  • EPOXI: Comet 103P/Hartley 2 Observations from a Worldwide Campaign
  • 2011
  • Ingår i: Astrophysical Journal Letters. - London : IOP. - 2041-8213 .- 2041-8205. ; 734:L1, s. 1-9
  • Tidskriftsartikel (refereegranskat)abstract
    • Earth- and space-based observations provide synergistic information for space mission encounters by providing data over longer timescales, at different wavelengths and using techniques that are impossible with an in situ flyby. We report here such observations in support of the EPOXI spacecraft flyby of comet 103P/Hartley 2. The nucleus is small and dark, and exhibited a very rapidly changing rotation period. Prior to the onset of activity, the period was ~16.4?hr. Starting in 2010 August the period changed from 16.6?hr to near 19?hr in December. With respect to dust composition, most volatiles and carbon and nitrogen isotope ratios, the comet is similar to other Jupiter-family comets. What is unusual is the dominance of CO 2 -driven activity near perihelion, which likely persists out to aphelion. Near perihelion the comet nucleus was surrounded by a large halo of water-ice grains that contributed significantly to the total water production.

  • Autosomal SNP typing of forensic samples with the GenPlex (TM) HID System: Results of a collaborative study
  • 2011
  • Ingår i: Forensic Science International. - : Elsevier. - 1872-4973 .- 1878-0326. ; 5:5, s. 369-375
  • Tidskriftsartikel (refereegranskat)abstract
    • The GenPlex (TM) HID System (Applied Biosystems - AB) offers typing of 48 of the 52 SNPforID SNPs and amelogenin. Previous studies have shown a high reproducibility of the GenPlex (TM) HID System using 250500 pg DNA of good quality. An international exercise was performed by 14 laboratories (9 in Europe and 5 in the US) in order to test the robustness and reliability of the GenPlex (TM) HID System on forensic samples. Three samples with partly degraded DNA and 10 samples with low amounts of DNA were analyzed in duplicates using various amounts of DNA. In order to compare the performance of the GenPlex (TM) HID System with the most commonly used STR kits, 500 pg of partly degraded DNA from three samples was typed by the laboratories using one or more STR kits. The median SNP typing success rate was 92.3% with 500 pg of partly degraded DNA. Three of the fourteen laboratories counted for more than two thirds of the locus dropouts. The median percentage of discrepant results was 0.2% with 500 pg degraded DNA. An increasing percentage of locus dropouts and discrepant results were observed when lower amounts of DNA were used. Different success rates were observed for the various SNPs. The rs763869 SNP was the least successful. With the exception of the MiniFiler (TM) kit (AB), GenPlex (TM) HID performed better than five other tested STR kits. When partly degraded DNA was analyzed, GenPlex (TM) HID showed a very low mean mach probability, while all STR kits except MiniFiler (TM) had very limited discriminatory power.

  • Comparison of primary human cytotoxic T-cell and natural killer cell responses reveal similar molecular requirements for lytic granule exocytosis but differences in cytokine production
  • 2013
  • Ingår i: Blood. - : American Society of Hematology. - 0006-4971 .- 1528-0020. ; 121:8, s. 1345-1356
  • Tidskriftsartikel (refereegranskat)abstract
    • Cytotoxic lymphocytes, encompassing cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells, kill pathogen-infected, neoplastic, or certain hematopoietic cells through the release of perforin-containing lytic granules. In the present study, we first performed probability-state modeling of differentiation and lytic granule markers on CD8(+) T cells to enable the comparison of bona fide CTLs with NK cells. Analysis identified CD57(bright) expression as a reliable phenotype of granule marker-containing CTLs. We then compared CD3(+)CD8(+)CD57(bright) CTLs with NK cells. Healthy adult peripheral blood CD3(+)CD8(+)CD57(bright) CTLs expressed more granzyme B but less perforin than CD3(-)CD56(dim) NK cells. On stimulation, such CTLs degranulated more readily than other T-cell subsets, but had a propensity to degranulate that was similar to NK cells. Remarkably, the CTLs produced cytokines more rapidly and with greater frequency than NK cells. In patients with biallelic mutations in UNC13D, STX11, or STXBP2 associated with familial hemophagocytic lymphohistiocytosis, CTL and NK cell degranulation were similarly impaired. Therefore, cytotoxic lymphocyte subsets have similar requirements for Munc13-4, syntaxin-11, and Munc18-2 in lytic granule exocytosis. The present results provide a detailed comparison of human CD3(+)CD8(+)CD57(bright) CTLs and NK cells and suggest that analysis of CD57(bright) CTL function may prove useful in the diagnosis of primary immunodeficiencies including familial hemophagocytic lymphohistiocytosis.

  • Glucose metabolism and body composition in young adults treated with TBI during childhood
  • 2011
  • Ingår i: Bone Marrow Transplantation. - : Springer Science and Business Media LLC. - 0268-3369 .- 1476-5365. ; 46:10, s. 1303-1308
  • Tidskriftsartikel (refereegranskat)abstract
    • After SCT in childhood, survivors may develop disorders of glucose metabolism. The role of obesity is controversial. We measured insulin sensitivity using the homeostasis model assessment (HOMA) and the frequently sampled i.v. glucose tolerance test (FSIVGTT), as well as body composition using dual-energy X-ray absorptiometry in 18 young adults median 18.2 years after SCT and compared them with matched controls. We also measured growth hormone (GH) secretion, and levels of leptin and adiponectin. HOMA showed insulin resistance in eight patients (44%), as opposed to none of the controls (P=0.008) and FSIVGTT showed a decreased sensitivity index in the patients (2.98 vs 4.54 mU/L/min, P=0.042). Dual energy X-ray absorptiometry showed a higher percentage fat mass in the patients (34.9 vs 24.3%, P=0.011), which correlated inversely with the sensitivity index (r=-0.52, P=0.032). The patients had a lower peak value of GH (GH(max) 9 vs 20.7 mU/L, P=0.002). Time post SCT correlated with percentage fat mass and inversely with GH(max). The patients had higher levels of leptin and lower levels of adiponectin, even after adjustment for fat mass. We propose that the decreased insulin sensitivity may primarily be explained by the adverse body composition, which may owe to long-standing GH deficiency.

  • Surface modification of PM gearlever
  • 2010
  • Ingår i: World Powder Metallurgy Congress and Exhibition, World PM 2010; Florence; Italy; 10 October 2010 through 14 October 2010. - : European Powder Metallurgy Association (EPMA). - 9781899072194 ; 3
  • Konferensbidrag (refereegranskat)abstract
    • A novel manufacturing concept for a gearlever has been evaluated by manufacturing of test components of two materials. Iron-based powders were used, and pressing and sintering was followed by a few different variants of hardening and surface densification. The components have been evaluated by means of component function testing, component fatigue testing, microstructure characterization, hardness measurements and residual stress determination. The gearlever is found to perform well in the function testing and shows also good fatigue properties that well match the demands for the intended application. The porosity in the sintered material is inhomogeneous, and some compaction-induced defects were observed in the material, which thus seems not to give any significant negative impact on the sintered product performance. The results show a doubling in hardness after hardening. If the materials were shot-peened (or shot-blasted) after hardening the surface hardness increased down to a depth of approximately 0.2 mm compared to the hardened material. The case-hardening should be applied after shot-peening to obtain significant effect on surface hardness within the outermost 0.1 mm. In principle no surface densification was created if shoot-peening was performed after hardening.

  • Combining CAR T cells and the Bcl-2 family apoptosis inhibitor ABT-737 for treating B-cell malignancy
  • 2013
  • Ingår i: Cancer Gene Therapy. - : Springer Science and Business Media LLC. - 0929-1903 .- 1476-5500. ; 20:7, s. 386-393
  • Tidskriftsartikel (refereegranskat)abstract
    • B-cell malignancies upregulate the B-cell lymphoma 2 (Bcl-2) family inhibitors of the intrinsic apoptosis pathway, making them therapy resistant. However, small-molecule inhibitors of Bcl-2 family members such as ABT-737 restore a functional apoptosis pathway in cancer cells, and its oral analog ABT-263 (Navitoclax) has entered clinical trials. Gene engineered chimeric antigen receptor (CAR) T cells also show promise in B-cell malignancy, and as they induce apoptosis via the extrinsic pathway, we hypothesized that small-molecule inhibitors of the Bcl-2 family may potentiate the efficacy of CAR T cells by engaging both apoptosis pathways. CAR T cells targeting CD19 were generated from healthy donors as well as from pre-B-ALL (precursor-B acute lymphoblastic leukemia) patients and tested together with ABT-737 to evaluate apoptosis induction in five B-cell tumor cell lines. The CAR T cells were effective even if the cell lines exhibited different apoptosis resistance profiles, as shown by analyzing the expression of apoptosis inhibitors by PCR and western blot. When combining T-cell and ABT-737 therapy simultaneously, or with ABT-737 as a presensitizer, tumor cell apoptosis was significantly increased. In conclusion, the apoptosis inducer ABT-737 enhanced the efficacy of CAR T cells and could be an interesting drug candidate to potentiate T-cell therapy.

  • New Ordovician-Silurian drill cores from the Siljan impact structure in central Sweden: an integral part of the Swedish Deep Drilling Program
  • 2012
  • Ingår i: Gff. - : Informa UK Limited. - 1103-5897 .- 2000-0863. ; 134:2, s. 87-98
  • Tidskriftsartikel (refereegranskat)abstract
    • New drill cores from the largest known impact structure in Europe, the relict of the Siljan meteorite crater, provide new possibilities to reconstruct Early Palaeozoic marine environments and ecosystems, and to document changes in sedimentary facies, sea level and palaeoclimate in Baltoscandia. The impact crater is an important target of the project "Concentric Impact Structures in the Palaeozoic" within the framework of the "Swedish Deep Drilling Program". Two core sections, Mora 001 and Solberga 1, have been analysed. The sedimentary successions of these core sections include strata of late Tremadocian through late Wenlock ages. Our preliminary studies show not only that several of the classical Palaeozoic units of Sweden are represented in the area, but also that other significantly different facies are preserved in the Siljan district. An erosional unconformity representing a substantial hiatus occurs between Middle Ordovician limestone and a Llandovery-Wenlock (Silurian) shale succession in the western part of the Siljan structure and suggests an extended period of uplift and erosion. This may be related to forebulge migration due to flexural loading by the Caledonian thrust sheet to the west. Thus, this part of Sweden, previously regarded as a stable cratonic area, presumably was affected by the Caledonian collision between Baltica and Laurentia.

  • Compound materials by PM-HIP
  • 2014
  • Ingår i: Powder Metallurgy. - 0032-5899 .- 1743-2901. ; 57:5, s. 341-347
  • Tidskriftsartikel (refereegranskat)abstract
    • There are many applications where compound materials can be of interest, for example when different properties are needed in different parts of a component. Compound materials can be produced by hot isostatic pressing (HIP) of powder metallurgical materials. One aspect that should be considered in the design is the quality of the interface between the two different material compositions. Diffusion during HIP can cause formation of brittle phases in the interface or deteriorate properties by diffusion of alloying elements. The present work shows results from a study where different steel types were joined (quench and temper steel/air hardening steel/bearing steel with a tool steel/corrosion resistant martensitic steel). The evaluation was performed by computational predictions and by small scale HIP experiments that were evaluated by microstructure analysis and chemical analysis. © 2014 Institute of Materials, Minerals and Mining.

  • Detailed modelling of the susceptibility of a thermally populated, strongly driven circuit-QED system
  • 2013
  • Ingår i: Journal of Physics B: Atomic, Molecular and Optical Physics. - : IOP Publishing. - 1361-6455 .- 0953-4075. ; 46:22
  • Tidskriftsartikel (refereegranskat)abstract
    • We present measurements and modelling of the susceptibility of a 2D microstrip cavity coupled to a driven transmon qubit. We are able to fit the response of the cavity to a weak probe signal with high accuracy in the strong coupling, low detuning, i.e., non-dispersive, limit over a wide bandwidth. The observed spectrum is rich in multi-photon processes for the doubly dressed transmon. These features are well explained by including the higher transmon levels in the driven Jaynes-Cummings model and solving the full master equation to calculate the susceptibility of the cavity.

  • Overview of the Martian atmospheric submillimetre sounder FIRE
  • 2012
  • Ingår i: Planetary and Space Science. - : Elsevier BV. - 0032-0633 .- 1873-5088. ; 63-64:SI, s. 62-82
  • Tidskriftsartikel (refereegranskat)abstract
    • We propose a submillimetre-wave atmospheric emission sounding instrument, called Far-InfraRed Experiment (FIRE), for the Japanese Martian exploration programme "Mars Exploration with Lander-Orbiter Synergy" (MELOS). The scientific target of FIRE/MELOS is to understand the dust suspended meteorology of the Mars. FIRE will provide key meteorological parameters, such as atmospheric temperature profiles for outside and inside dust storms, the abundance profile of the atmospheric compositions and their isotopes, and wind velocity profiles. FIRE will also provide the local time dependency of these parameters. The observational sensitivity of FIRE/MELOS is discussed in this paper. FIRE will explore the meteorological system of the Martian atmosphere including the interaction between its surface and atmosphere.

  • Mass concentration of Hirnantian cephalopods from the Siljan District, Sweden; taxonomy, palaeoecology and palaeobiogeographic relationships
  • 2011
  • Ingår i: Fossil Record. - : Pensoft Publishers. - 1435-1943. ; 14:1, s. 35-53
  • Tidskriftsartikel (refereegranskat)abstract
    • The Hirnantian Glisstjarn Formation (Normalograptus persculptus graptolite Biozone) is a succession of limestones and shales onlapping the Katian Boda Limestone in the Siljan District, Sweden. It contains a conspicuous, up to several decimeter thick bed densely packed with bipolarly oriented, orthoconic cephalopod conchs that can reach lengths of more than 120 cm. Conch fragmentation, bioereosion and the generally poor preservation of the conchs indicate time averaging and the conchs are tentatively interpreted as beached, and a result of winnowing. Ten nautiloid species were collected from the Glisstjarn Formation of which five are new: Dawsonoceras gregarium n. sp., Discoceras siljanense n. sp., Isorthoceras dalecarlense n. sp., Retizitteloceras rarum gen. et sp. n., and Transorthoceras osmundsbergense gen. et sp. n. The non-endemic taxa in most cases are known from elsewhere in Baltoscandia, except one species which is known from Siberia, and North America respectively. Proteocerid orthoceridans dominate the association, of which T. osmundsbergense is the predominant species. Oncocerids are diverse but together with tarphycerids very rare. Notable is the lack of many higher taxa, that are typical for other Late Ordovician shallow water depositional settings. Based on the taxonomical composition of the cephalopod mass occurrence it is interpreted as an indicator of eutrophication of the water masses in the area.

    • Mejare IA, Axelsson S, Davidson T, Frisk F, Hakeberg M, Kvist T, Norlund A, Petersson A, Portenier I, Sandberg H, Tran ae us S, Bergenholtz G. Diagnosis of the condition of the dental pulp: a systematic review. International Endodontic Journal, 45, 597613, 2012. Abstract The aim of this systematic review was to appraise the diagnostic accuracy of signs/symptoms and tests used to determine the condition of the pulp in teeth affected by deep caries, trauma or other types of injury. Radiographic methods were not included. The electronic literature search included the databases PubMed, EMBASE, The Cochrane Central Register of Controlled Trials and Cochrane Reviews from January 1950 to June 2011. The complete search strategy is given in an Appendix S1 (available online as Supporting Information). In addition, hand searches were made. Two reviewers independently assessed abstracts and full-text articles. An article was read in full text if at least one of the two reviewers considered an abstract to be potentially relevant. Altogether, 155 articles were read in full text. Of these, 18 studies fulfilled pre-specified inclusion criteria. The quality of included articles was assessed using the QUADAS tool. Based on studies of high or moderate quality, the quality of evidence of each diagnostic method/test was rated in four levels according to GRADE. No study reached high quality; two were of moderate quality. The overall evidence was insufficient to assess the value of toothache or abnormal reaction to heat/cold stimulation for determining the pulp condition. The same applies to methods for establishing pulp status, including electric or thermal pulp testing, or methods for measuring pulpal blood circulation. In general, there are major shortcomings in the design, conduct and reporting of studies in this domain of dental research.

  • 2012
  • Ingår i: International Endodontic Journal. - : Wiley. - 0143-2885 .- 1365-2591. ; 45:9, s. 783-801

    • Petersson A, Axelsson S, Davidson T, Frisk F, Hakeberg M, Kvist T, Norlund A, Mejare I, Portenier I, Sandberg H, Tranaeus S, Bergenholtz G. Radiological diagnosis of periapical bone tissue lesions in endodontics: a systematic review. International Endodontic Journal, 45, 783801, 2012. Abstract This systematic review evaluates the diagnostic accuracy of radiographic methods employed to indicate presence/absence and changes over time of periapical bone lesions. Also investigated were the leads radiographic images may give about the nature of the process and the condition of the pulp in nonendodontically treated teeth. Electronic literature search included the databases PubMed, Embase and CENTRAL from January 1950 to June 2011. All languages were accepted provided there was an abstract in English. The MeSH terms were Cone beam computed tomography (CBCT), Radiography, panoramic, Periapical diseases, Dental pulp diseases, Sensitivity and specificity, receiver operating characteristics (ROC) curve, Cadaver, Endodontics and Radiography dental. Two reviewers independently assessed abstracts and full text articles. An article was read in full text if at least one of the two reviewers considered an abstract to be potentially relevant. Altogether, 181 articles were read in full text. The GRADE approach was used to assess the quality of evidence of each radiographic method based on studies of high or moderate quality. Twenty-six studies fulfilled criteria set for inclusion. None was of high quality; 11 were of moderate quality. There is insufficient evidence that the digital intraoral radiographic technique is diagnostically as accurate as the conventional film technique. The same applies to CBCT. No conclusions can be drawn regarding the accuracy of radiological examination in identifying various forms of periapical bone tissue changes or about the pulpal condition.
