    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

    • The Advanced GAmma Tracking Array (AGATA) is a European project to develop and operate the next generation γ-ray spectrometer. AGATA is based on the technique of γ-ray energy tracking in electrically segmented high-purity germanium crystals. This technique requires the accurate determination of the energy, time and position of every interaction as a γ ray deposits its energy within the detector volume. Reconstruction of the full interaction path results in a detector with very high efficiency and excellent spectral response. The realisation of γ-ray tracking and AGATA is a result of many technical advances. These include the development of encapsulated highly segmented germanium detectors assembled in a triple cluster detector cryostat, an electronics system with fast digital sampling and a data acquisition system to process the data at a high rate. The full characterisation of the crystals was measured and compared with detector- response simulations. This enabled pulse-shape analysis algorithms, to extract energy, time and position, to be employed. In addition, tracking algorithms for event reconstruction were developed. The first phase of AGATA is now complete and operational in its first physics campaign. In the future AGATA will be moved between laboratories in Europe and operated in a series of campaigns to take advantage of the different beams and facilities available to maximise its science output. The paper reviews all the achievements made in the AGATA project including all the necessary infrastructure to operate and support the spectrometer. © 2011 Elsevier B.V. All rights reserved.

    • The aim of this paper is to describe on-going work in the integration of humans in automated manufacturing systems. The intention is to achieve a flexible manufacturing system to meet the rapid developing and changing of today’s industry. The approach is based on a control concept with multi-agents. Humans, which are considered as a valuable factor in industrial production, are proposed as flexible agent resources for the automated manufacturing system.A test case was performed on a manufacturing system where three different groups of humans where integrated in the system; inspection, carrier and recovery. The P-SOP agent generator was used to automatically generate IEC 61131-3 PLC control code for the system.

  • Vehicle-to-vehicle channel models with large vehicle obstructions

    • Vehicle-to-Vehicle (V2V) communication is an en-abler for improved traffic safety and congestion control. As for any wireless system the ultimate performance limit is determined by the propagation channel. A particular point of interest is the shadowing effect of large vehicles such as trucks and buses, as this might affect the communication range significantly. In this paper we present measurement results and model the propagation channel in which a bus acts as a shadowing object between two passenger cars. The measurement setup is based on a WARP FPGA software radio as transmitter, and a Tektronix RSA5106A real-time complex spectrum analyzer as receiver. We analyze the influence of the bus location and car separation distance on the large-scale path loss, shadowing, and small-scale fading. The main effect of the bus is that it is acting as an obstruction creating an additional 15-20 dB attenuation. A Nakagami distribution is found to describe the statistics of the small-scale fading, by using Akaike's Information Criterion and the Kolmogorov-Smirnov test. The distance-dependency of the path loss is analyzed, and a stochastic model is developed to reflect the impact.

A Nakagami distribution is found to well describe the statistics of the small-scale fading, by using Akaike's Information Criterion and the Kolmogorov-Smirnov test. The distance dependence of the path loss is analyzed and a stochastic model is developed.

    • Laser-induced breakdown spectroscopy (LIBS) was used in this study to measure quantitatively the sodium (Na) and potassium (K) release from burning coal particles under oxy-fuel combustion environments. A specially designed laminar premixed burner was employed to provide a postflame environment with different O-2 and CO2 concentrations, in which the effects of O-2 and CO2 on the release of Na and K during coal oxy-fuel combustion were studied systematically. For the devolatilization stage, neither O-2 nor CO2 had significant influence on the Na and K release. The release of Na and K during the char stage, however, changed significantly at different O-2 and CO2 concentrations. Under these experimental conditions, when the O-2 concentration increased from 3.9% to 10.6%, the peak concentration of Na at the char stage increased from 15.2 mg/m(3) to 33.7 mg/m(3), and the maximum concentration of K increased from 6.2 mg/m(3) to 11.7 mg/m(3). When the CO2 concentration increased from 35.8% to 69.4%, the release of Na and K was inhibited during the char stage, with the peak concentration decreasing from 8.9 mg/m(3) to 6.9 mg/m(3) for Na and from 3.7 mg/m(3) to 2.4 mg/m(3) for K. During the ash stage, the release of Na and K decreased with the O-2 concentration, whereas it increased with the CO2 concentration.

    • A new benzodithiophene (BDT)-based polymer, poly(4,8-bis(2-ethylhexyloxy)benzo[1,2-b:4,5-b]dithiophene vinylene) (PBDTV), was synthesized by Pd-catalyzed Stille-coupling method. The polymer is soluble in common organic solvents and possesses high thermal stability. PBDTV film shows a broad absorption band covering from 350 nm to 618 nm, strong photoluminescence peaked at 545 nm and high hole mobility of 4.84 x 10(-3) cm(2)/Vs. Photovoltaic properties of PBDTV were studied by fabricating the polymer solar cells based on PBDTV as donor and PC70BM as acceptor. With the weight ratio of PBDTV: PC70BM of 1:4 and the active layer thickness of 65 nm, the power conversion efficiency of the device reached 2.63% with V-oc = 0.71 V, I-sc = 6.46 mA/cm(2), and FF = 0.57 under the illumination of AM1.5, 100 mW/cm(2).

    • Genome function is dynamically regulated in part by chromatin, which consists of the histones, non-histone proteins and RNA molecules that package DNA. Studies in Caenorhabditis elegans and Drosophila melanogaster have contributed substantially to our understanding of molecular mechanisms of genome function in humans, and have revealed conservation of chromatin components and mechanisms(1-3). Nevertheless, the three organisms have markedly different genome sizes, chromosome architecture and gene organization. On human and fly chromosomes, for example, pericentric heterochromatin flanks single centromeres, whereas worm chromosomes have dispersed heterochromatin-like regions enriched in the distal chromosomal 'arms', and centromeres distributed along their lengths(4,5). To systematically investigate chromatin organization and associated gene regulation across species, we generated and analysed a large collection of genome-wide chromatin data sets from cell lines and developmental stages in worm, fly and human. Here we present over 800 new data sets from our ENCODE and modENCODE consortia, bringing the total to over 1,400. Comparison of combinatorial patterns of histone modifications, nuclear lamina-associated domains, organization of large-scale topological domains, chromatin environment at promoters and enhancers, nucleosome positioning, and DNA replication patterns reveals many conserved features of chromatin organization among the three organisms. We also find notable differences in the composition and locations of repressive chromatin. These data sets and analyses provide a rich resource for comparative and species-specific investigations of chromatin composition, organization and function.

    • Two novel molecules TABzPA and TATpPA with D-pi-A structure and large pi-conjugation have been synthesized via Wittig reaction. Unlike common molecules, which exhibit the aggregation-caused quenching (ACQ) phenomenon, TABzPA and TATpPA exhibit aggregation-induced emission enhancement (AIEE) activity: weak luminescence in common solvents but strong emission when aggregated as nanoparticles and solid powders. Due to their intramolecular charge transfer (ICT) attribution and AIEE features, TABzPA and TATpPA display bathochromic effects. Combining ICT and AIEE features, these molecules are intensely yellow solid emitters with high quantum efficiencies of about 23.2% and 24.1%. Moreover, TABzPA and TATpPA have excellent two-photon absorption (2PA) properties owing to good planarity and large pi-conjugation. The values of 2PA cross sections at 800 nm are 7590 GM and 7648 GM. The excellent optical properties of TABzPA and TATpPA pave the way for future potential applications in biophotonics and optoelectronics.

    • BACKGROUND: The FTO gene harbors the strongest known susceptibility locus for obesity. While many individual studies have suggested that physical activity (PA) may attenuate the effect of FTO on obesity risk, other studies have not been able to confirm this interaction. To confirm or refute unambiguously whether PA attenuates the association of FTO with obesity risk, we meta-analyzed data from 45 studies of adults (n=218,166) and nine studies of children and adolescents (n=19,268). METHODS AND FINDINGS: All studies identified to have data on the FTO rs9939609 variant (or any proxy [r(2)>0.8]) and PA were invited to participate, regardless of ethnicity or age of the participants. PA was standardized by categorizing it into a dichotomous variable (physically inactive versus active) in each study. Overall, 25% of adults and 13% of children were categorized as inactive. Interaction analyses were performed within each study by including the FTO×PA interaction term in an additive model, adjusting for age and sex. Subsequently, random effects meta-analysis was used to pool the interaction terms. In adults, the minor (A-) allele of rs9939609 increased the odds of obesity by 1.23-fold/allele (95% CI 1.20-1.26), but PA attenuated this effect (p(interaction) =0.001). More specifically, the minor allele of rs9939609 increased the odds of obesity less in the physically active group (odds ratio =1.22/allele, 95% CI 1.19-1.25) than in the inactive group (odds ratio =1.30/allele, 95% CI 1.24-1.36). No such interaction was found in children and adolescents. CONCLUSIONS: The association of the FTO risk allele with the odds of obesity is attenuated by 27% in physically active adults, highlighting the importance of PA in particular in those genetically predisposed to obesity.

    • An experimental study of methane + air flames doped with ammonia (4370 ppm of the fuel) has been performed. The goal of this work was to analyze formation of NOx from fuel-N under well-controlled conditions. The Heat Flux method was used for stabilization of non-stretched adiabatic flames on a perforated plate burner at atmospheric pressure. Laser-saturated fluorescence (LSF) and probe sampling were adopted to measure NO concentrations in the post-flame zone. LSF experiments include two series of measurements: in methane + air flames doped with NO and then in flames doped with NH3. In the lean flames seeded with NO, LSF measurements clearly deviates from the model predictions at higher concentrations of NO seeded, that strongly corroborates existence of the lean NO reburning. The modeling accurately predicts [NO] in the neat flame and shows no consumption of NO up to 170 ppm seeded. In (CH4 + NH3) + air mixtures the NO concentrations measured by LSF are in good agreement with the probe sampling results in the whole range of equivalence ratios 0.65

    • A series of new triarylamine-based platinum-acetylide complexes (WYs) have been designed and synthesized as new sensitizers for applications in dye-sensitized solar cells (DSSCs). With the aim of investigating the effect of a rigidifying donor structure on the photoelectrical parameters of the corresponding DSSCs, two new sensitizers, WY1 and WY2, with rigid and coplanar fluorene units as an electron donor, were prepared. Moreover, two sensitizers that contained triphenylamine units as an electron donor, WY3 and WY4, were also synthesized for comparison. The photo- and electrochemical properties of all of these new complexes have been extensively explored. We found that the dimethyl-fluorene unit exhibited a stronger electron-donating ability and better photovoltaic performance compared to the triphenylamine unit, owing to its rigidifying structure, which restricted the rotation of sigma bonds, thus increasing the conjugation efficiency. Furthermore, WY2, which contained a dimethyl-fluorene unit as an electron donor and bithiophene as a bridge, showed a relatively high open-circuit voltage (V-oc) of 640mV and a PCE of 4.09%. This work has not only expanded the choice of platinum-acetylide sensitizers, but also demonstrates the advantages of restricted rotation of donor sigma bonds for improved behavior of the corresponding DSSCs.

  • Population Genomics Reveal Recent Speciation and Rapid Evolutionary Adaptation in Polar Bears

    • Polar bears are uniquely adapted to life in the High Arctic and have undergone drastic physiological changes in response to Arctic climates and a hyper-lipid diet of primarily marine mammal prey. We analyzed 89 complete genomes of polar bear and brown bear using population genomic modeling and show that the species diverged only 479-343 thousand years BP. We find that genes on the polar bear lineage have been under stronger positive selection than in brown bears; nine of the top 16 genes under strong positive selection are associated with cardiomyopathy and vascular disease, implying important reorganization of the cardiovascular system. One of the genes showing the strongest evidence of selection, APOB, encodes the primary lipoprotein component of low-density lipoprotein (LDL); functional mutations in APOB may explain how polar bears are able to cope with life-long elevated LDL levels that are associated with high risk of heart disease in humans.

    • Comfort cooling demand continues to increase throughout the world Conventional cooling production results in high demand for electrical power during peak hours, leading to high emissions for producing cooling, and potential power shortages in electric grids With a cold storage, the peak power demand is effectively managed and enables free-cooling. This paper examines one concept using phase change materials (PCM) for storing of cold, where the cold carrier (water) is in direct contact with the PCM. This is in order to enable high power for charging and discharging while providing a high storage capacity. A theoretical model highlights important design parameters for reaching large storage and power capacity The capacity increases with the Packing Factor and temperature difference across the storage. For high power, the flow rate, temperature difference, and drop size is important parameters which is also verified in an experimental evaluation. The obtainable power is between 30 and 80 kW/m(3) storage Practical limitations of this concept are shown to be PCM-water bed expansion and non-uniform channeling due to asymmetric and unstable PCM shells.

    • A total of 155 rice plants were collected from ten mining areas in three provinces of China (Hunan, Guizhou and Guangdong), where most of mercury (Hg) mining takes place in China. During the harvest season, whole rice plants were sampled and divided into root, stalk & leaf, husk and seed (brown rice), together with soil from root zone. Although the degree of Hg contamination varied significantly among different mining areas, rice seed showed the highest ability for methylmercury (MeHg) accumulation. Both concentrations of total mercury (THg) and MeHg in rice plants were significantly correlated with Hg levels in soil, indicating soil is still an important source for both inorganic mercury (IHg) and MeHg in rice plants. The obvious discrepancy between the distribution patterns of THg and MeHg reflected different pathways of IHg and MeHg accumulation. Water soluble Hg may play more important role in MeHg accumulation in rice plants.

    • AOD slag samples from steel works of Outokumpu Stainless Company were used to study F-dissolution relating to treatments for volume stabilization. Results from the slag tests and sample characterizations indicate that the slag re-melting with or without reduction and granulation with either water or gas have rather small effects on F leaching, as well as formation of different C2S polymorphs. The chemical composition and cooling condition are the two important parameters to control F leaching from slag samples. These two parameters should be combined together in an optimum way by the steel industry to treat slags for F-immobilization.
