  • The Challenge of Transdisciplinary Research : A Case Study of Learning by Evaluation for Sustainable Transport Infrastructures

    • While transdisciplinary (TD) research is desired in order to solve real world sustainability issues, this may be challenging for both academic and non-academic participants. Supporting learning through evaluation, we analyzed a project aiming at sustainable transport infrastructures. After developing a TD research framework as a benchmark, two external independent evaluators interviewed all project researchers, representatives for end-users, and donors. The evaluators compared results with the framework, and evaluators and participants critically reflected on the results together. There were three inconsistencies relative to the framework: (1) limited understanding of TD research among project management, end-users, and most of the researchers; (2) no structured learning process among end-users; instead, they expressed very diverse opinions about what they expected from the project; (3) project leaders had limited understanding of the special challenges of TD research, did not fully understand the status of the project's social system, and thus did not act as facilitators of the required collaborative learning process. Non-academic participants saw themselves as customers and not as partners in the knowledge production process. We conclude that TD problem-solving research requires much time and needs facilitation and training. A preparatory phase with a lower level of funding would be helpful in preparing for TD processes.

  • Monitoring of the evolution of H2O vapor in the stratosphere of Jupiter over an 18-yr period with the Odin space telescope

    • Context. The comet Shoemaker-Levy 9 impacted Jupiter in July 1994, leaving its stratosphere with several new species, with water vapor (H2O) among them. Aims. With the aid of a photochemical model, H2O can be used as a dynamical tracer in the Jovian stratosphere. In this paper, we aim to constrain the vertical eddy diffusion (Kzz) at levels where H2O is present. Methods. We monitored the H2O disk-averaged emission at 556.936 GHz with the space telescope between 2002 and 2019, covering nearly two decades. We analyzed the data with a combination of 1D photochemical and radiative transfer models to constrain the vertical eddy diffusion in the stratosphere of Jupiter. Results. Odin observations show us that the emission of H2O has an almost linear decrease of about 40% between 2002 and 2019. We can only reproduce our time series if we increase the magnitude of Kzz in the pressure range where H2O diffuses downward from 2002 to 2019, that is, from ~0.2 mbar to ~5 mbar. However, this modified Kzz is incompatible with hydrocarbon observations. We find that even if an allowance is made for the initially large abundances of H2O and CO at the impact latitudes, the photochemical conversion of H2O to CO2 is not sufficient to explain the progressive decline of the H2O line emission, which is suggestive of additional loss mechanisms. Conclusions. The Kzz we derived from the Odin observations of H2O can only be viewed as an upper limit in the ~0.2 mbar to ~5 mbar pressure range. The incompatibility between the interpretations made from H2O and hydrocarbon observations probably results from 1D modeling limitations. Meridional variability of H2O, most probably at auroral latitudes, would need to be assessed and compared with that of hydrocarbons to quantify the role of auroral chemistry in the temporal evolution of the H2O abundance since the SL9 impacts. Modeling the temporal evolution of SL9 species with a 2D model would naturally be the next step in this area of study.

  • Phospholamban antisense oligonucleotides improve cardiac function in murine cardiomyopathy

    • Heart failure (HF) is a major cause of morbidity and mortality worldwide, highlighting an urgent need for novel treatment options, despite recent improvements. Aberrant Ca2+ handling is a key feature of HF pathophysiology. Restoring the Ca2+ regulating machinery is an attractive therapeutic strategy supported by genetic and pharmacological proof of concept studies. Here, we study antisense oligonucleotides (ASOs) as a therapeutic modality, interfering with the PLN/SERCA2a interaction by targeting Pln mRNA for downregulation in the heart of murine HF models. Mice harboring the PLN R14del pathogenic variant recapitulate the human dilated cardiomyopathy (DCM) phenotype; subcutaneous administration of PLN-ASO prevents PLN protein aggregation, cardiac dysfunction, and leads to a 3-fold increase in survival rate. In another genetic DCM mouse model, unrelated to PLN (Cspr3/Mlp−/−), PLN-ASO also reverses the HF phenotype. Finally, in rats with myocardial infarction, PLN-ASO treatment prevents progression of left ventricular dilatation and improves left ventricular contractility. Thus, our data establish that antisense inhibition of PLN is an effective strategy in preclinical models of genetic cardiomyopathy as well as ischemia driven HF.

  • Intraoperative MRI without an intraoperative MRI suite : a workflow for glial tumor surgery

    • BACKGROUND: Intraoperative MRI (iMRI) has emerged as a useful tool in glioma surgery to safely improve the extent of resection. However, iMRI requires a dedicated operating room (OR) with an integrated MRI scanner solely for this purpose. Due to physical or economical restraints, this may not be feasible in all centers. The aim of this study was to investigate the feasibility of using a non-dedicated MRI scanner at the radiology department for iMRI and to describe the workflow with special focus on time expenditure and surgical implications.METHODS: In total, 24 patients undergoing glioma surgery were included. When the resection was deemed completed, the wound was temporarily closed, and the patient, under general anesthesia, was transferred to the radiology department for iMRI, which was performed using a dedicated protocol on 1.5 or 3 T scanners. After performing iMRI the patient was returned to the OR for additional tumor resection or final wound closure. All procedural times, timestamps, and adverse events were recorded.RESULT: The median time from the decision to initiate iMRI until reopening of the wound after scanning was 68 (52-104) minutes. Residual tumors were found on iMRI in 13 patients (54%). There were no adverse events during the surgeries, transfers, transportations, or iMRI-examinations. There were no wound-related complications or infections in the postoperative period or at follow-up. There were no readmissions within 30 or 90 days due to any complication.CONCLUSION: Performing intraoperative MRI using an MRI located outside the OR department was feasible and safe with no adverse events. It did not require more time than previously reported data for dedicated iMRI scanners. This could be a viable alternative in centers without access to a dedicated iMRI suite.

  • Two years of neurosurgical intraoperative MRI in Sweden : evaluation of use and costs

    • BACKGROUND: The current shortage of radiology staff in healthcare provides a challenge for departments all over the world. This leads to more evaluation of how the radiology resources are used and a demand to use them in the most efficient way. Intraoperative MRI is one of many recent advancements in radiological practice. If radiology staff is performing intraoperative MRI at the operation ward, they may be impeded from performing other examinations at the radiology department, creating costs in terms of exams not being performed. Since this is a kind of cost whose importance is likely to increase, we have studied the practice of intraoperative MRI in Sweden.METHODS: The study includes data from the first four hospitals in Sweden that installed MRI scanners adjacent to the operating theaters. In addition, we included data from Karolinska University Hospital in Solna where intraoperative MRI is carried out at the radiology department.RESULTS: Scanners that were moved into the operation theater and doing no or few other scans were used 11-12% of the days. Stationary scanners adjacent to the operation room were used 35-41% of the days. For scanners situated at the radiology department doing intraoperative scans interspersed among all other scans, the proportion was 92%.CONCLUSION: Our study suggests that performing exams at the radiology department rather than at several locations throughout the hospital may be an efficient approach to tackle the simultaneous trends of increasing demands for imaging and increasing staff shortages at radiology departments.
