  • Interplanetary coronal mass ejection observed at STEREO-A, Mars, comet 67P/Churyumov-Gerasimenko, Saturn, and New Horizons en route to Pluto : Comparison of its Forbush decreases at 1.4, 3.1, and 9.9 AU

    • We discuss observations of the journey throughout the Solar System of a large interplanetary coronal mass ejection (ICME) that was ejected at the Sun on 14 October 2014. The ICME hit Mars on 17 October, as observed by the Mars Express, Mars Atmosphere and Volatile EvolutioN Mission (MAVEN), Mars Odyssey, and Mars Science Laboratory (MSL) missions, 44h before the encounter of the planet with the Siding-Spring comet, for which the space weather context is provided. It reached comet 67P/Churyumov-Gerasimenko, which was perfectly aligned with the Sun and Mars at 3.1 AU, as observed by Rosetta on 22 October. The ICME was also detected by STEREO-A on 16 October at 1 AU, and by Cassini in the solar wind around Saturn on the 12 November at 9.9AU. Fortuitously, the New Horizons spacecraft was also aligned with the direction of the ICME at 31.6 AU. We investigate whether this ICME has a nonambiguous signature at New Horizons. A potential detection of this ICME by Voyager 2 at 110-111 AU is also discussed. The multispacecraft observations allow the derivation of certain properties of the ICME, such as its large angular extension of at least 116 degrees, its speed as a function of distance, and its magnetic field structure at four locations from 1 to 10 AU. Observations of the speed data allow two different solar wind propagation models to be validated. Finally, we compare the Forbush decreases (transient decreases followed by gradual recoveries in the galactic cosmic ray intensity) due to the passage of this ICME at Mars, comet 67P, and Saturn.

  • Immune-mediated effects targeting hepatitis C virus in a syngeneic replicon cell transplantation mouse model

    • HCV is characterised by its ability to establish chronic infection in hepatocytes and to replicate in the presence of an inflammation. We mimicked this situation in vivo in immune-competent mice by syngeneic transplantation of HCV replicon-containing mouse hepatoma cells.DesignA total of 5 million H-2b positive Hep56.1D cells, carrying a subgenomic genotype (gt) 2a replicon (HCV replicon cells) or stably expressing comparable levels of the HCV NS3/4A protease/helicase complex (NS3/4A hepatoma cells), were injected subcutaneously into syngeneic H-2b-restricted mice. Kinetics of tumour growth, HCV RNA replication levels and HCV-specific immune responses were monitored. For immune monitoring, new H-2b-restricted cytotoxic T cell epitopes within the gt2a NS3/4A region were mapped. Immune mice were generated by DNA-based vaccination.ResultsHCV replicon and NS3/4A hepatoma cells generated solid tumours in vivo. Similar to what is seen in human HCV infection did HCV RNA replicate in the presence of inflammation. NS3/4A-specific CD8+ T cells seemed to transiently reduce HCV RNA levels. Both CD4+ and CD8+ T cells were required for protection against tumour growth. Vaccine-induced NS3/4A(gt2a)-specific T cells protected against HCV replicon tumours in wild-type, but not in HCV NS3/4A(gt1a)-transgenic mice with dysfunctional HCV-specific T cells. Importantly, as in human HCV infection, HCV replicon cells neither primed nor boosted a strong NS3/4A-specific T cell response.ConclusionSyngeneic transplantation of mouse HCV replicon cells into immune-competent animals mirrors many in vivo events in humans. This system is versatile and can be applied to any genetically modified H-2b-restricted mouse strain.

  • Oceanic and atmospheric modes in the Pacific and Atlantic Oceans since the Little Ice Age (LIA): Towards a synthesis

    • Understanding the variability of the oceanic and atmospheric modes from the Little Ice Age (LIA,similar to 1250 - 1850) to the present can help evaluate their behaviors under future warming scenarios. Numerous proxy-based reconstructions of the oceanic and atmospheric modes were presented. It is highly needed for a synthesis study to evaluate the existing reconstructions of the dominant oceanic and atmospheric modes since the LIA. We found that the El Nino-Southern Oscillation (ENSO) reconstructions are only robust on interannual and interdecadal scales, while the reconstructed Pacific Decadal Oscillation (PDO) and Atlantic Multi-decadal Oscillation (AMO) are robust on multi-decadal (50-100 years) timescales. We generated synthesized ENSO, PDO and AMO reconstructions as the average of the existing reconstructions on the most suitable timescales identified using timescale dependent correlation methods. In the 20th century, the interannual variability and periodicity of the ENSO and the multi-decadal periodicity of the PDO and AMO were most pronounced. The ENSO shows the strongest multi-decadal periodicity from mid-18th century onwards, while the multi-decadal periodicity of the PDO and AMO was particularly low in the 18th century. Multi-decadal variations of the AMO showed a prompt and positive response to solar irradiation, while the ENSO showed a lagged and negative response to solar irradiation from the 18th century to the present.   • Functional differences in hepatitis C virus nonstructural (NS) 3/4A- and 5A-specific T cell responses

    • The hepatitis C virus nonstructural (NS) 3/4A and NS5A proteins are major targets for the new direct-acting antiviral compounds. Both viral proteins have been suggested as modulators of the response to the host cell. We have shown that NS3/4A- and NS5A-specific T cell receptors confer different effector functions and that killing of NS3/4A-expressing hepatocytes is highly dependent on IFN-γ. We here characterize the functional differences in the T cell responses to NS3/4A and NS5A. NS3/4A- and NS5A-specific T cells could be induced at various frequencies in wild-type-, NS3/4A- and NS5A-transgenic mice. Priming of NS5A-specific T cells required a high DNA dose and was unlike NS3/4A dependent on both CD4+ and CD8+ T cells, but less influenced by CD25+/GITR+ regulatory T cells. The presence of IL-12 greatly improved specific CD8+ T cell priming by NS3/4A but not by NS5A, suggesting a less dependence of IFN-γ for NS5A. This notion was supported by the observation that NS5A-specific T cells could eliminate NS5A-expressing hepatocytes also in the absence of IFN-γ-receptor-2. This supports that NS3/4A- and NS5A-specific T cells become activated and eliminate antigen expressing, or infected hepatocytes, by distinct mechanisms and that NS5A-specific T cells show an overall less dependence of IFN-γ.

  • A comparison of very old patients admitted to intensive care unit after acute versus elective surgery or intervention

    • Background: We aimed to evaluate differences in outcome between patients admitted to intensive care unit (ICU) after elective versus acute surgery in a multinational cohort of very old patients (80 years; VIP). Predictors of mortality, with special emphasis on frailty, were assessed.Methods: In total, 5063 VIPs were induded in this analysis, 922 were admitted after elective surgery or intervention, 4141 acutely, with 402 after acute surgery. Differences were calculated using Mann-Whitney-U test and Wilcoxon test. Univariate and multivariable logistic regression were used to assess associations with mortality.Results: Compared patients admitted after acute surgery, patients admitted after elective surgery suffered less often from frailty as defined as CFS (28% vs 46%; p < 0.001), evidenced lower SOFA scores (4 +/- 5 vs 7 +/- 7; p < 0.001). Presence of frailty (CFS >4) was associated with significantly increased mortality both in elective surgery patients (7% vs 12%; p = 0.01), in acute surgery (7% vs 12%; p = 0.02).Conclusions: VIPs admitted to ICU after elective surgery evidenced favorable outcome over patients after acute surgery even after correction for relevant confounders. Frailty might be used to guide clinicians in risk stratification in both patients admitted after elective and acute surgery.

  • Long-term quality of life and postthrombotic syndrome in women after an episode of venous thromboembolism

    • To evaluate health-related quality of life after venous thromboembolism. Methods We conducted a cohort study, TEHS follow-up, including 1040 women with a first episode of venous thromboembolism and 994 women unexposed to venous thromboembolism. Patients were recruited from the “Thrombo Embolism Hormonal Study” (TEHS), a Swedish nation-wide case–control study on risk factors for venous thromboembolism in women 18–64 years of age. Quality of life was measured using SF-36 and VEINES-QoL/VEINES-Sym. Results On average there were no difference in mean SF-36 summary scales scores between exposed and unexposed women. Twenty percent of exposed women developed postthrombotic syndrome during follow-up. Women with postthrombotic syndrome had severely impaired quality of life with lower scores on all scales. Other predictors of low quality of life after venous thromboembolism were age, obesity, physical inactivity, and recurrent venous thromboembolism. Conclusion Long-term quality of life after venous thromboembolism in women was severely impaired among those developing postthrombotic syndrome, while quality of life in women not developing postthrombotic syndrome was similar to a control population.

    • The ion sensor of the Analyzer of Space Plasmas and Energetic Atoms (ASPERA-3) experiment detects accelerated ions during pulses of radio emissions from the powerful topside sounder: the Mars Advanced Radar for Subsurface and Ionosphere Sounding (MARSIS) onboard Mars Express. Accelerated ions (O-2(+), O+, and lighter ions) are observed in an energy range up to 800 eV when MARSIS transmits at a frequency close to the plasma frequency. Individual observations consist of almost monoenergetic ion beams aligned with the MARSIS antenna or lying in the plane perpendicular to the antenna. The observed ion beams are often accompanied by a small decrease in the electron flux observed by the electron sensor of Analyzer of Space Plasmas and Energetic Atoms 3. Observations indicate that the voltage applied to the antenna causes charging of the spacecraft to several hundreds of volts by the electrons of the ambient plasma. Positively charged ions are accelerated when the spacecraft discharges.
