    • Preeclampsia is a serious complication of pregnancy, affecting both maternal and fetal health. In genome-wide association meta-analysis of European and Central Asian mothers, we identify sequence variants that associate with preeclampsia in the maternal genome at ZNF831/20q13 and FTO/16q12. These are previously established variants for blood pressure (BP) and the FTO variant has also been associated with body mass index (BMI). Further analysis of BP variants establishes that variants at MECOM/3q26, FGF5/4q21 and SH2B3/12q24 also associate with preeclampsia through the maternal genome. We further show that a polygenic risk score for hypertension associates with preeclampsia. However, comparison with gestational hypertension indicates that additional factors modify the risk of preeclampsia.

    • Background Hyperlipidaemia is a major risk factor for cardiovascular disease, and atherosclerosis is the underlying cause of both myocardial infarction and stroke. We have previously shown that the Pro251 variant of perilipin-2 reduces plasma triglycerides and may therefore be beneficial to reduce atherosclerosis development. Objective We sought to delineate putative beneficial effects of the Pro251 variant of perlipin-2 on subclinical atherosclerosis and the mechanism by which it acts. Methods A pan-European cohort of high-risk individuals where carotid intima-media thickness has been assessed was adopted. Human primary monocyte-derived macrophages were prepared from whole blood from individuals recruited by perilipin-2 genotype or from buffy coats from the Karolinska University hospital blood central. Results The Pro251 variant of perilipin-2 is associated with decreased intima-media thickness at baseline and over 30 months of follow-up. Using human primary monocyte-derived macrophages from carriers of the beneficial Pro251 variant, we show that this variant increases autophagy activity, cholesterol efflux and a controlled inflammatory response. Through extensive mechanistic studies, we demonstrate that increase in autophagy activity is accompanied with an increase in liver-X-receptor (LXR) activity and that LXR and autophagy reciprocally activate each other in a feed-forward loop, regulated by CYP27A1 and 27OH-cholesterol. Conclusions For the first time, we show that perilipin-2 affects susceptibility to human atherosclerosis through activation of autophagy and stimulation of cholesterol efflux. We demonstrate that perilipin-2 modulates levels of the LXR ligand 27OH-cholesterol and initiates a feed-forward loop where LXR and autophagy reciprocally activate each other; the mechanism by which perilipin-2 exerts its beneficial effects on subclinical atherosclerosis.

    • Higher maternal vitamin D concentration during pregnancy is associated with better child mental health. Negative affectivity, an early-emerging temperamental trait, indicates an increased risk of psychopathology. We investigated if maternal early/mid-pregnancy 25-hydroxyvitamin D (25(OH)D) and neonatal cord blood 25(OH)D concentrations are associated with Negative affectivity in infancy. We studied term-born infants from the vitamin D Intervention in Infants study (VIDI, n = 777, follow-up rate 80%, Finland), and the Generation R Study (n = 1505, follow-up rate 40%, Netherlands). We measured maternal serum 25(OH)D at 6–27 weeks (VIDI) or 18–25 weeks (Generation R) of pregnancy, and cord blood 25(OH)D at birth (both cohorts). Caregivers rated infant Negative affectivity at 11.7 months (VIDI) or 6.5 months (Generation R) using the Revised Infant Behavior Questionnaire. Using linear regression, we tested associations between 25(OH)D and Negative affectivity adjusted for infant age, sex, season of 25(OH)D measurement, maternal age, education, smoking, and body-mass-index. Per 10 nmol/l increase in maternal early/mid-pregnancy 25(OH)D, infant Negative affectivity decreased by 0.02 standard deviations (95% confidence interval [CI] − 0.06, − 0.004) in VIDI, and 0.03 standard deviations (95% CI − 0.03, − 0.01) in Generation R. Cord blood 25(OH)D was associated with Negative affectivity in Generation R (− 0.03, 95% CI − 0.05, − 0.01), but not VIDI (0.00, 95% CI − 0.02, 0.02). Lower maternal 25(OH)D concentrations were consistently associated with higher infant Negative affectivity, while associations between cord blood 25(OH)D concentrations and Negative affectivity were less clear. Maternal vitamin D status during early- and mid-pregnancy may be linked with early-emerging differences in offspring behavior.

    • Bien que les mesures de teneur en eau soient largement utilisées dans l'industrie, les considérations métrologiques quant à cette mesure ne sont pas complètement abouties de sorte à fournir des mesures fiables et traçables au SI. Afin de remédier à ceci, le projet de recherche conjoint, Joint Research Project SIB64 “METefnet – Metrology for moisture in materials”, est actuellement en cours, et contribue au programme européen de recherche en métrologie European Metrology Research Programme. Le projet METefnet a pour objectifs de développer et d'améliorer l'approche métrologique de ce sujet. Ceci inclus notamment: le travail sur de nouvelles méthodes de référence pour évaluer la fraction massique en eau, l'amélioration des mesures mettant en œuvre la méthode primaire de type titration Karl Fischer, le développement de nouveaux matériaux de référence certifiés présentant une très bonne stabilité et permettant une traçabilité au SI, le développement de nouveaux étalons de transfert, la réalisation d'études visant à quantifier et réduire les effets liés à la prise d'échantillon, son transport et sa manipulation, le développement d'une nouvelle méthode pour étalonner les instruments mesurant l'humidité de surface, et l'amélioration des méthodes d'estimation d'incertitudes de ces mesures. Ce travail, réalisé dans le domaine de la métrologie de l'humidité au sein des matériaux, couvre à la fois le mesurande décrit comme étant spécifiquement la teneur en eau, seule, dans les matériaux, mais également un mesurande plus large pouvant inclure l'eau ainsi que d'autres liquides ou composés organiques volatiles; ceci afin de bien mettre en exergue la différence qui peut être observée entre ces deux mesurandes. Le projet global a pour objectif de soutenir une action de dissémination et de traçabilité au système SI des mesures de teneur en eau dans les matériaux avec une exactitude optimale et de développer une infrastructure métrologique cohérente pour ce type de mesures. Le travail technique ainsi que les dernières avancées vous sont ainsi présentées.

The core-level photoemission point to oxygen loss from only BaMnO3 changing some of Mn4+ to Mn3+ in it, transfer of some spectral weight to the highly localized Fehrenbacher-Rice states and an increased Mn 3d-O 2p hybridization. Magnetization measurements show that at low temperatures, the samples depict a canted antiferromagnetic ordering.

  • New Primary Standards for Establishing SI Traceability for Moisture Measurements in Solid Materials

    • A European research project METefnet addresses a fundamental obstacle to improving energy-intensive drying process control: due to ambiguous reference analysis methods and insufficient methods for estimating uncertainty in moisture measurements, the achievable accuracy in the past was limited and measurement uncertainties were largely unknown. This paper reports the developments in METefnet that provide a sound basis for the SI traceability: four new primary standards for realizing the water mass fraction were set up, analyzed and compared to each other. The operation of these standards is based on combining sample weighing with different water vapor detection techniques: cold trap, chilled mirror, electrolytic and coulometric Karl Fischer titration. The results show that an equivalence of 0.2 % has been achieved between the water mass fraction realizations and that the developed methods are applicable to a wide range of materials.

    • Preeclampsia (PE) is a complex pregnancy disorder. It is not extensively known how the metabolic alterations of PE women contribute to the metabolism of newborn. We applied liquid chromatography-mass spectrometry (LC-MS) based non-targeted metabolomics to determine whether the metabolic profile of plasma from umbilical cord differs between infants born to PE and non-PE pregnancies in the FINNPEC study. Cord plasma was available from 42 newborns born from PE and 53 from non-PE pregnancies. 133 molecular features differed between PE and non-PE newborns after correction for multiple testing. Decreased levels of 4-pyridoxic acid were observed in the cord plasma samples of PE newborns when compared to non-PE newborns. Compounds representing following areas of metabolism were increased in the cord plasma of PE newborns: urea and creatine metabolism; carnitine biosynthesis and acylcarnitines; putrescine metabolites; tryptophan metabolism and phosphatidylcholines. To our knowledge, this study is the first one to apply LC-MS based metabolomics in cord plasma of PE newborns. We demonstrate that this strategy provides a global picture of the widespread metabolic alterations associated with PE and particularly the elevated levels of carnitine precursors and trimethylated compounds appear to be associated with PE at birth.

    • We demonstrate magnetic droplet soliton pairs in all-perpendicular spin-torque nano-oscillators (STNOs), where one droplet resides in the STNO free layer (FL) and the other in the reference layer (RL). Typically, theoretical, numerical, and experimental droplet studies have focused on the FL, with any additional dynamics in the RL entirely ignored. Here we show that there is not only significant magnetodynamics in the RL, but the RL itself can host a droplet driven by, and coexisting with, the FL droplet. Both single droplets and pairs are observed experimentally as stepwise changes and sharp peaks in the dc and differential resistance, respectively. While the single FL droplet is highly stable, the coexistence state exhibits high-power broadband microwave noise. Furthermore, micromagnetic simulations reveal that the pair dynamics display periodic, quasi-periodic, and chaotic signatures controlled by applied field and current. The strongly interacting and closely spaced droplet pair offers a unique platform for fundamental studies of highly non-linear soliton pair dynamics.

    • The central Fennoscandia is characterized by the Archean-Proterozoic (AP) boundary and the Central Finland Granitoid Complex (CFGC), a roundish area of approximately 40,000 km2 surrounded by supracrustal belts. Deep seismic reflection profile FIRE 3A runs across these units, and we have re-interpreted the profile and crustal evolution along the profile using 1.92–1.85 Ga plutonic rocks as lithospheric probes. The surface part of the profile has been divided into five subareas: Archean continent (AC) in the east, AP, CFGC, boundary zone (BZ) and the Bothnian Belt (BB) in the west. There are 12 key samples from which zircons were studied for inclusions and analyzed (core-rim) by ion probe for U–Pb dating and oxygen isotopes, followed by analyzes for Lu–Hf by LA–MC–ICP–MS.The AC plutonic rocks (1.87–1.85 Ga) form a bimodal suite, where the proposed mantle source for the mafic rocks is 2.1–2.0 Ga metasomatized lower part of the Archean subcontinental lithospheric mantle (SCLM) and the source for the felsic melts is related plume-derived underplated mafic material in the lower crust. Variable degrees of contamination of the Archean lower crust have produced “subduction-like” Nb–Ta anomalies in spidergrams and negative εNd (T) values in the mafic-intermediate rocks. The felsic AC granitoids originate from a low degree melting of eclogitic or garnet-bearing amphibolites with titanite ± rutile partly prevailing in the residue (Nb–Ta fractionation) followed by variable degree of assimilation/melting of the Archean lower crust. The AP plutonic rocks (ca. 1.88 Ga) can be divided into I-type and A-type granitoids (AP/A), where the latter follow the sediment assimilation trend in ASI diagram, have high δ18O values (up to 8‰) in zircons and exhibit negative Ba anomalies (Rb–Ba–Th in spidergram), as found in sedimentary rocks. A mixing/assimilation of enriched mantle-derived melts with melts from already migmatized sedimentary rocks ± amphibolites is proposed. The CFGC is characterized by both I-type and A-type (CFGC/A) intermediate and felsic granitoids. The I-type granitoids are divided into two groups at ≥ 1885 Ma and ≤ 1882 Ma, where the latter overlap in age with the CFGC/A granitoids. Both I-type CFGC and CFGC/A granitoids are interpreted to have formed from mixing of Paleoproterozoic SCLM-derived melts with crustal melts from hydrous and dry intermediate-felsic igneous sources, respectively. The geochemistry, dominantly δ18O values below 6.5‰ in zircons and TDM (2.11–2.42 Ga) of the CFGC granitoids favor the occurrence of older crust (ca. 2.1–2.0 Ga) in their genesis. The BZ granitoids are similar in age but more juvenile with TDM ages between 1.94 Ga and 2.16 Ga. The 1.92 Ga granodiorite in the BB is correlated with juvenile gneissic tonalites and granodiorites found from the AP boundary.We suggest that the present high-velocity lower crust under the CFGC is composed of melt-extracted granulites (crustal source age ≥ 2.0 Ga) and mafic cumulates which both formed during 1.90–1.88 Ga arc magmatism. The ≤ 1.88 Ga stage represents the end of compression/transpression and is followed by 1.87–1.86 Ga buckling, forming the Bothnian Oroclines.

    • Neural networks provide an attractive framework to monitor the water table level and the volume of stored water in porous media from seismic data in an automated, fast and cost-efficient manner. In this work, a subsurface reservoir is modeled as a coupled three-dimensional poroviscoelastic-viscoelastic medium. The wave propagation from source to receiver(s) is numerically simulated using a nodal discontinuous Galerkin method coupled with an Adams-Bashforth time-stepping scheme on a graphics processing unit cluster. The wave field solver is used to generate databases for the neural network model to estimate the water table level and actual volume of water. We use a deconvolution-based approach to normalize the effect from the source wavelet. The results demonstrate the capacity of the fully connected neural network for estimating both the water table level and the volume of stored water in the porous storage reservoir from both synthetic and measured data.

    • Although postmenopausal hormone-replacement therapy (HRT) is known to prevent fractures, knowledge on the influence of long-term HRT on bone strength and its determinants other than areal bone mineral density is scarce. This study used a genetically controlled design with 24 monozygotic female twin pairs aged 54 to 72 years in which one cotwin was using HRT (mean duration 8 years) and the other had never used HRT. Estimated bone strength, cross-sectional area, volumetric bone mineral density, bone mineral mass, and cross-sectional density and mass distributions were assessed in the tibial shaft, distal tibia, and distal radius with peripheral computed tomography (pQCT). In the tibial shaft, HRT users had 9% [95% confidence interval (CI) 3%-15%] higher estimated bending strength than their nonusing cotwins. Larger cortical area and higher cortical bone mineral density accounted for this difference. The cortex was larger in the HRT users in the endocortical region. In the distal tibia, estimated compressive strength was 24% (95% CI 9%-40%) higher and in the distal radius 26% (95% CI 11%-41%) higher in the HRT users than in their nonusing cotwins owing to higher volumetric bone mineral density. No difference between users and nonusers was observed in total bone cross-sectional area in any measured bone site. The added mineral mass in the HRT users was distributed evenly within and between bone sites. In postmenopausal women, long-term HRT preserves estimated bone strength systemically by preventing bone mineral loss similarly in body weight-loaded and non-weight-loaded bone.     • Aims/hypothesis: Not all obese individuals display the metabolic disturbances commonly associated with excess fat accumulation. Mechanisms maintaining this ‘metabolically healthy obesity’ (MHO) are as yet unknown. We aimed to study different fat depots and transcriptional pathways in subcutaneous adipose tissue (SAT) as related to the MHO phenomenon.Methods: Sixteen rare young adult obesity-discordant monozygotic (MZ) twin pairs (intra-pair difference (∆) in BMI ≥3 kg/m2), aged 22.8–35.8 years, were examined for detailed characteristics of metabolic health (subcutaneous, intra-abdominal and liver fat [magnetic resonance imaging/spectroscopy]), OGTT, lipids, adipokines and C-reactive protein (CRP). Affymetrix U133 Plus 2.0 chips were used to analyse transcriptomics pathways related to mitochondrial function and inflammation in SAT.Results: Based on liver fat accumulation, two metabolically different subgroups emerged. In half (8/16) of the pairs (∆weight 17.1 ± 2.0 kg), the obese co-twin had significantly higher liver fat (∆718%), 78% increase in AUC insulin during OGTT and CRP, significantly more disturbance in the lipid profile and greater tendency for hypertension compared with the lean co-twin. In these obese co-twins, SAT expression of mitochondrial oxidative phosphorylation, branched-chain amino acid catabolism, fatty acid oxidation and adipocyte differentiation pathways were downregulated and chronic inflammation upregulated. In the other eight pairs (∆weight 17.4 ± 2.8 kg), the obese co-twin did not differ from the non-obese co-twin in liver fat (∆8%), insulin sensitivity, CRP, lipids, blood pressure or SAT transcriptomics.Conclusions/interpretation: Our results suggest that maintenance of high mitochondrial transcription and lack of inflammation in SAT are associated with low liver fat and MHO.

    • This study aimed to explore the association between maternal age and smoking during the second and third trimesters of pregnancy across socioeconomic groups and to evaluate the interacting effect of maternal age and socioeconomic status on smoking with a view to informing public health interventions.DesignThis is a register-based study.SettingsData from the Finnish Medical Birth Register were cross-linked with background data from Statistics Finland.ParticipantsThe information of 932 671 pregnant women who gave birth in Finland from 2000 to 2015.Main outcome measuresMaternal smoking during the second and third trimesters of pregnancy by occupation and maternal age.ResultsThe proportion of women who smoked during the second and third trimesters of pregnancy was 10.5%. Using women 30–34 years as the reference group, adjusted ORs (aOR) and 95% CIs for smoking were 6.02 (5.81 to 6.24) in women below 20 years and 2.77 (2.71 to 2.84) in women 20 to 24 years. The prevalence of smoking across socioeconomic groups compared with upper-level employees increased, peaking for women in manual occupations (aOR 3.39, 95% CI 3.25 to 3.52) and unemployed women (aOR 4.49, 95% CI 4.30 to 4.68). Significant interactions on the additive scale with the relative excess risk due to interaction >2 were found for unemployed women aged 25–29 years and for teenage mothers and mothers aged 20–24 years across all socioeconomic groups, but not for self-employed women.ConclusionsSmoking during the second and third trimesters of pregnancy was most common among teenage mothers across all socioeconomic groups. The association between maternal age and smoking differed by socioeconomic status for young mothers. Interventions should address a wider range of maternal risk factors among young mothers with low socioeconomic status and simultaneously target a broader number of women who smoke during the pregnancy.

    • We described the trend of fertility rates, age-specific fertility rates and associated factors in Finland over a 30-year period.DesignA descriptive population-based register study.SettingFertility data, including age at first birth, childlessness and educational levels were gathered from the Finnish Medical Birth Register and Statistics Finland.ParticipantsAll 1 792 792 live births from 1987 to 2016 in Finland.Main outcome measuresCompleted fertility rate, total fertility rate and age-specific fertility rate.ResultsThe total fertility rate of Finnish women fluctuated substantially from 1987 to 2016. Since 2010, the total fertility rate has gradually declined and reached the lowest during the study period in 2016: 1.57 children per woman. The mean maternal age at first birth rose by 2.5 years from 26.5 years in 1987 to 29 years in 2016. The proportion of childless women at the age of 50 years increased from 13.6% in 1989 to 19.6% in 2016. By considering the impact of postponement and childlessness, the effect on total fertility rates was between −0.01 and −0.12 points. Since 1987, the distribution of birth has declined for women under the age of 29 and increased for women aged 30 or more. However, start of childbearing after the age of 30 years was related to the completed fertility rate of less than two children per woman. The difference in completed fertility rate across educational groups was small.ConclusionsPostponement of first births was followed by decline in completed fertility rate. Increasing rate of childlessness, besides the mean age at first birth, was an important determinant for declined fertility rates, but the relation between women’s educational levels and the completed fertility rate was relatively weak.

    • To examine the association between maternal age and maternal obesity across socioeconomic groups and to determine whether socioeconomic status modifies the association between maternal age and maternal obesity with a view to informing public health policies. Methods: Data for this register-based study were sourced from the Finnish Medical Birth Register and Statistics Finland, using the information of 707,728 women who gave birth in Finland from 2004 to 2015. We used multivariable regression models to assess the association between maternal age and maternal obesity across socioeconomic groups. We further assessed interactions on both multiplicative and additive scales. Results: Across all socioeconomic groups, the adjusted odds ratio for the association between maternal age and maternal obesity increased, peaking for women 35 years or older. Using women below 20 years of age in the category of upper-level employees as a single reference group, in the category of upper-level employees, the adjusted odds ratio and 95% confidence intervals among women 35 years or older was 1.92 (1.39–2.64) for maternal obesity. Equally, the adjusted odds ratio and 95% confidence intervals in the category of long-term unemployed was 4.35 (3.16–5.98). Synergistic interactions on both multiplicative and additive scales were found across age and socioeconomic groups. Conclusions: The association between maternal age and maternal obesity was strongest among women 35 years or older with lower socioeconomic status. Population-level interventions that address maternal risk factors from teenage years are needed alongside individual-level interventions that target high-risk mothers in areas of low socioeconomic status and maternal obesity.

  • Tunable Mode Coupling in Nanocontact Spin-Torque Oscillators

    • Recent experiments on spin-torque oscillators have revealed interactions between multiple magneto-dynamic modes, including mode coexistence, mode hopping, and temperature-driven crossover between modes. The initial multimode theory indicates that a linear coupling between several dominant modes, arising from the interaction of the subdynamic system with a magnon bath, plays an essential role in the generation of various multimode behaviors, such as mode hopping and mode coexistence. In this work, we derive a set of rate equations to describe the dynamics of coupled magneto-dynamic modes in a nanocontact spin-torque oscillator. Expressions for both linear and nonlinear coupling terms are obtained, which allow us to analyze the dependence of the coupled dynamic behaviors of modes on external experimental conditions as well as intrinsic magnetic properties. For a minimal two-mode system, we further map the energy and phase difference of the two modes onto a two-dimensional phase space and demonstrate in the phase portraits how the manifolds of periodic orbits and fixed points vary with an external magnetic field as well as with the temperature.

    • It has been argued that if multiple spin wave modes are competing for the same centrally located energy source, as in a nanocontact spin torque oscillator, that only one mode should survive in the steady state. Here, the experimental conditions necessary for mode coexistence are explored. Mode coexistence is facilitated by the local field asymmetries induced by the spatially inhomogeneous Oersted field, which leads to a physical separation of the modes, and is further promoted by spin wave localization at reduced applied field angles. Finally, both simulation and experiment reveal a low frequency signal consistent with the intermodulation of two coexistent modes.
