  • Non-coding variability at the APOE locus contributes to the Alzheimer's risk

    • Alzheimer’s disease (AD) is a leading cause of mortality in the elderly. While the coding change of APOE-ε4 is a key risk factor for late-onset AD and has been believed to be the only risk factor in the APOE locus, it does not fully explain the risk effect conferred by the locus. Here, we report the identification of AD causal variants in PVRL2 and APOC1 regions in proximity to APOE and define common risk haplotypes independent of APOE-ε4 coding change. These risk haplotypes are associated with changes of AD-related endophenotypes including cognitive performance, and altered expression of APOE and its nearby genes in the human brain and blood. High-throughput genome-wide chromosome conformation capture analysis further supports the roles of these risk haplotypes in modulating chromatin states and gene expression in the brain. Our findings provide compelling evidence for additional risk factors in the APOE locus that contribute to AD pathogenesis.

    • Understanding the factors that affect water quality and the ecological services provided by freshwater ecosystems is an urgent global environmental issue. Predicting how water quality will respond to global changes not only requires water quality data, but also information about the ecological context of individual water bodies across broad spatial extents. Because lake water quality is usually sampled in limited geographic regions, often for limited time periods, assessing the environmental controls of water quality requires compilation of many data sets across broad regions and across time into an integrated database. LAGOS-NE accomplishes this goal for lakes in the northeastern-most 17 US states. LAGOS-NE contains data for 51101 lakes and reservoirs larger than 4 ha in 17 lake-rich US states. The database includes 3 datamodules for: lake location and physical characteristics for all lakes; ecological context (i.e., the land use, geologic, climatic, and hydrologic setting of lakes) for all lakes; and in situmeasurements of lake water quality for a subset of the lakes fromthe past 3 decades for approximately 2600–12 000 lakes depending on the variable. The database contains approximately 150000 measures of total phosphorus, 200 000 measures of chlorophyll, and 900 000 measures of Secchi depth. The water quality data were compiled from87 lake water quality data sets fromfederal, state, tribal, and non-profit agencies, university researchers, and citizen scientists. This database is one of the largest andmost comprehensive databases of its type because it includes both in situmeasurements and ecological context data. Because ecological context can be used to study a variety of other questions about lakes, streams, and wetlands, this database can also be used as the foundation for other studies of freshwaters at broad spatial and ecological scales

  • The Gaia-ESO Survey : Probes of the inner disk abundance gradient

    • Context. The nature of the metallicity gradient inside the solar circle (R-GC < 8 kpc) is poorly understood, but studies of Cepheids and a small sample of open clusters suggest that it steepens in the inner disk. Aims. We investigate the metallicity gradient of the inner disk using a sample of inner disk open clusters that is three times larger than has previously been studied in the literature to better characterize the gradient in this part of the disk. Methods. We used the Gaia-ESO Survey (GES) [Fe/H] values and stellar parameters for stars in 12 open clusters in the inner disk from GES-UVES data. Cluster mean [Fe/H] values were determined based on a membership analysis for each cluster. Where necessary, distances and ages to clusters were determined via comparison to theoretical isochrones. Results. The GES open clusters exhibit a radial metallicity gradient of -0.10 +/- 0.02 dex kpc(-1), consistent with the gradient measured by other literature studies of field red giant stars and open clusters in the range R-GC similar to 6-12 kpc. We also measure a trend of increasing [Fe/H] with increasing cluster age, as has also been found in the literature. Conclusions. We find no evidence for a steepening of the inner disk metallicity gradient inside the solar circle as earlier studies indicated. The age-metallicity relation shown by the clusters is consistent with that predicted by chemical evolution models that include the effects of radial migration, but a more detailed comparison between cluster observations and models would be premature.

    • Context. Stellar evolution models predict that internal mixing should cause some sodium overabundance at the surface of red giants more massive than similar to 1.5-2.0 M-circle dot. The surface aluminium abundance should not be affected. Nevertheless, observational results disagree about the presence and/or the degree of Na and Al overabundances. In addition, Galactic chemical evolution models adopting different stellar yields lead to very different predictions for the behavior of [Na/Fe] and [Al/Fe] versus [Fe/H]. Overall, the observed trends of these abundances with metallicity are not well reproduced.Aims. We readdress both issues, using new Na and Al abundances determined within the Gaia-ESO Survey. Our aim is to obtain better observational constraints on the behavior of these elements using two samples: i) more than 600 dwarfs of the solar neighborhood and of open clusters and ii) low-and intermediate-mass clump giants in six open clusters.Methods. Abundances were determined using high-resolution UVES spectra. The individual Na abundances were corrected for nonlocal thermodynamic equilibrium effects. For the Al abundances, the order of magnitude of the corrections was estimated for a few representative cases. For giants, the abundance trends with stellar mass are compared to stellar evolution models. For dwarfs, the abundance trends with metallicity and age are compared to detailed chemical evolution models.Results. Abundances of Na in stars with mass below similar to 2.0 M-circle dot, and of Al in stars below similar to 3.0 M-circle dot, seem to be unaffected by internal mixing processes. For more massive stars, the Na overabundance increases with stellar mass. This trend agrees well with predictions of stellar evolutionary models. For Al, our only cluster with giants more massive than 3.0 M-circle dot, NGC 6705, is Al enriched. However, this might be related to the environment where the cluster was formed. Chemical evolution models that well fit the observed [Na/Fe] vs. [Fe/H] trend in solar neighborhood dwarfs cannot simultaneously explain the run of [Al/Fe] with [Fe/H], and vice versa. The comparison with stellar ages is hampered by severe uncertainties. Indeed, reliable age estimates are available for only a half of the stars of the sample. We conclude that Al is underproduced by the models, except for stellar ages younger than about 7 Gyr. In addition, some significant source of late Na production seems to be missing in the models. Either current Na and Al yields are affected by large uncertainties, and/or some important Galactic source(s) of these elements has as yet not been taken into account.

  • The Gaia-ESO Survey: Insights into the inner-disc evolution from open clusters

    • Context. The inner disc, which links the thin disc with the bulge, has been somewhat neglected in the past because of the intrinsic difficulties in its study, among which crowding and high extinction. Open clusters located in the inner disc are among the best tracers of its chemistry at different ages and distances. Aims. We analyse the chemical patterns of four open clusters located within 7 kpc of the Galactic centre and of field stars to infer the properties of the inner disc with the Gaia-ESO survey idr2/3 data release. Methods. We derive the parameters of the newly observed cluster, Berkeley 81, finding an age of about 1 Gyr and a Galactocentric distance of similar to 5.4 kpc. We construct the chemical patterns of clusters and we compare them with those of field stars in the solar neighbourhood and in the inner-disc samples. Results. Comparing the three populations we observe that inner-disc clusters and field stars are both, on average, enhanced in [O/Fe], [Mg/Fe], and [Si/Fe]. Using the idr2/3 results of M67, we estimate the non-local thermodynamic equilibrium (NLTE) effect on the abundances of Mg and Si in giant stars. After empirically correcting for NLTE effects, we note that NGC 6705 and Be 81 still have a high [alpha/Fe]. Conclusions. The location of the four open clusters and of the field population reveals that the evolution of the metallicity [Fe/H] and of [alpha/Fe] can be explained within the framework of a simple chemical evolution model: both [Fe/H] and [alpha/Fe] of Trumpler 20 and of NGC 4815 are in agreement with expectations from a simple chemical evolution model. On the other hand, NGC 6705, and to a lesser degree Berkeley 81, have higher [alpha/Fe] than expected for their ages, location in the disc, and metallicity. These differences might originate from local enrichment processes as explained in the inhomogeneous evolution framework.

    • Considerable progress has been made in ecological and evolutionary genetics with studies demonstrating how genes underlying plant and microbial traits can influence adaptation and even 'extend' to influence community structure and ecosystem level processes. Progress in this area is limited to model systems with deep genetic and genomic resources that often have negligible ecological impact or interest. Thus, important linkages between genetic adaptations and their consequences at organismal and ecological scales are often lacking. Here we introduce the Sphagnome Project, which incorporates genomics into a long-running history of Sphagnum research that has documented unparalleled contributions to peatland ecology, carbon sequestration, biogeochemistry, microbiome research, niche construction, and ecosystem engineering. The Sphagnome Project encompasses a genus-level sequencing effort that represents a new type of model system driven not only by genetic tractability, but by ecologically relevant questions and hypotheses.

    • Knowledge of the maximum tyre-road friction coefficient can improve active safety systems by defining actuator boundaries and adaptable intervention thresholds. Estimation of the coefficient of friction based on tyre response measurements requires large level of force excitation. Under normal driving conditions, manoeuvres with large tyre utilizations are rare. This study investigates a method where wheel torques with opposite signs are applied to the front and rear axle simultaneously. This procedure allows for an intervention with large tyre excitations without disturbing the motion of the vehicle. The intervention is evaluated in simulations and experiments. Further, a method is proposed which does not require measurement of the vehicle longitudinal velocity. The results show that it is possible to estimate the current friction coefficient with the proposed method, although the assumption made in the proposed method makes the friction estimate sensitive to measurement noise on the wheel speed signal.

    • ObjectivesEslicarbazepine acetate (ESL) is a once-daily (QD) oral antiepileptic drug (AED) for focal-onset seizures (FOS). Pharmacokinetic (PK) and pharmacodynamic (PD) models were developed to assess dose selection, identify significant AED drug interactions, and quantitate relationships between exposure and safety and efficacy outcomes from Phase 3 trials of adjunctive ESL. MethodsEslicarbazepine (the primary active metabolite of ESL) population PK was evaluated using data from 1351 subjects enrolled in 14 studies (11 Phase 1 and three Phase 3 studies) after multiple oral doses ranging from 400 to 1200mg. Population PK and PD models related individual eslicarbazepine exposures to safety outcomes and efficacy responses. ResultsEslicarbazepine PK was described by a one-compartment model with linear absorption and elimination. The probability of a treatment-emergent adverse event (TEAE; dizziness, headache, or somnolence) was higher with an initial dose of ESL 800mg than with an initial dose of ESL 400mg QD. Body weight, sex, region, and baseline use of carbamazepine (CBZ) or lamotrigine were also found to influence the probability of TEAEs. Eslicarbazepine exposure influenced serum sodium concentration, standardized seizure frequency, and probability of response; better efficacy outcomes were predicted in patients not from Western Europe (WE; vs WE patients) and those not taking CBZ (vs taking CBZ) at baseline. ConclusionsPharmacokinetic and PK/PD modeling were implemented during the development of ESL for adjunctive treatment of FOS in adults. This quantitative approach supported decision-making during the development of ESL, and contributed to dosing recommendations and labeling information related to drug interactions.

    • To assess the long-term effect of bariatric surgery on the incidence of gout and hyperuricaemia in participants of the Swedish Obese Subjects (SOS) study.This report includes 1982 subjects who underwent bariatric surgery and 1999 obese controls from the SOS study, a prospective intervention trial designed to assess the effect of bariatric surgery compared with conventional treatment. None of the subjects had gout at baseline. An endpoint on gout incidence was created based on information on gout diagnosis and use of gout medications through national registers and questionnaires. Median follow-up for the incidence of gout was about 19years for both groups. Moreover, the incidence of hyperuricaemia over up to 20years was examined in a subgroup of participants having baseline uric acid levels <6.8mg/dL.Bariatric surgery was associated with a reduced incidence of gout compared with usual care (adjusted HR 0.60, 95% CI 0.48 to 0.75, p<0.001). The difference in absolute risk between groups was 3 percentage points at 15years, and the number of subjects needed to be treated by bariatric surgery to prevent one incident gout event was 32 (95% CI 22 to 59). The effect of bariatric surgery on gout incidence was not influenced by baseline risk factors, including body mass index. During follow-up, the surgery group had a lower incidence of hyperuricaemia (adjusted HR 0.47, 95% CI 0.39 to 0.58, p<0.001). The difference in absolute risk between groups was 12 percentage points at 15years, and the number of participants needed to be treated by bariatric surgery to prevent hyperuricaemia was 8 (95% CI 6 to 13).Bariatric surgery prevents gout and hyperuricaemia in obese subjects.NCT01479452; Results.

    • The terrestrial planets and the asteroids dominant in the inner asteroid belt are water poor. However, in the protoplanetary disk the temperature should have decreased below water-condensation level well before the disk was photo-evaporated. Thus, the global water depletion of the inner Solar System is puzzling. We show that, even if the inner disk becomes cold, there cannot be direct condensation of water. This is because the snowline moves towards the Sun more slowly than the gas itself. Thus the gas in the vicinity of the snowline always comes from farther out, where it should have already condensed, and therefore it should be dry. The appearance of ice in a range of heliocentric distances swept by the snowline can only be due to the radial drift of icy particles from the outer disk. However, if a planet with a mass larger than 20 Earth mass is present, the radial drift of particles is interrupted, because such a planet gives the disk a super-Keplerian rotation just outside of its own orbit. From this result, we propose that the precursor of Jupiter achieved this threshold mass when the snowline was still around 3 AU. This effectively fossilized the snowline at that location. In fact, even if it cooled later, the disk inside of Jupiter's orbit remained ice-depleted because the flow of icy particles from the outer system was intercepted by the planet. This scenario predicts that planetary systems without giant planets should be much more rich in water in their inner regions than our system. We also show that the inner edge of the planetesimal disk at 0.7 AU, required in terrestrial planet formation models to explain the small mass of Mercury and the absence of planets inside of its orbit, could be due to the silicate condensation line, fossilized at the end of the phase of streaming instability that generated the planetesimal seeds. Thus, when the disk cooled, silicate particles started to drift inwards of 0.7. AU without being sublimated, but they could not be accreted by any pre-existing planetesimals.
