for a replacement for fossil fuels, due to their depletion as well as an increased concern about our environment, 2nd generation bioethanol comes out as one of the most promising alternatives. There are challenges in several steps of lignocellulose processing – especially due to the formation of for yeast inhibitory compounds during pretreatment and hydrolysis. It has previously been shown that encapsulation of the yeast in membranes made of an alginate gel enables the yeast to survive otherwise toxic hydrolysates. The physiological changes arising from encapsulation are however largely unknown, although it has been shown that the macromolecular composition of the yeast changes during prolonged cultivation. In this study we have therefore performed a comparative proteomic study of yeast grown in capsules and in suspension in anaerobic batch cultivations.

    • Robust second generation bioethanol processes require microorganisms able to obtain high yields and production rates while fermenting inhibiting hydrolysates. However, tolerance towards inhibitors like, carboxylic acids, furan aldehydes and phenolic compounds, is still an issue and the factors contributing to improved tolerance are not well known. In this study, the constitutively flocculating Saccharomyces cerevisiae strain CCUG 53310, with good ability to ferment toxic hydrolysates, was compared with S. cerevisiae CBS 8066 in order to characterize the mechanisms of flocculation and the fermentative performance in different inhibitory media. The flocculation of CCUG 53310 depended on cell wall proteins and was partly inhibited by mannose. The flocculating cells also exhibited a significantly higher hydrophobicity than the cells of the non-flocculating strain CBS 8066, which might contribute to the flocculation. The flocculating strain was more tolerant to carboxylic acids and furan aldehydes, but more sensitive to phenolic compounds. Surprisingly, the expression increase of YAP1, ATR1 and FLR1, known to confer resistance against lignocellulose-derived inhibitors, upon addition of various inhibitors to the fermentation medium, was less in CCUG 53310 than in CBS 8066 in most cases. This indicates that the flocculating strain experienced the cultivation conditions as less stressful. The flocculation in itself is a likely cause of this by creating subinhibitory local levels of inhibitors for most cells, allowing the cells in flocs to experience a lower collective stress level.
