  • Dietary fat and breast cancer risk in the European Prospective Investigation into Cancer and Nutrition.

    • BACKGROUND:Epidemiologic studies have produced conflicting results with respect to an association of dietary fat with breast cancer.OBJECTIVE:We aimed to investigate the association between fat consumption and breast cancer.DESIGN:We prospectively investigated fat consumption in a large (n = 319,826), geographically and culturally heterogeneous cohort of European women enrolled in the European Prospective Investigation into Cancer and Nutrition who completed a dietary questionnaire. After a mean of 8.8 y of follow-up, 7119 women developed breast cancer. Cox proportional hazard models, stratified by age and center and adjusted for energy intake and confounders, were used to estimate hazard ratios (HRs) for breast cancer.RESULTS:An association between high saturated fat intake and greater breast cancer risk was found [HR = 1.13 (95% CI: 1.00, 1.27; P for trend = 0.038) for the highest quintile of saturated fat intake compared with the lowest quintile: 1.02 (1.00, 1.04) for a 20% increase in saturated fat consumption (continuous variable)]. No significant association of breast cancer with total, monounsaturated, or polyunsaturated fat was found, although trends were for a direct association of risk with monounsaturated fat and an inverse association with polyunsaturated fat. In menopausal women, the positive association with saturated fat was confined to nonusers of hormone therapy at baseline [1.21 (0.99, 1.48) for the highest quintile compared with the lowest quintile; P for trend = 0.044; and 1.03 (1.00, 1.07) for a 20% increase in saturated fat as a continuous variable].CONCLUSIONS:Evidence indicates a weak positive association between saturated fat intake and breast cancer risk. This association was more pronounced for postmenopausal women who never used hormone therapy.

  • Haplotype-based analysis of common variation in the acetyl-CoA carboxyiase alpha gene and breast cancer risk: A case-control study nested within the European prospective investigation into cancer and nutrition

    • A key fatty acid synthesis enzyme, acetyl-CoA carboxylase alpha(ACC-alpha), has been shown to be highly expressed in human breast cancer and other tumor types and also to specifically interact with the protein coded by one of two major breast cancer susceptibility genes BRCA1. We used a comprehensive haplotype analysis to examine the contribution of the ACC-alpha common genetic variation (allele frequency > 5%) to breast cancer in a case-control study (1,588 cases/2,600 controls) nested within the European Prospective Investigation into Cancer and Nutrition. We identified 21 haplotypetagging polymorphisms efficiently capturing common variation within 325 kb of ACC-alpha and surrounding sequences using genotype data from the HapMap project and our resequencing data. We found an effect on overall risk of breast cancer in homozygous carriers of one common haplotype [odds ratio (OR), 1.74; 95% confidence interval (95% CI), 1.03-2.94]. When the data were subdivided by menopausal status, we found statistical evidence of heterogeneity for two other common haplotypes (P value for heterogeneity = 0.016 and 0.045). In premenopausal women, the carriers of these haplotypes, compared with noncarriers, had an altered risk of breast cancer (OR, 0.70; 95% CI, 0.53-0.92 and OR, 1.35; 95% CI, 1.04-1.76). These findings were not significant after adjustment for multiple testing and therefore should be considered as preliminary and evaluated in larger independent studies. However, they suggest a possible role of the ACC-alpha common sequence variants in susceptibility to breast cancer and encourage studies of other genes involved in fatty acid synthesis.

    • Metabolizing enzymes, which often display genetic polymorphisms, are involved in the activation of compounds present in tobacco smoke that may be relevant to gastric carcinogenesis. We report the results of a study looking at the association between risk of gastric adenocarcinoma and polymorphisms in genes CYP1A1, CYP1A2, EPHX1, and GSTT1. A nested case-control study was carried out within the European Prospective Investigation into Cancer and Nutrition, developed in 10 European countries. The study includes 243 newly diagnosed cases of histologically confirmed gastric adenocarcinoma and 946 controls matched by center, age, sex, and date of blood collection. Genotypes were determined in nuclear DNA from WBCs. We found an increased risk of gastric cancer for homozygotes for C (histidine) variant in Y113H of EPHX1 (odds ratio, 1.91; 95% confidence interval, 1.19-3.07) compared with subjects with TC/TT. There was also a significant increased risk for smokers carrying at least one variant allele A in Ex7+129C > A (m4) of CYP1A1 and never smokers with null GSTT1 and allele A in the locus -3859G > A of CYP1A2. Most of these genes are involved in the activation and detoxification of polycyclic aromatic hydrocarbons, suggesting a potential role of these compounds in gastric carcinogenesis.

    • Java is a successful programming environment and its use has grown from little embedded applications until enterprise network servers based on J2EE. This intensive use of Java demands the validation of their fault tolerance mechanisms to avoid unexpected behavior of the applications at runtime. This paper describes the design and implementation of a fault injector for the "Exhaustif®" SWIFI tool. Aspecific fault model for java applications that include class corruption/substitution at loading time, method call interception and unexpected exception thrown is proposed. The injector uses the JVMTI (JavaVirtual Machine Tool Interface) to perform bytecode instrumentation at runtime to carry out the fault model previously defined. Finally a XML formalization of the specific Java fault model is proposed. This approach, JVMTI + XML fault model description, provides complete independency between the system under test and the fault injection tool, as well the interoperability with another SWIFI tools.

    • This paper presents Exhaustif®, a SWIFI fault injection tool for fault tolerance verification and the validation of embedded software in distributedheterogeneous systems. Exhaustif® mainly consists of two parts: EEM and FIK. Exhaustif® Executive Manager (EEM) is a GUI Java application to define the fault injection campaign that uses a SQL database to save the test results obtained from the System under Test (SUT) in order to carry out a post injection data analysis. FIK is under the command of EEM to cary out fault injections in applications running under diverse operating systems using pure SWIFI techniques. Exhaustif® carries out floating point register and memory corruptions using temporary triggers and uses an optimized routine interception mechanism to cany out argument and return value corruption with a minimal time overhead. Two experimentalFault Injector Kernels (FIK) under the RTEMS operating system for an EADS-Astrium SPARC ERC32-based MCM processor board and i386 standard PC mainboard have been developed.

    • Ghrelin, an endogenous ligand for the growth hormone secretagogue receptor, has two major functions: the stimulation of the growth hormone production and the stimulation of food intake. Accumulating evidence also suggests a role of ghrelin in cancer development. We conducted a case-control study on 1359 breast cancer cases and 2389 matched controls, nested within the European Prospective Investigation into Cancer and Nutrition, to examine the association of common genetic variants in the genes coding for ghrelin (GHRL) and its receptor (GHSR) with anthropometric measures, circulating insulin growth factor I (IGF-I) and insulin-like growth factor-binding protein 3 and breast cancer risk. Pair-wise tagging was used to select the 15 polymorphisms that represent the majority of common genetic variants across the GHRL and GHSR genes. A significant increase in breast cancer risk was observed in carriers of the GHRL rs171407-G allele (odds ratio: 1.2; 95% confidence interval: 1.0-1.4; P = 0.02). The GHRL single-nucleotide polymorphism rs375577 was associated with a 5% increase in IGF-I levels (P = 0.01). A number of GHRL and GHSR polymorphisms were associated with body mass index (BMI) and height (P between <0.01 and 0.04). The false-positive report probability (FPRP) approach suggests that these results are noteworthy (FPRP < 0.20). The results presented here add to a growing body of evidence that GHRL variations are associated with BMI. Furthermore, we have observed evidence for association of GHRL polymorphisms with circulating IGF-I levels and with breast cancer risk. These associations, however, might also be due to chance findings and further large studies are needed to confirm our results.

    • Objective To examine the association between anthropometry and endometrial cancer, particularly by menopausal status and exogenous hormone use subgroups. Methods Among 223,008 women in the European Prospective Investigation into Cancer and Nutrition (EPIC) study, there were 567 incident endometrial cancer cases during 6.4 years of follow-up. The analysis was performed with Cox proportional hazards modeling. Results Weight, body mass index (BMI), waist and hip circumferences and waist-hip ratio (WHR) were strongly associated with increased risk of endometrial cancer. The relative risk (RR) for obese (BMI 30- < 40 kg/m(2)) compared to normal weight (BMI < 25) women was 1.78, 95% CI = 1.41-2.26, and for morbidly obese women (BMI >= 40) was 3.02, 95% CI = 1.66-5.52. The RR for women with a waist circumference of >= 88 cm vs. < 80 cm was 1.76, 95% CI = 1.42-2.19. Adult weight gain of >= 20 kg compared with stable weight (+/- 3 kg) increased risk independent of body weight at age 20 (RR = 1.75, 95% CI = 1.11-2.77). These associations were generally stronger for postmenopausal than premenopausal women, and oral contraceptives never-users than ever-users, and much stronger among never-users of hormone replacement therapy compared to ever-users. Conclusion Obesity, abdominal adiposity, and adult weight gain were strongly associated with endometrial cancer risk. These associations were particularly evident among never-users of hormone replacement therapy.

    • This paper proposes an architecture for Personal Computers (PC) to avoid BIOS alteration and unauthorized access to resources. This proposal is based on results obtained from study of most popular PC platforms security mechanisms. Authentication controls which are established in PC platform in order to grant operating system booting or BIOS integrity of code mechanism incorporated to secureand avoid executing disallowed code are quite easy to break. The architecture described in the present work (Advanced Secure Architecture - ASA) increase the overall information and system security since prevents an unauthorized platform booting and it provides procedures for BIOS code authentication. On the other hand, ASA overcomes the users' authentication challenge in a corporative environment as well as it offers a very flexible way to specify the Personal Computers Corporation set that a user is allowed to access.

    • The present work describes how the concepts and foundations defined for multi-agent system technology can be applied in to a Wireless SensorNetwork (WSN), specifically it is focused on how multi-agent system technology's mechanisms and implementations could facilitate the development of systems based on WSN. In this respect, an architectural model where the above mentioned concepts, foundations and mechanisms come together is proposed, in order to define applications and services on WSN. Validation of the proposed architecture is made by means of its use in a perimeter security scenario ("tracking"). It is important to mention, that partial results of this work have been developed in the project PROPSI (Perimeter Protection by means of Wireless Sensor Networks).

    • A wireless sensor network (WSN) is a computer wireless network composed of spatially distributed and autonomous tiny nodes - smart dustsensors, motes -, which cooperatively monitor physical or environmental conditions. Nowadays these kinds of networks support a wide range of applications, such as target tracking, security, environmental control, habitat monitoring, source detection, source localization, vehicular and traffic monitoring, health monitoring, building and industrial monitoring, etc. Many of these applications have strong requirements for end-to-end delay and losses during data transmissions. In this work we have classified the main mechanisms that have been proposed to provide Quality of Service (QoS) in WSN at Medium Access Control (MAC) and network layers. Finally, taking into account some particularities of the studied MAC- andnetwork-layer protocols, we have selected a real application scenario in order to show how to choose an appropriate approach for guaranteeing performance in a WSN deployed application.

    • Nowadays WSNs support applications such as target tracking, environmental control or vehicles traffic monitoring. Generally, these applications have strong and strict requirements for end-to-end delaying and loosing during data transmissions. In this paper, we propose a practical scenario for application of the WSN field in order to illustrate selection of an appropriate approach for guaranteeing performance in a WSN-deployed application. The methodology we have used includes four major phases: 1) Requirements analysis of the application scenario; 2) QoS modelling in different layers of the communications protocol stack and selection of more suitable QoS protocols and mechanisms; 3) Definition of a simulation model based on an application scenario, to which we applied the protocols and mechanisms selected in phase 2; 4) Validation of decisions by means of simulation; and 5) analysis of results. This work has being partially developed in the framework of the CRISAL - M0700204174 project (partially funded by “Universidad Politécnica de Madrid” and “Comunidad de Madrid”, Spain).

    • Objective Most epidemiologic studies have suggested an increased risk of breast cancer with increasing alcohol intake. Using data from 274,688 women participating in the European Prospective Investigation into Cancer and Nutrition study (EPIC), we investigated the relation between alcohol intake and the risk of breast cancer. Methods Incidence rate ratios (IRRs) based on Cox proportional hazard models were calculated using reported intake of alcohol, recent (at baseline) and lifetime exposure. We adjusted for known risk factors and stratified according to study center as well as potentially modifying host factors. Results During 6.4 years of follow up, 4,285 invasive cases of breast cancer within the age group 35-75 years were identified. For all countries together the IRR per 10 g/day higher recent alcohol intake (continuous) was 1.03 (95% confidence interval (CI): 1.01-1.05). When adjusted, no association was seen between lifetime alcohol intake and risk of breast cancer. No difference in risk was shown between users and non-users of HRT, and there was no significant interaction between alcohol intake and BMI, HRT or dietary folate. Conclusion This large European study supports previous findings that recent alcohol intake increases the risk of breast cancer.
