  • Tumour necrosis factor inhibitors in Crohn's disease and the effect on surgery rates

    • Aim: Surgery is an important therapeutic option for Crohn's disease. The need for first bowel surgery seems to have decreased with the introduction of tumour necrosis factor inhibitors (TNFi; adalimumab or infliximab). However, the impact of TNFi on the need for intestinal surgery in Crohn's disease patients irrespective of prior bowel resection is not known. The aim of this work is to compare the incidence of bowel surgery in Crohn's disease patients who remain on TNFi treatment versus those who discontinue it. Method: We performed a nationwide register-based observational cohort study in Sweden of all incident and prevalent cases of Crohn's disease who started first-line TNFi treatment between 2006 and 2017. Patients were categorized according to TNFi treatment retention less than or beyond 1 year. The study cohort was evaluated with regard to incidence of bowel surgery from 12 months after the first ever TNFi dispensation. Results: We identified 5003 Crohn's disease patients with TNFi exposure: 3748 surgery naïve and 1255 with bowel surgery prior to TNFi initiation. Of these patients, 7% (n = 353) were subjected to abdominal surgery during the first 12 months after the start of TNFi and were subsequently excluded from the main analysis. A majority (62%) continued TNFi for 12 months or more. Treatment with TNFi for less than 12 months was associated with a significantly higher surgery rate compared with patients who continued on TNFi for 12 months or more (hazard ratio 1.26, 95% CI 1.09–1.46; p = 0.002). Conclusion: Treatment with TNFi for less than 12 months was associated with a higher risk of bowel surgery in Crohn's disease patients compared with those who continued TNFi for 12 months or more.

  • A novel infrared imager for studies of hydroxyl and oxygen nightglow emissions in the mesopause above northern Scandinavia

    • The paper describes technical characteristics and presents the first scientific results of a novel infrared imaging system (imager) for studies of nightglow emissions coming from the hydroxyl (OH) and molecular oxygen (O2) layers in the mesopause region (80–100 km) above northern Scandinavia. The OH imager was put into operation in November 2022 at the Swedish Institute of Space Physics in Kiruna (67.86° N, 20.42° E; 400 m altitude). The OH imager records selected emission lines in the OH(3-1) band near 1500 nm to obtain intensity and temperature maps at around 87 km altitude. In addition, the OH imager registers infrared emissions coming from the O2 IR A-band airglow at 1268.7 nm in order to obtain O2 intensity maps at a slightly higher altitude, around 94 km. This technique allows the tracing of wave disturbances in both horizontal and vertical domains in the mesopause region. Validation and comparison of the OH(3-1) rotational temperature with collocated lidar and Aura Microwave Limb Sounder (MLS) satellite temperatures are performed. The first scientific results obtained from the OH imager for the first winter season (2022–2023) are discussed.

    • On the night 16–17 August 2021, a balloon-borne experiment called Stratospheric Observations of Noctilucent Clouds (SONC) was successfully performed. A big scientific balloon, having onboard three automated cameras for studies of noctilucent clouds (NLC), was launched to 32.7 km altitude from Esrange (northern Sweden). All three NLC cameras and electronics were completely operational in the stratosphere for more than 10 h at low temperatures of about −30 °C. Two wide angle cameras registered an extended NLC field of about 1700 km long in the twilight sky sector from the north-west to the north-east of Esrange. NLC were of a moderate brightness and were located at high latitudes between 68° and 71°N. The NLC field was located in a cold area (138–142 K) below the frost point temperature (145–148 K) in the mesopause region that was confirmed by Aura/MLS satellite and Esrange lidar measurements. The balloon-borne NLC measurements were accompanied by ground-based lidar and radar measurements. The latter have registered Polar Mesosphere Summer Echoes (PMSE) in the same volume of the summer mesopause along with NLC observed from the stratosphere that has been performed for the first time above northern Scandinavia. We describe the technique and method of the NLC observation from the stratosphere as well as present the first scientific results of the SONC experiment.

    • Based on common volume rocket-borne measurements of temperature, densities of atomic oxygen and neutral air, we derived O(D-1) nighttime concentrations and corresponding Atmospheric band emission (762 nm). This is one of the first retrievals of the nighttime O(D-1) concentration. Recently, Kalogerakis, Sharma and co-workers have suggested a new production path of O(D-1) based on the reaction of vibrationally excited OH and O. We calculate Atmospheric band volume emission related to the population of O-2(b(1)Sigma(+)(g)) from O(D-1) and compare with total Atmospheric band emissions observed during the same rocket launch. This allows an estimation of the relative contribution of the new Kalogerakis-Sharma mechanism (KSM) to the total Atmospheric band emission. The concentration of O(D-1) due to KSM amounts to several tens cm(-3) with a peak around 95 km. The KSM gives an essential contribution to the total Atmospheric band volume emission (762 nm). Additionally, we illustrate analytically that the expressions for volume emission by the new KSM and the traditional two-step mechanism have similar functional dependences on the atmospheric concentrations of O and O-2. This causes an ambiguity, when interpreting Atmospheric band observations in terms of the one mechanism or the other.

  • Preclinical protein signatures in blood predict Crohn's disease and Ulcerative colitis several years before the diagnosis

    • Background: We aimed to identify protein signatures predictive of a future diagnosis of inflammatory bowel disease (IBD).Methods: We conducted a case-control study, nested within large population-based cohorts with biorepositories. Samples were obtained from individuals who later in life were diagnosed with IBD (preclinical cases) and compared with age and sex-matched individuals who remained free from IBD during follow-up (controls). Using proximity extension assays (Olink, Uppsala), we measured 176 proteins. We applied regularized logistic regression to identify protein signatures of preclinical disease in serum from the discovery cohort (n=312). Their performance was validated in an external preclinical cohort (n=222). The biological relevance of identified proteins was further assessed in an inception cohort (n=144). Finally, we used an IBD twin cohort (n=327) to examine the impact of genetic and shared environmental factors on identified proteins.Results: We identified 34 proteins associated with preclinical Crohn’s disease (CD) in the discovery cohort (Pfalse discovery rate <0.10), with 9 confirmed in the validation cohort (Pfalse discovery rate <0.05). For preclinical ulcerative colitis (UC), 45 proteins were identified and 12 validated (Fig. 1A-B). In the discovery cohort, a signature of 29 proteins differentiated preclinical CD cases from controls with an AUC of 0.85 (Fig. 1G). Its performance was confirmed when applied to the preclinical validation cohort (AUC=0.84, Fig. 1H). Moreover, the signature had excellent capacity to differentiate newly diagnosed CD from healthy controls in the inception cohort (AUC = 0.99, Fig. 1I). The preclinical UC signature had a significant, but albeit lower, predictive capacity in the discovery (AUC=0.77), validation (AUC=0.67) and inception cohort (AUC=0.90, Fig. 1G-I).15 of 17 proteins associated with preclinical IBD demonstrated significantly higher intra-pair correlation coefficients in healthy monozygotic- compared to dizygotic twin pairs, indicating an influence from genetic factors on the regulation of these protein markers. The preclinical signature for CD demonstrated an AUC of 0.87 when comparing twins with preclinical CD (n=10) to matched external healthy twins. However, its predictive capacity was lower when comparing preclinical CD twins with their healthy twin siblings (AUC=0.58), i.e., when accounting for genetic and shared environmental factors. The difference in AUC estimates in the twin cohort was not significant (P=0.07).

    • Global three-dimensional data are a key to understanding gravity waves in the mesosphere and lower thermosphere. MATS (Mesospheric Airglow/Aerosol Tomography and Spectroscopy) is a new Swedish satellite mission that addresses this need. It applies space-borne limb imaging in combination with tomographic and spectroscopic analysis to obtain gravity wave data on relevant spatial scales. Primary measurement targets are O-2 atmospheric band dayglow and nightglow in the near infrared, and sunlight scattered from noctilucent clouds in the ultraviolet. While tomography provides horizontally and vertically resolved data, spectroscopy allows analysis in terms of mesospheric temperature, composition, and cloud properties. Based on these dynamical tracers, MATS will produce a climatology on wave spectra during a 2-year mission. Major scientific objectives include a characterization of gravity waves and their interaction with larger-scale waves and mean flow in the mesosphere and lower thermosphere, as well as their relationship to dynamical conditions in the lower and upper atmosphere. MATS is currently being prepared to be ready for a launch in 2020. This paper provides an overview of scientific goals, measurement concepts, instruments, and analysis ideas.

  • Metapopulation dynamics over 25 years of a beetle, Osmoderma eremita, inhabiting hollow oaks

    • Osmoderma eremita is a species of beetle that inhabits hollows in ancient trees, which is a habitat that has decreased significantly during the last century. In southeastern Sweden, we studied the metapopulation dynamics of this beetle over a 25 year period, using capture-mark-recapture. The metapopulation size had been rather stable over time, but in most of the individual trees there had been a positive or negative trend in population development. The probability of colonisation was higher in well-connected trees with characteristics reflecting earlier successional stages, and the probability of extinction higher in trees with larger diameter (i.e. in later successional stages), which is expected from a habitat-tracking metapopulation. The annual tree mortality and fall rates (1.1% and 0.4%, respectively) are lower than the colonisation and extinction rates (5–7%), indicating that some of the metapopulation dynamics are due to the habitat dynamics, but many colonisations and extinctions take place for other reasons, such as stochastic events in small populations. The studied metapopulation occurs in an area with a high density of hollow oaks and where the oak pastures are still managed by grazing. In stands with fewer than ten suitable trees, the long-term extinction risk may be considerable, since only a small proportion of all hollow trees harbours large populations, and the population size in trees may change considerably during a decade.

    • Background: Improved mucosal immune profiling in active and quiescent colonic inflammatory bowel disease (IBD) is needed to develop therapeutic options for treating and preventing flares. This study therefore aimed to provide a comprehensive mucosal characterization with emphasis on immunological host response of patients with active ulcerative colitis (UC active), UC during remission (UC remission) and active colonic Crohn's disease (CD active).Methods: Colonic biopsies from 47 study subjects were collected for gene expression and pathway analyses using the NanoString host-response panel, including 776 genes and 56 immune-related pathways.Results: The majority of mucosal gene expression and signaling pathway scores were increased in active IBD (n=27) compared to healthy subjects (n=10). However, both active IBD and UC remission (n=10) demonstrated decreased gene expression and signaling pathway scores related to autophagy, alpha kinase-1 and IL-17 signaling pathways compared to healthy subjects. Further, UC remission was characterized by decreased scores of several signaling pathways linked to homeostasis along with increased mononuclear cell migration pathway score as compared to healthy subjects. No major differences in the colonic mucosal gene expression between CD active (n=7) and UC (n=20) active were observed.Conclusion: This study indicates that autophagy, alpha kinase-1 and IL-17 signaling pathways are persistently downregulated in UC irrespective of disease activity. Further, UC patients in remission present a unique mucosal environment, potentially preventing patients from reaching and sustaining true homeostasis. These findings may enable better comprehension of the remitting and relapsing pattern of colonic IBD and guide future treatment and prevention of flares.

  • Adsorption and Mass Transport in Zeolite Membranes

    • Zeolites are commonly used as adsorbents and catalysts in the industry due to their well-defined pores of molecular dimensions. Zeolites offer porous structure, which consists of interlinked alumina and silica tetrahedra with shared oxygen atoms. Zeolites can also be prepared as intergrown films on porous supports, which results in zeolite membranes. CHA and MFI are two promising zeolites that can be used as membranes for biogas and syngas separation and upgrading since their pore size is suitable.Membrane technology is considered an energy-lean gas separation method that offers a straightforward process with compact equipment and high efficiency. Compared with polymeric membranes, zeolite membranes offer higher permeance and stability due to their porous structure and ceramic nature. Since zeolite membranes are expensive and a higher flux would reduce the needed membrane area, thin membranes with high flux are of great interest. However, to enable the design of zeolite membrane processes, it is vital to enhance the fundamental understanding of the mass transport in the materials.In this study, zeolite membranes of different types, i.e., CHA and MFI, were evaluated for separation of various gas mixtures. MFI disc membranes were evaluated for the separation of equimolar CO2/H2 mixtures under both dry and humid conditions, as well as for the separation of ternary CH4/N2/He mixtures. High selectivity and high CO2 fluxes were observed during CO2/H2 separation under both dry and humid conditions. The MFI disc membrane also displayed a high performance for separation of ternary CH4/N2/He mixtures. The results indicated that MFI membranes are promising candidates for separation of CO2 from the gas mixtures and for helium recovery from natural gas. Tubular CHA membranes, with lengths of 10 and 50 cm, were also investigated for CO2/CH4 separation under industrially relevant conditions. A maximum CO2/CH4 separation selectivity of 198 combined with a CO2 permeance of 14×10-7 mol/(m2·s·Pa) was observed for humid gas. The results verified the feasibility of these membranes for industrial gas separations.After verifying the high performance of CHA zeolite membranes for gas separation under industrial conditions, CHA zeolite crystals with various Si/Al ratios were synthesized and the adsorption of CO2 and CH4 in the materials were studied. Subsequently, the mass transport through ultra-thin MFI and CHA zeolite disc membranes was measured and a model accounting for the adsorption and diffusion through the surface barriers and in the pores was developed. The model was successfully fitted to both single component and mixture permeation data. The fitted model indicates that the mass transport through ultra-thin membranes is controlled by the surface barriers. It revealed that the surface barrier is a surface diffusion process at the pore mouth with an activation energy that is higher than for the surface diffusion in the pores. Furthermore, the fitted model indicated that the high selectivity of CHA membranes is mostly due to a highly selective surface barrier and, to a lesser extent, is a result of adsorption selectivity.In the last part of the work, a process for upgrading biogas was designed by using the developed model. The process was compared with a process based on hollow fiber polymeric membranes. It was concluded that the zeolite membrane processes were much more compact and had a much lower demand for electricity than the polymeric membrane process.

  • Observational study of tofacitinib in ulcerative colitis in Sweden (ODEN) - Interim analysis of clinical and biomarker data

    • Background: Tofacitinib is a Janus kinase (JAK) inhibitor for the treatment of moderate to severe ulcerative colitis (UC). ODEN is an ongoing Swedish multicentre prospective observational study regarding effectiveness of tofacitinib in UC. In this interim analysis, we aimed to assess the clinical outcomes during the first 16 weeks.Methods: Patients with active UC were enrolled 2020-2023 when starting tofacitinib as per clinical indication. Inclusion criteria were fecal (F) calprotectin >250 mg/kg or Mayo endoscopic score ≥2. Data were collected using an electronic case report form linked to the Swedish National Quality Registry for Inflammatory Bowel Disease (SWIBREG). Data concerning inflammatory markers, endoscopic activity, partial (p) Mayo, extra intestinal manifestations, health-related quality of life measures, corticosteroid use, and colectomy rates were collected regardless of tofacitinib discontinuation. Information collected at week 8 and 16 is presented here. Intention-to-treat (ITT) analysis was applied and tofacitinib discontinuation was considered as treatment failure (i.e., no tofacitinib-induced clinical or laboratory response or remission). McNemar’s test was used for proportion differences.Results: The proportion of patients who previously had failed at least one biologic was 95% and at least two biologics, 62%. At inclusion, median p-Mayo was 5 and 39% of patients were on corticosteroids (Table 1a). Patients’ survival on drug is shown in Figure 1a. At week 8 and 16, 42% and 43%, respectively, achieved corticosteroid free clinical remission, Figure 1b. A 50% reduction in F-calprotectin was seen in 54% and 49% at week 8 and 16, respectively. The endpoint of Mayo endoscopic score 0 and/or F-calprotectin <100 mg/kg was achieved by 30% and 38% at week 8 and 16, respectively. Arthralgia frequency decreased significantly from baseline from 29% at inclusion to 13% and 11% at week 8 and 16 respectively. Three patients underwent colectomy the first 16 weeks (Table 1b).Conclusion: After 16 weeks of treatment with tofacitinib, a substantial proportion of previously treatment refractory UC patients were in clinical and endoscopic corticosteroid-free remission, and a distinct improvement in F-calprotectin levels was observed. In addition, a significant reduction in arthralgia was noted.

    • Background: Ulcerative colitis (UC) has a major impact on daily life. The Janus Kinas (JAK) inhibitor tofacitinib is effective in achieving remission in UC, but prospective real-world evidence concerning the effect on health-related quality of life (HRQoL) and fatigue are still scarce. Fatigue is a component of UC that is notoriously difficult to treat and not unambiguously related to inflammatory activity. ODEN is an ongoing Swedish multicentre prospective observational study of tofacitinib in UC. In this interim analysis, we assessed the effectiveness on HRQoL and fatigue during the first 16 weeks.Methods: Patients with UC and active inflammation were enrolled 2020-2023 when starting tofacitinib as per clinical indication. To measure various aspects of impairment of daily life, the validated questionnaires Short Health Scale (SHS), EQ-5D-5L [Swedish value set], and IBD-fatigue scale (IBD-F) were used. These data and information concerning clinical, biochemical, and endoscopic outcomes were collected in an e-CRF linked to the Swedish National Quality Registry for Inflammatory Bowel Disease (SWIBREG). For HRQoL outcomes, per protocol analysis was applied. Paired t-test and Wilcoxon’s signed-rank test were used for mean and median differences, respectively.Results: In total, 103 patients were included. Baseline data are shown in Table 1a. For patients still on tofacitinib treatment, all four dimensions of the SHS (symptoms, social function, disease related worry, and general well-being) improved significantly, Table 1b. A median decrease of one point from baseline was seen at week 8 in each of the parameters, which was maintained through week 16 with a tendency towards further improvement. EQ-5D-5L showed an impairment mainly in the aspects of pain/discomfort and ability to participate in common daily activities. Improvement in these dimensions was seen from baseline to week 16. The overall EQ-5D-5L index improved significantly from baseline (0.80) to week 8 (0.86) and week 16 (0.89), as did the EQ VAS 0-100 reflecting overall health (58, 71, and 74, respectively). A significant improvement in IBD-F part 1 and 2 was seen at week 8 and 16, Figure 1.Conclusion: This study demonstrates that tofacitinib treatment covariates with positive changes in a variety of measures of patients’ quality of life, including improvements in self-assessed overall wellbeing. Finally, fatigue significantly improved during tofacitinib treatment. Thus, tofacitinib treatment shows association with meaningful improvements in multiple aspects of quality of life during the first 16 weeks of treatment.

  • Direct ink writing of high-resolution cellulose structures

    • 3D printing is envisioned to play an important role in the production of membranes for e.g., water purification and bio-separation applications due to the prospect of creating new and cleverly designed structures. Among different 3D printing techniques, direct ink writing offers the opportunity to print a wide variety of materials with high-detail resolution. There is a range of parameters that need to be optimized in order to develop robust printing techniques at that scale. In this study, cellulose acetate (CA), which is a biocompatible material, has been used as an ink. In order to examine the printability and the possibility of printing features as small as a few mu m, nozzles with different diameters and inks with varying amounts and molecular weights of CA were investigated. Findings in this study indicate that, depending on the wetting on the underlaying structure, the nozzle's internal and external diameter affects the detail resolution of the printed structure. Different inks result in different widths of printed strands and generally a higher amount and higher molecular weights of CA results in higher detail resolution. However, too high amount of CA and molecular weight will increase the clogging risk in the nozzle. In this study, the internal size of the nozzle was 3 mu m, and by selecting a suitable ink, it was possible to print strands down to 1 mu m size and 6 mu m inter-strand distance in the air, bridging supports with limited sagging. Furthermore, wall structures consisting of 300 layers, corresponding to about 300 mu m in total height, were successfully printed.

    • Improved biomarkers are needed for pediatric inflammatory bowel disease. Here we identify a diagnostic lipidomic signature for pediatric inflammatory bowel disease by analyzing blood samples from a discovery cohort of incident treatment-naïve pediatric patients and validating findings in an independent inception cohort. The lipidomic signature comprising of only lactosyl ceramide (d18:1/16:0) and phosphatidylcholine (18:0p/22:6) improves the diagnostic prediction compared with high-sensitivity C-reactive protein. Adding high-sensitivity C-reactive protein to the signature does not improve its performance. In patients providing a stool sample, the diagnostic performance of the lipidomic signature and fecal calprotectin, a marker of gastrointestinal inflammation, does not substantially differ. Upon investigation in a third pediatric cohort, the findings of increased lactosyl ceramide (d18:1/16:0) and decreased phosphatidylcholine (18:0p/22:6) absolute concentrations are confirmed. Translation of the lipidomic signature into a scalable diagnostic blood test for pediatric inflammatory bowel disease has the potential to support clinical decision making.

    • Background: Better prognostic measures for ulcerative colitis (UC) could significantly advance patient care. While the prognostic capacity of circulating proteins in UC has been explored, the role of mucosal proteins remains largely unknown. We examined mucosal protein markers in patients with incident ulcerative colitis and evaluated their prognostic value.Methods: Biopsies from macroscopically inflamed colonic/rectal mucosa of adult patients in the Swedish inception cohort of IBD (SIC IBD) were obtained at diagnosis of UC. Patients were followed prospectively, and clinical data were recorded after 3 and 12 months. Disease course was categorised as indolent or aggressive at 12 months, based on a composite outcome of colectomy, hospital admission for active disease, treatment refractoriness towards ≥2 biological agents; the use of >2 courses of corticosteroids, or a cumulative dose of >2.5 g. Relative estimates of 162 protein markers were assessed in homogenised tissue supernatants, using proximity extension assay technology (Olink Proteomics, Uppsala, Inflammation and Oncology II panel). Mann-Whitney U test, with Benjamini-Hochberg correction was used to identify differentially regulated mucosal proteins in aggressive vs indolent disease course, with a 5% false discovery rate (FDR). Smoothly clipped absolute deviation regularised logistic regression models were used to identify prognostic signatures distinguishing aggressive from indolent disease course. Performance was estimated in a leave-one-out cross-validation and reported as the area under the receiver operating characteristic (ROC) curve (AUC).Results: 117 patients provided a macroscopically inflamed colonic/rectal biopsy at diagnosis of UC. Basic demographics and clinical characteristics are presented in Table 1. Relative protein levels of WFdc2 and CCL20 were significantly lower in lysates from patients developing an aggressive course vs patients developing an indolent course, while estimates of MMP1, CCL11, WISP-1, OPG, RSPO3 and VEGFR2 were higher (Figure 1A). Regularized logistic regression identified signatures restricted to 28 proteins, distinguishing aggressive from indolent UC courses, yielding an AUC of 0.68 (95% confidence interval (CI): 0.56-0.80) for left-out samples (Figure 1B). Incorporating extent of inflammation at diagnosis in the model improved the AUC to 0.71 (95% CI: 0.60-0.83).Conclusion: We identified prognostic mucosal protein signatures associated with future course of ulcerative colitis by analysing inflamed mucosal biopsies that were obtained at diagnosis. These protein markers may highlight pathways of relevance for ulcerative colitis outcomes.

    • Background: Mental health (MH) has been reported to be poorer among patients with inflammatory bowel disease (IBD) than general population. However, it is not known whether MH is more driven by inflammation itself or related to gastrointestinal (GI) symptoms. Also, the dynamics of MH following IBD diagnosis is not well understood.Methods: In the Swedish Inception Cohort of IBD (SIC-IBD), patients with Crohn’s disease (CD), ulcerative colitis (UC) and unclassified IBD (IBD-U) as well as symptomatic controls (SC) and healthy controls (HC) filled in Patient Health Questionnaire-9 (PHQ-9), a validated screening tool for depression. Patients completed PHQ-9 at diagnosis and at one year follow-up while the controls completed it once. Disease outcome was defined at one year based on requirement of advanced treatments/ surgery.Results: In total, 286 individuals (16 HC, 89 SC, 62 CD, 104 UC, 15 IBD-U) completed the questionnaire at baseline. HC had significantly lower PHQ-9 score, (fewer depressive symptoms), at baseline compared to all the other groups (p<0.01). The baseline PHQ-9 score was not significantly different between SC and CD, UC and IBD-U patients (p=0.06).At one year follow-up, 38 CD and 53 UC patients completed the PHQ-9. Between baseline and follow-up, UC patients had a significant drop in their PHQ-9 score (p<0.0000001), whereas CD patients did not have any significant change in their PHQ-9 score (p=0.06). Furthermore, UC patients had significantly lower PHQ-9 score compared with CD patients at follow-up (p=0.04, Figure 1).Baseline PHQ-9 score was not correlated with calprotectin at baseline neither in UC nor CD patients (p=0.7 and 0.5 respectively). Also, there was no positive correlation between PHQ-9 score change and calprotectin change in either UC or CD patients, and neither baseline nor follow-up PHQ-9 scores were significantly different in patients with poor or good outcome (p>0.05). When analysed separately by sex, there was still no correlation between baseline PHQ-9 score and outcome in neither CD nor UC patients (p>0.05)Conclusion: IBD patients have at the time of diagnosis more depressive symptoms than HC, but not different from SC. Depressive symptoms are more related to GI symptoms than IBD specific inflammation or disease outcome. UC patients show more of an improvement in their depressive symptoms one year after diagnosis than CD patients.

    • A first sounding rocket campaign dedicated to investigate the creation mechanism of Polar Mesosphere Winter Echoes (PMWE) was conducted in April 2018 from the north Norwegian Andøya Space Center (69 °N, 16 °E). Two instrumented sounding rockets were launched on 13th and 18th of April under PMWE and non-PMWE conditions, respectively. In this paper we give an overview of the PMWE sounding rocket mission. We describe and discuss some results of combined in situ and ground-based measurements which allow to verify existing PMWE theories. Our measurements ultimately show that: a) polar winter mesosphere is abounded with meteor smoke particles (MSP) and intermittent turbulent layers, b) all PMWE observed during this campaign can be explained by neutral air turbulence, c) turbulence creates small-scale structures in all D-region constituents, including free electrons; d) MSP ultimately influence the radar volume reflectivity by distorting the turbulence spectrum of electrons, e) the influence of MSP and of background electron density is just to increase SNR.

    • Inflammatory bowel disease [IBD] is a complex chronic disorder with no clear aetiology and no known cure. Despite recent advances in overall disease management and improved therapeutics, patients with IBD still experience a substantial burden. Furthermore, as the incidence continues to increase in developing areas of the world, it is expected that the burden of IBD to society will increase and exert tremendous pressure on healthcare systems worldwide. Therefore, new strategies to prevent the global increase of IBD are urgently required. Data are being progressively acquired on the period preceding disease diagnosis, which support the concept that IBD has a preclinical period that may reveal the triggers of disease and may be amenable to early intervention. Having a better knowledge of this preclinical period will increase the potential not only for improved understanding of disease pathogenesis and improved therapeutics, but also for disease prediction and prevention.

    • The ground-based measurements obtained from a lidar network and the 6-year OSIRIS (optical spectrograph and infrared imager system) limb-scanning radiance measurements made by the Odin satellite are used to study the climatology of the middle- and low-latitude sodium (Na) layer. Up to January 2021, four Na resonance fluorescence lidars at Beijing (40.5∘ N, 116.0∘ E), Hefei (31.8∘ N, 117.3∘ E), Wuhan (30.5∘ N, 114.4∘ E), and Haikou (19.5∘ N, 109.1∘ E) collected vertical profiles of Na density for a total of 2136 nights (19 587 h). These large datasets provide multi-year routine measurements of the Na layer with exceptionally high temporal and vertical resolution. The lidar measurements are particularly useful for filling in OSIRIS data gaps since the OSIRIS measurements were not made during the dark winter months because they utilize the solar-pumped resonance fluorescence from Na atoms. The observations of Na layers from the ground-based lidars and the satellite are comprehensively compared with a global model of meteoric Na in the atmosphere (WACCM–Na). The lidars present a unique test of OSIRIS and WACCM (Whole Atmosphere Community Climate Model), because they cover the latitude range along 120∘ E longitude in an unusual geographic location with significant gravity wave generation. In general, good agreement is found between lidar observations, satellite measurements, and WACCM simulations. On the other hand, the Na number density from OSIRIS is larger than that from the Na lidars at the four stations within one standard deviation of the OSIRIS monthly average, particularly in autumn and early winter arising from significant uncertainties in Na density retrieved from much less satellite radiance measurements. WACCM underestimates the seasonal variability of the Na layer observed at the lower latitude lidar stations (Wuhan and Haikou). This discrepancy suggests the seasonal variability of vertical constituent transport modelled in WACCM is underestimated because much of the gravity wave spectrum is not captured in the model.
