  • Comparison of the efficacy of salmeterol/fluticasone propionate combination in Japanese and Caucasian asthmatics

    • INTRODUCTION: The effect of ethnicity on the efficacy of salmeterol (S)+fluticasone propionate (FP) has not been examined in Japanese and Caucasian asthmatics. In this study, the efficacy of combination treatment with S and FP from a single inhaler (SFC) was compared with concurrent treatment with S and FP administration from separate inhalers (S+FP) in Japanese and Caucasian asthmatics. METHODS: This was a randomised, double-blind, crossover study in male and female Japanese (n=18) and Caucasian (n=17) asthmatics (50-100% predicted FEV(1); >35% reversibility in sGaw). Subjects received SFC (S 50mcg/FP 250mcgb.i.d.) and S+FP (S 50mcgb.i.d.+FP 250mcgb.i.d.) for 14 days. sGaw and FEV(1) were determined 0-12h after the first and last doses. RESULTS: Treatment with both SFC and S+FP produced marked bronchodilation, which was maintained 0-12h after the first dose. Baseline sGaw and FEV(1) increased up to 51% and 180mL, respectively, in Japanese subjects over 2 weeks of treatment, with similar improvements in Caucasian subjects. On Day 14 the 0-12h S+FP:SFC treatment ratios (90% CI) for sGaw AUC and peak were 1.05 (0.98, 1.12) and 1.05 (0.97, 1.14), respectively, in Japanese subjects, and 0.99 (0.92, 1.07) and 0.98 (0.89, 1.07), respectively, in Caucasian subjects, with no difference between the two ethnic groups. CONCLUSIONS: The finding of a similar significant bronchodilator response in Japanese and Caucasian asthmatics following concurrent and combination treatment with salmeterol and FP suggests that the therapeutic response to these agents is comparable and independent of ethnicity in Japanese and Caucasian asthma patients.

  • Tumor-specificity and type of cell death induced by phenoxazines

    • Phenoxazines have shown diverse biological activities, but tumor-specific cytotoxic activity has not been investigated. A total of 24 phenoxazine derivatives (WM1-24) was investigated for their relative cytotoxicity against human tumor cell lines vs. normal cells. WM7 and WM8 showed the highest tumor-specificity index of 4.3 and 4.8, respectively. Considerable difference in drug-sensitivity was found among these tumor cell lines. Human promyelocytic leukemia HL-60 cells showed the highest sensitivity to both WM7 and WM8, followed by human oral squamous cell carcinoma (HSC-2, HSC-3, HSC-4), and human gingival fibroblast (HGF), pulp cell (HPC) and periodontal ligament fibroblast (HPLF) were the most resistant. WM7 and WM8 induced little or no internucleosomal DNA fragmentation, and activated caspase-3 in HSC-2, HSC-4 and human glioblastoma T98G cells. These compounds failed to induce autophagic cell death, as judged by acridine orange and microtubule-associated protein I light chain 3 (LC3)-GFP assays. These results suggested that the higher cytotoxicity of WM7 and WM8 are derived from the positively-charged quaternary nitrogen substituents on the phenoxazine ring and the electron density of nitrogen at N12, and that inhibition of autophagy is not always coupled with apoplosis induction.

    • The thermal component of Mercury's electron population has never been measured. One scientific objective of the Plasma Wave Investigation consortium, PWI, is to determine the influence of the thermal plasma upon the formation and dynamics of the planetary magnetosphere, as a function of solar activity. The Active Measurement of Mercury's Plasma experiment, AM(2)p, has been proposed as part of PWI, to monitor the density and temperature of the thermal electron population, during the whole mission of the Mercury Magnetospheric Orbiter of BepiColombo. These two physical parameters will be deduced from the measurements of the self- and mutual-impedances of the MEFISTO (Mercury Electric Field In Situ Tool) double-sphere antenna, in a frequency range comprising the expected plasma frequency. The in situ measurement of the antenna impedance is also essential for calibrating the electric antenna which measures the natural waves; it will allow, in particular, the effective length of the antenna to be calculated as a function of frequency and plasma conditions. The purpose of this paper is to define the scientific objectives of AM(2)p, to explain the principle of the measurement, to describe the electronic device, and to show the ability of AM 2p to make reliable and accurate measurements of the thermal plasma density and temperature in the Hermean magnetosphere, as well as in the solar wind at heliocentric distances of 0.31-0.47 AU. The potential performance of this instrument has been evaluated using both an analytical approach and numerical simulations.

  • Adaptive optics imaging and optical spectroscopy of a multiple merger in a luminous infrared galaxy

    • We present near-infrared (NIR) adaptive optics imaging obtained with VLT/NACO and optical spectroscopy from the Southern African Large Telescope of a luminous IR galaxy (LIRG) IRAS 19115-2124. These data are combined with archival Hubble Space Telescope imaging and Spitzer imaging and spectroscopy, allowing us to study this disturbed interacting/merging galaxy, dubbed the Bird, in extraordinary detail. In particular, the data reveal a triple system where the LIRG phenomenon is dominated by the smallest of the components. One nucleus is a regular barred spiral with significant rotation, while another is highly disturbed with a surface brightness distribution intermediate to that of disc and bulge systems, and hints of remaining arm/bar structure. We derive dynamical masses in the range 3-7 × 1010Msolarfor both. The third component appears to be a 1-2 × 1010Msolar mass irregular galaxy. The total system exhibits HII galaxy-like optical line ratios and strengths, and no evidence for active galactic nucleus (AGN) activity is found from optical or mid-IR data. The star formation rate is estimated to be ~190Msolaryr-1. We also report a search for supernovae from NIR images separated by five months and search for superstar cluster candidates. We detect outflowing gas from the Bird mostly in the range 100-300 km s-1 using NaI D absorption features. Overall, the Bird shows kinematic, dynamical and emission-line properties typical for cool ultraluminous IR galaxies (ULIRGs). However, the interesting features setting it apart for future studies are its triple merger nature, and the location of its star formation peak - the strongest star formation, as revealed by Spitzer imaging, does not come from the two major K-band nuclei, but from the third irregular component. This is in contrast to the conventional view that the (U)LIRG phases are powered by infalling gas to the major nuclei of the merging spiral galaxies. Aided by simulations, we discuss scenarios where the irregular component is on its first high-speed encounter with the more massive components. Based on observations made with ESO Telescopes at the Paranal Observatory under programme 073.D-0406A, and with the Southern African Large Telescope (SALT).
