  • Hydraulic conductivity of fly ash : sewage sludge mixes for use in landfill cover liners

    • Secondary materials could help meeting the increasing demand of landfill cover liner materials. In this study, the effect of compaction energy, water content, ash ratio, freezing, drying and biological activity on the hydraulic conductivity of two fly ash - sewage sludge mixes was investigated using a 27-1 fractional factorial design. The aim was to identify the factors that influence hydraulic conductivity, to quantify their effects and to assess how a sufficiently low hydraulic conductivity can be achieved. The factors compaction energy and drying, as well as the factor interactions material×ash ratio and ash ratio×compaction energy affected hydraulic conductivity significantly (α = 0.05). Freezing on 5 freeze-thaw cycles did not affect hydraulic conductivity. Water content affected hydraulic conductivity only initially. The hydraulic conductivity data were modelled using multiple linear regression. The derived models were reliable as indicated by R2adjusted values between 0.75 and 0.86. Independent on the ash ratio and the material, hydraulic conductivity was predicted to be between 1.7 × 10-11 m s-1 and 8.9 × 10-10 m s-1 if the compaction energy was 2.4 J cm-3, the ash ratio between 20 and 75 % and drying did not occur. Thus, the investigated materials met the limit value for non-hazardous waste landfills of 10-9 m s-1.

    • Genotyping of Chlamydia trachomatis is limited by the low sequence variation in the genome, and no adequatemethod is available for analysis of the spread of chlamydial infections in the community. We have developeda multilocus sequence typing (MLST) system based on five target regions and compared it with analysis ofompA, the single gene most extensively used for genotyping. Sequence determination of 16 reference strains,comprising all major serotypes, serotypes A to L3, showed that the number of genetic variants in the fiveseparate target regions ranged from 8 to 16. The genetic variation in 47 clinical C. trachomatis isolates ofrepresentative serotypes (14 serotype D, 12 serotype E, 11 serotype G, and 10 serotype K strains) was analyzed;and the MLST system detected 32 variants, whereas 12 variants were detected by using ompA analysis.Specimens of the predominant serotype, serotype E, were differentiated into seven genotypes by MLST but intoonly two by ompA analysis. The MLST system was applied to C. trachomatis specimens from a population ofmen who have sex with men and was able to differentiate 10 specimens of one predominant ompA genotype Gvariant into four distinct MLST variants. To conclude, our MLST system can be used to discriminate C.trachomatis strains and can be applied to high-resolution molecular epidemiology.

  • Uneven distribution of ventilation in acute respiratory distress syndrome

    • INTRODUCTION: The aim of this study was to assess the volume of gas being poorly ventilated or non-ventilated within the lungs of patients treated with mechanical ventilation and suffering from acute respiratory distress syndrome (ARDS). METHODS: A prospective, descriptive study was performed of 25 sedated and paralysed ARDS patients, mechanically ventilated with a positive end-expiratory pressure (PEEP) of 5 cmH2O in a multidisciplinary intensive care unit of a tertiary university hospital. The volume of poorly ventilated or non-ventilated gas was assumed to correspond to a difference between the ventilated gas volume, determined as the end-expiratory lung volume by rebreathing of sulphur hexafluoride (EELVSF6), and the total gas volume, calculated from computed tomography images in the end-expiratory position (EELVCT). The methods used were validated by similar measurements in 20 healthy subjects in whom no poorly ventilated or non-ventilated gas is expected to be found. RESULTS: EELVSF6 was 66% of EELVCT, corresponding to a mean difference of 0.71 litre. EELVSF6 and EELVCT were significantly correlated (r2 = 0.72; P < 0.001). In the healthy subjects, the two methods yielded almost identical results. CONCLUSION: About one-third of the total pulmonary gas volume seems poorly ventilated or non-ventilated in sedated and paralysed ARDS patients when mechanically ventilated with a PEEP of 5 cmH2O. Uneven distribution of ventilation due to airway closure and/or obstruction is likely to be involved.
